AIM: To investigate the association between curative effects of interferon-α and partial human leucocyte antigen (HLA)Ⅱ alleles in chronic viral hepatitis B.METHODS: Sixty patients with chronic viral hepatitis B in Shanghai were treated with a standard course of treatment with interferon-α for 6 mo. HLA-DRB1, -DQA1, and -DQB1 alleles were detected by polymerase chain reaction-sequence specific primer (PCR-SSP) method. RESULTS: Frequencies of HLA-DRB1*04(P<0.025) and HLA-DQA1*0303 (P<0.01) in non-responders were significantly higher than those in partial and complete responders. Frequencies of HLA-DQAI*0505(P<0.025) and HLA-DQB1*0301(P<0.005) in partial and complete responders were significantly higher than those in non-responders.CONCLUSION: Non-response to interferon-α therapy is positively correlated with HLA-DRB1*04 and HLA-DQA1*0303, and negatively correlated with HLA-DQA1*0505 and -DQB1*0301 in patient with chronic viral hepatitis B.HLA Ⅱ genes of the identification alleles provide a method for evaluating outcome of interferon-α treatment. 相似文献

The docosahexaenoic (DHA), a ω-3 fatty acid, could play a beneficial inhibition of the incidence and progress of a series of human diseases including cancer. It has been report that DHA is involved in cell apoptosis. Recent studies show that the signal transduction pathway links with bcl-2, bax, caspase-3 and MMP-9 molecules. Therefore, we tested the relationship between DHA and cell apoptosis in human hepatocellular carcinoma cells (Bel-7402 cells). We show here that DHA induces Bel-7402 cells apoptosis after pre-treating cells with DHA. DHA down-regulates the protein expression of Bcl-2 and Bim mRNA level, and up-regulates caspase-3 activity and Bax expression level. We also found that DHA inhibits Bel-7402 cells migration. Basic on our studies, DHA may play a role in tumor invasion and survival. 相似文献

Thoracic aortic aneurysms are now routinely repaired with endovascular repair if anatomically feasible because of advantages in safety and recovery. However, intraoperative aneurysm rupture is a severe complication which may have an adverse effect on the outcome of treatment. Comprehensive preoperative assessment and considerate treatment are keys to success of endovascular aneurysm repair, especially during unexpected circumstances. Few cases have reported on intraoperative aortic rupture, which were successfully managed by endovascular treatment. Here, we present a rare case of an intraoperative aneurysm rupture during endovascular repair of thoracic aortic aneurysm with narrow neck and angulated aorta arch (coarctation-associated aneurysm), which was successfully treated using double access route approach and iliac limbs of infrarenal devices. OBJECTIVE: To elucidate whether endoxin is one of important factors involved in myocardial ischemia reperfusion (MIR) injury, the change of myocardial endoxin level was determined in rats with MIR injury model and the effects of anti-digoxin antiserum (ADA), an endoxin specific antagonist, on MIR injury were studied. METHODS: MIR injury model was obtained by ligating left anterior descending coronary artery 30 min followed by 45 min reperfusion. Sprague-Dawley rats were randomly divided into six groups of 10 rats, each. Sham group, MIR group, normal saline group, ADA 9, 18 and 36 mg.kg(-1). ECG was continuously recorded. After reperfusion left ventricular myocardium samples of ischemic area were processed immediately. Myocardial endoxin level, Na(+)-K(+)-ATPase, Ca(2+)-ATPase, Mg(2+)-ATPase activities, and intramitochondrial Ca(2+) content were measured. RESULTS: Myocardial endoxin level was significantly increased; Na(+)-K(+)-ATPase, Ca(2+)-ATPase, and Mg(2+)-ATPase activities were remarkably decreased; intramitochondrial Ca(2+) content was remarkably raised; ST segments of ECG were significantly elevated and occurrence and scores of ventricular arrhythmias were significantly increased in early stage of reperfusion in rats with MIR. In all groups with ADA, myocardial endoxin level was remarkably decreased; Na(+)-K(+)-ATPase, Ca(2+)-ATPase and Mg(2+)-ATPase activities were drastically increased; intramitochondrial Ca(2+) content was declined; ST segments and ventricular arrhythmias were improved. CONCLUSION: Myocardial endoxin level was increased in MIR, which implies that the elevated endoxin may be one of major factors inducing MIR injury. This postulate is supported by the observation that ADA has protective and therapeutic effects against MIR injury probably by antagonizing the action of endoxin. The underlying mechanism may be ascribed to restoration of energy metabolism, and attenuation of intracellular Ca(2+) overload. 相似文献

目的:研究高脂血症、高脂血症性脂肪肝大鼠血浆及肝组织ADM2/IMD的变化,探讨ADM2/IMD在高脂血症、高脂血症性脂肪肝病理过程中的意义.方法:雄性SD大鼠16只,随机均分为对照组与高脂组.大鼠被高脂喂食造模4 wk后,观察大鼠肝脏病理学变化,测定血脂(TG,TC,HDL-C,LDL-C)及肝功能(ALT,AKP);放免法测定血浆及肝组织匀浆ADM2/IMD含量.结果:高脂组体质量、肝质量及肝指数(肝质量/体质量)均显著高于对照组(P<0.01)肝组织呈轻中度脂肪变.高脂组TG、TC、LDL-C、ALT及AKP均显著高于对照组(0.29±0.05 mmol/L vs 0.18±0.09 mmol/L.2.49±0.35 mmol/L vs 1.25±0.16 mmol/L.1.69±0.18 mmol/L vs 0.49±0.06 mmol/L.25.46±5.14 kU/ L vs 13.15±2.83 kU/L,416.1±42.7 U/L vs 281.2±47.3 U/L;P<0.01).HDL-C则低于对照(0.65±0.11 mmol/L vs 0.75±0.10 mmol/L,P<0.01).血浆ADM2/IMD水平两组间差异无统计学意义(P>0.05);但高脂组肝组织匀浆ADM2/IMD水平较对照组高115%(26.21±11.2 pg/mg·pro vs 12.18±2.9 pg/mg·pro,P<0.01).结论:ADM2/IMD可能与大鼠饮食性高脂血症和高脂血症性脂肪肝病理进程有关. 相似文献

公众对自杀的态度量表的编制及在社区和大学学生中的应用 总被引:3,自引:1,他引:2
目的:编制公众对自杀的态度量表(the Scale of Public Attitudes about Suicide,SPAS),并在社区和大学学生中检验其信效度。方法:采用多阶段分层随机取样,抽取山东农村居民604例、天津城市居民548例及重庆的大学学生626例,用SPAS进行调查。首次调查后5~8天,顺序选择1063例被试进行重测。将受教育程度≥9年(n=580)的社区居民随机分为自评(n=243)和由调查员提问并填写问卷(n=337),将自评的大学学生随机分为署名(n=321)和匿名(n=305)。结果:(1)SPAS包含47个条目,分为7个分量表。(2)自杀问题的社会重要性分量表的内部一致性和重测一致性偏低(Cronbachα=0.48,ICC=0.59);其他6个分量表的内部一致性α在0.62~0.87之间,重测一致性ICC在0.62~0.82之间。(3)农村居民、城市居民和学生之间,除对自杀的正性态度分量表得分外,其余6个分量表得分的差异均有统计学意义(均P<0.05),农村组得分最高而学生组得分最低。自评组和由调查员提问并填写组相比,除自杀行为的自我不可控制性和对自杀的正性态度2个分量表外,其余5个分量表得分均是后者高于前者(均P<0.05)。署名和匿名组各分量表得分差异均无统计学意义(均P>0.05)。结论:公众对自杀的态度量表是一个可靠的评估国内社会对自杀的态度的工具。 相似文献

目的观察脑苷肌肽(cattle encephalon glycoside and ignotin,CEGI)对体外分离的rd小鼠视网膜细胞的保护作用,旨在探求CEGI对视网膜以及视神经损伤相关疾病的治疗效果,为临床用药提供客观依据和指导。方法采用体外分离rd小鼠视网膜细胞悬液,用MTT和台盼蓝染料排斥法观察CEGI和神经生长因子(NGF)对细胞活性的影响。结果 MTT实验示CEGI8.00mg·L-1、0.04g·L-1、0.20g·L-1和NGF0.04mg·L-1、0.20mg·L-1对视网膜细胞活性有促进作用,尤其以CEGI0.04g·L-1和NGF0.20mg·L-1最为明显,细胞增殖率分别为130.0%±21.8%和124.0%±13.3%。若将二者组合使用,可使细胞增长率达到132.0%±9.1%,与药物空白对照组100.0%±10.1%相比差异均有统计学意义。台盼蓝染料排斥试验结果显示0.04g·L-1CEGI组活细胞比例在除5min外的各时间点,都明显高于对照组,而0.20mg·L-1NGF组也可保护视网膜细胞,主要以短时间点为主(5min~1d),与对照组相比差异均有统计学意义(均为P<0.05)。结论 CEGI有促进视网膜细胞生长发育作用,对rd小鼠视网膜细胞有一定的保护作用。 相似文献
