枝管藻多糖对实验性高脂血大鼠血脂和过氧化水平的影响   总被引:13,自引:1,他引:13  
目的:观察枝管藻多糖对实验性高脂血大鼠血脂和过氧化水平的影响。方法:以Wistar大鼠为研究对象,用高脂饲料建立高脂血症大鼠模型,分别ig枝管藻多糖和烟酸肌醇酯(枝管藻多糖的剂量为150mg·kg~(-1)·d~(-1),300mg·kg~(-1)·d~(-1),烟酸肌醇酯的剂量为 100mg·kg~(-1)·d~(-1)),连续40d。于实验的第 30天和第40天,各采血一次,分别测定血清 TC,TG,LDL-C,HDL-C,MDA,ROS含量和 SOD活性。结果:枝管藻多糖能显著抑制们喂高脂饲料大鼠的 TG(P<0.05)水平的升高,显著降低高脂血症大鼠的血清 TC(P<0.05)和TG(P<0.05)含量,但对血清LDL-C和HDL-C含量的变化几乎无影响;明显降低高脂血大鼠血清ROS,MDA含量,提高血清 SOD活性(P<0.01或 P<0.05)。结论:枝管藻多糖能够显著改善高脂血大鼠体内过氧化状态,但其抗高脂血效果低于烟酸肌醇酯。  相似文献

目的观察中药罗汉果对实验性血脂紊乱SD大鼠血清总胆固醇(TC)、低密度脂蛋白(LDL-c)、三酰甘油(TG)及高密度脂蛋白(HDL-c)水平的影响。方法 50只雄性SD大鼠(体质量180g-220g)适应性饲养7d后随机分为空白对照组和造模组,造模组上午8时至下午18时给予普通饲料喂养,18时至次日上午8时高脂饲料喂养,共4周。4周后禁食12h断尾采血测两组空腹血清TC、LDL-c、TG、及HDL-c水平,造模组血脂各指标显著升高具有统计学意义者,确定为模型建立成功。将造模成功的模型组大鼠再随即分为药物实验组(20只,给予含生药4g/(kg d)罗汉果水溶液灌胃)和模型对照组(20只),后者继续高脂饲料喂养。至第八周后测血清TC、LDL-c、TG、及HDL-c水平。结果实验组血清TC)和LDL-c)的水平显著低于对照组(P<0.05);TG水平较对照组有下降趋势,HDL-c水平较对照组有上升趋势。结论中药罗汉果可显著降低实验大鼠血清TC和LDL-c,可通过调节血脂代谢紊乱发挥抗动脉粥样硬化作用。  相似文献

阿托伐他汀和吉非贝齐治疗老年人高脂血症的疗效比较   总被引:1,自引:0,他引:1  
目的:比较阿托伐他汀和吉非贝齐治疗老年人高脂血症的疗效。方法:40例老年高脂血症患者随机分为阿托伐他汀(治疗组)组和吉非贝齐组(对照组),每组20例。阿托伐他汀口服10mg/d,吉非贝齐口服60Omg,每日2次,疗程均为4周。结果:治疗4周末血总胆固醇(TC)、甘油三酯(TG)及低密度脂蛋白胆固醇(LDL-c)与基线水平比较,两组均有明显下降(P<0.05,P<0.01或P<0.001);高密度脂蛋白胆固醇(HDL-c)有明显升高(P<0.01);治疗组降低TC及 LDL-c疗效明显优于对照组(P<0.01和P<0.001),降低TG及升高HDL-c疗效和对照组无明显差异(P>0.05)。两组均未见明显副作用。结论:阿托伐他汀在降低TC,TG,LDL-c及升高HDL-c方面有很好疗效,无明显副作用,适用于老年患者。  相似文献

目的研究动脉粥样硬化高脂饲料对小鼠体重、血糖血脂水平及动脉粥样硬化斑块形成的作用,并且明确禁食时间对血脂检测结果的影响。方法对10周龄雄性ApoE基因敲除(ApoE-/-)小鼠分别给予高脂饲料及普通饲料,4个月后观察动脉粥样硬化斑块情况;对10周龄雄性C57BL/6J小鼠分别给予普通饲料4周、普通饲料2周+高脂饲料2周、高脂饲料4周,考察动物的体重、肝脏、血糖、血脂水平,并分析正常饮食条件下取材前禁食12、6 h及不禁食条件下对血脂检测结果的影响。结果给予高脂饲料的ApoE-/-小鼠动脉斑块面积显著增加(P<0.01);给予高脂饲料的C57BL/6J小鼠的体重增加(P<0.01),肝脏脂肪样变;血糖、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-c)、高密度脂蛋白胆固醇(HDL-c)水平显著升高(P<0.01),而三酰甘油(TG)水平明显下降(P<0.01)。相比于禁食12 h,禁食6 h及不禁食条件下检测到的三酰甘油水平明显升高(P<0.01)。结论动脉粥样硬化高脂饲料可加速ApoE-/-小鼠形成动脉粥样硬化斑块,显著升高血糖、TC及LDL-c水平,但明显降低TG值。禁食时间可影响血清TG的检测结果。  相似文献

目的探讨海带对高脂血症大鼠血脂的调节作用机制。方法雌性Wistar大鼠40只,高脂饲料喂养建立高脂血症动物模型,海带粉饲料喂养干预治疗。血生化法检测大鼠血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)、高密度脂蛋白(HDL)水平及脂蛋白酯酶(LPL)和肝脂酶(HL)的活性。结果海带治疗组动物血清TG和TC水平较治疗前和模型组显著下降(P<0.05)。LDL水平显著低于模型组(P<0.05),而HDL、LPL和HL水平均显著高于模型组(P<0.05)。结论海带可能通过增强LPL和HL的活性,影响TG、TC、LDL和HDL等血脂组分的代谢,而发挥调节血脂水平的作用。  相似文献

目的 探讨乌贼墨对实验性高脂血症大鼠血脂代谢和抗氧化能力的影响。方法 饲喂高脂饲料建立高脂血症大鼠模型,同时灌胃乌贼墨,剂量分别为50和200mg·kg^-1,连续30d。分别测定大鼠血清总胆固醇(TC),甘油三酯(TG),低密度脂蛋白胆固醇(LDL—c),高密度脂蛋白胆固醇(HDL-c)和一氧化氮(NO)含量,以及大鼠血清和肝脏丙二醛(MDA)含量,超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH—Px)活性。结果乌贼墨能显著性降低模型大鼠血清中的TC,TG和LDL-c含量,升高HDL-C,降低动脉粥样硬化指数(LDL-c/HDL-c);显著提高高脂血症大鼠血清中NO含量;显著降低血清及肝组织中MDA含量,提高SOD和GSH—Px活性。结论乌贼墨可通过调节机体血脂代谢、提高机体内源性抗氧化酶活性,降低机体的过氧化水平,有效预防高脂血症的形成。  相似文献

目的:研究地龙活性蛋白(Earthworm active protein,EWAP)对实验性高脂血症SD大鼠的调脂作用并探讨可能的降脂作用机制。方法:通过高脂饲料及维生素D2联合应用法建立SD大鼠高脂血症模型,以EWAP进行治疗28 d,末次给药后测定大鼠血清中总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL-c)、低密度脂蛋白(LDL-c)水平、脂蛋白脂酶(LPL)、肝脂酶(HL)活性、粪便胆汁酸(FBA)含量以及大鼠体质量变化。结果:EWAP高、中剂量组与模型组比较,TC、TG、LDL-c水平显著降低,同时HL,LPL活性及粪便中FBA排泄量均显著提高,并且EWAP高剂量组大鼠体质量相比模型组增加趋势明显。结论:EWAP对实验性高脂血症大鼠血脂代谢有调节作用,其作用机制可能与提高脂蛋白脂酶、肝脂酶活性,加强肠内胆固醇代谢转化有关。  相似文献

目的建立高脂血症合并血管钙化大鼠实验模型。方法在灌胃38 d高脂乳剂的基础上连续3 d给予大剂量维生素D3注射液(1×106U.kg-1)灌服,继续高脂乳剂灌胃28 d处理大鼠,观察血脂、钙、磷以及胸主动脉的形态学变化。结果在高脂乳剂-大剂量维生素D3-高脂乳剂相继灌胃处理后,大鼠血清总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-c)均明显升高(P<0.01),高密度脂蛋白胆固醇(HDL-c)无明显变化,但HDL-c/TC显著降低(P<0.01);血钙明显上升(P<0.05),主动脉形成较典型的粥样硬化斑块。结论采用高脂乳剂-大剂量维生素D3-高脂乳剂相结合的方法可以成功建立高脂血症合并血管钙化大鼠模型。  相似文献

目的:研究丹红脂肝清胶囊对复合因素所致的脂肪肝的治疗作用。方法:大鼠在小剂量四氯化碳致肝损伤的基础上,合并应用高脂饮食复制大鼠脂肪肝模型。于造模成功后分别给予不同浓度的药物,以血清及肝脏甘油三酯、总胆固醇的含量,血清超氧化物歧化酶和丙二醛及肝脏游离脂肪酸(freefatty acids,FFA)含量为衡量标准,观察药物的治疗效果。结果:与大鼠脂肪肝模型组相比较,丹红脂肝清胶囊高剂量组(0.4 g.kg-1)血清TC、肝脏TG有显著降低(P<0.05);丹红脂肝清胶囊低、中、高剂量组(0.1、0.2、0.4 g.kg-1)血清TG、肝脏FFA含量有显著降低(P<0.01,P<0.05);丹红脂肝清胶囊中、高剂量组(0.20、.4 g.kg-1)肝脏TC、血清MDA有显著降低(P<0.01,P<0.05);丹红脂肝清胶囊低、中、高剂量组(0.1、0.2、0.4 g.kg-1)血清SOD有显著升高(P<0.01,P<0.05)。丹红脂肝清胶囊胶囊高、中、低剂量组病理形态学改变较模型组均有明显改善。结论:丹红脂肝清胶囊对复合因素引起的脂肪肝有很好的治疗作用。  相似文献

Dosiinpartner (DSP) is a newly developed dietary functional food to help control weight. The aim of this study was to evaluate whether DSP combined with a high-fat (HF) diet could influence body weight, fat accumulation, and plasma glucose levels. Mice were fed for 8 weeks with normal diet, HF diet, and HF+10% or 20% DSP diet. Body weight was recorded at 1 week, and plasma levels of total cholesterol, low-density lipoprotein (LDL) cholesterol, high-density lipoprotein (HDL) cholesterol, and glucose were analyzed at the end of the study. Weight increases in the 10% or 20% DSP group were significantly less than in the HF diet group (p<0.05). Plasma total cholesterol and LDL cholesterol levels decreased by 48.3% and 26.8% in the 10% DSP group and by 42.9% and 34.9% in the 20% DSP group, respectively. However, the HDL cholesterol level was unchanged. Glucose levels also decreased by 80.6% in the 10% DSP group but was almost the same in the HF and 20% DSP groups. Our findings indicate that DSP may be beneficial in the regulation of high-fat diet-induced overweight and other complications such as circulatory disorders and diabetes mellitus.  相似文献

1. We have previously identified strong inhibitory effects of Kabosu (Citrus sphaerocarpa Hort.) juice precipitate (KJP) on cholesterol elevation in stroke-prone spontaneously hypertensive rats (SHRSP) fed a cholesterol diet. In the present study, to elucidate the hypocholesterolaemic mechanism, we examined the effect of dietary KJP on lipid metabolism by using SHRSP fed a cholesterol-free diet. 2. Compositions of the experimental diet containing 10% KJP powder were adjusted to those of the control diet. Seven-week-old male SHRSP were fed control or experimental diet for 2 weeks with free access to the diet and water. 3. Serum levels of cholesterol, phospholipid and triglyceride of the KJP group were significantly reduced, which was due to decreases in the very low-density lipoprotein (VLDL), low-density lipoprotein (LDL) and high-density lipoprotein (HDL) fractions. 4. Serum concentrations of apolipoproteins A-I and E (apoA-I and E) of the KJP group were significantly lower than those of the control group, whereas no significant differences were observed in serum apoB and apoA-IV between the two groups. 5. In liver, there were no significant differences in the contents of lipids or relative liver weight between the two groups. The activity of microsomal cholesterol 7alpha-hydroxylase of the KJP group tended to increase, whereas that of microsomal acyl-coenzyme A : cholesterol acyltransferase was significantly reduced, compared with the control group. 6. These results indicate that dietary KJP produces reductions of serum lipid levels, which are due to reductions in VLDL, apoE HDL and apoA-I HDL, and may promote catabolism and excretion of hepatic cholesterol in SHRSP fed a cholesterol-free diet.  相似文献

This study was designed to determine the effects of a novel hydrophilic phytostanol analog, FM-VP4, on total plasma cholesterol, total plasma triglyceride, low-density lipoprotein (LDL) cholesterol, and high-density lipoprotein (HDL) cholesterol concentrations after acute oral administration to gerbils. Gerbils were administered a standard gerbil diet for 4 continuous weeks, and daily water and food intake was monitored and replaced. The diet contained either no FM-VP4 (control) or FM-VP4 at the following concentrations: 0.25, 0.50, 1.0, or 2.0% w/w; six gerbils were fed each diet formulation. After 4 weeks of receiving a single diet formulation, blood was obtained from each gerbil by cardiac puncture and the animals were sacrificed humanely. Plasma obtained from this blood was analyzed for total cholesterol, total triglyceride, and HDL cholesterol levels by standard enzymatic and precipitation techniques. LDL cholesterol levels were calculated using the Friedewald equation. Administration of dietary FM-VP4 resulted in significant decreases in total plasma cholesterol and LDL cholesterol concentrations compared with controls. Dietary FM-VP4 at concentrations of 1% and 2% (w/w) decreased total plasma cholesterol by 3.4 mmol/L compared with controls. This decrease was entirely due to the loss of cholesterol from the LDL pool because LDL cholesterol was decreased by 3.3 and 3.2 mmol/L after 1% and 2% (w/w) FM-VP4, respectively. There were no significant changes in plasma triglyceride or HDL cholesterol concentrations after the administration of FM-VP4. Animals administered 1% or 2% (w/w) FM-VP4 also had significantly lower body weight after 4 weeks of treatment compared with the other groups. However, no unusual behavior was observed in these animals. No major differences in daily water or food intake were observed throughout the study. These findings indicate that FM-VP4 decreases total and LDL cholesterol concentrations.  相似文献

This study evaluated the influence of silymarin (SM) and polyphenolic fraction (PF) of silymarin on cholesterol absorption in rats fed on high cholesterol diet (HCD). HCD induced a remarkable increase in hepatic, plasma, VLDL and LDL cholesterol, a decrease in HDL cholesterol and an elevation in triacylglycerol (TAG) levels in plasma, VLDL and in the liver. SM and PF were administered as dietary supplements (1.0%) in HCD for 18 days. Intestinal cholesterol absorption was measured by dual-isotope plasma ratio method, which calculates percent of cholesterol absorption from the ratio of two labelled cholesterol doses, one given intragastrically (14C) and one intravenously (3H). Silymarin and PF significantly reduced cholesterol absorption in rats fed on HCD and caused significant decreases in plasma and VLDL cholesterol and content of cholesterol and TAG in the liver. The level of HDL cholesterol was significantly increased after silymarin, but not after administration of PF. The levels of TAG in plasma and VLDL were not affected by either silymarin or PF. These results suggest that the inhibition of cholesterol absorption caused by silymarin and its polyphenolic fraction could be a mechanism contributing to the positive changes in plasma cholesterol lipoprotein profile and in lipid content in liver.  相似文献

The hypoglycemic and hypocholesterolemic effects of high and low molecular weight chitosan were evaluated in streptozotocin (STZ)-induced diabetic rats. Rats were divided into three groups of normal rats (Experiment I) and three groups of diabetic rats (Experiment II). The first group received a cellulose (control) diet, the second group received a low MW (1.4 x 10(4)Da) chitosan diet and the third group received a high MW (1.0 x 10(6)Da) chitosan diet. All three diets were containing 0.5% cholesterol. Experiment I: rats fed with high MW or low MW chitosan diet had increased high density lipoprotein (HDL) cholesterol. However, chitosan did not affect plasma glucose in normal rats. Experiment II: significantly decreased plasma glucose and total cholesterol and increased HDL cholesterol and fecal cholesterol excretion were observed in diabetic rats fed with high MW chitosan diet than animals fed with cellulose diet. However, no statistical significant difference in plasma glucose and total cholesterol was observed in diabetic rats fed with low MW chitosan. The total content of SCFAs in cecum was significantly increased and the ratio of acetate to propionate was slight but significantly decreased in diabetic rats after consuming high MW chitosan diet. The activities of hepatic hexokinase were significantly increased and the intestinal disaccharidases including sucrase and maltase were significantly decreased in normal and diabetic rats fed with high MW chitosan diet. Results obtained from the present study demonstrated the potential of high MW chitosan in reducing hyperglycemia and hypercholesterolemia in STZ-induced diabetic rats.  相似文献

Flax and pumpkin seeds are a rich source of unsaturated fatty acids, antioxidants and fibers, known to have anti-atherogenic and hepatoprotective activities. These effects were evaluated in Wistar rats fed with 1% cholesterol diet. The study was performed on 30 male rats divided into three groups: a control group (CD), CD-chol group fed diet with 1% cholesterol and MS-chol group fed diet enriched with flax and pumpkin seed mixture. In CD-chol group, total cholesterol TC, triacylglycerol TG in plasma and liver, plasma LDL-C, atherogenic index AI and LDL/HDL ratio significantly increased. In MS-chol group lipid parameters decreased significantly, plasma and liver fatty acid composition showed an increase of PUFAs (ALA and LA), and MUFAs (oleic and eicosaenoic acid) and a decrease of SFA (palmitic and stearic acid). In plasma and liver of MS-chol group, malondialdehyde levels decreased and the efficiency of antioxidant defense system was improved compared to CD-chol group. Liver histological sections showed lipid storage in hepatocytes of CD-chol group and an improvement was noted in MS-chol group. Our results suggested that flax and pumpkin seed mixture had anti-atherogenic and hepatoprotective effects which were probably mediated by unsaturated fatty acids present in seed mixture.  相似文献

在高脂饲料中加入6%的琉璃苣油饲喂Wistar♂大鼠3wk,其大鼠血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、TC/HDL-C、LDL-C/HDL-C增加值和高密度脂蛋白胆固醇(HDL-C)、高密度脂蛋白亚组分Ⅱ胆固醇(HDL2-C)、HDL2-C与高密度脂蛋白亚组分Ⅲ胆固醇HDL3-C比值及卵磷脂胆固醇酰基转移酶(LCAT)活性的下降显著低于单纯食猪油的高脂对照组,琉璃苣油组大鼠血清LDL-C水平低于月见草油组。高脂摄入所致大鼠肝脂质和肝过氧化脂质(LPO)水平增高,谷胱甘肽过氧化物酶(GSH-Px)活性下降,琉璃苣油致肝脂质过氧化作用低于月见草油。  相似文献

Tomatoes contain the steroidal glycoalkaloid tomatine, which has been reported to form strong, insoluble complexes with cholesterol in vitro. To determine whether tomatine can reduce dietary cholesterol absorption and plasma levels of cholesterol and triglycerides, we fed hamsters a high-fat, high-cholesterol diet with 0.05-0.2% added tomatine in the diet. The tomatine diets induced lowering of serum low-density lipoprotein (LDL) without changing high-density lipoprotein (HDL) cholesterol. Compared to the control diets, four- to fivefold more labeled dietary cholesterol and coprostanol was excreted in the feces of the tomatine-fed hamsters. The amount of cholesterol excreted in the feces corresponded to the amount of tomatine in the diet. These observations suggest that due to the formation of an insoluble tomatine-cholesterol complex and its excretion in the feces, very little dietary tomatine is absorbed from the digestive tract into the blood stream. They are also consistent with the reported low oral toxicity of tomatine compared to other glycoalkaloids.  相似文献

PURPOSE: To study the effect of piperine, an alkaloid, on thyroid hormones and apolipoproteins in high-fat-diet (HFD) and antithyroid drug-induced hyperlipidemic rats. EXPERIMENTAL: Male Wistar rats were first divided into two groups, control diet and high-fat diet (HFD) and then subdivided into four subgroups of ten animals each. The animals were treated with the following regimens for 10 weeks: 1% carboxymethyl cellulose; 10 mg carbimazole (CM)/kg body weight; 10 mg CM + 40 mg piperine/kg body weight, and 10 mg CM + 2 mg atorvastatin /ATV//kg body weight. Lipid profiles, hormone levels, and apolipoprotein levels were studied in all groups. RESULTS: HFD and/or CM administration significantly elevated the plasma levels of total cholesterol, VLDL, LDL, triglycerides, free fatty acids, and phospholipids, but significantly reduced the HDL levels. Moreover, CM administration significantly reduced apo A-I levels and T3, T4 and testosterone levels while significantly elevating plasma apo B, thyroid stimulating hormone (TSH) and insulin levels. The simultaneous administration of piperine and HFD significantly reduced plasma lipids and lipoproteins levels, except for HDL, which was significantly elevated. Piperine supplementation also improved the plasma levels of apo A-I, T3, T4, testosterone, and I and significantly reduced apo B, TSH, and insulin to near normal levels. CONCLUSIONS: The data presented here provide evidence that piperine possesses thyrogenic activity, thus modulating apolipoprotein levels and insulin resistance in HFD-fed rats, opening a new view in the management of dyslipidemia by dietary supplementation with nutrients.  相似文献

目的观察两种旨可平软胶囊内容物对高脂血症SD大鼠模型的调血脂作用。方法SD大鼠用高脂饲料喂养45 d,采血检测胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)和高密度脂蛋白(HDL)水平,血脂升高达到试验要求后随机分组,分为模型对照组,旨可平Ⅰ低、中、高剂量组,旨可平Ⅱ低、中、高剂量组,并以采用标准饲料喂养试验动物设为空白对照组。在继续高脂饲料喂养的同时给予上述药物治疗15 d,再次采血检测上述血脂水平。结果高剂量旨可平Ⅰ可在一定程度上降低高脂大鼠TC水平;旨可平Ⅰ低、中、高3个剂量对高脂大鼠血清HDL均显示出不同程度的降低作用,且呈一定的量效关系,但仅在高剂量下对高脂大鼠血清LDL水平显示出一定的降低作用。旨可平Ⅱ低、中、高3个剂量对高脂大鼠血清TC与LDL水平显示出明显的降低作用,且具有量效关系。在各剂量水平下,与旨可平Ⅰ相比,旨可平Ⅱ对于高脂大鼠血清TC与LDL的降低作用更加显著;在低、中两个剂量水平下,旨可平Ⅱ较旨可平Ⅰ对于血清TC,TG,LDL 3项指标的降低作用更加显著,且中剂量旨可平Ⅱ可升高HDL水平。结论对高脂SD大鼠模型,旨可平Ⅰ和Ⅱ样品对血清TC与LDL均显示出较好的降低作用,但旨可平Ⅱ对HDL和TG的降低作用旨可平Ⅰ较弱。综合多种指标结果,旨可平Ⅱ较旨可平Ⅰ相比,其降脂作用强度较高。  相似文献
