目的: 采用生物信息学方法研究宫颈鳞状细胞癌(CESC)组织中差异性表达的miRNA并预测其靶基因,以期找到影响CESC发生发展及可用于肿瘤标志物的miRNA。 方法: 癌症基因组图谱 (TCGA)数据库中下载3例CESC组织及3例配对正常组织,用统计学方法对差异性表达的基因及miRNA进行筛选,通过靶基因预测网站对差异性表达的miRNA进行靶基因预测,运用KEGG数据库分析靶基因参与的肿瘤相关信号通路。 结果: 与同源正常组织比较,CESC组织有18个miRNA和1180个基因有差异性表达(P<0.05),其中有15个miRNA和411个基因上调,3个miRNA和770个基因下调,采用靶基因预测网站有7个miRNA与预测到的靶基因呈负向调控关系,6个上调miRNA的靶基因下调,1个下调miRNA的靶基因上调,靶基因集中在Wnt、MAPK、P53和cAMP等肿瘤相关信号通路。 结论: 差异性表达的miRNA包括hsa-miR-27a、hsa-miR-148b、hsa-miR-185、hsa-miR-200a、hsa-miR-200b、hsa-miR-221和hsa-mir-133b,差异性表达的miRNA及其靶基因在CESC的发生发展过程中起重要作用,有可能成为诊断CESC的肿瘤标志物。  相似文献

目的 筛选脾虚证患者血清miRNA表达谱进行生物信息学分析,从miRNA水平探讨脾虚证的发病机制和辨证分型规律。方法 选择高脂血症脾虚证和脾胃湿热证患者各4例,以及健康志愿者5名,提取其血清RNA,进行miRNA定量PCR芯片检测。筛选脾虚证患者血清miRNA表达谱,进行层次聚类、靶基因预测和功能注释等生物信息学分析。结果 筛选出脾虚证患者9个候选血清miRNA,其中6个表达上调,3个表达下调;候选miRNA的表达在脾虚证患者、脾胃湿热证患者及健康志愿者3组样本间存在差异。京都基因与基因组百科全书(KEGG)通路分析结果显示,6个上调miRNA调控的83个靶基因显著富集于7个通路,主要涉及细胞因子-细胞因子受体相互作用通路、病原虫感染性疾病、免疫/炎症相关的信号通路和胰腺癌等;3个下调miRNA调控的365个靶基因显著富集于5个通路,主要涉及神经营养因子信号通路、磷脂酰肌醇信号通路、RNA运输、磷酸肌醇代谢和氨基酸代谢等。结论 本研究筛选出的9个脾虚证患者候选miRNA可作为脾虚证临床辨证的潜在血清标志物,为脾虚证患者发病机制的认识和研究提供依据。  相似文献

目的 构建高血压脑出血患者脑组织差异表达miRNA,并进行生物信息学分析。方法 选取行高血压脑出血手术患者脑血肿腔壁脑组织标本16例,同期选择重型颅脑损伤且具有高血压病史患者脑组织标本16例作为对照组。通过Trizol法提取标本总RNA,筛选出差异表达的miRNA。利用生物信息学技术分析差异表达miRNA的特点,通过String数据库得到靶基因蛋白互作分析结果,Cytoscape软件进行miRNA-mRNA核心调控网络可视化。结果 实验组与对照组相比较共筛选出差异表达miRNA 43个(P<0.05),其中上调miRNA共7个,下调miRNA共36个。对差异miRNA的靶基因进行KEGG/GO富集分析,得到与高血压脑出血相关的信号通路:cAMP信号通路、ErbB信号通路、Calcium信号通路、Wnt信号通路(P<0.05)。构建miRNA-mRNA可视化网络图,发现关联度最高的miRNA为:hsa-miR-1303、hsa-miR-1972、hsa-miR-362-3p、hsa-miR-101-3p、hsa-miR-378d, PPI蛋白互作分析发现连接度最高的前三个基因为:MAPK1、GRB2和RAC1。结论 差异miRNA可能在自发性高血压脑出血的发病机制中具有重要作用,可能为自发性高血压脑出血的治疗及预防提供潜在治疗靶点及方向。  相似文献

目的:探讨高尿酸血症(HUA)患者外周血miRNA表达谱及ceRNA网络构建。方法:对5例HUA患者及5名体检健康者外周血单个核细胞miRNA表达水平进行检测并测序,筛选出差异表达的miRNA分子。利用miRwalk、miRBD、miRTarBase和targetscan数据库预测miRNA的靶基因;circbank数据库预测circRNA-miRNA的靶向结合关系。通过DAVID和Metascape数据库对靶基因的基因功能和通路富集分析。利用Cytoscape构建主要信号通路的circRNA-miRNA-mRNA调控网络。结果:在HUA患者与体检健康者中,共发现47种已知miRNA和4种新miRNA差异表达,其中已知差异表达miRNA有25种上调,22种下调。差异表达miRNA的靶基因主要富集在癌症和细胞衰老相关信号通路。基于ceRNA理论构建的癌症相关信号通路circRNA-miRNA-mRNA调控网络,涉及96个circRNA节点、13个miRNA节点及212个mRNA节点。结论:HUA患者外周血miRNA表达谱与健康体检者存在差异,且差异表达miRNA靶基因与癌症的发生发展密切...  相似文献

【摘要】 目的 对microRNA 200(miR 200)家族成员的靶基因进行预测和生物信息学分析,为进一步探讨miR 200家族的功能及其调节机制提供证据。方法 运用靶基因预测软件TargetScan、PicTar2和miRanda分别预测miR 200家族9个成员的靶基因,再分别取各自在三个软件下预测所得靶基因的交集,然后利用miRTarbase数据库获取这9个miRNA经过至少3种实验验证并且靶向关系类型为具有功能的miRNA 靶基因互作的靶基因,将软件预测所得结果与靶向关系预测结果取并集,作为所纳入每个miRNA的靶基因集合,然后分别对靶基因集合进行生物学过程的功能富集,信号通路及蛋白质互作网络分析。结果 miR-200家族的预测靶基因富集于多个生物学过程,其生物学过程主要富集于:RNA聚合酶Ⅱ启动子的转录负调控、DNA模板转录负调控、RNA聚合酶Ⅱ启动子的转录调控等;信号通路主要富集于:肿瘤、胶质瘤、黑色素瘤、肺小细胞癌等多种肿瘤和EB病毒感染、乙型肝炎等感染相关通路,蛋白质互作网络显示9个成员的预测靶基因编码蛋白质间存在复杂的相互作用,靶基因编码蛋白质“RAC1”、“SRC”、“RHOA”、“CREBBP”、“CTNNB1”在互作网络中具有核心地位。结论 200家族中的成员通过调控靶基因进而调节下游靶蛋白参与肿瘤、感染等病理生理进程。  相似文献

目的比较甲状腺微小乳头状癌患者与甲状腺良性结节患者血浆小分子RNA(miRNA)表达谱的差异,初步分析差异miRNA靶基因的功能。方法收集并分离5例甲状腺微小乳头状癌和5例甲状腺良性结节患者的血浆,采用miRNA-seq方法筛选出两组差异表达miRNA,进一步采用mirdbV5和TaregetScan7.1对差异表达miRNA进行靶基因预测,并运用GeneOntology(GO)分析和KEGG信号通路数据库初步分析靶基因功能。结果与甲状腺良性结节相比,甲状腺微小乳头状癌共有11个miRAN表达有显著差异,其中上调3个,下调8个;3个上调的miRNA通过两个软件共同预测到靶基因76个,8个下调miRNA共同预测到靶基因298个。上调miRNA靶基因GO分析提示与蛋白质修饰和细胞分解代谢有关。下调的miRNA靶基因GO分析结果提示与代谢以及生物合成有关。KEGG分析提示差异miRNA与RNA降解以及HIF-1信号通路相关。结论甲状腺微小乳头状癌与甲状腺良性结节miRNA表达谱存在明显差异,这些差异表达的miRNA可能与蛋白质修饰、细胞代谢以及生物合成、HIF-1信号通路有关。  相似文献

目的初步筛选髓母细胞瘤中差异表达显著的微RNA(miRNA)并探求其生物学功能。方法应用基因芯片获得差异表达的miRNAs,对其中差异显著的目的miRNA行实时定量PCR验证,再利用生物信息学方法预测其潜在靶基因以及靶基因所富集的生物学通路。结果 miR-130a在髓母细胞瘤中显著下调,生物信息学分析表明,血小板衍生的生长因子受体α(PDGFRA)为其靶基因,且该基因与Ras/MAPK通路有关。结论 Ras/MAPK通路在髓母细胞瘤中上调,可能是miR-130a下调引起PDGFRA上调所致。  相似文献

目的 运用生物信息学方法研究复发转移乳腺癌组织中的差异表达MicroRNAs,预测其在乳腺癌预后中的分子调控网络。方法 从GEO数据库中获取乳腺癌组织MiRNA表达谱,包括131例10年内无远处转移复发者,79例10年内有复发者。GEO2R在线分析工具筛选差异表达MiRNA,靶基因预测验证数据库预测miRNA靶基因,使用DAVID工具的基因功能注释和通路富集方法对靶基因进行分析。TF-miRNA调控数据库寻找其上游转录因子,Cytoscape软件构建互作网络。结果 复发转移组乳腺癌组织共筛选到差异表达的miRNA 5个,其中表达上调3个,表达下调2个。得到4个相关上游转录因子HIF1A、EGR1、FOXM1、ESR2,与差异miRNA共调控154个的靶基因。靶基因功能集中在BMP信号通路,TGF-β信号通路,细胞骨架组装功能和Rho GTPases信号通路。结论 差异性表达的miRNA包括hsa-miR-210、hsa-miR-33a、hsa-miR-32、hsa-miR-135a、hsa-miR-30a-3p,利用生物信息学方法,能够有效获取信息,为乳腺癌的治疗及预后监测的新策略研究开辟新思路。  相似文献

目的  研究缺氧对肺腺癌A549细胞中microRNA(miRNA)表达谱的影响,分析靶基因参与的生物学功能和通路。方法  基于miRNA芯片技术和生物信息学分析方法,分析缺氧条件和正常氧条件下培养的A549细胞中差异表达的miRNA,并预测该miRNA的靶基因,分析靶基因参与的生物学功能和通路。结果  本研究共筛选出14个差异表达miRNA,包括9个在缺氧细胞中上调miRNA和5个下调miRNA。上调和下调miRNA的靶基因都参与DNA转录、核染色质修饰等功能,并且都参与Wnt信号、转化生长因子-β信号和丝裂原活化蛋白激酶信号通路。结论  缺氧的肺腺癌A549细胞中miRNA表达谱发生显著改变,一些差异表达miRNA对某些基因具有调控作用。

目的:基于生物信息学方法构建多囊卵巢综合征(PCOS)表达的miRNA及miRNA靶基因的调控网络。方法:从GEO数据库的GPL16543平台下载miRNA基因芯片数据集GSE72274,用GEO2R筛选差异表达的miRNA,应用在线数据库miRWalk分析预测miRNA差异表达的靶基因。对靶基因进行GO富集分析和KEGG信号通路富集分析。应用Cytoscape软件绘制miRNA及其相关靶基因调控图。通过STRING网站对靶基因进行PPI网络构建,并使用CytoHubba插件进行Hub基因分析。结果:共筛选出差异miRNA一共38个,其中32个上调,6个下调。预测差异miRNA的靶基因得到393个,GO分析发现差异的靶基因主要参与RNA聚合酶Ⅱ启动子对转录的调节、RNA聚合酶Ⅱ启动子对转录的正调节、蛋白结合、金属离子结合、D等生物学过程。KEEG分析表明其主要参与TGF-β信号通路和Hedgehog信号通路。成功构建miRNA相关靶基因调控网络,调控网络中的miRNA包括:hsa-miR-135b-5p、hsa-miR-199a-3p、hsa-miR-32-5p。通过CytoHubba...  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

目的:评价使用安心颗粒对急诊经皮冠状动脉介入术(PPCI)术后生活质量的影响.方法:将160例接受PPCI的急性ST段抬高型心肌梗死患者随机分为安心颗粒组(术前顿服安心颗粒8.8g,术后安心颗粒4.4 g/次,每日2次)和对照组(仅接受基础药物治疗).所有患者均服用阿司匹林、氯吡格雷和阿托伐他汀.分别在入院时、出院前1d、出院后180 d时,应用心肌梗死多维度量表(MIDAS)、中文版SF-36评价量表对患者生活质量评分.并观察术后30 d以内的出血并发症、血小板减少症发生情况.结果:入院时和出院前1d,两组患者的心肌梗死MIDAS、SF-36量表评分比较无差异(P>0.05);出院后180 d时,与对照组比较,安心颗粒组MIDAS、SF-36评分明显减低(P<0.05);组内与入院时比较,两组出院前1d、出院后180 d时,MIDAS、SF-36评分均降低(P<0.05).两组患者在随访期间均无大量出血、少量出血、重度和极重度血小板减少症发生,安心颗粒组有4例、对照组有7例发生不明显出血(P>0.05).两组发生轻度血小板减少症的患者数比较无差异(P>0.05).结论:PPCI使用安心颗粒,能改善急性ST段抬高型心肌梗死患者的生活质量,且不增加出血风险.  相似文献

Objectives To explore serum cytokines levels (including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v) and their significance in patients with acute coronary syndrome (ACS) and the subsequent follow-ups, with attempt to estimate the role of various serum inflammatory markers in the diagnosis and assessment of ACS.Methods The study population include 40 patients with acute myocardial infarction (AMI), 40 patients with unstable angina pectoris (UAP), and 40 controls. Among the 80 patients, 60 patients attended a follow up 4 months later. Serum inflammatory markers including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v were measured by enzyme linked immunosorbent assay.Results Serum IL- 1 β, sIL-2R, IL-6, TNF-α were significantly higher in AMI group or UAP group compared to the control group and became significantly lower 4 months later in the follow-up patients. Serum levels of IFN-v shows no significant difference between AMI group or UAP group and controls, also showing no significant change when measured in follow up patients. There was no correlation between serum creatine kinase-MB isoenzyme levels and serum inflammatory markers either in UAP or AMI group. Furthermore, when divided into two subgroups using Wagner's QRS scoring system in the AMI group, there is no difference of each serum inflammatory marker between ≤ 6 scores group and > 6 scores group.Conclusion Serum levels of certain inflammatory markers may have some diagnostic value for ACS, and can be a useful marker reflecting disease stability.  相似文献
