Objective To establish animal model of fatty liver similar with human pathophysiology used for the study of imaging quantitative diagnosis or verification of medicine curative effect. Methods Forty-five New Zealand white rabbits were used in this study. They were divided randomly into five groups : 10 rabbits in normal control group, 10 rabbits in experiment group 1,10 rabbits in experiment group 2,10 rabbits in experi-ment group 3,and remaining 5 rabbits were in a supplement group. Rabbits in normal control group were given free access to normal food and water for 9 weeks. Rabbits in the experimental groups were fed a high-fat and high-sucrose diet for 3,6,and 12 weeks,respectively. Rabbits in the supplement group were fed the same diet as the experimental groups. Liver specimen stained with HE and Sudan Ⅳ was examined histological. Results The success rate of hepatic steatotie models was 87 % (39 of 45 ) ;mortality was 13 % (6 of 45 ). There were 10 nor-mal livers, 10 mildly fatty livers, 9 moderately fatty livers, and 10 severely fatty livers in results (stained with HE), and the rate of conforming to Sudan Ⅳ was 75%. Conclusion The animal model of fatty liver with differ-ent severity can be established by being fed a high fat, high sucrose and alcohol diet.  相似文献

目的建立近似人病理生理过程的动物脂肪肝模型,使之适用于影像学定量诊断或药物疗效的验证研究。方法健康新西兰大白兔45只,随机分为四组:正常对照组10只,普通饲料及水喂养9周;实验1、2、3组,各10只,采用高脂高糖饲料+酒精饮料分别喂养3周、6周和12周;补充组5只,给予实验组相同的喂养和处理。采用HE染色和苏丹Ⅳ染色对动物肝脏进行组织学检查。结果实验成模率为87%(39/45),死亡率为13%(6/45),出现正常肝10只,轻度脂肪肝10只,中度脂肪肝9只,重度脂肪肝10只(HE染色),与苏丹Ⅳ染色Vv值分级的诊断符合率约为75%。结论综合运用高脂高糖饲料+酒精饮料可成功建立近似人病理生理过程的动物脂肪肝模型。  相似文献

目的比较生化、超声、CT、1H MRS检查在脂肪肝诊断中的临床价值。方法选用45只健康新西兰大白兔,采用高脂高糖饲料加酒精饮料建立不同程度脂肪肝模型,应用生化、超声、CT及1H MRS多种临床诊断手段,观测多个变量指标,并进行对照研究。组间差异采用单因素方差分析和LSD检验,相关性采用等级相关分析。结果在所选择的18个观测指标中,血清谷草转氨酶、胆固醇、甘油三脂、超声衰减系数、肝CT值、肝/竖脊肌密度比率、1H MRS脂肪峰值、脂肪波峰下面积、脂/水波峰下面积比率、脂肪百分比等10个指标存在组间差异性,且与脂肪肝分级具有显著性相关,其中血清胆固醇、超声衰减系数、肝CT值和1H MRS脂肪曲线下面积分别是生化、超声、CT、MRS诊断方法中与脂肪肝分级相关系数最高的观测指标,相关系数分别为0.886、0.483、-0.764、0.558。结论在脂肪肝的临床诊断中,生化、CT检查的应用价值高于超声波和1HMRS检查。  相似文献

目的 寻找和筛选脂肪肝定量诊断的最佳预测指标,建立多指标联合定量诊断脂肪肝的新方法.方法 选用45只健康新西兰大白兔,采用高脂高糖饲料加酒精饮料建立不同程度脂肪肝模型,应用大体观测、血生化、超声(US)、计算机断层扫描(CT)及质子磁共振波谱(1H-MRS)多种临床诊断手段,对22个观测指标进行筛选,寻找脂肪肝定量诊断的最佳预测指标,并建立脂肪肝综合定量诊断的数学模型;通过ROC曲线分析比较多指标联合应用与单指标应用诊断脂肪肝的价值.结果 肝脏湿重、肝脏指数、肝CT值、肝/腰大肌密度比率、1H-MRS脂肪峰值、脂肪波峰下面积、脂/水波峰下面积比率、脂肪百分比、超声衰减系数、谷草转氨酶(AST)、胆固醇(TC)、甘油三脂(TG)等12个指标存在组间差异性,且与脂肪肝分级诊断具有显著性相关,其中肝脏指数、肝CT值和血TC为脂肪肝分级诊断的最佳预测指标,数学模型为Y=1.98+3.91×10-2X1+0.37X2-2.84×10-2X3(Y:脂肪肝分级;X1:血TC;X2:肝脏指数;X3:肝CT值);ROC分析显示多指标联合应用(Y)诊断脂肪肝的价值优于单指标应用.结论 联合应用多种诊断手段,综合考虑多个最佳预测指标的综合诊断方法,能够为临床快速、准确地判断脂肪肝程度提供帮助.  相似文献

目的 利用ROC曲线比较生化、超声、CT、质子磁共振波谱(1H MRS)定量诊断脂肪肝的应用价值.方法 选用45只健康新西兰大白兔,高脂高糖饲料加酒精饮料建立不同程度脂肪肝模型,应用生化、超声、CT及1H MRS多种.临床诊断方法,共观测18个变量指标,在每种诊断方法所观测的指标中,选取与脂肪肝分级相关性最高的指标,并以此进行受试者操作特征(ROC)曲线分析和ROC曲线下面积Z检验,比较这几种诊断方法的应用价值.结果 在所有18个观测指标中,血清胆固醇、超声衰减系数、肝CT值和1H MRS脂肪波峰下面积分别是生化、超声、CT、1H MRS诊断方法中与脂肪肝分级相关性最高的观测指标,相关系数分别为0.886(P=0.000)、0.483(P=0.002)、-0.764(P=0.000)、0.558(P=0.001);ROC曲线分析和Z检验显示:上述各指标ROC曲线下面积分别为0.981,0.581,0.810,0.713,除1H MRS脂肪波峰下面积与超声衰减系数、肝CT值之间差异无统计学意义外(P>O.05),其他两两之间差异均有统计学意义(P1H MRS>超声;生化测定血清胆固醇水平对判断脂肪肝程度可能具有重要的意义,值得进一步研究.  相似文献

目的比较STEAM和PRESS序列在脂肪肝定量诊断中的应用价值。方法选用45只健康新西兰大白兔,采用高脂高糖饲料加酒精饮料建立不同程度脂肪肝模型,采用STEAM和PRESS两种脉冲序列对同一位置、同一体素进行磁共振氢波谱扫描,共观察和测定9个指标,并逐一进行配对对照研究。结果经STEAM和PRESS两种定位方法测定,肝内水化学位移、脂肪化学位移、脂/水峰值比率、脂/水波峰下面积比率、脂肪百分比组间均数无显著性差异(P〉0.05);而肝内水峰峰值、脂峰峰值、水波峰下面积、脂波峰下面积组间均数存在显著性差异(P〈0.05),即STEAM序列下上述各指标均数明显低于PRESS序列,信号相对损失率平均约47.9%;且STEAM序列下各观测指标与脂肪肝病理分级的相关性普遍低于PRESS序列。结论PRESS较STEAM序列更适用于肝内脂肪含量的定量研究。  相似文献

目的筛选和寻找脂肪肝分级诊断的最佳预测指标。方法45只健康新西兰大白兔,采用高脂高糖饲料加酒精饮料建立不同程度脂肪肝模型,应用肉眼观测、血生化、超声、计算机断层扫描(CT)及氢磁共振波谱(1H MRS)多种临床诊断手段,观测22个指标并进行对照研究。结果在所选择的22个观测指标中,肝脏湿重、肝脏指数、肝CT值、肝/腰大肌密度比率、1HMRS脂肪峰值、脂肪波峰下面积、脂/水波峰下面积比率、脂肪百分比、超声衰减系数、谷草转氨酶(AST)、血清胆固醇(CHOL)、三酰甘油(TG)等12个指标总体均数存在组间差异性,且与脂肪肝分级具有显著性相关,其中肝脏指数、肝CT值和血清胆固醇为脂肪肝分级诊断的最佳预测指标,相关系数分别为0.709,-0.764,0.886,判断脂肪肝分级的回归方程Y=1.975+3.906×10^-2X1+0.369X2-2.84×10^-2X3,Y:脂肪肝分级,X1:血清胆固醇,X2:肝脏指数,X3:肝CT值。结论联合应用多种诊断手段,综合考虑多个最佳预测指标的诊断方法,能够为临床快速、准确地判断脂肪肝程度提供帮助。  相似文献
