PDGFR-β反义寡核苷酸对大鼠血管平滑肌细胞增殖的影响   总被引:1,自引:0,他引:1  
目的 研究血小板衍化生长因子 β受体 (PDGFR- β)反义寡核苷酸 (AODN)对血管平滑肌细胞 (VSMC)增殖的影响 .方法 建立大鼠血管平滑肌细胞体外增殖模型 .将细胞分为反义寡核苷酸组、正义寡核苷酸组、无义寡核苷酸组和空白对照组 ,其中反义组按浓度又分为 1,2 .5 ,5 ,10和 15μmol· L- 1 5小组 .在加药后 2 4,48和 72 h以流式细胞仪测定细胞周期 ,免疫细胞化学染色分析细胞核增殖抗原的表达 ,MTT(四唑盐 )比色法测定细胞增殖活力 .结果  10μmol·L- 1 PDGFR- β AODN干预 VSMC48h后 ,处于 DNA合成前期 (G0 / G1 )细胞数分数 (0 .70 )高于空白对照组 (0 .0 7,P <0 .0 5 ) ;PCNA免疫细胞化学染色发现以 5~ 15μmol· L- 1PDGFR- β AODN干预 48h后 VSMC各组胞核染色强度为2 0 .4%~ 6 .4%明显弱于空白组 (73.8% ) ,正义组 (72 .9% )和无义组 (75 .6 % ) (P<0 .0 5 ) ;10μmol· L- 1 PDGFR-β AODN作用 48h后 VSMC的增殖抑制率为 6 6 .7% ,高于正义组 (8.1% ) ,错义组 (11.8% )和 5 μmol· L- 1 AODN组 (4 5 .4% ) (P<0 .0 5 ) ;同时 10μmol· L- 1 AODN组 48h增殖抑制率与 2 4h(9.1% )相比明显增加 (P<0 .0 5 ) .结论  PDGFR- β AODN对VSMC增殖有明显抑制作用 ,并且呈时间和浓度依赖性 .  相似文献

目的探讨韧粘素 (TN)反义寡核苷酸 (ODN)对培养大鼠血管平滑肌细胞 (VSMC)增殖的抑制作用。方法采用细胞计数及快速竞争性逆转录聚合酶链反应 (RT -PCR)等方法 ,观察不同剂量 ( 5、10、2 0 μg/ml)TN反义ODN和正义ODN对体外培养大鼠VSMC的增殖以及TNmRNA表达水平的影响。 结果TN反义ODN可有效抑制大鼠血管平滑肌细胞的增殖 ,作用呈剂量依赖性 (反义组 5、10、2 0 μg/ml剂量细胞计数分别为 10 5/ml× ( 4 .99± 0 .2 1)、( 4 .0 8± 0 .14 )、( 2 .85± 0 .39) ,对照组为 10 5/ml× ( 5 .95± 0 .41) ,(P <0 .0 5 ) ;TN反义ODN亦可下调TNmRNA表达水平 (反义组 0 .32± 0 .0 5 ,对照组为 0 .5 2± 0 .0 8;P <0 .0 1) ;而TN正义ODN则无上述作用。 结论TN反义ODN可能通过下调TNmRNA表达水平而抑制血管平滑肌细胞的增殖。  相似文献

Ⅰ型内皮素受体反义寡核苷酸抑制人血管平滑肌细胞增殖   总被引:3,自引:0,他引:3  
目的 研究Ⅰ型内皮素 (ET -1)受体 (ETA)反义寡核苷酸 (ODN)对血管平滑肌细胞 (VSMCs)增殖的影响。方法 培养人桡动脉 (humanradialartery ,HRA)的VSMCs ,通过四唑盐 (MTT)比色实验测定细胞的增殖 ,进而分别采用放射受体结合分析法和放射免疫法检测ETA蛋白表达和ET -1自分泌的情况。结果 ETA -ODN显著抑制VSMCs增殖 ,而且具有浓度和时间依赖性。ETA -ODN( 15 μmol L)具有时间依赖抑制ETA表达。ETA -ODN( 15 μmol L)作用 2 4、48h ,VSMCs的上清液ET -1水平以及VSMCs平均ET -1分泌水平 ,较对照组均显著降低 (P <0 .0 5 )。作用 72h ,ET -1分泌上清液水平继续降低 (P <0 .0 5 ) ,但是VSMCs平均ET -1分泌水平不但没有降低 ,反而高于对照组水平 (P <0 .0 5 )。结论 ETA -ODN通过减弱ETA表达和调节ET -1的自分泌抑制HRA的VSMCs增殖 ,提示对临床上血管移植术后的再狭窄有防治作用。  相似文献

目的 探讨全反式维甲酸 (ATRA)对培养大鼠主动脉血管平滑肌细胞增生及P2 1蛋白表达水平的影响。方法 血管平滑肌细胞 (VSMC)采用组织贴块法培养。ATRA(2 .5×10 -6mol·L-1)作用于经 2 0 %胎牛血清刺激后的血管平滑肌细胞 ,2 4h后分别行 3H TdR掺入实验测定和P2 1蛋白印迹检测P2 1蛋白表达。结果 ATRA明显抑制血管平滑肌细胞DNA合成和促进P2 1蛋白表达。结论 ATRA促进P2 1蛋白表达是抑制VSMC增生的原因之一  相似文献

目的 研究全反式维甲酸 (ATRA)对培养大鼠主动脉血管平滑肌细胞 (VSMC)增殖及细胞周期素E(Cy clinE)蛋白表达水平的影响。方法 血管平滑肌细胞采用组织贴块法培养。ATRA(2 .5× 10 - 6 mol·L- 1 )作用于经 2 0 %胎牛血清刺激后的VSMC ,2 4h后分别行3H TdR掺入实验测定和蛋白印迹检测CyclinE蛋白表达。 结果 ATRA明显抑制ATRA的DNA合成 (P <0 .0 1)和CyclinE蛋白的表达 (P <0 .0 5 )。结论 ATRA抑制CyclinE蛋白的表达是抑制VSMC增殖的原因之一。  相似文献

目的研究反义c-myb寡聚脱氧核苷酸(oligodeoxy-nucleotides,ODN)对大鼠自体移植静脉内膜增生的影响.方法移植静脉外周涂加有反义c-mybODN的F127凝胶,于术后1周及2周测量平均厚度、观察血管内膜内c-myb蛋白表达情况.结果手术后1周和2周,移植静脉内膜的平均厚度、内膜中c-myb蛋白的表达在反义c-mybODN组均减少,并存在量效关系(P<0.01).而只用F127胶组及正义ODN组均无抑制作用.结论反义c-mybODN对移植静脉内膜增生有抑制作用,并存在量效关系.此作用可能是通过抑制血管内膜内细胞c-myb蛋白的表达来实现的.  相似文献

目的 研究胞质泛素蛋白连接酶1(KPC1)在大鼠颈动脉球囊损伤后的蛋白表达情况,探讨其在新生内膜形成过程中的生物学功能.方法 将成年雄性SD大鼠(450 ~ 500 g)随机分为假手术组和颈动脉球囊损伤模型组,建模后以左侧损伤动脉为实验组,同时以假手术组的左侧正常颈动脉为对照组.实验组大鼠于损伤后1d、3d、7d、14d、28d分别处死并取材,采用Western blot检测KPC1蛋白、内膜增殖细胞核抗原(PCNA)表达情况,以qRT-PCR检测KPC1的mRNA表达,苏木精-伊红(HE)染色后比较内膜增生程度;采用体外培养血管平滑肌细胞(VSMC)建立增殖模型,运用Western blot及qRT-PCR检测其KPC1蛋白、KPC1 mRNA表达特征.结果 Western blot结果显示,颈动脉球囊损伤后实验组KPC1蛋白表达水平明显低于正常对照组(P<0.05),而PCNA蛋白表达量明显高于正常对照组(P<0.05);qRT-PCR检测结果显示,实验组KPC1的mRNA水平明显低于对照组(P<0.05);HE染色结果示颈动脉球囊损伤后1d、3d时光镜下血管内膜增生不明显;7d时光镜下可见新生内膜,增生的VSMC向内膜下迁移,管腔开始狭窄;14d时,可见明显新生内膜,VSMC排列紊乱,内弹力膜断裂,管腔明显狭窄,内膜厚度超过中膜;28d时,血管内膜继续不规则增生,血管腔狭窄程度继续加重,狭窄超过50%.体外培养并模拟VSMC增殖,提取细胞进行Western blot和qRT-PCR检测显示,KPC1蛋白、KPC1 mRNA表达在血小板源性生长因子-BB刺激增殖后开始逐渐减低.结论 抑制KPC1基因表达可促进损伤内膜增生,其作用机制可能与促进VSMC增殖有关.  相似文献

目的:探讨血管内皮生长因子受体2(KDR)反义寡核苷酸(asONs)在体外对人乳癌细胞生长的抑制作用.方法:采用MTT及Western-blot方法检测4条KDR反义寡核苷酸(asON1、asON2、asON3、asON4)在体外对人乳癌细胞系MCF-7细胞增殖及KDR蛋白表达的抑制作用;asON4作用于MCF-7细胞后,Western-blot检测信号蛋白Bcl-2、Bax、P53表达的变化.结果:asON1、asON2、asON3、asON4在体外以剂量依赖方式抑制了MCF-7细胞的增殖,asON1、asON2、asON3、asON4以0.8 μmol/L剂量给药54 h后,均显著抑制了MCF-7细胞KDR蛋白表达.asON4作用54 h后,MCF-7细胞Bcl-2表达下降,Bax、P53的表达无明显改变.结论:KDR asONs在体外可抑制乳癌细胞的生长.  相似文献

目的 观察反义Smad3腺病毒载体转染增生型血管平滑肌细胞(VSMC)后对其增殖能力的影响.方法 构建大鼠颈总动脉急性球囊损伤模型.取损伤处血管组织,组织块贴壁法培养增生型VSMC,反义Smad3腺病毒载体转染(实验组).以空载病毒作为阴性对照组,以无病毒载体作为空白对照组.抽提转染后细胞总RNA,RT-PCR检测转染后各组Smad3 mRNA表达;MTT比色法检测转染后细胞增殖能力的变化.结果 与阴性对照组和空白对照组比较,转染后实验组Smad3 mRNA表达明显减少;转染后第2、3、5天,实验组细胞增殖能力明显降低(P<0.05),转染后第7天各组细胞增殖能力差异无统计学意义(P>0.05).结论 反义Smad3腺病毒载体转染对增生型VSMC具有增殖抑制作用,为通过基因修饰阻断TGF-β1信号转导从而预防增生性血管病变奠定了基础.  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

目的:评价使用安心颗粒对急诊经皮冠状动脉介入术(PPCI)术后生活质量的影响.方法:将160例接受PPCI的急性ST段抬高型心肌梗死患者随机分为安心颗粒组(术前顿服安心颗粒8.8g,术后安心颗粒4.4 g/次,每日2次)和对照组(仅接受基础药物治疗).所有患者均服用阿司匹林、氯吡格雷和阿托伐他汀.分别在入院时、出院前1d、出院后180 d时,应用心肌梗死多维度量表(MIDAS)、中文版SF-36评价量表对患者生活质量评分.并观察术后30 d以内的出血并发症、血小板减少症发生情况.结果:入院时和出院前1d,两组患者的心肌梗死MIDAS、SF-36量表评分比较无差异(P>0.05);出院后180 d时,与对照组比较,安心颗粒组MIDAS、SF-36评分明显减低(P<0.05);组内与入院时比较,两组出院前1d、出院后180 d时,MIDAS、SF-36评分均降低(P<0.05).两组患者在随访期间均无大量出血、少量出血、重度和极重度血小板减少症发生,安心颗粒组有4例、对照组有7例发生不明显出血(P>0.05).两组发生轻度血小板减少症的患者数比较无差异(P>0.05).结论:PPCI使用安心颗粒,能改善急性ST段抬高型心肌梗死患者的生活质量,且不增加出血风险.  相似文献

Objectives To explore serum cytokines levels (including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v) and their significance in patients with acute coronary syndrome (ACS) and the subsequent follow-ups, with attempt to estimate the role of various serum inflammatory markers in the diagnosis and assessment of ACS.Methods The study population include 40 patients with acute myocardial infarction (AMI), 40 patients with unstable angina pectoris (UAP), and 40 controls. Among the 80 patients, 60 patients attended a follow up 4 months later. Serum inflammatory markers including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v were measured by enzyme linked immunosorbent assay.Results Serum IL- 1 β, sIL-2R, IL-6, TNF-α were significantly higher in AMI group or UAP group compared to the control group and became significantly lower 4 months later in the follow-up patients. Serum levels of IFN-v shows no significant difference between AMI group or UAP group and controls, also showing no significant change when measured in follow up patients. There was no correlation between serum creatine kinase-MB isoenzyme levels and serum inflammatory markers either in UAP or AMI group. Furthermore, when divided into two subgroups using Wagner's QRS scoring system in the AMI group, there is no difference of each serum inflammatory marker between ≤ 6 scores group and > 6 scores group.Conclusion Serum levels of certain inflammatory markers may have some diagnostic value for ACS, and can be a useful marker reflecting disease stability.  相似文献
