目的 观察c—myc反义寡核苷酸对胃癌MKN-45细胞株增殖的抑制作用。方法 用四甲基偶氮唑盐法评价脂质体介导c-myc反义寡核苷酸对细胞增殖的影响,免疫细胞化学ABC方法检测转染后c—myc蛋白表达;荧光显微镜观察细胞凋亡;流式细胞仪分析细胞凋亡。结果 c-myc基因反义寡核苷酸转染MKN-45细胞后,可以显著抑制细胞增殖,其抑制作用具有剂量依赖性;流式细胞仪分析结果显示,转染后能诱导胃癌细胞凋亡,Go/G1期细胞增多;免疫细胞化学显示c—myc蛋白水平明显降低,阳性表达率为64.9%±5.68%。结论 c-myc基因反义寡核苷酸能明显抑制胃癌MKN-45细胞增殖、诱导细胞凋亡和下调c-myc蛋白水平。  相似文献

目的: 观察反义c-myc寡核苷酸对大鼠气道平滑肌细胞周期的影响. 方法: 大鼠气道平滑肌细胞原代培养后,以Lipofectin将反义、正义和错配c-myc寡核苷酸导入平滑肌细胞,流式细胞技术检测细胞周期,免疫组织化学方法检测cyclin D1和cyclin E的表达. 结果: 反义c-myc寡核苷酸可明显抑制大鼠气道平滑肌细胞通过S期,正义及错配c-myc寡核苷酸对细胞周期无明显影响;反义c-myc寡核苷酸抑制了大鼠气道平滑肌细胞cyclin D1和cyclin E的表达. 结论: 反义c-myc寡核苷酸可抑制大鼠气道平滑肌细胞通过S期,抑制cyclin D1和cyclin E 的表达.  相似文献

目的 观察反义c-myc寡核苷酸对大鼠气道平滑肌增殖的抑制作用。方法 大鼠气道平滑肌细胞原代培养,4-12代用于实验。利用Lipofectin将正义、反义及错配c-myc寡核苷酸导入大鼠气道平滑肌细胞,MTT法检测不同寡核苷酸对大鼠气道平滑肌细胞增殖的抑制作用,RT-PCR检测c-myc mRNA的表达,免疫组织化学染色检测c-myc蛋白的表达。结果 反义c-myc寡核苷酸可抑制大鼠气道平滑肌细胞增殖,且这种抑制作用呈浓度依赖性;反义c-myc寡核苷酸可降低c-myc mRNA的表达,同时降低了c-myc mRNA的翻译。结论 反义c-myc寡核苷酸可抑制大鼠气道平滑肌的增殖。  相似文献

目的 :探讨氨茶碱对大鼠气道平滑肌细胞 (ASMCs)增殖抑制和促凋亡作用及在哮喘发生、发展中的机制 .方法分离大鼠气道平滑肌 ,在含有 10 0mL·L -1胎牛血清的DMEM中培养 ,第 4~ 6代细胞用于实验 ,细胞用不同浓度氨茶碱 (0 ,5 0 ,10 0 ,2 0 0 ,30 0 ,4 0 0mg·L -1)诱导培养12~ 72h ;用光学显微镜观察其形态学凋亡改变 ;用琼脂糖电泳分析DNA片段 .结果 :①氨茶碱以时间和浓度依赖的方式降低存活的细胞数 ;②当氨茶碱浓度 >5 0mg·L -1时 ,光镜下可见细胞核皱缩、碎裂 ,随着药物浓度增加 ,凋亡小体形成 ;③当氨茶碱浓度 >5 0mg·L -1时 ,琼脂糖电泳显示了典型的梯状条带 .结论 :实验表明氨茶碱可以诱导体外大鼠ASMCs发生凋亡样改变  相似文献

目的 研究c—myc反义核苷酸对体外培养的淋巴瘤细胞系CA46基因表达及诱导凋亡的作用。方法 c—myc反义核苷酸(ASPO)为人工合成的针对c—myc基因的一段外源性基因片段,以无义的寡核苷酸作为对照,导入CA46细胞后观察其基因表达及诱导凋亡的作用。流式细胞仪检测c—myc基因产物表达,荧光染色观察CA46细胞的凋亡形态,流式细胞仪定量检测凋亡百分率,TUNEL检测加用VP16后的CA46原位凋亡。结果 c—myc反义核苷酸(ASPO)作用CA46细胞后,RT—PCR示c—myc mRNA表达降低。荧光染色ASPO组细胞胞体缩小,胞膜完整,核质浓缩,出现黄绿色不规则或局部致密荧光,凋亡细胞约占23.3%,而SPO组及空白组未见明显凋亡细胞,细胞呈均匀绿色荧光。流式细胞仪检测仅用VP16 10mg/L 3d,ASPO作用2d后加VP16 10mg/L 1d及ASPO作用3d均出现凋亡峰,凋亡率分别为16%,72%及52%,而SPO组及空白但均未见凋亡峰。流式细胞仪检测CA46细胞c—myc蛋白为95.3%,经ASPO作用后降为23.7%,同时SPO组c—myc表达率为90.6%,TUNEL检测ASPO组凋亡细胞为25.5%,加用VP16后的CA46凋亡率为36.7%。结论 体外实验中CA46特异地被c—myc反义核酸抑制生长,细胞出现凋亡表现,c—myc mRNA癌基因表达降低。  相似文献

研究两个反义核酸(18mer,21mer)对人白血病HL—60细胞中c-myc基因表达的抑制作用。用HL-60活细胞计数,RNA含量的RT-PCR测定和c-myc基因蛋白的免疫组化染色检查。结果:两个反义寡核苷酸都能抑制HL-60细胞的增殖,增殖抑制率为11%~75%。反义核酸还抑制c-mycmRNA和Myc蛋白的表达,而对c-mcy基因正常表达的红白血病细胞K562的增殖和PHA刺激的人淋巴细胞的增殖没有影响。结论:反义寡核苷酸能抑制c-myc基因的过度表达。  相似文献

目的:探讨核仁素反义寡核苷酸对细胞增殖与凋亡的影响。方法:采用反义寡核苷酸技术以抑制C2C12细胞中核仁素的表达后,用MTT法检测细胞增殖状况,流式细胞术及DNA琼脂糖凝胶电泳检测细胞凋亡。结果:免疫印迹结果显示,反义寡核苷酸导入细胞后24 h,核仁素的表达受到明显抑制,同时反义寡核苷酸处理组细胞的增殖能力亦明显受到抑制,细胞凋亡百分率显著升高,并能检测到清晰的“梯状条带”。而正义及随机核酸导入细胞后不能降低核仁素的表达,对细胞增殖及凋亡均无明显影响。结论:核仁素表达下调能抑制C2C12细胞增殖并能触发C2C12细胞凋亡。  相似文献

应用相差显微镜、电子显微镜、流式细胞术和DNA电泳等方法观察MGC-803胃癌细胞系凋亡的发生。结果显示30μmoL/L bcl-2反义寡核苷酸、2μg/ml顺铂、20μg/ml卡铂、4μg/ml阿霉素、4μg/ml丝裂霉素处理胃癌细胞,24h后观察到凋亡小体出现、染色质浓缩、凋亡峰及DNA梯状条带等凋亡特征性改变。  相似文献

目的探讨hTERT反义寡核苷酸抑制端粒酶活性对乳腺癌细胞增殖的影响。方法用脂质体介导反义寡核苷酸转染MCF-7乳腺癌细胞株,采用TRAP-ELISA法检测转染后细胞端粒酶的活性,观察其前后的变化。MTT法检测细胞活性,流式细胞仪观察细胞周期及细胞凋亡率变化。结果转染细胞的端粒酶活性及细胞生长都明显受到抑制。流式细胞仪检测到细胞凋亡峰,细胞阻滞于S期,正义寡核苷酸则无此作用。结论端粒酶hTERT为靶点的反义寡核苷酸可明显抑制乳腺癌细胞的增殖,且具有促凋亡作用。  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献

目的 探讨猪肺磷脂注射液联合经鼻持续气道正压通气(NCPAP)对呼吸衰竭早产儿的临床疗效及肌酸激酶同工酶活性(CK-MB)的影响.方法 选取呼吸衰竭早产儿80例,分为观察组和对照组各40例.对照组采用NCPAP给氧治疗,观察组给予NCPAP给氧联合猪肺磷脂气管内给药.观察两组患儿治疗前及治疗12h、24 h后PaO2、PaCO2、血氧饱和度(SaO2)、pH的变化情况,检测治疗前及治疗5d后血清CK-MB水平;评估两组患儿的临床治疗效果.结果 两组患儿PaO2、PaCO2、SaO2、pH比较,差异均有统计学意义(P<0.05),其中观察组治疗后的PaO2、SaO2、pH均高于对照组,PaCO2则低于对照组.两组的PaO2、SaO2、pH均随观察时间延长而升高(P<0.05),PaCO2均随观察时间的延长而降低(P<0.05).观察组治疗有效率为87.5%,显著高于对照组的70.0% (P <0.05).治疗5d后两组患儿血清CK-MB水平均较前降低(P<0.05),且观察组明显低于对照组(P<0.05).结论 猪肺磷脂注射液气管内给药联合NCPAP可以显著降低呼吸衰竭早产儿CK-MB的含量,提高治疗有效率,起到很好的呼吸循环支持作用.  相似文献

Evidence obtained from randomized controlled trials (RCTs) has been generally accepted as the gold standard in the evaluation of clinical effectiveness. Readers need to understand the trial design, implementation, results, analysis and interpretation, so as to fully Jnderstand the results of RCTs. Thus, the investigators of RCTs have to report these items in a complete, accurate and clear manner. Since 1998, we have conducted several evaluations on the reporting quality of RCTs published in Chinese journals on traditional Chinese medicine (TCM) and results have shown that there is an urgent need for higher quality RCTs on TCM.  相似文献

Ankylosing spondylitis is a chronic and progressive disorder with inflammation mainly involving the central axis joints. It mainly affects the cervical spine and the lumbosacral area, with the pathogenesis closely related to the kidney and the Governor Vessel (GV). TCM holds that the syndrome is deficiency in origin and excess in superficiality, which is due to insufficiency of the kidney, deficiency of GV, and blocking of the channels with the invasion of exogenous evil, leading to poor circulation of qi and blood and malnutrition of the bones, muscles and joints. The TCM method of tonifying the kidney and strengthening GV to regulate circulation of qi and blood and check the arthralgia pain should be adopted, with the Kidney-Tonifying and GV Strengthening Decoction (益肾强督汤) prescribed.  相似文献
