The endothelial cells of the corneas of rabbits preserved by desiccation for vari-ous duration showed no hexagonal form of normal cells but broad bean-like nuclci afterdouble staining with alizarin red and trypan blue.Scanning electron microscopic examina-tion revealed interdigitation at the intercellular junction together with intercellularvacuoles and cellular edema.Transmission electron microscopic examination showedincomplete nuclear membrane,karyopyknosis,vacuoles and remnant organellae in thecytoplasm.Partial penetrating keratoplasties were done in 37 rabbits(37 eyes)with pre-served corneas.The grafts remained transparent for a minimum of 4 days to a maximumof over 1 year,and 7 grafts remained transparent for more than 30 days.Staining ofthe transparent grafts revealed normal hexagonal endothelial cells in mosaic arrange-ment.We assumed that after transplantation of the preserved cornea the endothelial cellsregained their activities owing to the nutrient effect of the aqueous humor.  相似文献

目的:观察无活性内皮细胞的冷冻保存角膜和使用Optisol中期保存液的新鲜角膜应用于直径小于3mm角膜组织缺损的周边角膜穿孔伤病人,行急诊治疗性角膜移植术后效果的比较。方法:回顾性分析2008年3月至2012年6月,因各种原因导致的周边角膜穿孔,在本院急诊行治疗性角膜移植术的患者10例(10眼),角膜穿孔范围均小于3mm,其中使用冷冻保存角膜6例(6眼),使用新鲜角膜4例(4眼)。术后均随访6个月以上。结果:所有患者均获得临床治愈,眼球完整性得以保持,术后前房形成良好,其中冷冻保存角膜组5眼及新鲜角膜组3眼术后随访6个月角膜保持透明,冷冻角膜及新鲜角膜组各有1眼发生术后混浊,新生血管长入。结论:无活性内皮细胞的冷冻保存角膜和新鲜角膜均可用于角膜穿孔伤患者的急诊治疗性角膜移植术,术后效果相近,可不行二次光学角膜移植手术,保持角膜的透明。冷冻保存角膜满足了急诊手术的需要,也缓解了新鲜角膜紧缺的现状。  相似文献

88例角膜移植,出院时植片透明愈合率65.9%,2~38个月期间随访到65例,植片透明率70.8%。其中用长期保存脱水角膜进行板层角膜移植65例,随访到44例,植片透明率为81.8%。在紧急情况下,为挽救眼球可用保存的全厚角膜暂代新鲜角膜行眼前节外膜重建术。  相似文献

目的 观察大鼠角膜组织中树突状细胞的分布,以了解树突状细胞在穿透性角膜移植排斥反应中的作用.方法 制作大鼠穿透性角膜移植模型,观察角膜透明度、新生血管.采用免疫组织化学方法检测OX-62 树突状细胞在角膜的分布.结果 术后角膜植片均出现不同程度的新生血管、角膜水肿、混浊、基质增厚.排斥反应发生率100%.正常角膜组织中未见OX-62 树突状细胞分布,在发生急性排斥反应的角膜上皮层中有阳性细胞表达.脾脏阳性细胞表达与文献报道一致.结论 OX-62 树突状细胞出现在移植后发生排斥的角膜组织中,角膜组织中的树突状细胞可能参与角膜移植后的免疫反应.  相似文献

目的 :观察深低温长期保存角膜穿透性移植治疗感染性角膜病的临床效果。方法 :应用深低温保存 1~ 60月的健康人尸角膜为真菌性角膜炎 10例、10眼和病毒性角膜炎 4例、4眼行穿透性角膜移植术。结果 :随诊 1~ 19月 ,13例角膜植片透明 ( 92 .85% ) ,1例半透明 ,无 1例原发病复发 ,视力均有提高。结论 :深低温长期保存的角膜是治疗急性致盲性角膜病的重要储备材料 ,有重要的临床应用价值。  相似文献

目的 探讨冻存羊膜移植联合应用糖皮质激素对早期重度眼表碱烧伤的疗效。方法 将重度眼表碱烧伤病人34例(38只眼)分为两组,手术组28眼在早期应用糖皮质激素的基础上行冻存羊膜移植术,对照组10眼早期应用糖皮质激素。结果 随访3~12个月,手术组所有病眼术后视力均有不同程度提高,20眼角膜水肿减轻,21眼角膜遗留不同程度的斑翳,4眼角膜恢复透明,1眼持续混浊、水肿并发生角膜溃疡,2眼发生睑球粘连。对照组3眼角膜溃疡,6眼角膜白斑伴角膜血管化,4眼发生不同程度睑球粘连。结论 早期羊膜移植并糖皮质激素治疗严重眼表碱烧伤具有较好的效果。  相似文献

OBJECTIVE: To construct artificial rabbit corneas with autologous skin epidermal stem cells and allogenic stromal cells in vitro and promote healing of corneal wounds. METHODS: Skin epidermal stem cells were isolated from autologous skin samples. Keratocytes were isolated from newborn cornea biopsies. The cells were combined with acellular porcine corneal stroma scaffold to construct artificial corneas. Then the constructed artificial corneas were used to repair severe vision loss caused by complete loss of corneal epithelial stem cells. RESULTS: Cultured skin epidermal stem cells and keratocytes were in good growth conditions. Cultured artificial corneas consisted of multiplayer epithelial cells growing on stroma equivalent consisting of stromal matrix with incorporated keratocytes. The in vitro constructed artificial corneas were histologically similar to normal rabbit corneas. Three months after transplantation, the cornea wounds were healed and the rabbit cornea became transparent. CONCLUSION: The artificial corneas were constructed successfully in vitro and can be used to repair severe vision loss caused by complete loss of corneal epithelial stem cells.  相似文献
