查尔酮对人胃癌BGC823细胞生物学行为影响的初步研究   总被引:1,自引:0,他引:1  
目的:研究查尔酮对人胃癌BGC823细胞生长抑制、细胞周期阻滞及诱导细胞凋亡的作用,为寻找预防肿瘤药物提供一定的线索.方法:细胞生长抑制采用MTT比色法检测,细胞周期阻滞用流式细胞仪检测,细胞凋亡用流式细胞仪及琼脂糖凝胶电泳检测.结果:浓度为6.25、12.5、25和50μmol/L的查尔酮呈剂量依赖性抑制BGC823细胞增殖.查尔酮诱导细胞凋亡率是对照的2.71、2.79、4.74倍.琼脂糖凝胶电泳可观测到12.5μmol/L、25μmol/L有DNA"梯子"现象.结论:查尔酮可以抑制BGC823细胞生长,并呈剂量-反应关系.查尔酮有将BGC823细胞增殖周期阻滞在G0/G1期趋势,查尔酮可以抑制肿瘤细胞的生长和增殖.  相似文献

目的:研究脱氧胆酸对人胃癌BGC-823细胞的影响,并探讨其作用机制。方法采用细胞培养、MTT法、HE染色、Giemsa 染色、Hoechst33258荧光染色、AO/EB荧光染色和流式细胞术法,检测脱氧胆酸对人胃癌BGC-823细胞生长抑制效应和凋亡效应及其机制。结果脱氧胆酸可明显抑制人胃癌BGC-823细胞的增殖,0.7 mmol/L脱氧胆酸处理48 h后抑制率达81.5%;经脱氧胆酸处理后,细胞发生皱缩、细胞体积变小、核染色质浓缩、核碎裂、细胞出芽以及凋亡小体形成等显著的凋亡特征。细胞周期检测出细胞被阻滞在G2/M期;线粒体膜电位明显下降;AnnexinV-FITC/PI双染法检测出细胞凋亡率达40.35%。结论脱氧胆酸能够有效诱导人胃癌BGC-823细胞发生凋亡,且可能是通过线粒体途径诱导细胞凋亡,从而可为胃癌等相关恶性肿瘤疾病的治疗和凋亡机制的进一步研究提供重要理论基础和科学依据。  相似文献

目的观察小檗碱对人胃癌细胞BGC-823凋亡的诱导作用,以探讨具抗癌作用机理.方法采用体外细胞培养技术,以lO~40 mg/L小檗碱作用于培养的BGC-823细胞48 h后,进行光镜、琼脂糖凝胶电泳、流式细胞仪和TUNEL法的观察和分析.结果小檗碱作用后,光镜、TUNEL观察到细胞呈现凋亡特征,琼脂糖凝胶电泳显示细胞DNA呈梯状降解,流式细胞仪分析出现典型亚二倍体峰.结论小檗碱可诱导人胃癌细胞BGC-823细胞凋亡,这可能为小檗碱抗癌的机理之一.  相似文献

目的 探讨枸杞多糖(LBP-X)诱导人类慢性髓系白血病K562细胞株凋亡。方法 用吖啶橙(AO)荧光染色观察细胞结构的变化,用MTT、比色法测定抑制率,琼脂糖凝胶电泳和流式细胞仪检测法分析细胞凋亡。结果 LBP-X可明显抑制K562细胞的生长,凋亡率呈时间和剂量依赖性。LBP-X作用48h后,荧光显微镜下可见细胞核不规则,浓缩成大小不等的团快,核碎裂呈新月形改变,琼脂糖凝胶电泳可见DNA梯带。流式细胞仪分析图上可见明显的凋亡。结论 LBP-X能诱导K562细胞的凋亡,并呈一定的时间浓度依赖关系。  相似文献

目的:研究6,8-二-三氟甲基-7-乙酰氧基白杨素(9dFMAChR))抑制体外培养人卵巢癌细胞系CoC1细胞增殖和诱导凋亡作用及机制。方法:体外培养CoC1细胞,MTT比色法测定dFMAChR对CoC1细胞增殖活性的影响;AO/EB染色法荧光显微镜观察dFMAChR诱导CoC1凋亡细胞的形态学改变;DNA凝胶电泳确证dFMAChR诱导CoC1细胞凋亡作用;Western blotting法分析dFMAChR对CoC1细胞酪蛋白激酶CK2α蛋白表达和活性的影响。结果:MTT比色法结果显示,dFMAChR有效抑制CoC1细胞增殖活性,呈剂量依赖性;其IC50值为11.8μM。AO/EB染色荧光显微镜观察dFMAChR处理后,部分CoC1细胞呈现典型凋亡细胞形态特征;dFMAChR(10.0μM)处理CoC1细胞48 h,琼脂糖凝胶电泳出现“梯形”DNA条带。Western blotting分析结果表明:dFMAChR下调酪蛋白激酶CK2α表达,呈浓度和时间依赖性。结论:dFMAChR具有抑制人卵巢癌CoC1细胞增殖和诱导细胞凋亡作用;作用机制与其抑制酪蛋白激酶CK2α表达有关。  相似文献

目的: 研究细胞黏附肽(RGD)对人胃癌BGC823细胞生长增殖的影响,探讨RGD诱导BGC823细胞凋亡的作用。方法: 将体外培养的 BGC823细胞分为12.5、25.0、50.0、100.0 mg·L-1RGD处理组和对照组,RGD处理BGC823细胞24、48、和72 h后,MTT法检测不同浓度RGD作用不同时间对BGC823细胞生长增殖的抑制作用,分别利用光镜、透射电镜、免疫组织化学法、流式细胞术和凋亡细胞的DNA片段方法观察凋亡细胞的形态学变化以及定性、定量检测细胞凋亡。结果: MTT检测,RGD各剂量组在作用24、48和72 h时, BGC823细胞的抑制率均高于对照组(P<0.01),且随着RGD浓度增加BGC823细胞的抑制率增加;50.0 mg·L-1 RGD处理BGC823细胞48 h后,光镜下显示,细胞体积变小,胞核深染,胞浆发红,出现很多的凋亡细胞;透射电镜显示,细胞表面微绒毛消失,异染色质趋边凝聚、聚集在核膜上,形成新月形帽状结构,甚至核膜消失,细胞核碎裂,可见凋亡小体。流式细胞术检测,BGC823细胞发生凋亡,且凋亡峰随着浓度的增加而增高;电泳结果显示,BGC823细胞内的DNA片段断裂为大小不等的小片段,呈现出很清晰的梯状降解条带;免疫组织化学染色结果显示,50.0 mg·L-1RGD处理BGC823细胞48 h后 Survivin的表达量较对照组明显减少,经图像分析检测,实验组平均灰度值
高于对照组(P< 0.01),提示实验组细胞发生凋亡。结论: RGD通过诱导BGC823细胞凋亡抑制肿瘤细胞的增殖,其RGD诱导BGC823细胞凋亡的作用机制可能通过线粒体引发这一途径。  相似文献

目的研究香加皮水提取物(CPE)诱导人胃癌细胞BGC-823凋亡及其作用机制。方法采用G iem sa染色观察细胞凋亡形态学变化;电子显微镜观察凋亡细胞的超微结构变化;流式细胞术和琼脂糖凝胶电泳方法检测BGC-823细胞凋亡率、细胞周期和细胞凋亡的DNA水平变化;RT-PCR方法检测细胞凋亡相关基因bcl-2、bax和surv iv in mRNA表达水平变化;免疫细胞化学方法检测bcl-2、bax和surv iv in蛋白表达的变化。结果经CPE作用后,人胃癌细胞BGC-823出现明显的细胞凋亡形态学变化及超微结构改变,细胞DNA琼脂糖凝胶电泳呈现梯形图。经250μg/mL CPE处理48 h后,多数BGC-823细胞被阻滞在G2/M期,而且细胞发生明显的凋亡变化,BGC-823细胞凋亡率可达18.9%。CPE可抑制BGC-823细胞bcl和surv iv in mRNA及蛋白的表达,促进baxmRNA及蛋白的表达。CPE可明显延长S180荷瘤小鼠生存期,且具有剂量依赖性。结论CPE通过阻滞BGC-823细胞于G2/M期及诱导BGC-823细胞凋亡发挥抗肿瘤作用,其作用机制与抑制细胞的bcl-2和surv iv in基因mRNA及蛋白表达、促进bax基因和蛋白的表达有关。  相似文献

目的探讨冬凌草甲素对白血病NB 4细胞的诱导凋亡作用及其机制。方法以不同质量浓度(8、16、24和32μm o l/L)的冬凌草甲素作用于体外培养的NB 4细胞。应用四氮唑蓝(M TT)比色法检测细胞生长抑制率,流式细胞术(FCM)检测细胞凋亡率;采用Hoechst 33258荧光染色法和琼脂糖凝胶电泳观察细胞凋亡;采用蛋白印迹(W estern b lot)和比色法测定细胞凋亡前后caspase-3表达水平及其活性的变化。结果冬凌草甲素(16μm o l/L以上浓度)对白血病NB 4细胞具有显著的增殖抑制及诱导凋亡作用,并且呈现出一定的时间-效应和剂量-效应关系。Hoechst 33258荧光染色观察到典型的核浓缩、核碎裂等细胞凋亡的形态学变化,琼脂糖凝胶电泳观察到细胞凋亡时的DNA“梯形”条带;W estern b lot检测结果表明32 kD a的caspase-3酶原被激活,出现20 kD a的亚单位活化片段,同时在细胞凋亡过程中,caspase-3活性显著增高。结论冬凌草甲素能够通过激活caspase-3的表达而诱导NB 4细胞发生凋亡,可为冬凌草甲素进一步应用于临床治疗急性白血病提供实验依据。  相似文献

目的观察斑蝥酸钠体外对人胃癌BGC823细胞凋亡的影响。方法应用MTT法及流式细胞仪检测、分析斑蝥酸钠对人胃癌BGC823细胞的生长和凋亡的影响。结果不同剂量斑蝥酸钠作用24 h均可使人胃癌BGC823细胞生长明显抑制(P<0.01),并出现典型的凋亡峰,细胞增殖阻滞于G1期。结论斑蝥酸钠对人胃癌BGC823细胞生长有抑制作用,可诱导胃癌细胞凋亡。   相似文献

CASE HISTORY A female patient, 46 years old, head of the foreign affairs department of a certain university in Beijing, paid her first visit on October 9, 2006, with the chief complaint of vomiting for one month. She got vomiting after meals in early September. Before that, she had discomfortable sensation in the stomach due to angry with others, but she didn't pay much attention. Later, it developed into vomiting after eating. After the vomiting, the discomfort would be relieved, but with slight hypodynamia. She was once diagnosed as having 'neurogenic vomiting'. Having taken some western and Chinese drugs, the above symptoms were a little bit improved, but she would have nausea upon eating and with regurgitation. OBJECTIVE: To observe therapeutic effects of the comprehensive therapy of acupuncture-moxibustion and Chinese Tuina for treatment of insomnia due to deficiency of both the heart and spleen. METHODS: 92 cases were divided randomly into the treatment group (treated by acupuncture-moxibustion and Chinese Tuina) and the control group (treated by acupuncture-moxibustion). RESULTS: The therapeutic effect of the treatment group was obviously superior to that of the control group (the CHI2 test showed P < 0.01). CONCLUSIONS: The comprehensive therapy of acupuncture-moxibustion and Chinese Tuina can give marked therapeutic effects for treatment of insomnia due to deficiency of both the heart and spleen.  相似文献

Objective: To investigate if there are the CK-19, PDX-1, Nestin, Ngn3 positive cells in the donor islets of different purity in rats. Methods: Thirty male adult SD rats were randomly divided into 3 groups. Islets were isolated using digestion by ductal injection of collagenase. Group Ⅰ (n=10): Separating cell preparations were not purified, Group Ⅱ(n=10): Islet sediment was purified with 25% Ficoll400 ,Group Ⅲ (n=10): Islet sediment was purified with 25% and 11% Ficoll-400. The levels of protein of CK-19, PDX-1, Nestin and Ngn3 were detected by immunohistochemistry and the mRNA of CK-19, PDX-1, Nestin, Ngn3 was amplified by RT-PCR. Results: After two different purification methods applied, three islet preparations of different purities were obtained. The difference of islet purity was significant among various groups (P<0.05). Compared with group Ⅱ and group Ⅲ,the protein and mRNA of CK-19, PDX-1, Nestin,Ngn3 were both higher in group Ⅰ; group Ⅲ was poorly expressed. Conclusions: The three different islet purity donor islet have different CK-19, PDX-1, Nestin, Ngn3 positive cells within them, indicating that there are some islet stem cells in the purified donor islet.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
