IL-12协同B7-1诱导机体抗肿瘤免疫的实验研究 总被引:4,自引:0,他引:4
目的研究白细胞介素-12(Interleukin-12,IL-12)协同共刺激分子B7-1,联合诱导机体抗肿瘤免疫对大鼠卵巢上皮癌的治疗作用并探讨其机理.方法用携带有小鼠IL-12和B7-1的逆转录病毒表达载体感染大鼠卵巢癌细胞株NuTu-19,建立高表达细胞株NuTu-19/IL-12、NuTu-19/B7-1及双基因共表达细胞株NuTu-19/IL-12-B7-1,并以转染空载体pLXSN的细胞NuTu-19/Neo为对照.用经丝裂霉素C处理的各种基因修饰的肿瘤细胞免疫动物,观察卵巢癌腹腔转移模型动物生存期,及其诱导细胞毒性T淋巴细胞(cytotoxic lymphocyte,CTL)杀伤活性的作用.结果经各种基因修饰的肿瘤细胞免疫后,大鼠脾淋巴细胞增殖能力有不同程度的提高,CTL杀伤同源肿瘤细胞的活性明显增强,IL-12和B7-1基因联合修饰的肿瘤细胞免疫动物对模型动物生存期的延长具有显著意义(P<0.05).结论IL-12和B7-1基因联合修饰的肿瘤细胞可以刺激机体CTL增殖、成熟,增强CTL对肿瘤细胞的识别和杀伤活性等,两者联合具有明显的协同效应.联合免疫基因治疗作为一种新的卵巢癌治疗方法,值得深入研究. 相似文献

Cell-cell adhesion molecules and signaling intermediates and their role in the invasive potential of prostate cancer cells 总被引:1,自引:0,他引:1
PURPOSE: The highly variable natural history of prostate carcinoma may be reflected in heterogeneity of invasive potential between tumors. MATERIALS AND METHODS: We have examined two prostate cancer cell lines of low invasive potential (CAHPV10 and PZHPV7) and three cell lines of high invasive potential (DU-145, PC-3, LNCapFGC), to determine whether specific adhesion molecule profiles correlated with their invasive behavior. RESULTS: Using an in vitro invasion assay, we demonstrated that DU-145, LNCapFGC and PC-3 cells were highly invasive compared with CA-HPV-10 and PZ-HPV-7 cells. LNCapFGC cells expressed high levels of E-cadherin, alpha-, beta- and gamma-catenin, desmoglein, desmoplakin and GSK3beta using immunoblotting. This was, in general, comparable to immunohistochemical staining. PC-3 cells had no E-cadherin or alpha-catenin, but expressed a high level of the HGF/SF receptor c-Met. In contrast, DU-145 cells were found to express E-cadherin and low levels for all other protein molecules, except c-Met. The DU-145 cell line also lacked alpha-catenin expression. In CA-HPV-10 and PZ-HPV-7 cells, there was no detection of APC, PECAM-1, P-cadherin or Wnt-1. DU-145, LNCapFGC and PC-3 cells formed cell-cell aggregates, which were reduced by inclusion of anti-E-cadherin antibody and the motogen HGF/SF. CONCLUSION: These results show that prostate cancer cells exhibit a diverse expression of cell-cell adhesion molecules and their signaling intermediates. The expression of these adhesion molecules bears an important relationship with the invasive phenotype of these cells. 相似文献

目的 探讨西洛他唑对高糖大鼠肾组织VCAM - 1的影响及对肾脏的可能保护作用。方法 雄性SD大鼠,链脲佐菌素(STZ)制备糖尿病大鼠模型。观察西洛他唑12周后对血糖、糖化血红蛋白、肾重/体重比值、肾脏VCAM - 1mRNA及蛋白表达的影响。结果 糖尿病对照组及各治疗组肾重/体重比值均明显增加(P <0 . 0 1);VCAM - 1mRNA表达在糖尿病组明显升高,药物治疗后有显著改善( P <0. 0 1);Westernblot各组均有VCAM - 1表达,糖尿病组较正常组表达增强,药物治疗后呈下降趋势。结论 STZ诱发的糖尿病大鼠肾脏VCAM - 1表达增加,西洛他唑降可低糖尿病大鼠肾组织VCAM - 1mRNA和蛋白表达,对糖尿病肾脏具有保护作用。 相似文献

Bothrops asper venom (BaV) causes systemic and local effects characterized by an acute inflammatory reaction with accumulation of leukocytes and release of endogenous mediators. In this study, the effects of BaV on the release of the cytokines IL-1, IL-6 and TNF-alpha and the eicosanoids LTB4 and TXA2 in the peritoneal cavity of mice were analyzed. We also investigated the participation of beta2 integrin chain, l-selectin, LFA-1, ICAM-1 and PECAM-1 adhesion molecules in the BaV-induced leukocyte accumulation. Levels of proinflammatory cytokines IL-6 and TNF-alpha, as well as eicosanoids LTB4 and TXA2 were significantly increased after BaV injection (250 microg/kg), whereas no increment in IL-1 was observed. Anti-mouse l-selectin, LFA-1, ICAM-1, PECAM-1 and beta2 integrin chain monoclonal antibodies resulted in a reduction of neutrophil accumulation induced by BaV injection compared with isotype-matched control injected animals. These data suggest that BaV is able to induce the activation of leukocytes and endothelium to express adhesion molecules involved in the recruitment of neutrophils into the inflammed site. Furthermore, these results showed that BaV induces the release of cytokines and eicosanoids in the local of the venom injection; these inflammatory mediators may be important for the initiation and amplification of the inflammatory reaction characteristic from Bothrops sp envenomation. 相似文献

[目的]探讨正常小鼠组织与结肠癌(C26)小鼠黏附分子CD44v6和E-cadherin表达的关系.[方法]应用SP免疫组织化学方法检测结肠癌小鼠和正常小鼠组织中CD44v6和E-cadherin的表达情况,以组织标本的平均密度(MOD)表示.[结果]结肠癌小鼠组织中CD44v6的MOD为0.1103±0.0020,而对照组为0.0868±0.0187;结肠癌小鼠中CD44v6表达水平比正常小鼠对照组明显增强(P<0.01);结肠癌小鼠组织中E-cadherin的MOD为0.1100±0.0034,正常对照为0.1426±0.0015,结肠癌小鼠比正常小鼠明显降低(P<0.01),且CD44v6表达水平与E-cadherin表达水平呈负相关(P=0.000).[结论]结肠癌的生长、侵袭可能与黏附分子异常表达相关,检测CD44v6和E-cadherin表达可作为判断结肠癌生长和侵袭倾向的参考指标. 相似文献

目的 :探讨丹参有效成分丹酚酸B在心脏微血管内皮细胞 (CMEC)缺氧 /复氧损伤中的保护作用。方法 :通过心脏微血管内皮细胞体外培养技术 ,建立缺氧预适应 (HPC)模型并给予丹酚酸B ,应用酶联免疫方法检测细胞间粘附分子 1(ICAM 1)表达。结果 :在CMEC缺氧 3h ,复氧 6h后ICAM 1表达升高 ;缺氧预适应和应用丹酚酸B对CMEC进行预处理则能有效降低CMECICAM 1的表达。结论 :丹酚酸B在CMEC缺氧 /复氧损伤中可产生与缺氧预适应类似的保护效果。 相似文献

Cementum is a mineralized tissue that acts to connect the periodontal ligament to the tooth root surface. Its composition is very much like bone, being comprised mainly of type I collagen, inorganic mineral and noncollagenous proteins, however the origin of the cells and factors necessary for cementum formation have yet to be elucidated.Our laboratory has focused on the role that adhesion molecules, and their cell surface receptors, play in the formation of cementum and tooth root. In order to study this, we used a mouse molar as a model system. This system enabled us to study the formation of four distinct mineralized tissues; bone, cementum, dentin and enamel at various stages of their development. For these studies, we initiated experiments to examine potential cementoblast progenitor cells, in vitro. As a first step, we show that dental papilla and dental follicle cells, n vitro, obtained from molar tissues at day 21 of development, induce mineralized nodules, in vitroIn addition, we obtained tissues from mice where defects in root development may exist and determined bone sialoprotein (BSP) protein expression, a mineralized tissue specific adhesion molecule, in such tissues. As discussed here, we found that osteopetrotic (op/op) mice have delayed and/or defective root development and BSP does not localize in the dental tissues, at day 33 of development. In addition, dentin formation was defective and odontoblasts appeared immature, based on morphological examination. In contrast, the day 33 control molars demonstrated positive staining for BSP localized to root cementum, with normal formation of dentin. 相似文献

Abstract The inner polymeric surface of an ELISA titration well is plasma-modified and coated with different surfactant molecules. The titration of neurodegenerative proteins markers (prion, Tau and β-synuclein), previously demonstrated as more efficient with such modified tubes, is related to the adhesion behaviour of these proteins and their corresponding capture antibodies. The adhesion process is studied in terms of anchoring and specific mechanisms. The proteins and antibodies binding onto such modified surfaces is related to the substrate hydrophilic character calculated from the angle contact measure, to the polymer surface charge measured through the streaming potential determination at different pH and the inner surface roughness determined from AFM images.
