实验证实3H-1,2二氢-1-吡咯里嗪酮(A)及5-(4-氯苯甲酰基)3H-1,2-二氢-1-吡咯里嗪酮(B)具有明显的抗炎镇痛作用。在此基础上,设计合成了A的12个Mannich碱衍生物C(1-12)及B的缩氨脲D1和肟D2,这14个化合物均未见文献报道。药理实验证明,这些化合物大多具有抗炎镇痛活性,其中化合物C1、C2、C3、C9活性最为显著。  相似文献

The purposes of this study were to localize the position of spinal center of sympathetic nerve which controls the pulmonary circulation, and to evaluate the role of pulmonary sympathetic nerve in hypoxic pulmonary vasoconstriction (HPV) in pithed rat model. The sympathetic preganglionic fibers arising from C7–T10 segments were stimulated electrically in succession. During stimulation the pulmonary vascular resistance (PVR) was increased in all segments tested, most significantly in C7-T4 (about 28 % above control value), obviously higher than that in T5-T10. The higher the stimulated spinal segments, the larger the ratio of ΔPVR/ΔSVR. Alveolar hypoxia (12%O2–88% N2) could induce HPV in pithed rat. In the presence of hypoxia, stimulation of T1–T3 segments caused a double increment in PVR and -Pp, as compared with those during normoxia (P<0.05). The data show that 1) the spinal center of sympathetic nerve regulating the vasomotion of pulmonary circulation is located in the C7-T4 segments; 2) the excitation of sympathetic nervous system during hypoxia could enhance HPV.  相似文献

目的 :研究急性等容血液稀释 (ANHD)对体循环及肺循环的影响 ,指导临床ANHD的应用。方法 :10只健康成年杂种犬 ,麻醉后气管插管 ,控制呼吸 ,正常体温下 ,每只犬行三个水平的ANHD ,即中度 (HD1Hct约为 19% )、深度 (HD2 Hct约为 15 % )、极深度 (HD3 Hct约为 9% ) ,然后回输自家血 ,在血液稀释前、稀释后 30min、回输血后 15min分别测血液流变学、体循环和肺循环各指标。结果 :HD时血粘度 η渐下降。HD1、HD2 时 ,心输出量 (CO)和心脏指数 (CI)分别升高 41%和 48% ;心率 (HR)无明显变化 ,总外周阻力 (SVR)渐下降 ,平均动脉压 (MAP)渐下降 ;肺小动脉阻力 (PVR)渐下降 ,而平均肺动脉压力 (MPA)、肺动脉楔入压 (PCWP)、中心静脉压 (CVP)无明显变化。HD3 时 ,CO(CI)不再升高反而降低 ,HR仍无明显变化 ,SVR和MAP继续下降 ;PVR继续下降 ,MPA无明显变化 ,PCWP、CVP有所升高 ,回输血后各指标均有所回升。整个稀释过程中 ,心搏血量SV和心搏指数SVI,左室每搏作功指数LVSWI、右室每搏作功指数RVSWI的变化与CO(CI)的变化相似。结论 :中度、深度ANHD时 ,机体代偿 ,CO(CI)及SV(SVI)升高 ,SVR、PVR降低 ,有利于循环 ;极深度ANHD时 ,CO(CI)及SV(SVI)不再升高反而降低 ,MAP下降明显 ,MPA无明显变化 ,对体循环影响大 ,对肺?  相似文献

观察了吸入0.004%的一氧化氮(NO)对急、慢性缺氧大鼠血流动力学、缺氧性肺血管收缩反应(HPV)、血气及高铁血红蛋白(MetHb)的影响。结果表明:(1)常氧吸入NO时能明显降低慢性缺氧大鼠肺动脉平均压(Ppa)和肺血管阻力(PVR),但对正常大鼠的Ppa和PVR无明显影响;(2)慢性缺氧大鼠急性缺氧时HPV较正常大鼠弱,吸入NO不但降低两者的急性缺氧肺动脉高压,且完全逆转两者的HPV;(3)吸入NO对急、慢性缺氧大鼠体循环血流动力学、血气及MetHb含量无明显影响。提示吸入NO能选择性降低,急、慢性缺氧性肺动脉高血压,且逆转HPV。  相似文献

目的应用彩色多普勒超声心动图评价腺苷急性血管扩张试验在先天性室间隔缺损合并重度肺动脉高压中的应用价值。方法选取先天性室间隔缺损并重度肺动脉高压患者68例,肘静脉注射腺苷75μg.kg-1.min-1,应用彩色多普勒超声心动图检测主动脉平均压、肺动脉平均压、肺动脉收缩压、肺动脉舒张压、肺血管阻力、体循环阻力、肺循环阻力/体循环阻力比值、肺动脉/主动脉收缩压比值、肺循环/体循环血流量比值,注射5min后,与用药前比较,肺血管阻力下降>30%和肺动脉平均压下降>10%为急性血管扩张试验阳性。结果肺动脉平均压、肺动脉压、肺血管阻力、肺循环阻力/体循环阻力比值、肺动脉/主动脉收缩压比值用药后较用药前均明显下降(P<0.05),肺循环/体循环血流量比值明显增加(P<0.05),体循环阻力下降不明显(P>0.05),其中39例急性血管扩张试验阳性者,通过临床药物降压治疗有效后成功手术;29例急性血管扩张试验阴性者继续降压治疗无效并非手术适应证而行保守和对症治疗。结论腺苷选择性作用于肺血管,在先天性室间隔缺损合并重度肺动脉高压中,是急性血管扩张试验中较好的药物。  相似文献

Summary: The effects of 3, 4-Dihydroxyacetophenone (3. 4-DHAP) on cytosolic free calcium[Ca^2 ], in pulmonary artery endothelia (PAECs) and smooth muscle cells (PASMCs) during acute hypoxia were studied. Porcine pulmonary artery endothelial and smooth muscle cells (PASMCs)were cultured primarily, and they were divided into 4 groups: groups incubated under normoxia or hypoxia and those with or without treatment with 3. 4-DHAP. The [Ca^2 ]i of both PAECs and PASMCs was measured by determining the fluorescence of fura 2 AM on spetrofluorometer. Our results showed that hypoxia caused significant elevation of [Ca^2 ]i. in both PAECs and PASMCs,3, 4-DHAP could attenuate the hypoxic elevation of [Ca^2 ]i only in PASMCs but not in PAECs. It is concluded that 3, 4-DHAP decreases the hypoxic elevation of [Ca^2 ]i in PASMCs. This might contribute to its inhibitory effect on hypoxic pulmonary vasoconstriction.  相似文献

Summary In order to study the effect of nitric oxide (NO) on the expression of hypoxia-inducible factor-1 alpha (HIF-1α) mRNA in hypoxic pulmonary hypertension (HPH) rats, 30 healthy male Wistar rats were randomly divided into normoxic control group, chronic hypoxic group and hypoxia plus L-argine (L-Arg) group. The animal model of HPH was developed. The mean pulmonary arterial pressure (mPAP) was measured by inserting a microcatheter into the pulmonary artery. The HIF-1α mRNA expression levels were detected by in situ hybridization (ISH) and semiquantitative RT-PCR. It was found that after 14 days hypoxia, the mPAP in normoxic control group (17.6±2.7 mmHg,l mmHg=0.133 kPa) was significantly lower than that in chronic hypoxic group (35.8 ±6.1 mmHg,t=0.2918,P<0.05) and mPAP in chronic hypoxic group was higher than that in hypoxia plus L-argine group (24.4±3.8 mmHg,t=0.2563,P<0.05). ISH showed that the expression of HIF-1α mRNA in the intraacinar pulmonary arteriolae (IAPA) in normoxic control group (0.1076±0.0205) was markedly weaker than that in chronic hypoxic group (0.3317±0.0683,t=3.125,P<0.05) and that in chronic hypoxic group was stronger than that in hypoxia plus L-argine group (0.1928±0.0381,t=2.844,P<0.05). RT-PCR showed that the content of HIF-1α mRNA in chronic hypoxic group (2.5395±0.6449) was 2.16 times and 1.75 times higher than that in normoxic control group (1.1781±0.3628) and hypoxia plus L-argine group (1.4511±0.3981), respectively. It is concluded that NO can reduce the mPAP by the inhibition of the expression of HIF-1α mRNA, which may be one of the mechanisms through which NO affects the pathogenesis of HPH. Summary The aim of this study was to investigate the role of sympathetic nerve and adrenal gland in the changes in hemodynamics and hypoxic pulmonary vasoconstriction (HPV) induced by cigarette smoking in Wistar rats. Data showed that an increment of pulmonary vascular resistance (PVR) induced by alveolar hypoxia was partially prevented by prazosin (α1-receptor blocker) and prazosin combined with propranolol (β-receptor blocker), but not significantly affected by propranolol or adrenalectomy. Cigarette smoking tould increase PVR and potentiate HPV, whereas the potentiation of HPV was partially inhibited by prazosin and strengthened by propranolol, but not affected by adrenalectomy. It is suggested that the sympathetic nerve excitation caused by cigarette smoking may play a role in the strengthening of HPV.  相似文献

OBJECTIVE: To evaluate effect of tetrandrine (Tet) on chronic hypoxic pulmonary by observing expression of bFGF in lung tissue of rat with chronic hypoxic pulmonary hypertension. METHODS: 30 rats were divided into 3 groups: hypoxia group, hypoxia + Tet group and control group. On the 21st day of hypoxia, the hemodynamic parameters were measured and the intra acinar pulmonary arteries (IAPA) changes were studied under light microscopes. Immunohistochemical staining with monoclonal antibody against human recombinant bFGF was performed in the paraffin section of rat lung. RESULTS: Tetrandrine could reduce pulmonary artery mean pressure (mPAP) and pulmonary vascular resistance (PVR), (P < 0.01), decrease the expression of bFGF in wall of IAPA, and inhibit hypoxic structural remodeling of IAPA. CONCLUSION: Tetrandrine could decrease the expression of bFGF in wall of IAPA and in that connection to inhibit remodeling of IAPA and pulmonary hypertension.  相似文献

目的 观察汉防已甲素 ( tetrandrine,Tet)对慢性低氧大鼠肺组织碱性成纤维细胞生长因子( b FGF)表达的影响 ,探讨 Tet降低低氧性肺动脉高压的机制。方法 建立低氧性肺动脉高压大鼠模型 ,检测血流动力学 ,观察肺腺泡内动脉 ( IAPA)结构的变化 ,用免疫组化法检测肺组织 b FGF表达。结果  Tet能明显降低肺动脉压 ,抑制肺腺泡内动脉壁 b FGF表达 ,阻抑肺腺泡内动脉重建。结论  Tet通过抑制 b FGF表达 ,从而阻抑肺腺泡内动脉重建是其降低肺动脉压的机制之一  相似文献
