抗乙型肝炎病毒表面抗原单克隆抗体的研究——12株杂交瘤细胞的建立及其分泌抗体的分析

本文报道12株分泌抗乙型肝炎病毒表面抗原单克隆抗体(HBsAg McAb)杂交瘤细胞的建株及其 McAb 性质的分析结果,用 ID、CEP、PHA 和 ELISA 等四种方法均测得较高水平的McAb。杂交瘤细胞培养液中 McAb 的 PHA 效价为1:80～1:320,由杂交瘤诱生的小鼠腹水 McAb的 ID 和 CEP 效价可高达1:3,200～6,400,PHA 和 ELISA 滴度分别为10~(-4)～10~(-5)和10~(-5)～5×10~(-7)。将这些腹水以 PBS 稀释到 10~(-5),分别与~(125)I-HBsAg(2×10~4cpm)结合,其结合率为52.8～68.4%.12个 McAb 的特异性均良好,其中1个为抗“HBs/d”亚型抗体,其余的均为抗“HBs/a”型特异性抗体。这些 McAb 均属小鼠 IgG_1亚类。选出其中3个 McAb 用于制备 HBsAg 诊断试剂,获得良好结果。     

分泌抗人甲胎蛋白单克隆抗体杂交瘤细胞株的建立

用人甲胎蛋白(AFP)免疫BALB/c小鼠的脾细胞与SP2/O骨髓瘤细胞融合,获得两株分泌抗人AFP单克隆抗体(McAb)的杂交瘤细胞株(G2和G10)。用酶联免疫吸附试验(ELISA)检测细胞培养上清液和小鼠腹水的特异性抗体效价,分别为2～4×10~(-3)和10~(-6)～10~(-7)。两株杂交瘤细胞染色体的众数为104～108条。经体外连续传代及冻存半年复苏后,仍具有分泌抗体能力,说明其性能稳定。两株所分泌的抗体,均属IgGl型。     

抗胰岛素的单克隆抗体

用猪胰岛素免疫的 BALB/C 小鼠脾细胞,与小鼠骨髓瘤细胞 NS-1融合,获得两株分泌抗猪胰岛素单克隆抗体的杂交瘤细胞,命名为4B9和3B2。其中,以4B9细胞的抗体分泌能力最强。其培液抗体的 ELISA 效价为10~(-2)～10~(-3);注入同系小鼠腹腔诱生的腹水抗体效价达10~(-6)～10~(-7)。其分泌抗体属 IgG_1亚类;对猪胰岛素的亲和常数(Ka)为5.86×10~9L/mole;与用 DNA 重组技术制备的人胰岛素,呈交叉反应。     

抗绿脓杆菌Ⅱ群10117株及Ⅳ群10119株McAb杂交瘤细胞株的筛选和鉴定

用SP2/0骨髓瘤细胞分别与绿脓杆菌Ⅱ群10117株和Ⅳ群10119株免疫BALB/c小鼠脾细胞融合,各获得三株连续分泌抗绿脓杆菌单克隆抗体(McAb)的杂交瘤细胞株10117A_1、B_3、D_5及10119A_6、B_4、C_3。经染色体、IFA、ELISA 鉴定分析,证明是能分泌高效价McAb的杂交瘤细胞株。McAb类和亚类鉴定分别为IgG_? IgG_2b和IgM。交叉反应初步表明上述McAb具有较高的型特异性。     

分泌本周氏蛋白单克隆抗体杂交瘤细胞系的建立

本文报告了以人的本周氏蛋白(BJK)免疫BALB/C小鼠脾细胞与SP2/o—Ag14小鼠骨髓瘤细胞在聚乙二醇(PEG)MW400作用下融合,并经7次克隆后获得二株抗人的BJK杂交瘤细胞系(E_6,H_4)。经染色体核型分析证实为杂交瘤细胞。本单克隆抗体经标准兔抗小鼠IgG亚类血清鉴定,属小鼠IgG_2a,IgG_2b。两株细胞经液氮冻存、复苏仍能正常分泌抗体。用酶联免疫吸附试验(ELISA)测定植瘤小鼠的腹水及其血清的抗体滴度分别为10~4—10~5,10~3—10~4。     

鸡白血病/肉瘤病毒杂交瘤细胞株的建立及其单抗特性分析

用提纯的RSV-1抗原免疫BALB/C小鼠,将其脾细胞与SP2/0瘤细胞进行融合,获得10株能稳定分泌特异性抗体的杂交瘤细胞系。用间接ELISA IFA、AGP对其生物学特性进行了分析,10株MCA均属小鼠免疫球蛋白IgG_1,R_5C_(21)a、R_5C_(22a)、R_7C_(32)、R_7C(34)四株MCA与RAV-l、RAV-2、SR-RSV-A、Pr-RSV-C、RAV-50病毒株均有强交叉反应,其余6     

抗猴逆转病毒Ⅰ型(SRV-1)杂交瘤细胞株的建立及应用

以SRV-1蔗糖梯度离心纯化抗原免疫SPF级的BALB/c小鼠,用SP2/0骨髓瘤细胞与免疫小鼠脾细胞融合。应用间接免疫荧光法(IFA)检测抗体,抗原为感染SRV-110d的Raji细胞。筛选出两株SRV-1小鼠杂交瘤细胞株及单克隆抗体(McAb)C_4和C_(15)。两株杂交瘤细胞经3次克隆后分别注入同品系的小鼠腹腔,所产生的腹水经IFA和ELISA法测定其滴度为1:800～1:1600。经琼脂免疫双扩散法鉴定,C_4McAb     

抗ANXA2单克隆抗体的制备及初步鉴定

目的制备ANXA2特异性单克隆抗体(McAb),为研究ANXA2的生物学功能和检测该蛋白提供有力的抗体工具。方法用经IPTG诱导表达的基因重组PGEX-4T-1-ANXA2抗原免疫BALB/c小鼠,利用常规杂交瘤技术和间接ELISA法筛选抗ANXA2杂交瘤细胞,用ELISA法鉴定所得单抗腹水的效价及Western blot鉴定McAb特异性。单抗纯化和Eu3+标记后,分别包被聚苯乙烯板及作为铕标单抗,利用时间分辨荧光免疫分析法(TRFIA)筛选出检测ANXA2蛋白的最佳配伍单抗。结果经细胞融合、筛选、克隆化,获得了16株可分泌高效价单抗的杂交瘤细胞,间接ELISA显示16株McAb与空载体PGEX-4T-1均未发生交叉反应,抗体特异性良好,16株抗体腹水效价为1∶8 000~1∶256 000,培养上清效价为1∶256~1∶512,纯化的腹水单抗经配对试验,2D6和9H9-Eu3+配对最佳。结论获得16株能稳定分泌高特异性抗ANXA2的杂交瘤细胞,能用于后续研究。     

脂蛋白脂肪酶单克隆抗体的制备及特征分析

用牛奶纯化的脂蛋白脂肪酶(LPL),一分子量为56KD,免疫Balb/c小鼠,利用杂交瘤技术,建立了30余株分泌抗朱LPL单克隆抗体的杂交瘤细胞。对其中2E_5、2G_(10)、6D_7、8D_2、7F_4进行特征分析表明:①其抗体类别均为IgG,亚类分别为IgG_1、TgG_(2b)、IgG_(2b)、IgG_1、IgG_1;②抗体效价(细胞培养上清)5×10~(-2)——2×10~(-3);③5种单抗可认为识别三个不同的抗原表位;④Western—blot及Dot—blot结果显示各单抗均能识别纯化的LPL,与人奶均无交叉反应,与牛奶显示不同的结合特性。     

抗脱氧雪腐镰刀茵烯醇单克隆抗体的制备及鉴定

目的 制备抗脱氧雪腐镰刀茵烯醇(DoN)单克隆抗体(McAb)并对其进行初步鉴定.方法 采用小剂量长周期的免疫方案,以DON-牛血清白蛋白(BSA)偶联物免疫雌性BALB/c小鼠,采用细胞融合的方法制备杂交瘤细胞株,酶联免疫吸附试验(ELISA)检测上清中McAb,对抗体分泌阳性的细胞株进行克隆化,直至抗体阳性率100%,用竞争抑制ELISA法进一步检测McAb的特异性,腹水诱生法制备大量McAb,并用饱和硫酸铵对其进行纯化,用抗体亚类鉴定试剂盒对该McAb亚类进行鉴定,BCA蛋白测定试剂盒测定蛋白含量,ELISA检测McAb的滴度、参考工作稀释度和亲和常数.结果 DON-BSA免疫的BALB/c小鼠血清效价为1∶256000,与BSA有强烈的交叉反应.细胞融合后,ELISA筛选抗体分泌阳性的杂交瘤细胞株,经3轮克隆化,建立了1株能稳定分泌抗DON-BSA McAb的杂交瘤细胞株3G5,腹水诱生法制备了大量的McAb.该McAb属IgG1,IgG含量为6.06 g/L,抗体滴度为1∶500000,参考工作稀释度为1∶64 000,抗体亲和常数为1.62×109.用间接竞争抑制ELISA测得校正曲线线性范围9.8～10 000 ng/mL,线性方程Y=-0.272 6X+0.225 9(r=0.930 9).结论 获得了分泌抗DON-McAb的杂交瘤细胞株,该单克隆抗体灵敏度高,特异性强,可用于制备高质量国产DON-ELISA检测试剂盒.

Acupuncture Treatment of Vomiting

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. Because of the fear for vomiting, she did not dare to have food intake, with body weight reduction of 6 kilos in one month.     

Qiu Baoguo''''s Experience in Treating Consumptive Syndromes after Radio-Chemotherapies

Radiotherapy and chemotherapy are the important modern medical therapies for malignant tumors,yet they can also bring about serious local and systemic toxic side reactions so to decrease the patient;'s life quality,manifested by a series of consumptive symptoms.Having engaged in the combined work of Chinese and western medicine for nearly 50 years,the research fellow Qiu Baoguo in Henan Provincial Academy of TCM has developed his unique views on the TCM study of consumptive syndromes.The author of this essay had once the fortune tO follow Dr.Qiu in clinic,and specially would like to introduce in the following Dr.Qiu's experience in treating consumptive syndromes after radio-chemotherapies for patients with malignant tumor.     

Insomnia Due to Deficiency of Both the Heart and Spleen Treated by Acupuncture-Moxibustion and Chinese Tuina

OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.     

Analysis of CK-19,PDX-1,Nestin,Ngn3 in different donor islet purity in rats

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P＜0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.

Acupuncture Combined with Spinal Tui Na for Treatment of Primary Dysmenorrhea in 30 Cases

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.
