目的 研究胚胎大鼠脊髓源性神经干细胞的培养和在体外骨髓基质细胞诱导其分化的情况。方法 从孕龄13d的胚胎大鼠脊髓组织中分离获得神经干细胞,采用含表皮生长因子(EGF)及碱性成纤维细胞生长因子(bFGF)的无血清限定性培养基培养,同时进行诱导分化,观测脊髓神经干细胞向胆碱能神经元分化的情况,用细胞免疫荧光化学方法鉴定分化结果。结果 可从胚胎脊髓中分离得到大量的神经于细胞,通过限定性培养基培养可获得于细胞球,经从骨髓基质细胞培养液中获得的限制性培养液在体外可被诱导分化,用细胞免疫荧光化学法鉴定,可见有胆碱能神经元生成。结论 胚胎大鼠脊髓神经干细胞在添加EGF与bFGF的限定性培养基中可以增殖并长期保持稳定的性状,在体外可以被诱导分化为胆碱能神经元。  相似文献

目的:探讨兔骨髓基质干细胞的体外培养技巧,观察免骨髓基质干细胞的体外生长特性,为用于骨组织工程的功能细胞提供实验基础。方法:自兔髂骨抽取骨髓,分离骨髓基质干细胞,分别采用常规培养液及含矿化液的培养液进行培养,绘制细胞生长曲线,对细胞行碱性磷酸酶染色,计算细胞碱性磷酸酶染色阳性率,观察细胞在体外长期培养时钙结节形成情况。结果:第1-3代细胞生长曲线基本相同,加入矿化液后细胞生长速度明显减慢,但细胞表现出成骨细胞特性,其碱性磷酸酶染色阳性率大大提高,在体外长期不传代培养时可形成钙结节。结论:兔骨髓基质干细胞在体外适当的培养条件下可以向成骨细胞方向分化,可作为骨组织工程的种子细胞.  相似文献

目的:采用体外全骨髓培养法培养SD大鼠骨髓基质干细胞(BMSCs)并进行鉴定。方法:采用全骨髓贴壁法分离扩增大鼠骨髓基质干细胞,采用流式细胞仪检测细胞表面分子CD29、CD34、CD45的表达。结果:全骨髓培养法能迅速、有效地培养和获得大量骨髓基质干细胞,流式细胞仪检测显示CD29阳性率为99.0%,CD34阳性率1.5%,CD45阳性率0.8%。结论 :全骨髓贴壁培养法是获得骨髓基质干细简捷、高效、经济的途径,可以获得较纯的BMSCs。  相似文献

目的:分离培养兔骨髓基质干细胞并探讨其体外生物学特性。方法:通过梯度密度离心法分离兔骨髓基质干细胞,体外培养、扩增,观察细胞的大体形态和超微结构;通过绘制细胞生长曲线、MTT实验来研究细胞的增殖能力争代谢活性。结果:兔骨髓基质干细胞呈梭形。胞质内有丰富的粗面内质网和线粒体,具有很强的增殖能力和代谢活性。结论:本实验方法取材方便,能较容易地获得大量的兔骨髓基质干细胞。  相似文献

骨髓基质干细胞定向分化成心肌样细胞的基础研究   总被引:2,自引:0,他引:2  
目的:骨髓基质干细胞在体外能定向分化成心肌样细胞。本研究旨在将分离出来的骨髓基质干细胞标记后注射到心壁内,观察其能否分化成心肌样细胞。方法:将骨髓基质干细胞体外培养扩增后用Brd U标记,将标记后的骨髓基质干细胞注射入结扎冠状动脉左前降支远端的大鼠左心室壁内,假手术组仅注射等量的培养液(元细胞)。结果:反复传代可获得大量纤维状的骨髓基质干细胞,Brd U对骨髓基质干细胞的标记率约为41.4l%。手术组细胞注射部位心脏组织中部分细胞内可见抗心室肌肌凝蛋白重链单克隆抗体(MHC)、抗Brd U抗体及抗心房利钠肽抗体、抗Brd U抗体双染色阳性颗粒,假手术组仅为单染色阳性颗粒。结论:移植的骨髓基质干细胞能在心脏提供的特殊微环境下分化成心肌样细胞。  相似文献

目的观察大鼠骨髓基质细胞在体外培养的条件,为诱导骨髓基质分化形成神经干细胞、神经元,以及移植鼠骨髓基质细胞治疗帕金森病大鼠奠定基础。方法取60~90g SD大鼠,体外培养骨髓基质细胞,利用倒置相差显微镜观察原代及不同传代细胞生长形态特点,并应用流式细胞仪鉴定原代及不同传代细胞表型。结果培养的骨髓基质细胞第3代,间充质干细胞的细胞表面标志CD90表达阳性的细胞已占到92.9%。造血干细胞的表面标志CD45表达阳性的细胞已从原代的73.4%降低到2.3%。结论传3代的细胞适合于细胞诱导分化,可作为理想细胞移植治疗的细胞来源。  相似文献

目的 探讨体外培养、扩增骨髓基质干细胞的方法,诱导骨髓基质干细胞向神经元样细胞定向分化。方法 采用全骨髓法培养成年SD大鼠股骨骨髓基质干细胞。传至第5代,用全反式维甲酸诱导分化,于诱导后第5小时行神经细胞特异性抗原MAP-2和胶质细胞特异性抗原GFAP鉴定。结果 成年鼠骨髓基质干细胞经全反式维甲酸诱导30min后可见神经元样细胞出现,经MAP-2免疫组化鉴定阳性。结论 (1)骨髓基质干细胞在体外扩增迅速,纯化需时短。(2)骨髓基质干细胞在全反式维甲酸诱导下向神经元样细胞分化。  相似文献

目的:探讨人骨髓基质干细胞的克隆化培养及其对体外造血干细胞扩增的影响。方法:取正常肋骨分离骨髓,制备单个核细胞贴壁培养,约5d后取集落样生长的梭形细胞扩增培养,传代、将分选的脐血造血干/祖细胞接种到克隆培养的人骨髓基质干细胞及其他培养条件液上,比较不同培养条件及不同代次骨髓基质干细胞对造血干/祖细胞扩增能力及集落形成能力的影响。结果:人骨髓基质干细胞能扩增达20代以上,经流式细胞仪检测证实基质干细胞,单纯细胞因子组和细胞因子加基质干细胞组能有效扩增造血细胞,而后者有维持长期造血的作用。结论:该克隆培养方法能有效扩增骨髓基质干细胞,培养的细胞有体外支持长期造血的作用。  相似文献

[摘要] 目的 通过研究在体外培养大鼠骨髓基质干细胞(rBMSCs)及其成骨生物学特性,探讨构建骨组织工程种子细胞的方法。方法 通过全骨髓贴壁培养法分离大鼠骨髓基质干细胞,应用含地塞米松、维生素C、β2甘油磷酸钠的诱导培养液定向诱导传代细胞向成骨细胞分化,并检测碱性磷酸酶活性及钙结节茜素红染色反应,进行成骨细胞鉴定。结果 全骨髓贴壁培养下,骨髓基质干细胞增殖能力强,生长迅速,呈成纤维细胞样生长。诱导条件下细胞的碱性磷酸酶活性明显增高,并且钙结节茜素红染色反应阳性。结论 采用全骨髓贴壁培养法,骨髓基质干细胞分离培养及体外扩增更为简便迅速,诱导条件下成骨能力肯定,可为骨组织工程构建提供大量种子细胞。  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
