目的研究慢性牙周炎牙龈卟啉单胞菌感染患者NLRP3炎症小体与炎症反应及焦亡指标的相关性。 方法选择300例慢性牙周炎患者,按照牙龈卟啉单胞菌感染情况分为阳性组和阴性组。比较两组牙周健康指标、牙周组织NLRP3、凋亡相关斑点样蛋白(ASC)、Caspase-1 mRNA和GSDMD-N蛋白表达水平,以及龈沟液白细胞介素(IL)-1β、IL-18、肿瘤坏死因子-α(TNF-α)水平。采用Pearson检验进行相关性分析。 结果阳性组菌斑指数(PLI)、牙龈指数(GI)、附着丧失(AL)、牙周袋深度(PD)、牙周组织NLRP3、ASC、Caspase-1 mRNA、GSDMD-N蛋白,以及龈沟液IL-1β、IL-18、TNF-α水平均高于阴性组(P<0.05)。NLRP3、ASC、Caspase-1 mRNA表达水平与PLI、GI、AL、PD、GSDMD-N蛋白、IL-1β、IL-18、TNF-α水平均呈正相关(P<0.05)。 结论慢性牙周炎患者牙龈卟啉单胞菌感染加重病情、炎症反应及细胞焦亡,其作用可能与激活NLRP3炎症小体有关。  相似文献

目的 探讨丙泊酚作用核苷酸结合寡聚化结构域样受体蛋白3(NLRP3)/凋亡相关斑点样蛋白(ASC)/半胱氨酸天冬氨酸蛋白酶-1(caspase-1)通路对A549细胞焦亡的影响。方法 超低吸附培养法建立肺癌A549细胞三维培养模型,建立模型后采用不同浓度丙泊酚作用肺癌A549细胞,CCK-8法检测丙泊酚对A549细胞增殖的影响;酶联免疫吸附法检测肺癌A549细胞上清炎性细胞因子白细胞介素(IL)-18、IL-1β、IL-6表达水平变化;蛋白免疫印迹法检测各组焦亡相关蛋白NLRP3、ASC、caspase-1、焦孔素蛋白N端(GSDMD-N)、IL-1β表达水平。结果 三维培养下,A549细胞间有聚拢黏附趋势,培养12 h即可清晰观察到细胞球。与空白对照组相比,随着丙泊酚浓度增加,肺癌A549细胞存活率依次降低(P <0.05);肺癌A549细胞上清炎性细胞因子IL-18、IL-1β、IL-6、焦亡相关蛋白NLRP3、ASC、caspase-1、GSDMD-N、IL-1β表达水平依次增加(P <0.05)。结论 超低吸附培养法成功建立肺癌A549细胞三维培养模型,丙泊酚通过激...  相似文献

目的:探索原发性高血压(EH)患者外周血单核细胞上NLRP3/Caspase-1介导的细胞焦亡与疾病的关系及Pannexin-1(Panx-1)半通道对其的调控。方法:采集EH患者及健康受试者外周血,收集外周血浆,ELISA法检测外周血浆中IL-1β含量;免疫磁珠法分选单核细胞,并通过RT-qPCR、Western blot法测定外周血单核细胞上Panx-1、NLRP3炎症体相关分子、效应分子IL-1β及焦亡蛋白GSDMD的表达。体外培养两组人外周血单核细胞,使用免疫荧光法检测Panx-1的表达及定位。在体外培养的EH患者单核细胞上,使用Panx-1半通道抑制剂丙磺舒及特异性siRNA进行预处理,再给予NLRP3炎症体通路激活剂脂多糖(LPS)活化细胞,CCK-8法检测细胞活力,ELISA法分析培养上清中IL-1β含量,Western blot法检测单核细胞上各目的蛋白表达变化。结果:与健康受试组相比,EH组患者外周血浆中IL-1β含量升高;外周血单核细胞上Panx-1、NLRP3炎症体相关分子及GSDMD的mRNA及蛋白表达量上调;单核细胞上Panx-1蛋白表达增强且主要定位于细胞膜...  相似文献

目的研究卡维地洛对氧化型低密度脂蛋白(ox-LDL)诱导内皮细胞中NLRP3炎症小体介导焦亡及炎症的调控作用。方法人脐静脉内皮细胞(HUVEC)分为对照组、ox-LDL组、低剂量组(0.01 mmol/L)、高剂量组(0.1 mmol/L);转染阴性对照腺病毒(Ad-NC)或过表达NLRP3腺病毒(Ad-NLRP3)后分为Ad-NC组、Ad-NC+ox-LDL组、Ad-NC+高剂量组、Ad-NLRP3+高剂量组。检测各组细胞活力和白细胞介素(IL)-1β、IL-18含量,以及GSDMD-N、NLRP3、Cleaved Caspase-1表达水平。 结果ox-LDL组细胞活力低于对照组,IL-1β、IL-18及GSDMD-N、NLRP3、Cleaved Caspase-1均高于对照组(P<0.05)。低剂量组和高剂量组细胞活力高于ox-LDL组,IL-1β、IL-18及GSDMD-N、NLRP3、Cleaved Caspase-1均低于ox-LDL组(P<0.05)。转染NLRP3腺病毒后,Ad-NLRP3+高剂量组和Ad-NC+ox-LDL组细胞活力低于Ad-NC+高剂量组,IL-1β、IL-18及GSDMD-N、NLRP3、Cleaved Caspase-1均高于Ad-NC+高剂量组(P<0.05)。 结论卡维地洛显著抑制ox-LDL诱导HUVEC中NLRP3炎症小体介导的细胞焦亡及炎症反应。  相似文献

目的探讨瞬时受体电位香草素受体4型通道蛋白(TRPV4)抑制剂HC067047对小鼠膝骨关节炎软骨组织的影响。 方法将30只小鼠均分为假手术组、模型组、HC067047组。假手术组仅切开右侧膝关节髌韧带内侧皮肤,不予处理膝关节囊内结构;模型组、HC067047组手术切除小鼠板股韧带及内侧半月板前角以构建膝骨关节炎,分别用生理盐水、HC067047每天10 mg/kg灌胃。qRT-PCR检测各组小鼠软骨组织中TRPV4 mRNA表达水平。番红固绿染色评估关节软骨受损程度,HE染色分析膝关节软骨下骨的骨量变化。Western blotting检测各组软骨组织中细胞焦亡标志蛋白TRPV4、Caspase-1、核苷酸结合寡聚化结构域样受体家族热蛋白结构域蛋白3(NLRP3)、凋亡相关斑点样蛋白(ASC)、GSDMD-N蛋白水平;ELISA、Western blotting分别检测血清、软骨组织中炎症因子水平。 结果与假手术组比较,模型组TRPV4、NLRP3、ASC、Caspase-1、GSDMD-N、IL-1β、IL-18水平显著升高(P<0.05)。与模型组比较,HC067047组NLRP3、ASC、Caspase-1、GSDMD-N、IL-1β、IL-18水平显著降低(P<0.05)。HC067047能明显改善关节软骨形态及降低骨关节炎受损程度,并能维持软骨下骨的骨量。 结论TRPV4抑制剂HC067047能抑制软骨组织细胞焦亡,改善膝骨关节炎小鼠关节软骨的退变。  相似文献

目的 研究胃癌细胞中NLRP3炎性小体的表达及其对增殖、侵袭和迁移的影响。并初步探讨NLRP3介导caspase-1依赖的焦亡信号通路在胃癌发展中的作用。 方法qRT-PCR和Western blotting法分别检测人胃黏膜上皮细胞(GES-1)和人胃癌细胞(MKN45及MGC803)中NLRP3、caspase-1表达;随后下调NLRP3,检测胃癌细胞NLRP3、caspase-1 mRNA和蛋白的表达;Western blotting检测GSDMD、白细胞介素(IL)-18、IL-1β蛋白表达;ELISA法检测细胞培养液上清液中IL-1β、IL-18含量;乳酸脱氢酶(LDH)实验、流式细胞术检测细胞焦亡;CCK-8法及细胞周期试剂盒检测细胞增殖情况;Transwell和划痕实验检测胃癌细胞侵袭和迁移能力。 结果NLRP3、caspase-1在胃癌细胞中较GES-1表达增高(P < 0.01)。下调NLRP3后,胃癌细胞LDH释放率及焦亡率显著降低(P < 0.01);GSDMD、IL-1β、IL-18表达降低(P < 0.01);细胞增殖阻滞在G1期(P < 0.01);细胞增殖以及侵袭迁移能力显著减弱。 结论NLRP3在胃癌中高表达,其表达影响胃癌细胞的增殖、侵袭和迁移以及其介导的caspase-1依赖焦亡信号通路关键蛋白的表达。  相似文献

【目的】观察黄芩和黄芩苷对脂多糖(LPS)/三磷酸腺苷(ATP)诱导的人脐静脉内皮细胞(HUVECs)NIMA相关蛋白激酶7(NEK7)和Nod样受体蛋白3(NLRP3)小体表达的影响。【方法】将HUVECs随机分为空白对照组、模型组、阿托伐他汀组、黄芩苷组和黄芩血清组,按分组相应药物预处理细胞24 h,然后用1μg/mL LPS刺激细胞24 h,再用5.5 mmol/L ATP刺激细胞4 h后检测各指标。四甲基偶氮唑盐(MTT)法检测细胞活力,均相竞争法测定细胞上清液中内皮素1(ET-1)的含量,硝酸还原酶法测定细胞上清液中总硝酸盐(NOx)水平,蛋白免疫印迹法检测细胞内NEK7、NLRP3、凋亡相关斑点样蛋白(ASC)、裂解型胱天蛋白酶1(cleaved-Caspase-1)、白细胞介素1β(IL-1β)蛋白的表达。【结果】与模型组比较,黄芩血清组、黄芩苷组、阿托伐他汀组LPS/ATP诱导的细胞活力升高,ET-1水平降低、NOx水平升高,NEK7、NLRP3、ASC、cleaved-Caspase-1、IL-1β的蛋白表达水平降低(P<0.05或P<0.01),且黄芩血清组、黄芩苷组的作用效果优于阿托伐他汀组。【结论】黄芩和黄芩苷可以抑制LPS/ATP诱导HUVECs的NLRP3炎症小体激活引起的细胞焦亡而保护内皮,其机制可能与调控NEK7表达有关。  相似文献

目的 研究NLRP3炎症小体及其特异性抑制剂Mcc950对新生儿坏死性小肠结肠炎(NEC)的作用及机制.方法 将120只1日龄新生SD乳鼠随机分为对照组、NEC组和Mcc950治疗组,每组40只.通过人工喂养、缺氧复氧、冷刺激加脂多糖(LPS)灌胃建立NEC动物模型.采用HE染色观察各组肠组织病理表现,并对肠组织行损伤评分;采用蛋白质印迹技术和实时荧光定量PCR(RT-qPCR)检测炎症因子IL-1β和IL-18、炎症小体NLRP3、细胞焦亡主要标志分子Caspase-1和GSDMD的蛋白及mRNA表达水平.结果 与对照组相比,NEC组大鼠体重减轻,肠上皮细胞排列紊乱,部分绒毛坏死、脱落,黏膜层、黏膜下层及固有层可见水肿分离,相应的肠组织损伤评分升高,IL-lβ、IL-18、NLRP3、Caspase-1和GSDMD蛋白及mRNA表达水平升高,差异有统计学意义(P<0.05).与NEC组相比,Mcc950治疗组大鼠体重下降趋势变缓,肠组织病理损伤减轻,肠组织损伤评分降低,IL-1β、IL18、NLRP3、Caspase-1和GSDMD表达降低,差异有统计学意义(P<0.05).结论 成功建立NEC模型,Mcc950可抑制NLRP3介导的炎症及细胞焦亡,从而在NEC中发挥保护作用.  相似文献

目的探讨CXC趋化因子配体14(CXCL14)对高糖暴露环境中脂肪细胞焦亡的影响。方法利用“鸡尾酒法”诱导3T3-L1细胞分化为成熟脂肪细胞,用5.5 mmol/L低糖(NG)或25 mmol/L高糖(HG)葡萄糖培养基培养脂肪细胞24 h;HG环境下用不同浓度CXCL14处理3T3-L1细胞不同时间。Western blot检测消化道皮肤素(GSDMD)、核苷酸结合寡聚化结构样受体蛋白3(NLRP3)、天冬氨酸蛋白水解酶-1(Caspase-1)蛋白、白细胞介素-6(IL-6)蛋白表达水平。实时荧光定量PCR检测GSDMD、NLRP3、白细胞介素-1β(IL-1β) mRNA转录水平。LDH测定试剂盒检测脂肪细胞上清液中乳酸脱氢酶(LDH)活力;Annexin V-FITC荧光检测细胞死亡情况;CCK-8法检测各组细胞增殖活力。结果高糖环境下脂肪细胞焦亡发生率升高,CXCL14处理可提高脂肪细胞增殖活力,但脂肪细胞焦亡相关指标却受到CXCL14浓度梯度的不同影响。50 nmol/L CXCL14处理可降低高糖环境下脂肪细胞GSDMD、NLRP3、IL-1β mRNA以及NLRP3蛋白的表达,下调脂肪细胞LDH活力,减少Annexin V-FITC荧光染色细胞死亡率,但25、100、200 nmol/L CXCL14对其焦亡的指标却呈反向趋势。并且50 nmol/L CXCL14干预后脂肪细胞NLRP3、Caspase-1蛋白随干预时间的延长呈先下降再上升趋势。结论CXCL14对高糖环境中脂肪细胞焦亡的影响与其浓度存在相关性。  相似文献

目的探讨mi R-16-5p对缺氧复氧诱导的大鼠正常肝细胞系BRL-3A焦亡的作用以及作用机制。方法通过细胞缺氧复氧的方法构建肝缺血再灌注损伤(HIRI)的体外模型,通过流式细胞术检测细胞焦亡率;通过荧光实时定量PCR(RT-qPCR)检测mi R-16-5p的表达水平;运用mi R-16-5p mimic在细胞中过表达mi R-16-5p并研究mi R-16-5p对细胞焦亡的影响;通过蛋白免疫印迹实验(Western blot)检测焦亡相关蛋白caspase1、IL-1β、IL-18的蛋白表达水平;通过生物信息学分析和双荧光素酶报告基因实验预测并鉴定mi R-16-5p与NLRP1的靶向关系;通过过表达和干扰NLRP1,研究NLRP1及mi R-16-5p/NLRP1轴对细胞焦亡的影响。结果与空白对照组相比,模型组焦亡率极显著上升(P0.01),mi R-16-5p表达量显著降低(P0.05)。过表达mi R-16-5p极显著抑制细胞焦亡(P0.01)并抑制焦亡相关蛋白caspase1、IL-1β、IL-18的表达(P0.01或P0.05)。双荧光素酶报告基因实验显示NLRP1是mi R-16-5p的靶基因,过表达mi R-16-5p显著降低了NLRP1的m RNA和蛋白水平(P0.01或P0.05)。此外,过表达NLRP1逆转了mi R-16-5p对细胞焦亡的抑制作用(P0.01)和对caspase1、IL-1β、IL-18表达的抑制作用(P0.01或P0.05)。结论 MiR-16-5p通过靶向NLRP1来抑制caspase1、IL-1β、IL-18的表达,从而改善缺氧复氧引起的BRL-3A细胞焦亡。  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
