骨质疏松症是一种以骨量低下,骨微结构损坏,导致骨脆性增加,易发生骨折为特征的全身性骨病。随着老龄化的加剧,老年人健康问题尤其是骨密度下降引起的骨质疏松越来越引起重视,但是目前原发性骨质疏松症的基础研究和临床诊治主要集中在"肾主骨,生髓"方面,肾虚是其主要病机,而肝对本病的研究和治疗相对较少。原发性骨质疏松症是一个复杂的、多因素导致的疾病,中医整体观要求我们不能一味地治疗某一症状或某一脏腑,要结合传统医学和现代医学分析其病因病机,辨证施治。从中医学肝对骨质疏松的认识到肝与骨质疏松的基础研究再到滋补肝肾的临床疗效,都说明肝对骨质疏松的重要性。补肾兼顾治肝,及早调肝养肝有益于防治骨质疏松症。 相似文献

In an animal model of type I diabetes, the non-obese diabetic (NOD) mouse, the influence of the antioxidant lipoic acid (LA) on the development of diabetes was investigated. Acceleration of diabetes development with cyclophosphamide (CY) resulted in 60% diabetic animals with severely infiltrated islets within 1–3 weeks. Daily administration of lipoic acid for 20 or 30 days around cyclophosphamide treatment suppressed the incidence of diabetes to 30% (P<0.05) and 33%, respectively. Semiquantitative analysis of islet infiltration showed a reduction of severe intraislet infiltration and an increase in the percentage of islets with mild peri-insular and periductular infiltrates (from 8.4 to 29.6 and 25.9%, respectively, P<0.01) after lipoic acid treatment. These results show that the protective effect of lipoic acid on diabetes development correlates with partial suppression of islet inflammation. The anti-inflammatory action of lipoic acid may be due to its ability to scavenge oxygen radicals and to suppress nitric oxide production. 相似文献

目的验证Cyclin D1及其下游信号蛋白对成骨细胞增殖的调控作用,探讨健骨颗粒促进成骨细胞增殖的作用机制。方法培养成骨样细胞株UMR-106,运用CRISPR/Cas9技术敲减Cyclin D1基因后,将细胞分为基因敲减+生理盐水组(组1)、基因敲减+健骨颗粒组(组2)、病毒空载体细胞(组3,阴性对照)。未行基因敲减的细胞分为正常细胞+生理盐水组(组4)和正常细胞+健骨颗粒组(组5)。分别用生理盐水血清或健骨颗粒含药血清干预各组细胞24、48、72 h,通过CCK8法检测细胞增殖情况,利用Real time PCR检测Cyclin D1、Cyclin E、CDK 2、CDK 4、Rb及E2F-1基因的表达水平。结果经Western-bolt检测表明基因敲减的成骨细胞Cyclin D1蛋白表达明显下降。CCK8法检测结果显示,与组4相比,组5增殖速度变快(P0. 05),组3增殖速度无差异(P0. 05),组1和组2增殖速度均减慢(P0. 05);与组5相比,组2增殖速度减慢(P0. 01);与组1相比,组2增殖速度无统计学意义(P0. 05)。Real time PCR检测结果显示,与组4相比,组5 Cyclin D1、CDK2、CDK4、Cyclin E、Rb及E2F-1 mRNA的表达明显增高(P0. 05或P0. 01),组3表达无统计学意义(P0. 05),组1和组2表达明显降低(P0. 05或P0. 01);与组5相比,组2增殖速度减慢(P0. 01);与组1相比,组2无统计学意义(P0. 05)。结论 Cyclin D1及其下游信号蛋白是调控成骨细胞增殖的关键因素;健骨颗粒可以通过调节Cyclin D1及其下游信号蛋白促进成骨细胞增殖。 相似文献

目的:建立同时测定莲子心中莲心碱、异莲心碱及甲基莲心碱含量的HPLC方法,并测定了10个不同产地、批次的建莲莲子心生物碱的含量。方法:Platisil C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-水(55∶45),乙二胺调pH 9.80;检测波长282 nm,流速1 mL·min-1。结果:莲心碱在3.9~115.9 mg·L-1(r=0.999 8)、异莲心碱在4.0~120.7 mg·L-1(r=0.999 8)、甲基莲心碱9.7~290.2 mg·L-1(r=0.999 8)呈现良好的线性关系。平均回收率分别为100.24%,99.17%,100.10%,RSD分别为0.78%,1.31%,1.40%。福建产的10个不同产地、批次的建莲莲子心的莲心总碱的含量多数在1.50%~2.00%。结论:该方法简便、灵敏、准确、可靠。测定的10个产地批次的建莲莲子心中,以产自福建建宁的几何莲子芯茶样品中莲心总碱含量最高,可达2.28%。 相似文献

目的:探讨半夏泻心汤人含药血清对幽门螺杆菌(Helicobacter pylori,HP)感染人胃黏膜上皮GES-1细胞转化生长因子-β(TGF-β)/信号转导蛋白(Smad)信号通路的影响,以揭示该方治疗消化性溃疡可能的作用机制。方法:GES-1细胞分为空白组,HP感染模型组,健康人血清组,雷贝拉唑血清组,用药前血清组,10%半夏泻心汤血清组,20%半夏泻心汤血清组。除空白组外,其余各组均接种HP造成幽门螺杆菌感染模型,采用雷贝拉唑、半夏泻心汤含药血清及用药前、健康人血清进行干预,24 h后收集细胞及其上清液,噻唑蓝(MTT)比色法检测细胞增殖情况;蛋白质免疫印迹法(Western blot)检测Smad2/3,Smad7,TGF-β1及磷酸化Smad2/3(p-Smad2/3),磷酸化Smad7(p-Smad7)蛋白的表达。结果:与空白组比较,HP可明显抑制GES-1细胞的增殖,降低GES-1细胞内Smad2/3,TGF-β1及p-Smad2/3蛋白表达,增加Smad7及p-Smad7蛋白表达(P0.05,P0.01);与HP感染模型组比较,雷贝拉唑血清组及10%,20%半夏泻心汤血清组可明显降低HP对GES-1细胞增殖的抑制,增加GES-1细胞内Smad2/3,TGF-β1,p-Smad2/3蛋白表达,并明显降低Smad7,p-Smad7蛋白表达(P0.05,P0.01);与用药前血清组比较,加入雷贝拉唑、半夏泻心汤含药血清后均可降低HP对GES-1细胞增殖的抑制,其中雷贝拉唑血清组及20%半夏泻心汤血清组可增加GES-1细胞内Smad2/3,TGF-β1,p-Smad2/3蛋白表达,降低Smad7,p-Smad7蛋白表达(P0.05,P0.01)。结论:半夏泻心汤能够减轻HP诱导GES-1细胞的损伤,促进GES-1细胞增殖,治疗HP相关性消化性溃疡,提高溃疡愈合质量,其机制可能与其调控TGF-β/Smad信号通路有关。 相似文献

BACKGROUND: Different training methods have different effects on primary knee osteoarthritis.
OBJECTIVE: To compare the effects of uphill or downhill running on articular cartilage degeneration and inflammatory responses of knee osteoarthritis rats.
METHODS: 108 female Sprague-Dawley rats were randomly divided into six groups: normal control group, ovariectomized group, uphill running group (run & up), downhill running group (run & down), uphill running model group (model & up), downhill running model group (model & down). The rats in the normal control group were raised routinely with no treatment; rats in the ovariectomized group were ovarientomized and raised in routine way; rats in the run & up group and run & down group were subjected to uphill running training on the slop +15° or downhill running training on the slop -15°; rats in the model & up group and model & down group were subjected to uphill running training on the slop +15°or downhill running training on the slop -15° after ovarientomized. Training programs were as follows: 28 m/min, 60 minutes per day, 6 days per week.
RESULTS AND CONCLUSION: Compared with the other five groups, in the model & up group, the Mankin’s score was significantly increased, indicating that articular cartilage degeneration occurred, and the peak of carboxy-terminal telopeptide of type II collagen contained in the urine had passed, at 4 weeks after modeling. At 6 weeks, the Mankin’s score of the model & down group was increased to reach the degeneration standard, but it was still lower than that of the model & up group; the expression levels of tumor necrosis factor-α and interleukin-1β in the synovial fluid were significantly higher in the model & up and model & down groups than the other four groups. At 8 weeks, the Mankin’s score and the expression levels of tumor necrosis factor-α and interleukin-1β were further increased in the model & up and model & down groups, especially in the model & up group. These results indicate that treadmill running exercise after ovariectomized can result in articular cartilage injury and local inflammatory responses; compared with the downhill running training, the uphill running training can cause cartilage injury earlier and more efficiently. 相似文献
