葛根素葡萄糖注射液对犬实验性心肌血的影响   总被引:5,自引:0,他引:5  
目的:观察葛根素葡萄糖注射液对犬急性心肌缺血的影响。方法:结扎犬冠状动脉左前降支靠成急性心肌缺血模型,葛根素葡萄糖注射液83mg.kg^-1连续4h静脉洋,观察心外膜电图,酶及心肌病理染色(N-BT)的变化。结果:葛根素葡萄糖注射液可明显改善心电图指标,限制心肌梗死范围,降低血清肌酸激酶(CK)肌征同工酶 (CK-MB),AST,乳酸脱氢酶(LDH)水平。结论:葛根素葡萄糖注对犬实验性心肌缺血有较好的治疗作用。  相似文献

目的观察白藜芦醇苷注射液对急性心肌梗死模型犬的影响。方法将犬麻醉后,采用冠状动脉左前降支结扎造成心肌梗死模型,分别给予白藜芦醇苷注射液2.5,5.0,10.0 mg.kg-1,测定犬心肌梗死范围并检测犬血清乳酸脱氢酶(LDH)、肌酸激酶(CK)的活性。结果白藜芦醇苷静脉注射可显著降低心肌梗死模型犬心肌缺血的程度,缩小心肌缺血的范围;显著减小梗死范围,降低血清LDH、CK升高的程度;减轻心肌缺血时的细胞损害。结论白藜芦醇苷注射液对急性心肌梗死模型犬心肌细胞有明显的保护作用。  相似文献

目的观察赤苷脉通注射液对犬急性心肌缺血损伤的保护作用。方法采用麻醉犬冠状动脉结扎造成心肌梗死模型,用心外膜标测法测定心肌梗死程度,硝基四氮唑蓝染色法测定心肌梗死面积,并测定血清乳酸脱氢酶(LDH)、肌酸激酶(CK)活性。结果赤苷脉通注射液(4,2,1mg·kg^-1)减轻犬冠脉结扎所致ST段升高总mV数,减小心肌梗死面积,抑制血清LDH、CK活性的升高。结论赤苷脉通注射液对麻醉犬急性心肌缺血损伤有保护作用。  相似文献

目的研究欣通胶囊对冠状动脉结扎诱发犬急性心肌梗死的心肌酶的影响,为开发新药提供实验依据。方法采用结扎犬冠状动脉的造模方法,建立急性心肌缺血模型,观察欣通胶囊对模型犬血浆中谷草转氨酶(AST)、肌酸激酶(CK)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)的影响,观察欣通胶囊对急性心肌缺血的保护作用。结果欣通胶囊可以降低急性心肌梗死模型犬血浆中AST、CK、CK-MB、LDH水平。结论欣通胶囊对犬急性心肌梗死模型有一定的保护作用。  相似文献

目的:观察红花黄色素对犬急性心肌缺血以及血流动力学的影响。方法采用犬冠状动脉结扎心肌缺血动物模型,注射给予红花黄色素,观察其对心肌缺血程度、心肌缺血范围及心肌缺血梗死程度的影响,对血清肌酸激酶及血清乳酸脱氢酶的影响及对血流动力学的影响。结果红花黄色素能减轻心肌缺血程度,降低血清肌酸激酶及乳酸脱氢酶。增加冠脉流量,增加心输出量,对心肌耗氧量无明显影响。结论红花黄色素对犬心肌缺血梗塞具有明显的改善作用。  相似文献

目的观察降纤酶联合葛根素葡萄糖注射液对急性脑梗死的治疗效果。方法将脑梗死患者随机分成降纤酶联合葛根素葡萄糖注射液治疗组与降纤酶对照组。对照组应用应用降纤酶第一天10U,第3,5天各5U;治疗组在此基础上应用葛根素葡萄糖注射液250ml静滴每日一次,连用14天。治疗前后对神经功能缺损进行评分。结果治疗组显效率74.3%,明显高于对照组67.9%,总有效率96.3%亦明显高于对照组85.7%(P<0.05)。结论降纤酶联合葛根素葡萄糖注射液治疗急性脑梗死能够明显改善临床症状,降低致残率。  相似文献

目的观察参芍颗粒对犬急性心肌缺血的影响.方法结扎犬冠状动脉前降支,造成急性心肌缺血模型,观察不同剂量对模型犬心肌缺血不同时间血清乳酸脱氢酶(LDH),磷酸肌酸激酶(CPK)和心肌梗死程度的影响.结果参芍颗粒可明显缩小犬N-BT染色所显示的梗死面积,降低心肌缺血模型犬血清的LDH和CPK.结论参芍颗粒能明显改善急性心肌缺血模型犬的心肌缺血程度,缩小心肌梗死范围,具有保护心肌的作用.  相似文献

目的观察葛根素注射液在5%葡萄糖注射液(聚丙烯瓶)中被吸附的情况。方法以同等实验下玻璃瓶为对照,以紫外分光法测定输液中葛根素的含量变化。结果在实验条件下,塑料瓶中葛根素含量无明显变化。结论以5%葡萄糖注射液为溶媒时,聚丙烯塑料瓶对葛根素注射液无吸附性。  相似文献

目的研究参芎葡萄糖注射液对大鼠心肌缺血的保护及治疗作用,为其临床应用提供科学依据。方法采用冠状动脉结扎致大鼠心肌梗死疾病模型,比较用药组和模型组心电图J点、血清肌酸激酶、血清乳酸脱氢酶、心肌梗死百分率和心肌组织形态学的变化。结果参芎葡萄糖注射液能减轻缺血引起的心肌细胞损伤,明显降低结扎冠状动脉导致的大鼠心电图J点升高,减少心肌梗死百分率,降低心肌缺血大鼠血清肌酸激酶及乳酸脱氢酶活力。结论参芎葡萄糖注射液对心肌缺血有保护作用。  相似文献

目的:探讨葛根素注射液引起急性溶血的机制。方法:用Beagle犬和豚鼠进行葛根素注射液的体内外溶血试验如下:制剂的溶媒丙二醇和pH值对体外Beagle犬红细胞的溶血作用;静脉注射葛根素对豚鼠超氧化物歧化酶(SOD)、丙二醛(MDA)的含量及致敏豚鼠红细胞溶血率的影响;葛根素对Beagle犬和豚鼠红细胞的体外溶血作用和细胞形态的影响。结果:葛根素注射液最大浓度时(10g/L)的pH值为4.48,可致Beagle犬红细胞溶血;而同pH值的0.9%氯化钠注射液未见引起Beagle犬红细胞溶血。含5%和10%丙二醇的葛根素注射液可引起Beagle犬红细胞溶血;单纯5%和10%丙二醇溶液未见引起Beagle犬红细胞溶血。豚鼠静脉注射葛根素注射液组与0.9%氯化钠组比较,红细胞膜MDA含量明显下降(P〈0.01),未见引起氧化性溶血。葛根素注射液致敏豚鼠后与致敏前比较,未见红细胞溶血率升高,未见抗原抗体复合物产生。豚鼠静脉注射葛根素注射液能引起红细胞皱缩甚至破裂。体外非免疫性溶血试验显示葛根素注射液能直接致Beagle犬和豚鼠红细胞溶血,且与剂量相关。结论:葛根素注射液引起的溶血属于药物非免疫性溶血,可能与葛根素注射液直接作用于红细胞,引起红细胞稳定性改变有关。  相似文献

目的:观察丹酚酸胶囊的抗心肌缺血作用。方法:56只体重为(12±2)kg的健康Beagle犬,雌雄兼用,随机分为假手术组,模型对照组、高(100mg/kg)、中(50mg/kg)、低(25mg/kg)剂量的丹酚酸胶囊组,丹参片组(275mg/kg),地奥心血康胶囊组(100mg/kg)。采用结扎冠状动脉前降支法致Beagle犬急性心肌缺血,经十二指肠给药后,用心外膜电图标测法计算心肌缺血程度,硝基四氮唑蓝(NBT)组织化学染色法测定心肌梗死面积,检测血清中磷酸肌酸激酶(CK)和乳酸脱氢酶(LDH),观察丹酚酸胶囊对心肌缺血损伤的保护作用。结果:丹酚酸胶囊可明显降低心肌缺血程度(∑-ST),减少心肌缺血范围(N-ST),缩小心肌梗死面积(MIS),并能显著降低血清CK和LDH活力,明显减轻心肌细胞损伤程度。结论:丹酚酸胶裳对急性心肌缺血犬具有明显的保护作用。  相似文献

采用心外膜电图标测心肌缺血范围及程度,以定量组织学方法(N-BT染色法)测定心肌梗死范围,同时测定血清酶活性及血液流变性的变化,研究了心舒口服液对麻醉犬心肌缺血、心肌梗死、血清AKP、LDH及血液流变学的影响。结果表明:心舒口服液能明显改善犬急性心肌缺血和缩小心肌梗死范围,显著抑制心肌缺血及心肌梗死所引起的血清AKP活性升高,有效地降低低切速下的血液粘度,抑制血小板粘附性和聚集性,改善血液流变性。  相似文献

目的 观察水杉总黄酮对麻醉犬急性心肌梗死的保护作用。方法 采用结扎犬冠状动脉左前降支的方法造成急性心肌梗死模型 ,十二指肠给药 ,测定犬冠脉循环及心肌耗氧量参数 ,心肌缺血程度和缺血范围 ,梗死面积 ,血清CK、LDH活性及FFA含量。结果 水杉总黄酮能降低冠脉阻力 ,增加冠脉流量 ,减少心肌耗氧量 ,减轻心肌缺血程度和缺血范围 ,缩小心肌梗死面积 ,降低血清CK、LDH活性及FFA含量。结论 水杉总黄酮能改善冠脉循环 ,减少心肌耗氧量 ,纠正心肌缺血时FFA代谢紊乱 ,对缺血心肌具有明显保护作用。  相似文献

Prostacyclin (PGI2), a potent vasodilatory substance that effectively inhibits platelet aggregation is under clinical investigation for the treatment of angina pectoris. We studied the effects of PGI2 on transient drug-induced myocardial dysfunction in anesthetized, thoracotomized dogs. Regional myocardial function was assessed using two pairs of piezoelectric transducers and a multidimensional measuring gauge; one pair was implanted in the distribution area of the left circumflex branch (LCX) and the other in the distribution area of the descending branch (LAD) of the left coronary artery. After intravenous injection of isoproterenol (ISO), which led to an increase in systolic shortening in the LCX and LAD areas, a critical stenosis was performed on the LCX and maintained at this level. Intravenous ISO injection now induced regional dysfunction in the LCX-dependent segment with the occurrence of systolic bulging. Infusion of PGI2 at a dosage of 100 ng/kg/min caused a decrease in arterial blood pressure and an increase in heart rate, but had no effect on regional myocardial function. ISO-induced myocardial dysfunction in the critically stenosed LCX segment, which was observed before PGI2 administration, was not ameliorated, but rather aggravated, by PGI2. It may be concluded that PGI2 does not improve normal or ISO-stimulated myocardial function in pressure-dependent perfused areas of the heart.  相似文献

目的:观察注射用丹红对犬冠状动脉结扎造成心肌梗塞模型的治疗作用。方法:采用麻醉犬冠状动脉左前降支结扎造成心肌梗塞模型,测定犬心外膜电图,硝基四氮唑兰(N—BT)染色确定心肌梗塞面积和血清肌酸激酶(creatine kinase,cK)及乳酸脱氢酶(lactate dehydrogenase,LDH)测定。结果:注射用丹红2、6、12g生药/kg静注可显著降低犬冠脉结扎后心肌缺血的程度,显著缩小心肌梗塞范围,显著降低冠脉结扎犬ST段升高总毫伏(mV)数(∑-ST)和sT段升高总点数(N-ST),显著降低血清LDH、CK升高的程度,作用强度与剂量有一定的关系。结论:注射用丹红能显著减轻犬心肌梗塞时的心肌缺血程度和范围,对犬心肌梗塞有治疗作用。  相似文献

目的:研究刺五加总皂苷对犬急性心肌梗死的保护作用。方法:30条犬麻醉后结扎冠状动脉前降支造成急性心肌梗死模型,随机分为5组:生理盐水组、丹参注射液组(0.6mL/kg)、刺五加总皂苷高、中、低剂量组(30、15、7.5mg/kg),记录心外膜电图,检测血清乳酸脱氢酶(LDH)、肌酸磷酸激酶(CPK)活性,计算心肌梗死面积(MIS)。结果:刺五加总皂苷可明显降低实验性心肌梗死犬的ST段升高总毫伏数(∑ST)及ST段升高超过2mV总点数(N—ST),缩小MIS,可降低犬心肌梗死后引起的血清LDH、CPK活性升高。结论:刺五加总皂苷对犬急性心肌缺血有较好的保护作用。  相似文献

Summary The effect of nitroglycerin (N.G.) on the size of myocardial infarction was studied in 12 open-chest dogs with chloralose-urethane anesthesia by means of epicardial ECG-mapping and compared with the influence of simple ligature of the same coronary artery branch. In order to identify the specific effect of N.G. on the heart and the coronary circulation without contribution of the peripheral circulation in 6 dogs aortic pressure was stabilized by a clamp placed around the descending aorta and heart rate was kept constant by atrial pacing.Nitroglycerin (0.5 mg/kg i.v.) infused during the occlusion period of 20 min resulted in a decrease of mean arterial pressure for 27 mm Hg (p<0.005) and left ventricular enddiastolic pressure for 2 mm Hg (p<0.05) indicating a diminishment in oxygen demand. Mean coronary blood flow was reduced to a similar extent (18%) as in the control infarction (22%). Compared to simple occlusion no significant alteration in the extent of ST-elevation in the epicardial electrogram was observed during N.G.-infusion even in the presence of hypotension. In the stabilized group with constant afterload (mean arterial pressure) and heart rate nitroglycerin induced a slight, but not significant decrease of the enddiastolic left ventricular pressure compared to the control group. However, coronary blood flow and ST-elevation were unaffected. Thus, neither in the unstabilized nor in the stabilized group the extent of myocardial injury after ligature of a coronary artery branch were influenced by nitroglycerin. Obviously the improvement in the oxygen demand during nitroglycerin is out balanced by the reduction in oxygen supply so that in effect myocardial infarction size is not altered by N.G. as estimated from epicardial ECG.With support of the Deutsche Forschungsgemeinschaft.  相似文献

目的:观察超微粉通心络对犬急性心肌梗死的作用。方法:结扎犬冠状动脉前降支中段制备犬急性心肌梗死模型;采用氯化三苯基四氮唑(TTC)染色法和心外膜电图测定心肌梗死面积、缺血程度;测定血氧饱和度和血氧分压,计算氧利用率。结果:超微粉通心络明显减小TTC染色法所显示的心肌梗死范围,显著降低心肌梗死犬氧利用率;心外膜电图数据显示,超微粉通心络组与给药前及与模型组比较都明显降低犬心肌缺血程度,犬心肌缺血范围明显减小。结论:超微粉通心络可恢复心肌梗死犬氧利用率,对犬急性心肌梗死有明显的保护作用。  相似文献

目的观察复方降香胶囊(RCC)对犬实验性心肌梗死和大鼠急性心肌缺血的保护作用以及对小鼠常压耗氧量的影响。方法通过结扎犬左冠状动脉前降支产生急性心肌梗死模型,行心外膜电图标测和定量组织学N BT染色法,研究给药后对心肌缺血的影响;大鼠采用异丙肾上腺素(Iso)所致急性心肌缺血模型,观察给药后心电图的变化,测量给予Iso后心肌超氧化物歧化酶(SOD)活性与丙二醛(MDA)的含量;小鼠给予Iso后,观察常压整体耗氧量。结果RCC能显著降低冠脉结扎犬ST段升高总毫伏(mV)数(ΣST)和ST段升高总点数(N ST),减小N BT染色的梗塞面积,拮抗Iso引起的大鼠急性心肌缺血心电图ST段移位程度,增强缺血心肌SOD活性、降低MDA含量,降低小鼠常压下的耗氧量。结论RCC能对冠脉结扎犬的心肌梗死产生明显保护作用,具有抗Iso引起的大鼠急性心肌缺血作用和降低小鼠耗氧作用。  相似文献

1 The antidysrhythmic, haemodynamic and metabolic effects of a new prospective antianginal and antidysrhythmic agent, N-(3-3-dipenylpropyl)-alpha-methyl-beta-cyclohexylethylamine hydrochloride (MG 8926), have been compared with the chemically related substance, prenylamine, in anaesthetized greyhounds and guinea-pigs. 2 When given intravenously 20 min beforehand, both MG 8926 and prenylamine (5 mg/kg) significantly suppressed the early dysrhythmias induced by coronary artery ligation in anaesthetized greyhounds. At a dose of 1 mg/kg, MG 8926 also protected anaesthetized guinea-pigs from dysrhythmias induced by ouabain infusions. 3 In dogs pretreated with MG 8926, metabolic changes indicative of myocardial ischaemia (increased PCO2 and potassium efflux, decreased oxygen content and pH) were less marked than those occurring in control animals. 4 Evidence was obtained that MG 8926, when given either before or after coronary occlusion, was capable of decreasing the severity of myocardial ischaemia as assessed by ST-segment changes in epicardial electrocardiograms.  相似文献
