Bronchial asthma is a chronic inflammatory disease of the airways that is characterized by lymphocyte, eosinophil, and mast cell infiltration of the submucosa along with mucous gland hyperplasia and subepithelial fibrosis. The inflammatory response in asthma is tightly associated with airway hyperresponsiveness (AHR) to antigen-specific and nonspecific stimuli. Central to the process of airway inflammation in asthma is the T lymphocyte. Thus, T-cell recruitment and differentiation are critical elements in the evolution of the asthmatic state. Besides adhesion molecules such as selectins and integrins, chemokines are thought to be involved in the multi-step process of extravasation by the triggering of integrins and chemotactic attraction of cell subsets. The diversity of chemokines that potentially attract different cell populations makes them interesting candidates for the regulation of effector cell recruitment. Objective:To observe effects of Liandou Qingmai Recipe(连豆清脉方) on endothelin-1(ET-1),nitric oxide(NO),interleukin-6(IL-6) and IL-10 levels in patients with coronary heart disease.Methods:Total 101 cases with coronary heart disease were randomly divided into a treatment group(n=45) treated by Liandou Qingmai Recipe and a standard treatment group(control group,n=56),with a normal group of 16 health persons set up.Changes of ET-1,NO,IL-6 and IL-10 levels were measured before treatment and after treatment for two weeks.And the data were analyzed by SPSS 16.0 statistic software.Results:Before treatment,the levels of ET-1,IL-6 and IL-10 levels were significantly higher and NO was significantly lower in the patients with coronary heart disease than those in the normal group(90.7±12.7 ng/L vs 41.8±13.5 ng/L,9.17±0.18 ng/L vs 1.10±0.08 ng/L,1.94±0.26 ng/L vs 1.09±0.06 ng/L,and 92.2±17.7 μmol/L vs 124.5±27.2 μmol/L;all P<0.05),with no significant differences in the levels of ET-1,NO,IL-6 and IL-10 between the treatment group and the control group(P>0.05);After treatment,ET-1 and IL-6 significantly decreased in the treatment group and the control group,and NO increased in the treatment group;And IL-6 level was significantly lower and NO level was higher in the treatment group than those in the control group(4.48±1.22 ng/L vs 5.13±1.85 ng/L,117.4±22.3 μmol/L vs 92.4±17.1 μmol/L;both P<0.05);There was a positive correlation between IL-6 and IL-10,and a negative correlation between NO and IL-10(r=0.142,r=-0.152;both P<0.05).Conclusion:Liandou Qingmai Recipe can decline IL-6,IL-10 and ET-1 levels,and raise NO level in patients with coronary heart disease on the basis of standard treatment,so as to inhibit endothelial inflammatory response,improve vascular endothelial function,with stronger anti-AS action;And vascular endothelial lesion is related with inflammatory response.  相似文献

To investigate the effect of interleukin-1β (IL-1β) on IA and IK currents in cultured murine trigeminal ganglion (TG) neurons, whole-cell patch clamp technique was used to record the IA and IK currents before and after 20 ng/mL IL-1β perfusion. Our results showed that 20 ng/mL IL-1β inhibited IA currents (18.3±10.7)% (n=6, P<0.05). IL-1β at 20 ng/mL had no effect on G-V curve of IA but moved the H-infinity curve V0.5 from -36.6±6.1 mV to -42.4±5.2 mV (n=5, P<0.01). However, 20 ng/mL IL-1β had effect on neither the amplitude nor the G-V curve of IK. IL-1β was found to selec- tively inhibit IA current in TG neurons and the effect may contribute to hyperalgesia under various in- flammatory conditions.  相似文献

Objective: Monocytes/macrophages, proinflammatory cytokines and chemokines are important in the pathogenesis of glomerulonephritis. Interleukin (IL) -13 has been shown to exert potent anti-inflammatory properties. This study was designed to investigate the effect of IL-13 on the expression of proinflammatory cytokines, chemokines and profibrogenic cytokines and the involved molecular mechanism in cultured human mesangial cells (HMCs). Methods: The expressions of proinflammatory cytokines, chemokines and profibrogenic cytokines were determined by ribonuclease protection assay (RPA). Activity of nuclear factor-kappa B (NF-κB) and activa- tor protein-1 (AP-1) was examined by electrophoretic mobility shift assay (EMSA). NF-κB subunit p65 nuclear transportation and c-Jun N-terminal kinase (JNK) activity were assayed by immunoblot. Results: Recombinant IL-13 inhibited tumor necrosis factor-α (TNF-u), IL-1α, IL-1β, monocyte chemoattractant protein-1 (MCP-1), IL-8, and transforming growth factor-β1 (TGF-β1) mRNA expressions in a dose-dependent manner. Lipopoly- sacchorides (LPS) dramatically increased NF-κB DNA binding activity of HMCs, which was inhibited by IL-13 in a dose-dependent manner. LPS-activated NF-κB contained p50 and p65 dimers, but not c-Rel subunit. IL-13 blocked LPS-induced NF-κB subunit p65. LPS stimulated JNK/AP-1 activation, which was inhibited by IL-13 in a dose-dependent manner. Conclusion: IL-13 inhibits proinflammatory cytokines, chemokines, and profibrogenic cytokines synthesis by blocking NF-κB and JNK/AP-1 activation. These observations point to the importance of IL-13 in the modulation of inflammatory processes in the renal glomerulus.  相似文献

摘要 目的 骨关节炎(OA)是人类多发性关节炎最常见的形式。许多遗传因素与OA有关。据报道,白细胞介素-6(IL-6)基因多态性与膝骨关节炎(KOA)相关。本研究的目的是,确定不同种群中功能性白细胞介素-6(IL-6)启动子-174C/G(rs1800795)的多态性是否易患膝骨关节炎。 方法 对IL-6基因多态性和KOA之间的相关性进行Meta分析。通过PubMed和中国生物医学数据库电子检索选择分析研究对象。选择含有IL-6-174G/C基因型频率的病例-对照研究,并运用OR的95%置信区间进行相关性评估。 结果 总体结果表明,在所有遗传模型(加性模型: OR=1.146, 95%CI:0.933~1.407, P = 0.194;隐性模型: OR =1.113, 95%CI: 0.799~1.55, P = 0.526;显性模型: OR=1.189, 95%CI =0.918~1.54, P = 0.19)中,基因型(CC/CG/GG)与膝骨关节炎患病风险都不相关。同样,在种族亚组(高加索人、亚洲人)分析,结果均为阴性。对称漏斗图Begg-test(P=0.317)显示缺乏发表偏倚。 结论 在亚洲人和高加索人种群中,本meta分析不支持rs1800795基因型与膝骨关节炎风险性增加的相关性。为了进一步验证rs1800795多态性与KOA的相关性,需要大量世界各地的参与者进行前瞻性研究,才能得出全面的、真实的结论。  相似文献

Human interleukin-15(IL-15) is a proinflammatory cytokine to suppress neutrophil apoptosis,which is a potential therapeutic agent.The modulatory effect of TNFα was investigated in IL-15-induced suppression of human neutrophil apoptosis.TNFα was shown to reverse the ability of IL-15 to delay neutrophil apoptosis within certain time course.Moreover,this reverse effect by TNFα might be associated with a reduction of the expression of the anti-apoptotic Bcl-Xl protein detected by Western blotting.It is concluded that TNFα can be used to modulate IL-15-induced suppression of neutrophil apoptosis within certain time course.  相似文献

The killing effects of lymphocytes on Hela cells expressing interleukin-12 (IL-12) in vitro were explored. By using gene transfection technique, full length IL-12 gene was transfected into Hela cells. The expression of IL-12 in Hela cells was detected quantitatively by ELISA; Changes in killing effects of lymphocytes on Hela cells expressing IL-12 were observed by MTT. It was found that Hela cells could express IL- 12 between 24 h and 72 h after transfection. Killing activity of lymphocytes on Hela cells expressing IL-12 was significantly enhanced. It was concluded by cell transfection technique, Hela cells could express 1L-12 and were more easily killed by lymphocytes.  相似文献
