目的 通过网络药理学的方法进行预测,再深一步进行动物实验验证来研究柴胡疏肝散治疗CAG的作用机制。方法 首先在TCMSP数据库中检索柴胡疏肝散的所有活性成分与药物靶点;通过收集PharmGkb、OMIM、GeneCards和DrugBank数据库中收录的慢性萎缩性胃炎的相关靶点。将药物靶点与疾病靶点进行映射筛选出交集靶点,将得到的交集靶点构建PPI网络与活性成分-共同靶点网络,并对其进行GO和KEGG富集分析。最后利用Vina软件进行分子对接实验验证,并通过免疫印迹法验证柴胡疏肝散对两种受体蛋白EGFR和STAT1的影响。结果 最终筛选得到柴胡疏肝散活性成分104个,潜在靶点238个,与慢性萎缩性胃炎的交集靶点52个;GO与KEGG富集分析分别得到2166条目和148条目,主要涉及到JAK-STAT信号通路、TNF信号通路、HIF-1信号通路等;分子对接结果显示EGFR、STAT1两个靶点能够与核心活性成分能够自发结合成较为稳定的构像;免疫印迹法实验证明柴胡疏肝散能够降低大鼠胃黏膜组织EGFR和STAT1蛋白表达。结论 通过网络药理学和实验验证,发现柴胡疏肝散可能通过调节EGFR和STAT1蛋白表达来共同调控胃黏膜细胞增殖与凋亡,进而发挥着治疗慢性萎缩性胃炎的效果,为深入进行柴胡疏肝散治疗慢性萎缩性胃炎的作用机制研究提供新思路和新方法。  相似文献

Non-alcoholic fatty liver disease (NAFLD) is one of the most common complications of a metabolic syndrome caused by excessive accumulation of fat in the liver. Orthosiphon stamineus also known as Orthosiphon aristatus is a medicinal plant with possible potential beneficial effects on various metabolic disorders. This study aims to investigate the in vitro inhibitory effects of O. stamineus on hepatic fat accumulation and to further use the computational systems pharmacology approach to identify the pharmacokinetic properties of the bioactive compounds of O. stamineus and to predict their molecular mechanisms against NAFLD. Methods: The effects of an ethanolic extract of O. stamineus leaves on cytotoxicity, fat accumulation and antioxidant activity were assessed using HepG2 cells. The bioactive compounds of O. stamineus were identified using LC/MS and two bioinformatics databases, namely the Traditional Chinese Medicine Integrated Database (TCMID) and the Bioinformatics Analysis Tool for the Molecular Mechanism of Traditional Chinese Medicine (BATMAN-TCM). Pathway enrichment analysis was performed on the predicted targets of the bioactive compounds to provide a systematic overview of the molecular mechanism of action, while molecular docking was used to validate the predicted targets. Results: A total of 27 bioactive compounds corresponding to 50 potential NAFLD-related targets were identified. O. stamineus exerts its anti-NAFLD effects by modulating a variety of cellular processes, including oxidative stress, mitochondrial β-oxidation, inflammatory signalling pathways, insulin signalling, and fatty acid homeostasis pathways. O. stamineus is significantly targeting many oxidative stress regulators, including JNK, mammalian target of rapamycin (mTOR), NFKB1, PPAR, and AKT1. Molecular docking analysis confirmed the expected high affinity for the potential targets, while the in vitro assay indicates the ability of O. stamineus to inhibit hepatic fat accumulation. Conclusion: Using the computational systems pharmacology approach, the potentially beneficial effect of O. stamineus in NAFLD was indicated through the combination of multiple compounds, multiple targets, and multicellular components.  相似文献

目的 基于“伏风暗瘀宿痰”小儿哮喘病机新说,采用网络药理学及实验验证的方法,探索搜风愈喘方拆方“祛宿痰方”治疗儿童哮喘的作用机制,验证中医“宿痰”病机与西医细胞外基质改变病理之间的交通性。方法 通过TCMSP数据库建立“祛宿痰方”的有效成分和靶点数据集,利用OMIM、GeneCards、DrugBank、TTD疾病数据库建立哮喘疾病靶点数据集,利用Cytoscape软件取交集并构建“祛宿痰方”与哮喘的蛋白质互作网络,筛选关键靶点蛋白。利用Metascape数据库进行基因本体(Gene ontology,GO)分析以及京都基因和基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)富集分析。复制卵蛋白(OVA)诱导的哮喘大鼠模型,对核心通路及关键靶点进行实验验证。结果 共得到“祛宿痰方”治疗哮喘的靶点98个,包括IL-13、TP53、TGF-β1、VEGF-A、MMP9等,KEGG得到与哮喘相关的通路287条(P<0.05),包括NF-κB信号通路、PI3K-AKT信号通路、IL-17信号通路等。GO结果显示与哮喘相关生物进程包括炎症反应、细胞外基质调控、氧化应激、血管生成等。动物实验证实“祛宿痰方”可下调大鼠肺组织中p-NF-κB-P65磷酸化水平,抑制NF-κB信号通路的激活,降低IL-13、TGF-β1 mRNA表达量(P<0.05),减少哮喘大鼠肺组织中炎症细胞浸润、黏液产生,从而延缓哮喘的进程。结论 “祛宿痰方”可抑制NF-κB信号通路的激活,降低肺组织中IL-13、TGF-β1 mRNA表达量,可能通过抑制炎症反应、调控细胞外基质等途径作用于哮喘,中医“宿痰”病机与西医细胞外基质改变病理之间存在一定的的交通性。  相似文献

目的 基于网络药理学和分子对接技术探究黄芪-赤芍配伍对治疗慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)的作用机制。方法 利用TCMSP,Pharmmaper数据库,筛选黄芪-赤芍治疗COPD的活性成分和潜在靶点;结合Genecards数据库挖掘的COPD相关靶点,对黄芪-赤芍药对与COPD靶点进行PPI网络构建,交互处理得到黄芪-赤芍药对治疗COPD的关键靶点,并进行GO分析和KEGG通路富集分析;并采用分子对接技术将主要活性成分与TNF-α(肿瘤坏死因子),IL-6(白细胞介素6)等进行分子对接;最后利用A549炎症细胞与人脐静脉内皮细胞缺氧损伤模型进行体外细胞实验对结果加以验证。结果 黄芪-赤芍药对中44个有效成分作用于COPD,核心成分为:槲皮素、山奈酚、丁子香萜、芍药苷、(2R,3R)-4-methoxyl-distylin、二氢异黄酮;黄芪-赤芍药对通过IL6、PTGS2、TNF等113个靶蛋白,调控Ras、PI3KAkt、IL-17等多条信号通路治疗COPD,且分子对接结果显示槲皮素、山奈酚、丁子香萜、芍药苷与IL-6、PTGS2、TNF大分子蛋白有良好的结合性,体外细胞试验证实,槲皮素与山奈酚均能减少IL-8,MMP-9炎症因子的分泌,具有不同程度的抗炎效果;芍药苷有明显的扩血管、抗血栓之效。结论 黄芪-赤芍药对治疗COPD具有多成分、多靶点、多通路、整体调节的作用特点。初步揭示了黄芪-赤芍药对通过抑制炎症反应、调节上皮细胞生长增强保护屏障等预测出黄芪-赤芍药对治疗COPD的潜在作用机制,以期为其活性成分的药效物质基础提供理论研究和思路。  相似文献

MicroRNAs (miRNAs) play important roles in prostate cancer development. However, it remains unclear how individual miRNAs contribute to the initiation and progression of prostate cancer. Here we show that a basal layer‐enriched miRNA is required for prostate tumorigenesis. We identify miR‐205 as the most highly expressed miRNA and enriched in the basal cells of the prostate. Although miR‐205 is not required for normal prostate development and homeostasis, genetic deletion of miR‐205 in a Pten null tumor model significantly compromises tumor progression and does not promote metastasis. In Pten null basal cells, loss of miR‐205 attenuates pAkt levels and promotes cellular senescence. Furthermore, although overexpression of miR‐205 in prostate cancer cells with luminal phenotypes inhibits cell growth in both human and mouse, miR‐205 has a minimal effect on the growth of a normal human prostate cell line. Taken together, we have provided genetic evidence for a requirement of miR‐205 in the progression of Pten null‐induced prostate cancer.  相似文献

Hepatocellular carcinoma (HCC) ranks the sixth place of most common cancers. Meanwhile, it is the tertiary mortality cause of cancer. There is no effective therapeutic method to prevent and treat the liver cancer. Sinomenine is a kind of Chinese traditional medicine herbal, it is reported that it can inhibit the viability of several cancer cells. The study is to explore whether sinomenine is also able to inhibit the cell viability of HCC and its potential mechanism. The IC50 of sinomenine in BEL-7402 cells was 5.351 mmol/L, and the IC50 of sinomenine in SMMC-7721 cells was 6.204 mmol/L. The gene expression results showed the relative expression of FGF2, CCND2, DCN, F3, MMP7, NRG1, HMGB1, TRIM29, HAS2, EHF, CTGF, PLK2 were down-regulated, and the relative expression of VEGF A, CITED2, NUPR1, DDX58, IRF9, NAMPT, MMP1, NDRG1, HMGA2, PPARGC1A, IFIT2, PARP9, HEY1, LOX, ETV1, ISG15, BACH, CYLD were up-regulated. Moreover, the IPA analysis results suggested that IFIT3, IFIT1, OAS1, MX1, IRF9, IFI6, IFITM1, ISG15 were up-regulated in BEL-7402 cells treated with sinomenine by activating IFNA2. The findings presented in this study may provide a promising method for the prevention and treatment of liver cancer.  相似文献

David Marr's three-level method for completely understanding a cognitive system and the importance he attaches to the computational level are so familiar as to scarcely need repeating. Fewer seem to recognize that Marr defends his famous method by criticizing the “reductionistic approach.” This sets up a more interesting relationship between Marr and reductionism than is usually acknowledged. I argue that Marr was correct in his criticism of the reductionists of his time—they were only describing (cellular activity), not explaining (cognitive functions). But a careful metascientific account of reductionistic neuroscience over the past two decades reveals that Marr's criticisms no longer have force. Contemporary neuroscience now explains cognition directly, although in a fashion—causal-mechanistically—quite different than Marr recommended. So while Marr was correct to reject the reductionism of his day and offer an alternative method for genuinely explaining cognition, contemporary cognitive scientists now owe us a new defense of Marr's famous method and the advantages of its explanations over the type now pursued successfully in current reductionist neuroscience. There are familiar reasons for thinking that this debt will not be paid easily.  相似文献

摘要:目的 基于Hippo信号通路核心基因mRNA表达,探索具有补肾填精壮骨之效的金刚丸治疗去卵巢(ovariectomized,OVX)大鼠骨质疏松症的疗效机制。方法 通过OVX法建立绝经后骨质疏松症(postmenopausal osteoporosis,PMOP)大鼠模型,分正常组、假手术组、模型组、金刚丸高剂量组、金刚丸中剂量组、金刚丸低剂量组、仙灵骨葆对照组、骨化三醇对照组。灌胃12周后,通过X射线骨密度仪检测骨密度、镜下观察股骨头显微形态结构、ELISA法检测血清ALP、实时定量RT-PCR检测骨组织Mst2、Lats1、Taz mRNA表达。结果 ①与正常组比较,模型组股骨骨密度显著降低(P<0.01)、骨微结构显著破坏、血清ALP显著降低(P<0.01)、骨组织Mst2、Lats1 mRNA表达显著升高(P<0.01)、Taz mRNA表达显著降低(P<0.01);②与模型组比较,除金刚丸低剂量组外,各给药组的骨密度均显著升高(P<0.01),各给药组骨微结构破坏均得到改善、血清ALP均显著升高(P<0.01)、骨组织Mst2、Lats1 mRNA表达均显著降低(P<0.01)、Taz mRNA表达均显著升高(P<0.01),均以金刚丸高剂量组最为显著。结论 骨组织Hippo信号通路核心基因Mst2、Lats1 mRNA表达上调,Taz mRNA表达下调可能是PMOP的发病机制之一;金刚丸可能通过下调骨组织Hippo信号通路核心基因Mst2、Lats1 mRNA表达、上调Taz mRNA表达的机制,有效防治PMOP。  相似文献
