目的探讨缺氧诱导因子(HIF-1α)的表达与胃癌组织中血管生成的相互关系。方法对48例手术切除后的胃癌标本进行连续切片和HIF-1α、VEGF蛋白的免疫组织化学检测,用CD34单克隆抗体标记血管内皮细胞并计数微血管密度(MVD)。结果在48例胃癌组织中,HIF-1α阳性表达32例(66.7%),VEGF阳性表达29例(60.4%),胃癌组织的MVD值为42.5土14.7,均与正常胃黏膜组比较,差异有显著性。HIF-1α的表达与VEGF的表达、MVD值呈正相关。结论HIF-1/αVEGF通路在胃癌血管生成过程中起重要作用。 相似文献

目的 探讨合成化合物脂蛋白酯酶活化剂NO-1886对高糖高脂喂养小型猪肾脏蛋白激酶C表达的影响.方法 广西巴马小型猪共15只,按体重随机分为3组,5只/组:正常对照组,喂正常猪饲料;高糖高脂组,喂高糖高脂饲料;加药组,喂加入1.0%NO-1886的高糖高脂饲料.动物分栏喂养,每日投食3次,日粮为体重的4%,自由饮水.实验期为5个月.实验前和实验过程中每月末分别抽取血样,葡萄糖氧化酶法检测空腹血糖;GPO-PAP酶法测定血浆三酰甘油;放射免疫法测血浆胰岛素.第5个月从左肾下极的相同部位取部分组织(包括皮质和髓质),HE染色以观察细胞显微结构,同时免疫组化染色;免疫组织化学方法和Western blot方法分别检测肾脏蛋白激酶C表达情况.结果 使用NO-1886组的葡萄糖、胰岛素和三酰甘油水平显著下降;肾脏细胞显微结构结构基本正常;免疫组织化学方法和Western blot方法检测的结果同时说明NO-1886可以降低高糖高脂饮食所致巴马小型猪肾脏组织中蛋白激酶C的表达.结论 NO-1886可以通过降低糖尿病肾病中的高血糖和高血脂,下调巴马小型猪肾脏组织中蛋白激酶C的表达. 相似文献

目的:研究补中益气法对甲状腺功能减退(甲减)大鼠心肌肌球蛋白重链α和β(myosin heavy chain alpha and beta,α-MHC和β-MHC)mRNA表达的影响。方法:将甲减Wistar大鼠随机分为模型对照组、L-T4组、补中益气汤组。实时定量RT-PCR法测心肌α-MHC和β-MHC mRNA。结果:给处理因素8周,L-T4组、补中益气汤组的α-MHCmRNA的表达分别为正常组的1.77倍、2.32倍,补中益气汤组较L-T4组上升明显(P<0.05);L-T4组、补中益气汤组β-MHCmRNA的表达分别为正常组的2.61倍、1.17倍,补中益气汤组下调明显(P<0.05)。结论:补中益气法在甲减心肌损伤的修复中起重要作用,其机制与其提高心肌α-MHC mRNA的表达、降低β-MHCmRNA的表达有关。 相似文献

Objective To investigate the changes in reactive oxygen species (ROS) and dimethylarginine dimethylaminohydrolase-asymmetric dimethylarginine (DDAH-ADMA) system in the process of endothelial cell senescence after exposure to high glucose. Methods The human umbilical vein endothelial cells (HUVECs) were cultured with different concentrations of glucose, e.g. 5. 5 mmol/L (normal level),and high levels as 11. 0, 22. 0 and 33. 0 mmol/L. for 48 hours, respectively. Subsequently, SA-β-gal staining was used to evaluate senescence of cells. Telomerase activity was detected by polymerase chain reactionenzyme linked immunosorbent assay (PCR-ELISA). The intracellular ROS level was measured by flow cytometry. The ADMA concentration and DDAH activity were determined with high-performance liquid chromatography. Results Compared with normal glucose concentration group, after the endothelial cells were treated with high glucose concentration (11. 0 - 33. 0 mmol/L) for 48 hours, the number of SA-β-gal positive cells was increased significantly [(7.00±1. 73)%, (12. 67±2. 03)%, (16. 00±2. 26)% vs. (4. 00±1.33)%, P>0.05, P<0.05, P<0.05] and the telomerase activity was inhibited dramatically [(91. 32±4.01)%, (78. 44±3. 78)%, (56. 04±3. 35)% vs. 100%, all P<0. 05]. 目的探讨短期持续皮下胰岛素注射治疗(CSII)对初诊2型糖尿病(T2DM)患者血糖控制的影响。方法采用病例对照研究,对58例空腹血糖10 mmol/L的初诊T2DM患者,分为CSII治疗组和每天多次胰岛素注射治疗组(MSII治疗组),每组29例,比较两组治疗前后血糖控制效果、胰岛素用量等。结果两组血糖均显著下降(P0.01);血糖达标胰岛素日最大用量CSII组〔(0.60±0.15)U/kg〕较MSII组〔(0.89±0.21)U/kg〕明显减少(P0.01);血糖达标平均控制天数CSII组〔(6.3±2.1)d〕与MSII组〔(9.5±4.9)d〕相比明显减少(P0.01)。结论 CSII较MSII能更快速有效控制血糖。 相似文献

Objective To clarify the effect of iodine intake on serum thyroglobulin (Tg). Methods A 5-year prospective study was conducted in the 3 different iodine intake areas in China [Panshan (miht deficiency) ,Zhangwu (more than adequate) and Huanghua (excess)]. A total of 3 099 people with normal serum levels of Tg in 1999 were followed and 2 448 of these participants were feasible to be observed in 2004 and included in the present study. The serum levels of Tg, thyraglobulin antibody(TgAb), thyroid peroxidase antibody(TPOAb) and TSH, thyroid volume, family and personal histories of thyroid diseases were measured and inquried. The general linear model (GLM) was used to explore the determinants of Tg. Results Among the study population at baseline, serum Tg were significantly different in three areas [7.5 (4.4-13. 1) μg/L at Panshan, 6.8 (3.6-11.2)μg/L at Huanghua, 5.9 (3.2-10.7) μg/L at Zhangwu, P<0.01]. They were associated with age, sex and the rate of positive TgAb, abnormal thyroid volume, abnormal TSH and positive personal history of thyroid diseases, in order to control the effects of confounding factors, the data from 1856 subjects with thyroid-related indexes all in normal range and without personal history of thyroid diseases were analyzed to clarify the effect of iodine intake on Tg. The serum Tg among three areas were significantly different in both 1999 and 2004, they were all increased in 5 years with significant augment (△ Tg) among the three areas[3.1 (-0.2-8.0) μg/L at Panshan, 3.5 (0.5-9.0)μg/L at Huanghua vs 2. 5(0.3-6.1) μg/L at Zhangwu,P<0.01]. The GLM analysis revealed that age, Tg and TSH levels at baseline were the determinants of △Tg in addition to iodine intake. Conclusion Iodine intake is a dominant determinant of serum Tg.
