目的:探讨γδT细胞在系统性红斑狼疮疾病中的免疫保护作用。方法:检测17例正常人群和44例系统性红斑狼疮患者外周血γδT细胞及亚群的表达情况。以TCRγδ单克隆抗体固相法体外选择性扩增获得17例活动期系统性红斑狼疮患者和10例正常人群外周血TCRγδ细胞系。测定γδT细胞毒活性。用正常人群γδT细胞系与异体活动期SLE患者外周血单核细胞(PMBC)以1:5、1:10比例共同培养48小时,活动期SLE患者PMBC作对照孔,观察γδT细胞系对活动期SLE患者PMBC活化和凋亡、细胞因子分泌的影响。结果:SLE患者的γδT细胞及亚群数量明显减少(P〈0.05)。IL-2可显著增强γδT细胞的细胞毒作用。正常人γδT细胞系与异体SLE活动期患者PMBC细胞共同培养,共同培养二组SLE患者PMBC的CD69表达和凋亡率均较对照组明显下降(P〈0.01)。共同培养二组培养上清IL-10水平较对照组明显升高,具有明显差异(P〈0.05)。结论:SLE患者γδT细胞数及亚群较正常人群明显下降,γδT细胞对SLE外周血淋巴细胞的凋亡和活化具有抑制性作用,并使IL-10分泌水平升高,表明γδT细胞在SLE发病中具有保护性的免疫调节作用。  相似文献

目的:检测系统性红斑狼疮(SLE)患者血清白细胞介素15(IL-15)水平及外周血单个核细胞(PBMC)IL-15mRNA表达,并进一步分析其临床意义。方法:IL-15检测采用ELISA方法;PBMCIL-15mRNA表达采用原位杂交法检测。结果:①SLE组患者血清IL-15水平显著高于正常对照组(P〈0.01),活动期SLE患者血清IL-15水平显著高于缓解期患者(P〈0.05)。②发生临床肾损害者IL-15水平明显高于无肾损害者(P〈0.05),出现血清抗dsDNA抗体阳性、低补体C3血症、高IgG血症者血清IL-15水平均分别显著高于无上述表现者。③SLE患者PBMCIL-15mRNA表达量明显高于正常对照组(P〈0.05),活动期SLE显著高于缓解期(P〈0.05)。④SLE患者PBMC培养上清IgG、IgM和抗dsDNA抗体浓度均显著高于正常对照组;SLE患者PBMCIL-15mRNA表达量与细胞培养上清的IgG及抗dsDNA抗体滴度均呈正相关关系(分别为r=0.645和r=0.715,P〈0.05),而与IgM.无相关关系(r=0.451,P〉0.05)。⑤SEE患者PBMCIL-15mRNA表达量与血清IL-15水平呈正相关关系(r=0.726,P〈0.05)。结论:SLE患者存在外周血IL-15蛋白和基因表达异常,且与其分泌免疫球蛋白和自身抗体有关,提示IL-15可能参与SLE的病理生理过程。  相似文献

目的 研究手足口病(HFMD)患者外周血淋巴细胞CD95、HLA-DR活化分子表达的变化及其意义.方法 以健康儿童作为对照,采用流式细胞术,检测58例手足口病患者外周血淋巴细胞CD95标志、T细胞亚群及其HLA-DR抗原表达.结果 HFMD患者外周血CD3^+T细胞的百分率(63.82±7.74)%与健康对照组比较差异有统计学意义(P〈0.001),CIM^+T细胞的百分率(34.29±7.33)%明显低于正常对照组(P〈0.005);HLA-DR^+淋巴细胞(28.30±7.61)%比正常对照组显著增高(P〈0.005);CD8^+T细胞表达HLA-DR水平(1.34±1.12)%明显高于对照组(P〈0.005);淋巴细胞CD95表达差异无统计学意义(P〉0.05).结论 HFMD患者存在细胞免疫功能紊乱和淋巴细胞异常激活现象,T淋巴细胞的激活以CD8^+T细胞为主,其在抗病毒感染中起着重要作用.  相似文献

目的:通过检测SLE患者外周血淋巴细胞ERα的表达及外周血淋巴细胞功能的变化,探讨SLE发生过程中性激素对淋巴细胞功能的影响。方法:收集33例SLE患者和31例正常对照者血样,采用ELISA法检测外周血雌二醇(Estradiol,E2)、睾酮(Testosterone,T)水平,MTT法检测ConA刺激T淋巴细胞增殖反应,RT-PCR法检测外周血单个核细胞(Peripheral blood mononuclear cells,PBMCs)、T淋巴细胞和B淋巴细胞雌激素受体α(Estragen receptor,ERα)mRNA的表达。结果:与健康对照者相比较,SLE患者外周血T淋巴细胞增殖的刺激指数(Stimulatingindex,SI)降低(P〈0.01),E2水平升高,T降低,但P〉0.05,E2/T比值明显升高(P〈0.01),淋巴细胞ERαmRNA的表达均明显升高(P〈0.01)。结论:SLE患者性激素水平的改变可通过淋巴细胞ERα,影响淋巴细胞的功能,参与SLE的发生。  相似文献

目的:研究CD226在SLE患者外周血T淋巴细胞亚群上的表达,以阐明CD226抗原在SLE患者体内T细胞活化中作用以及与SLE发病的关系。方法:31例SLE患者和30例健康志愿者外周血单个核细胞,在体外培养72h后,三色荧光标记的单克隆抗体染色,利用流式细胞仪测定T细胞亚群细胞表面CD226抗原的表达。结果;SLE患者组的CD3^ 、CD4^ 、CD8^ T淋巴细胞上CD226^ 表达均高于正常对照组(P<0.01);活动期SLE组、静止期SLE组的CD3^ 、CD4^ 、CD8^ T细胞上CD226^ 表达均显著高于对照组(P<0.01),而活动期与静止期患者之间T细胞亚群上CD226^ 表达则无显著性差异(P>0.05)。SLE患者CD3^ 、CD4^ 、CD8^ T细胞CD226^ 抗原表达水平与SLEDAI之间无明显相关性(P>0.05)。结论:SLE患者体内存在T细胞亚群异常活化;活动期、静止期SLET淋巴细胞CD226^+表达均增高,CD226^ 可能参与了SLE的免疫发病。  相似文献

目的探讨SLE患者免疫功能紊乱与淋巴细胞凋亡信号传导途径异常之间的关系。方法应用流式细胞术测定SLE患者淋巴细胞表面Fas、FasL及细胞质中活化caspase-3的表达率,并测定凋亡细胞百分率(AnnexinV^+PI^-)和坏死细胞百分率(AnnexinV^+PI^+)。应用ELISA方法测定血清中抗核小体抗体浓度。结果与健康对照组相比,稳定期和活动期SLE患者组淋巴细胞中凋亡细胞和坏死细胞百分率均显著增加(P〈0.05),淋巴细胞表面Fas、FasL及细胞质中活化caspase-3的表达率也显著增加(P〈0.05)。与稳定期SLE患者组相比,活动期SLE患者组淋巴细胞中坏死细胞百分率显著增加(P〈0.05),凋亡细胞百分率略有增加但无统计学意义(P〉0.05)。活动期患者组淋巴细胞表面Fas、FasL以及细胞质中活化caspase-3的表达率略有增加但无统计学意义(P〉0.05)。活动期SLE患者组抗核小体抗体浓度显著高于健康对照组和稳定期患者组(P〈0.05)。SLE患者凋亡细胞百分率和活化caspase-3的表达率与补体C3浓度水平呈负相关关系(P〈0.05)。结论Fas信号传导通路在SLE患者淋巴细胞凋亡紊乱中发挥了重要作用。caspase-3的活化是早期提示淋巴细胞凋亡的重要信号。SLE患者淋巴细胞凋亡活化程度与疾病活动程度和免疫效应功能紊乱密切相关,而淋巴细胞凋亡异常程度与抗核小体抗体水平的高低密切相关。淋巴细胞凋亡加速在SLE患者免疫病理损伤加重和免疫细胞调控紊乱中扮演了重要角色。  相似文献

目的:观察清络通痹颗粒(QLT)对类风湿关节炎(RA)患者外周血T淋巴细胞表达破骨细胞分化因子(RANKL)的影响。方法:采用流式细胞术检测RA患者及健康志愿者外周血T淋巴细胞表达RANKL情况;同时检测了添加清络通痹颗粒含药血清后RA患者外周血T淋巴细胞表达RANKL的水平。结果:活动期RA患者外周血T淋巴细胞RANKL表达率显著高于健康者(P〈0.01);添加清络通痹颗粒含药血清后RA患者外周血T淋巴细胞RANKL表达的水平明显下降(P〈0.05或〈0.01)。结论:清络通痹颗粒有可能通过下调T淋巴细胞RANKL的表达,从而抑制破骨细胞(OC)的分化。  相似文献

目的:通过检测乙型肝炎患者外周血单核细胞各亚群分布情况以及HLA—DR表达水平,探讨慢性乙型肝炎患者外周血中CD14^+CD16^+单核细胞及HLA-DR的表达特点,以及与疾病进程之间的关系。方法:利用流式细胞仪(FCM)检测50例慢性乙型肝炎患者,其中21例免疫耐受(IT)患者、29例免疫活化(IA)患者,及31例正常人外周血CD14、CD16、HLA—DR的表达。同时调查肝功能、乙肝五项和血清HBVDNA等相应临床资料。结果:免疫活化组(IA)的CD14^+CD16^+亚群比率明显高于正常对照组和免疫耐受组(IT)(P〈0.01);单核细胞CD14^+CD16^+亚群HLA.DR表达明显高于CD14^+CD16^+亚群(P〈0.01);乙型肝炎患者CD14^+CD16^+单核细胞比率与ALT呈正相关(r=0.876,P〈0.01),与HBVDNA载量成负相关(r=-0.267,P〈0.01)。结论:CD14^+CD16^+单核细胞可能在乙型肝炎患者外周血中参与调节针对HBV的免疫反应,CD14^+CD16^+单核细胞与HBV复制和肝脏炎症之间存在相关性,因此检测单核细胞CD14^+CD16^+亚群及HLA-DR水平对于了解乙型肝炎患者疾病进程具有重要的临床应用价值。  相似文献

使用密度离心法分离SLE患者和正常人外周血单个核细胞 (PBMC ) ,采用流式细胞术检测B淋巴细胞白细胞分化抗原 4 0 (CD4 0 )的表达水平 ,进行SLE患者 (活动期和缓解期 )和正常人之间的比较 ;并进行B淋巴细胞CD4 0表达水平和血清抗dsDNA抗体水平及狼疮活动指数 (SLEDAI)的相关分析。结果表明 ,活动期SLE患者外周血B淋巴细胞比例 (% )和其表达CD4 0的比例 (% )均明显高于缓解期SLE患者和对照组 ,其表达CD4 0的平均荧光强度 (MFI)在活动期SLE患者最高 ,缓解期SLE患者稍低 ,对照组最低 ;相关分析结果表明 ,活动期SLE患者B淋巴细胞CD4 0的表达比例 (% )和强度 (MFI)均与血清抗dsDNA抗体及SLEDAI呈正相关 ,后两者呈高度相关 ;缓解期SLE患者B淋巴细胞表达CD4 0的强度 (MFI)和SLEDAI呈正相关。CD4 0在活动期SLE患者B淋巴细胞的表达增加 ,其水平与疾病活动度有关。  相似文献

A further analysis of already published data supports the position that retardates of low ability level less frequently have retarded siblings, retarded parents, and parents low in occupational level than do retardates higher in ability level. 1. Recovery of responsiveness of single cells in lateral geniculate nucleus of rat has been determined in both P and I cells. There are three types of recovery curve among P cells; (a) early recovery, (b) early partial recovery followed by depression and then complete recovery, (c) prolonged depression followed by cyclic recovery. Type (c) is by far the commonest recovery curve. In contrast to the spike in a P cell, the synaptic potential recovers to its full amplitude in about 20 msec. All I cells exhibit similar rapid recovery curves after a prolonged depression.2. Conditioning stimuli applied to visual cortex also produce a prolonged depression in most P cells but I cells can be re-excited at short intervals from cortex. Decortication does not prevent the prolonged depression of the multineuronal response produced by optic nerve stimulation.3. A neuronal model is proposed to explain these observations. It is supposed that I cells (interneurones) are innervated by axon collaterals of the P cells (principal cells, projecting to visual cortex) and that the I cells exert an inhibitory influence on the P cells.  相似文献

Eighteen families in which both parents had refractions within the range of +4·0 D to −4·0 D and axial lengths seen in emmetropia (22·3-26·0 mm) showed coefficients of correlation of the order 0·5 indicative of polygenic inheritance. Such coefficients were seen for axial length (0·407) and for the cornea (0·487), but not for the lens (which is known to be yoked to the axial length). No such coefficients were seen in 19 families in which one of the parents had axial length outside the emmetropic range (nine families with long axes and 10 with short axes).

The pattern of polygenic inheritance for emmetropia (completely correlated optical components) and errors of refraction up to 4·0 D (inadequately correlated components: correlation ametropia) follows that seen in stature and other measurable characters. In contrast the high refractive errors with their abnormal axial lengths (component ametropia) are—like the extremes in stature—pathological anomalies with monofactorial inheritance.

It was established, in experiments on isolated spinal ganglia of adult rats in concluons of intracellular recording, that dopamine (1 M/liter) elicits depolarized responses in 61% of neurons, hyperpolarized in 20% of neurons, and depolarized-hyperpolarized in 19% of neurons. The depolarized responses are associated with the activation of D1 dopamine receptors, and are governed by the shift of cAMP-dependent cation (sodium) channels to the conducting state. The hyperpolarized responses are triggered by the activation of D2 dopamine receptors, which by means of HTP-binding protein convert the potassium channels to the conducting state.
