Myocardial infarction (MI) is one of the diseases with high fatality rate. Berberine (BBR) is a monomer compound with various biological functions. And some studies have confirmed that BBR plays an important role in alleviating cardiomyocyte injury after MI. However, the specific mechanism is unclear. In this study, we induced a model of MI by ligation of the left anterior descending coronary artery and we surprisingly found that BBR significantly improved ventricular remodeling, with a minor inflammatory and oxidative stress injury, and stronger angiogenesis. Moreover, BBR inhibited the secretion of Wnt5a/β-catenin pathway in macrophages after MI, thus promoting the differentiation of macrophages into M2 type. In summary, BBR effectively improved cardiac function of mice after MI, and the potential protective mechanism was associated with the regulation of inflammatory responses and the inhibition of macrophage Wnt5a/β-catenin pathway in the infarcted heart tissues. 目的 定性分析治疗痤疮的临床验方痤疮消洗液中的化学成分。方法 采用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF-MS)技术,选用Acquity HSS T3色谱柱(2.1 mm×100 mm,1.8 μm),0.1%甲酸乙腈和0.1%甲酸水为流动相进行梯度洗脱,流速为0.3 mL/min,质谱使用ESI离子源,正负离子模式扫描采集数据。通过精确质谱信息及元素组成分析,结合Scifinder数据库、对照品信息及相关文献,对痤疮消洗液中的化学成分进行推断和鉴定。结果 痤疮消洗液中鉴定出59个化合物,它们分别为蒽醌类、环烯醚萜类、有机酸类、黄酮类化合物。结论 UPLC-Q-TOF-MS技术可快速、灵敏地鉴定痤疮消洗液中的化学成分,为探讨药效物质基础及控制制剂质量提供了实验依据。 相似文献

MiRNAs play an important role in regulating tumor migration and invasion, and abnormal expression of miRNAs occurs in various kinds of human cancers. In this essay, it is reported that the level of miRNA-218 decreases in metastatic breast cancer cells, moreover, miRNA-218 suppresses breast cancer cells migration and invasion through binding Robo1 (one of Slit receptors) to its 3′UTR. MiRNA-218 restoration suppresses Robo1 expression and inhibits breast cancer cells invasion and migration. What the results describe is that the function of Robo1 regulated by miRNA-218 may provide a new strategy for inhibiting migration and invasion of breast cancer cells. 相似文献

Introduction:Several studies reported that traditional Chinese mind-body exercises showed beneficial effects on improving anxiety and depression of patients with low back pain (LBP) in recent years. However, the effects of traditional Chinese mind-body exercises on improving psychological disorders of patients with LBP remain controversial. Most previous reviews only focused on the effects of traditional Chinese mind-body exercises for LBP on pain and dysfunction. Therefore, the present systematic review and meta-analysis will be conducted to evaluate the evidence on psychological effects of traditional Chinese mind-body exercises for LBP.Methods and analysis:The electronic databases (PubMed, Embase, MEDLINE, Cochrane Central Register of Controlled Trials, Web of Science, China Knowledge Resource Integrated Database, and Wanfang Data) will be searched. The search will include all documents from their inception to February 2021. The Physiotherapy Evidence Database scale will be used for quality assessment of eligible studies. Risk of bias of eligible studies will also be assessed by Cochrane tool. The meta-analysis will be conducted using the Review Manager Version 5.3 software. The Higgins I2 statistic will be performed to examine for heterogeneity. The subgroup analysis will be conducted based on different types of traditional Chinese mind-body exercises, different intervention time, and different outcomes. Quality of evidence will be assessed using the Grades of Recommendation, Assessment, Development and Evaluation.Ethics and dissemination:No ethical statement will be required for the performance of this review and meta-analysis. Purpose
Identifying necrotic myocardium in ischemic regions is of great importance for risk stratification and clinical decision-making. However, rapid noninvasive imaging of necrotic myocardium is still challenging. This study sought to evaluate the potential of 1,4-naphthoquinones to rapidly visualize necrotic myocardium and the possible mechanisms of necrosis avidity.Procedures
Six 1,4-naphthoquinones were radiolabeled with iodine-131 and the necrosis avidity was estimated in mouse models with muscular necrosis by gamma counting and autoradiography. The necrotic myocardium imaging property and biodistribution of [131I]naphthazarin (6) were determined in rat models with re-perfused myocardial infarction. A possible mechanism of necrosis avidity was explored by in vitro DNA-binding and in vivo blocking experiments.Results
The radiochemical purities of the six radiotracers were greater than 95 %. The uptakes in necrotic muscles of all six radiotracers were higher than those in viable muscles, and [131I]naphthazarin (6) showed the highest necrotic-to-viable ratio and necrosis-to-blood ratio at all tested time points. The necrotic myocardium could be clearly visualized by single-photon emission computed tomography/x-ray computed tomography (SPECT/CT) using [131I]naphthazarin (6) as early as 3 h post-injection. Post-mortem biodistribution showed the uptake of [131I]naphthazarin (6) in necrotic myocardium was 11.67-fold higher than that in viable myocardium. Absorption spectra and emission spectra suggested naphthazarin (6) could bind to DNA through intercalation. The uptake of [131I]naphthazarin (6) in necrotic muscle could be significantly blocked by excessive ethidium bromide (a typical DNA intercalator) and cold naphthazarin (6) with 63.49 and 71.96 % decline at 3 h post-injection in vivo, respectively.Conclusions
1,4-Naphthoquinones retained necrosis avidity and [131I]naphthazarin (6) rapidly visualized necrotic myocardium. The necrosis avidity mechanism of [131I]naphthazarin (6) may be attributed to its binding with exposed DNA in necrotic tissues.10.
类风湿关节炎患者丙二醛修饰型低密度脂蛋白及低密度脂蛋白循环免疫复合物水平分析 总被引:1,自引:0,他引:1
目的分析类风湿关节炎患者丙二醛修饰型低密度脂蛋白、低密度脂蛋白循环免疫复合物及血脂水平,探讨其与心血管病高发生率的关系。方法选择类风湿关节炎患者55例,其中合并心血管疾病患者(并发症组)13例,单纯类风湿关节炎患者(单纯组)42例;正常对照组60例。采用酶联免疫吸附试验分别测定血浆丙二醛修饰型低密度脂蛋白和低密度脂蛋白循环免疫复合物水平,同时对受检者血脂和炎症指标进行检测。结果两组类风湿关节炎患者除载脂蛋白B高于对照组外(P<0.05),其它脂质和载脂蛋白水平无明显改变;并发症组与单纯组间脂质和载脂蛋白水平相似。并发症和单纯组血浆丙二醛修饰型低密度脂蛋白(分别为187.81±90.89和102.01±57.73mg/L)和低密度脂蛋白循环免疫复合物水平(分别为2.58±1.69和1.87±0.74AU)均明显高于对照组(分别为32.65±27.00mg/L和1.21±0.38AU,P<0.01),且并发症组高于单纯组(P<0.01或0.05)。低密度脂蛋白循环免疫复合物与C反应蛋白正相关(r=0.301,P=0.026),丙二醛修饰型低密度脂蛋白与血沉趋于相关(r=0.263,P=0.057)。结论类风湿关节炎患者丙二醛修饰型低密度脂蛋白和低密度脂蛋白循环免疫复合物水平显著升高,参与动脉粥样硬化的发生、发展过程。 相似文献
