目的探讨紧密连接蛋白的闭合蛋白(Occludin)和紧密粘连蛋白-1(ZO-1)在不同程度脾破裂大鼠血清及回肠黏膜中的表达及意义。方法将48只健康大鼠随机分成脾破裂轻度组、中度组、重度组和假手术组(Sham组)4组,每组12只,前3组采用开腹手术方式制作不同程度脾破裂大鼠模型,Sham组仅接受开、关腹手术。观测大鼠腹腔内情况;ELISA法检测各组大鼠血清Occludin和ZO-1的含量;HE染色法观察回肠黏膜组织病理学改变;免疫组化染色法检测回肠黏膜组织中Occludin及ZO-1的表达情况[以光密度(OD值)表示]。结果Sham组腹腔内无渗出及出血;轻度组腹腔内平均约1.6ml积血,脾脏创面少许渗出;中度组腹腔内平均约3.5ml积血,肠管颜色较苍白,脾脏创面渗出较明显;重度组腹腔内平均约4.3ml积血,肠管苍白明显,肠管间部分粘连,脾脏创面渗出明显。病理学检查显示中度组和重度组大鼠的回肠黏膜损伤较Sham组及轻度组明显为重。与Sham组相比,中、重度组血清中Occludin及ZO-1含量、回肠黏膜组织中Occludin及ZO-1的OD值均有所升高或降低(均P<0.05);与轻度组相比,中、重度组血清中Occludin及ZO-1含量、回肠黏膜组织中Occludin及ZO-1的OD值均有所升高或降低(均P<0.05)。结论血清中Occludin及ZO-1含量增高可作为临床上观测脾破裂及肠黏膜损伤程度的一个可靠指标。  相似文献

目的 探讨胎盘间充质干细胞(P-MSCs)移植对重症急性胰腺炎(SAP)大鼠肠屏障功能的保护作用.方法 24只健康成年雄性SD大鼠,随机均分为SHAM组、SAP组与SAP+P-MSCs组.采用4%牛磺胆酸钠胆胰管逆行注射建立SAP模型,术后6 h经尾静脉输注P-MSCs(1×106个·100 g-1).P-MSCs移植24 h后处死大鼠并取材,采用HE染色与透射电镜技术评估肠道上皮损伤程度,采用ELISA法测定血清D-乳酸、内毒素、IL-1β、TNF-α、CRP浓度与肠道IL-1β、TNF-α与KGF含量.观察P-MSCs在SAP大鼠肠道中分布情况、胰腺与肠系膜淋巴结中细菌移位情况.采用Western-blot法测定肠道中ZO-1与Occudin蛋白表达情况.结果 相比SAP组,SAP+P-MSCs组肠道损伤明显减轻并且有更多P-MSCs定植;血清D-乳酸、内毒素、IL-1β、TNF-α与CRP浓度显著降低(P<0.05);肠道中IL-1β、TNF-α含量显著降低(P<0.05),KGF含量显著升高(P<0.05);胰腺与肠系膜淋巴结组织培养获得的细菌菌落数明显减少(P<0.05);肠道中ZO-1与Occludin蛋白表达量显著升高(P<0.05).结论 外源性P-MSCs移植可减轻SAP大鼠肠道损伤与全身炎症反应,并可显著改善肠道屏障功能.  相似文献

目的研究黄芪多糖对溃疡性结肠炎(UC)大鼠结肠黏膜的修复作用及对肠道炎症反应、黏膜屏障的影响,旨在阐明黄芪多糖治疗UC的分子机制。方法将60只Wister雄性大鼠随机分为空白组,模型组,柳氮磺胺吡啶组,黄芪多糖高、中、低剂量组,每组10只。采用三硝基苯磺酸诱导UC大鼠模型,空白组及模型组予生理盐水灌胃,各药物组分别予柳氮磺胺吡啶及高、中、低剂量的黄芪多糖灌胃,连续灌胃7 d后,取结肠组织进行病理组织学观察、评分和髓过氧化物酶(MPO)活性测定,并采用酶联免疫吸附法(ELISA)测定结肠组织中肿瘤坏死因子-α(TNF-α)、表皮生长因子(EGF)和转化生长因子-β(TGF-β)的含量,通过免疫印迹实验测定结肠紧密连接蛋白Occludin及ZO-1的表达。结果黄芪多糖可剂量依赖性地降低UC大鼠结肠病理组织学评分,结肠组织中的MPO活性及TNF-α、TGF-β含量,升高EGF含量及Occludin、ZO-1蛋白表达水平(P0.05或P0.01),改善大鼠结肠黏膜的病理改变。结论黄芪多糖对UC的治疗作用与减轻肠道炎症反应、促进肠道黏膜修复、改善肠道黏膜屏障功能有关。  相似文献

目的探讨醋酸钠林格液复苏基础上应用乳酸菌对创伤失血性休克大鼠肠黏膜屏障的作用。方法取30只SD大鼠数字法随机分为创伤失血性休克未复苏组(THS组,n=10),醋酸钠林格液复苏组(AR组,n=10),醋酸钠林格液联合乳酸菌复苏组(AL组,n=10),其中AL组大鼠建立休克模型前在正常喂养的基础上加服用乳酸菌素片1周(按照体积面积换算,大鼠每次服用剂量108 mg/kg,每天3次)。THS、AR组及AL组制备成休克模型[平均动脉压维持(35±5)mmHg],AL组及AR组于休克后60 min应用醋酸钠林格液进行30 min液体复苏,复苏后观察4 h取大鼠回肠组织,THS组不予复苏,于休克观察4 h后取回肠组织。利用ELISA检测大鼠外周血肿瘤坏死因子α(TNF-α)、白细胞介素(IL-6)、IL-4、及IL-10含量;应用Western blotting法检测回肠组织ZO-1、Claudin-1、TLR4、p38磷酸化及JNK磷酸化蛋白相对表达水平。结果与THS组相比,AL组大鼠外周血TNF-α、IL-6含量降低,IL-4、IL-10含量升高,AR组TNF-α含量降低,差异均有统计学意义(P<0.05);与AR组比较,AL组大鼠外周血TNF-α含量降低,差异均有统计学意义(P<0.05),其余各细胞因子水平AR组和AL组比较差异无统计学意义(P>0.05)。回肠组织中,与THS组相比,AR组大鼠外周血TLR4、P-P38、P-JNK含量降低,AL组ZO-1、Claudin-1含量升高,差异均有统计学意义(P<0.05);与AR组相比,AL组ZO-1、Claudin-1含量升高,TLR4、P-JNK含量降低,差异均有统计学意义(P<0.05);回肠组织病理学结果显示AL组回肠组织损伤程度轻于THS组与AR组。结论在醋酸钠林格液复苏创伤失血性休克基础上,应用乳酸菌可能进一步抑制TLR4-p38MAPK/JNK炎性信号通路的表达,逆转休克造成的促炎因子和抗炎因子的表达失衡,减轻了创伤失血性休克肠道损伤。  相似文献

目的〓观察傣百解对肠道黏膜屏障功能的保护作用及作用机制。方法 IEC-6细胞分为空白对照组、LPS组、傣百解不同剂量组、LPS+傣百解不同剂量组,分别检测傣百解干预后对IEC-6细胞及LPS诱导后IEC-6细胞模型的活力和炎症因子的水平。另取昆明种小鼠随机分为正常对照组、LPS组、LPS+傣百解高、低剂量组,连续给药1周后,建立小鼠肠道屏障损伤模型,分别检测血清中D-乳酸含量和结肠中TNF-α、IL-1β、IL-6的mRNA表达水平,并观察结肠病理变化。结果 傣百解在30~480 μg/mL范围内对正常IEC-6细胞活力无明显影响,且能对抗LPS对IEC-6细胞活力的抑制作用,降低LPS诱导后IEC-6细胞中的IL-1β、TNF-α、IL-6水平。小鼠肠道损伤模型结果显示傣百解能够降低模型小鼠血清中D-乳酸含量和结肠中IL-6、IL-1β、TNF-α mRNA表达水平,同时减少结肠黏膜的损伤。结论 傣百解对肠道黏膜屏障功能具有保护作用,其作用机制可能与提高IEC-6细胞活力及减轻炎症反应有关。  相似文献

目的 探讨全胃肠外营养对幼鼠肠道炎症损伤的作用及机制。方法 选取18只6~8周龄雄性SD幼鼠,随机分为正常组、对照组和观察组,每组6只。观察组幼鼠行右颈静脉置管后给予全胃肠外营养治疗,并禁食、水;对照组幼鼠行右颈静脉置管后给予生理盐水治疗;正常组不予以置管。正常组和对照组幼鼠均喂食标准鼠粮和水,14 d后处死幼鼠,获取肠道组织。采用HE染色观察各组幼鼠肠道组织的病理变化,TUNEL染色观察各组幼鼠小肠上皮细胞凋亡情况,实时荧光定量聚合酶链反应(qRT-PCR)和Western blotting检测各组幼鼠肠道组织中的肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-10(IL-10)、Occludin和ZO-1的mRNA和蛋白表达,免疫组织化学检测各组幼鼠肠道组织中的Occludin、ZO-1的表达。结果 观察组幼鼠的肠道组织均出现不同程度的损伤;与正常组和对照组比较,观察组肠上皮细胞凋亡率升高(P <0.05),TNF-α、IL-1β的mRNA和蛋白表达升高(P <0.05),而IL-10、Occludin、ZO-1的mRNA和蛋白表达降低(P <0.05);免疫组织化学结果显示,Occludin、ZO-1表达下降(P <0.05)。结论 持续2周的全胃肠外营养可导致幼鼠肠道功能受损,促炎因子增加,肠道机械屏障功能受损,尽可能恢复肠内营养对于减少肠道并发症具有重要意义。  相似文献

目的 探讨黄芪对于大黄诱导腹泻大鼠肠道炎症、肠道屏障和肠道菌群的治疗作用,并阐明其可能的作用机制。 方法 50只大鼠随机分为对照组、模型组、阳性对照组(2.468 g·kg-1参苓白术散)、低剂量(1.35 g·kg-1)黄芪组和高剂量(2.70 g·kg-1)黄芪组,每组10只。采用大黄灌胃结合饮食不节复制大鼠腹泻模型,检测各组大鼠体质量、粪便含水量和腹泻评分;HE染色观察各组大鼠结肠组织病理形态表现,Western blotting法检测各组大鼠结肠组织中蛋白酶激活受体2(PAR-2)、磷酸化p38(p-p38)、肌球蛋白轻链激酶(MLCK)、磷酸化肌球蛋白轻链(p-MLC)、闭锁蛋白1(ZO-1)、闭合蛋白(Occludin)、Toll样受体4(TLR4)、肿瘤坏死因子受体相关因子6(TRAF6)、髓样分化因子88(MyD88)和核因子κB(NF-κB)蛋白表达水平;采用酶联免疫吸附测定(ELISA)法检测各组大鼠血清中胃动素(MTL)、胃泌素(GAS)、肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、白细胞介素10(IL-10)、丙二醛(MDA)和分泌型免疫球蛋白A(SIgA)水平及超氧化物歧化酶(SOD)活性;16S r DNA 基因测序分析各组大鼠肠道菌群情况。 结果 与对照组比较,模型组大鼠体质量明显降低(P<0.01),粪便含水量和腹泻评分明显升高(P<0.01),血清中MTL和GAS水平明显升高(P<0.01);与模型组比较,阳性对照组、低剂量黄芪组和高剂量黄芪组大鼠体质量明显升高(P<0.01),粪便含水量和腹泻评分明显降低(P<0.01),血清中MTL和GAS水平明显降低(P<0.01)。HE染色,与对照组比较,模型组大鼠结肠组织上皮黏膜损伤,有大量炎症细胞浸润,腺体排列紊乱;与模型组比较,阳性对照组和高剂量黄芪组大鼠结肠组织上皮黏膜结构较规则,炎症细胞浸润较少,未见明显腺体紊乱,低剂量黄芪组大鼠结肠组织上皮黏膜结构可见轻微损伤,有部分炎症细胞浸润,腺体排列较整齐。Western blotting法检测,与对照组比较,模型组大鼠结肠组织中PAR-2、MLCK、TLR4、TRAF6、MyD88、NF-κB、p-p38和p-MLC蛋白表达水平明显升高(P<0.01),ZO-1和Occludin蛋白表达水平明显降低(P<0.01);与模型组比较,阳性对照组、低剂量黄芪组和高剂量黄芪组大鼠结肠组织中PAR-2、MLCK、TLR4、TRAF6、MyD88、NF-κB、p-p38和p-MLC蛋白表达水平明显降低(P<0.05或P<0.01),ZO-1和Occludin蛋白表达水平明显升高(P<0.01)。与对照组比较,模型组大鼠血清中TNF-α、IL-1β、IL-6和MDA水平明显升高(P<0.01),IL-10和SIgA水平及SOD活性明显降低(P<0.01);与模型组比较,阳性对照组、低剂量黄芪组和高剂量黄芪组大鼠血清中TNF-α、IL-1β、IL-6和MDA水平明显降低(P<0.01),IL-10和SIg A水平及SOD活性明显升高(P<0.01)。肠道菌群检测,在门水平上,与对照组比较,模型组大鼠肠道菌群中变形菌门和疣微菌门丰度明显升高(P<0.01);与模型组比较,低和高剂量黄芪组大鼠肠道菌群中变形菌门和疣微菌门丰度明显降低(P<0.01),高剂量黄芪组大鼠肠道菌群中ε-变形菌门丰度明显升高(P<0.01)。 结论 黄芪对大黄诱导的腹泻模型大鼠有治疗作用,其机制可能与通过TLR4/TRAF6/NF-κB信号通路抑制肠道炎症,进而修复肠道屏障,恢复肠道菌群组成有关。  相似文献

目的 观察苓桂术甘汤对慢性心力衰竭模型大鼠心肌组织肿瘤坏死因子-α(TNF-α)蛋白及mRNA表达、血清核因子-κB(NF-κB)、白细胞介素-1β(IL-1β)水平的影响,探讨苓桂术甘汤防治慢性心衰的作用机制。方法 采用冠状动脉结扎法制备慢性心衰大鼠模型,造模4周后将模型大鼠随机分为模型组,卡托普利(4.375 mg/kg)阳性对照组,苓桂术甘汤低、中、高剂量(生药2.1、4.2、8.4 g/kg)组,另设假手术组,每天给药1次,连续给药4周。Western blotting、RT-PCR法分别检测各组大鼠心肌组织TNF-α蛋白及mRNA的表达,ELISA法检测血清中NF-κB、IL-1β水平。结果 与假手术组相比,模型组大鼠心肌组织TNF-α蛋白及mRNA表达增强,血清NF-κB、IL-1β水平显著升高(P<0.01);与模型组相比,苓桂术甘汤及卡托普利均能显著抑制模型大鼠心肌组织TNF-α蛋白及mRNA表达、降低模型大鼠血清NF-κB、IL-1β水平(P<0.05、0.01)。结论 苓桂术甘汤干预慢性心衰的机制与其调节细胞因子网络有关。  相似文献

目的 观察盐酸戊乙奎醚(PHC)对脂多糖(LPS)致大鼠急性肾损伤的作用,并探讨其对内毒素血症大鼠紧密连接闭锁小带蛋白(ZO-1)和紧密连接闭锁蛋白(Occludin protein)表达的影响.方法 成年雄性SD大鼠随机分为对照组(C组)、LPS组(LPS组)、PHC低剂量组(L-PHC)和PHC高剂量组(H-PHC),每组10只,后3组注入LPS (5.0 mg/kg),30min后,L-PHC和H-PHC组分别注入PHC 0.3、0.9 mg/kg,24h后取外周血和肾组织,检测血清肌酐(Scr)、尿素氮(BUN)、TNF-α和IL-1β含量;蛋白免疫印迹法检测ZO-1和occludin蛋白的表达.结果 LPS组肾脏炎症明显,而L-PHC和H-PHC组肾脏炎症较弱;与C组比较,各组血浆TNF-α、IL-1β、Scr和BUN含量均显著升高(P<0.05),而L-PHC和H-PHC组其含量比LPC组明显降低(P<0.05);ZO-1和occludin蛋白表达水平LPS组升高,L-PHC和H-PHC组比LPS组下降(P<0.05).结论 PHC可减轻脓毒症诱发的AKI,调控ZO-1和occludin蛋白的表达可能是其发挥保护作用的机制之一.  相似文献

目的 探讨蛋白质二硫键异构酶A3(PDIA3)对溃疡性结肠炎(UC)小鼠的保护作用及其可能机制.方法 将60只C57BL/6小鼠随机分成正常对照组(Control组)、模型组(Model组)、空载慢病毒组(Lv-NC组)和PDIA3过表达慢病毒组(Lv-PDIA3组),每组15只.采用连续7d自由饮用3%葡聚糖硫酸钠(DSS)溶液建立UC小鼠模型,造模成功后开始注射慢病毒干预.慢病毒干预7d后结束实验,对各组小鼠进行疾病活动指数(DAI)评分,HE染色观察结肠组织病理学变化,ELISA法检测血清中炎症因子白介素1β(IL-1β)、白介素6(IL-6)、肿瘤坏死因子α(TNF-α)以及肠黏膜屏障通透性标志物D-乳酸(D-Lac)、二胺氧化酶(DAO)含量,qRT-PCR检测结肠组织中PDIA3 mRNA水平,Western blot检测结肠组织中PDIA3、ZO-1、Occludin、Claudin-1、IκBα、p-NF-κB p65 (Ser536)、NF-κB p65蛋白表达水平.结果 与Control组比较,Model组小鼠结肠组织受损严重,DAI评分、血清中IL-1β、IL-6、TNF-α、D-Lac、DAO含量以及结肠组织中IκBα和p-NF-κB p65 (Ser536)蛋白表达水平均升高,而ZO-1、Occludin和Claudin-1等蛋白表达水平降低,差异有统计学意义(P<0.01);与Model组比较,Lv-PDIA3组小鼠结肠组织受损程度减轻,DAI评分、血清中IL-1β、IL-6、TNF-α、D-Lac、DAO含量以及结肠组织中IκBα和p-NF-κB p65(Ser536)蛋白表达水平降低,同时ZO-1、Occludin和Claudin-1蛋白表达水平增加,差异有统计学意义(P<0.01),而Lv-NC组差异无统计学意义.结论 过表达PDIA3能明显改善UC小鼠肠黏膜屏障功能,其机制可能与抑制NF-κB信号通路的激活有关.  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
