目的研究生存素反义核酸诱导胰腺癌细胞株PANC-1的凋亡和增强对阿霉素敏感性的影响.方法我们应用构建的生存素反义核酸真核表达载体电转染PANC-1细胞;RT-PCR检测生存素mRNA表达、细胞计数和MTT实验测定转染后细胞对阿霉素敏感性;流式细胞仪检测细胞凋亡率.结果生存素反义核酸可抑制PANC-1细胞中生存素mRNA的表达,并能抑制PANC-1细胞生长,提高PANC-1对阿霉素敏感性,流式细胞仪检测凋亡率明显提高.结论生存素反义核酸能增强阿霉素诱导的PANC-1细胞凋亡,联合生存素反义核酸和阿霉素可以改善胰腺癌的治疗效果.  相似文献

目的研究香加皮水提取物(CPE)诱导人胃癌细胞BGC-823凋亡及其作用机制。方法采用G iem sa染色观察细胞凋亡形态学变化;电子显微镜观察凋亡细胞的超微结构变化;流式细胞术和琼脂糖凝胶电泳方法检测BGC-823细胞凋亡率、细胞周期和细胞凋亡的DNA水平变化;RT-PCR方法检测细胞凋亡相关基因bcl-2、bax和surv iv in mRNA表达水平变化;免疫细胞化学方法检测bcl-2、bax和surv iv in蛋白表达的变化。结果经CPE作用后,人胃癌细胞BGC-823出现明显的细胞凋亡形态学变化及超微结构改变,细胞DNA琼脂糖凝胶电泳呈现梯形图。经250μg/mL CPE处理48 h后,多数BGC-823细胞被阻滞在G2/M期,而且细胞发生明显的凋亡变化,BGC-823细胞凋亡率可达18.9%。CPE可抑制BGC-823细胞bcl和surv iv in mRNA及蛋白的表达,促进baxmRNA及蛋白的表达。CPE可明显延长S180荷瘤小鼠生存期,且具有剂量依赖性。结论CPE通过阻滞BGC-823细胞于G2/M期及诱导BGC-823细胞凋亡发挥抗肿瘤作用,其作用机制与抑制细胞的bcl-2和surv iv in基因mRNA及蛋白表达、促进bax基因和蛋白的表达有关。  相似文献

目的:研究缺氧诱导因子-1α(H IF-1α)反义寡核苷酸(ASODN)转染对人胶质瘤细胞增殖及凋亡的影响,探讨以H IF-1α为靶点进行反义寡核苷酸转染治疗胶质瘤的有效性。方法:设计合成针对H IF-1α的寡核苷酸片段.实验分6组:空白对照组、脂质体组、正义组和反义组(1.0μmol/l、2.5μmol/l和5.0μmol/l)。利用脂质体包裹瞬时转染人胶质瘤细胞系U251,MTT法检测细胞增殖;采用AO/EB染色及末端标记法(TUNEL)检测细胞凋亡的变化。结果:转染后各组细胞增殖抑制率分别为(1.30±0.05)%、(1.98±0.35)%、(2.47±0.48)%、(22.30±2.08)%、(33.60±4.19)%和(56.20±4.22)%。转染H IF-1α反义寡核苷酸组较各对照组相比细胞增生下降,凋亡增加(P<0.01)。结论:H IF-1α反义寡核苷酸转染人胶质瘤U251细胞系可以抑制细胞增生并诱导胶质瘤细胞凋亡,H IF-1α有望成为一个极具潜力的肿瘤治疗的靶点。  相似文献

目的 研究VEGF-C反义寡核苷酸对胰腺癌原位种植瘤模型中PANC-1细胞凋亡的影响.方法 建立人胰腺癌细胞株PANC-1裸鼠原位种植瘤自发性淋巴结转移模型,分离原代培养原位灶和淋巴结转移灶中的胰腺癌细胞,并应用荧光定量PCR、流式细胞术检测VEGF-C反义寡核苷酸转染对其凋亡能力的影响.结果 VEGF-C 反义寡核苷酸抑制胰腺癌细胞VEGF-C的表达后,可促进淋巴结转移灶中PANC-1细胞的凋亡,而对原位灶中胰腺癌细胞无明显影响(P>0.05).结论 VEGF-C反义寡核苷酸能显著促进淋巴结转移灶中胰腺癌细胞的凋亡.  相似文献

目的 研究人类端粒酶反义核酸对K-562细胞增殖的影响.方法 采用脂质体介导的基因转染的方法将端粒酶反义核酸导入K-562细胞,用荧光染色观察端粒酶反义核酸诱导K-562细胞凋亡的量效和时效关系,用电镜、DNA琼脂糖凝胶电泳和流式细胞术检测细胞凋亡.结果 端粒酶反义核酸对K-562生长有抑制作用,细胞凋亡率呈剂量和时间依赖性.正义核酸元此效应.结论 端粒酶反义核酸可诱导K-562细胞凋亡,它有可能成为白血病治疗的新靶点.  相似文献

目的 研究α1,4半乳糖糖基转移酶 (α1,4GalT)基因硫代反义核酸对脑胶质瘤细胞系SWO -3 8细胞的增值与凋亡的影响及其与Bcl -2和Bax表达的关系。方法 采用脂质体介导的基因转染方法将人工合成α1,4GalT基因反义核酸转入SWO -3 8细胞中。应用RT -PCR检测对α1,4GalTmRNA表达的影响 ,应用克隆形成实验检测对细胞增殖的作用 ,应用流式细胞术 (FCM)和琼脂糖凝胶电泳检测细胞凋亡的发生 ,应用FCM检测Bax和Bcl -2表达的变化。结果 反义核酸转染后 ,经RT -PCR检测证实α1,4GalTmRNA基因的表达下降 ;克隆形成试验显示细胞生长明显受到抑制 (P <0 .0 1) ;FCM检测表明 ,导入α1,4GalT基因反义核酸的SWO -3 8细胞的凋亡明显增加 (P <0 .0 1) ,Bcl -2蛋白表达显著下降 (P <0 .0 1) ,而Bax蛋白表达显著提高 (P <0 .0 1)。结论 α1,4GalT基因硫代反义核酸可诱导脑胶质瘤细胞系SWO -3 8细胞凋亡 ,其机制与Bcl-2和Bax基因调控有关  相似文献

目的 研究VEGF-C反义寡核苷酸对胰腺癌原位种植瘤模型中PANC-1细胞凋亡的影响.方法 建立人胰腺癌细胞株PANC-1裸鼠原位种植瘤自发性淋巴结转移模型,分离、原代培养原位灶和淋巴结转移灶中的胰腺癌细胞,并应用real time定量PCR、流式细胞术检测VEGF-C反义寡核苷酸转染对其凋亡能力的影响.结果 VEGF-C反义寡核苷酸抑制胰腺癌细胞VEGF-C的表达后,淋巴结转移灶组PANC-1细胞凋亡率为(15.2±2.7)%,显著高于空白组、SODN组(P<0.05);原发灶组PANC-1细胞凋亡率为(5.5±2.7)%,与空白组、SODN组的差异无统计学意义(P>0.05);并且淋巴结转移灶组PANC-1细胞的凋亡率高于原发灶组,差异有统计学意义(P<0.05).结论 VEGF-C表达下调能显著诱导淋巴结转移灶中胰腺癌细胞的凋亡.  相似文献

目的 :观察Survivin反义核酸对人卵巢癌细胞系SKOV3的生长及Survivin基因表达的影响。方法 :应用基因重组技术 ,将在真核细胞中能稳定表达反义Survivin的pcDNA3 SVVas经脂质体LipofectamineTM2 0 0 0 介导转染人卵巢癌细胞株SKOV3,经G4 1 8筛选得到抗性克隆 (SKOV3/SVVas) ,同时以空载体pcDNA3转染SKOV3作为对照(SKOV3/neo) ;绘制细胞生长曲线 ;采用免疫组化、RT PCR和流式细胞仪检测SKOV3、SKOV3/neo及SKOV3/SVVas细胞的内源性SurvivinmRNA及蛋白的表达和凋亡细胞。结果 :与SKOV3及SKOV3/neo细胞比较 ,SKOV3/SVVas细胞增殖和代谢能力均明显降低 ,Survivin蛋白阳性表达率及mRNA相对表达量明显降低 (P <0 .0 1 ) ,细胞凋亡率显著增加 (P <0 .0 1 )。结论 :稳定表达的反义Survivin能够有效抑制SKOV3细胞内源性Survivin蛋白的表达 ,诱导SKOV3细胞凋亡。  相似文献

目的 :探讨Survivin反义核酸对拓扑替康 (TPT)诱导卵巢癌细胞SKOV3凋亡的影响。方法 :在光镜下观察Survivin反义核酸与TPT联合应用后SKOV3凋亡的形态学改变 ,通过DNA电泳检测SKOV3细胞凋亡的情况 ;应用MTT法检测SKOV3细胞对TPT的敏感性。结果 :Survivin反义核酸可明显促进TPT诱导的SKOV3凋亡 ,呈现出特征性的DNA梯状带 ;TPT对SKOV3、SKOV3/neo、SKOV3/SVVas细胞的IC50 值分别为 (1 6 0 .0 0± 6 .36 ) μg/L、(1 5 8.0 0±6 .32 ) μg/L、(4 2 .0 0± 2 .31 ) μg/L ,其差异具有统计学意义 (P <0 .0 1 )。 结论 :Survivin反义核酸可明显促进TPT诱导的卵巢癌细胞SKOV3凋亡 ,提高卵巢癌细胞对TPT的敏感性  相似文献

目的研究survivin反义寡核苷酸对人肝癌耐药细胞株的增殖和凋亡情况,以及对阿霉素化疗敏感性的影响。方法将人肝癌耐药细胞系SMMC-7721/ADM分为脂质体转染组、阿霉素组、正义寡核苷酸转染组、正义寡核苷酸转染 阿霉素组、反义寡核苷酸转染组、反义寡核苷酸转染 阿霉素组共6组。MTT法检测细胞相对存活率,流式细胞仪分析细胞凋亡率变化,RT-PCR和Westernblot印迹法检测细胞survivinmRNA和蛋白表达。结果survivin-ASODN作用后的人肝癌耐药细胞SMMC-7721/ADM的细胞凋亡率明显增高(P<0.05)。survivinmRNA和survivin蛋白表达:脂质体转染对照组、阿霉素组、正义寡核苷酸转染对照组、正义寡核苷酸转染对照 阿霉素组之间survivin蛋白表达无明显差异(P>0.05)。400ng/mlsurvivingASODN组和400ng/mlASODN 阿霉素组survivinmRNA较其他4组显著降低(P<0.05),但其两组之间表达无明显差异(P>0.05)。结论survivin反义寡核苷酸能降低肝癌耐药细胞survivin表达,增强人肝癌耐药细胞对阿霉素的化疗敏感性。  相似文献

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.  相似文献

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.  相似文献

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P<0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
