目的 采用分子生物学方法评价羟基磷灰石/二氧化锆(HA/ZrO2)复合材料颗粒对兔骨髓间充质干细胞(MSCs)增殖及成骨分化的影响.方法 按不同比例将纳米HA粉体与纳米ZrO2粉体混合在高温下烧结制备HA/ZrO2复合材料颗粒,分离和培养兔MSCs.MTT法检测复合材料颗粒悬液对兔MSCs增殖促进作用,ALP法测定碱性磷酸酶活性,RT-PCR检测细胞Ⅰ型胶原(collagen Ⅰ),骨钙蛋白(osteocalcin),骨桥蛋白(osteopontin)mRNA表达.结果 HA和含有HA的复合材料颗粒均对细胞增殖有一定促进作用.Vonkossa染色显示复合材料和纯HA颗粒可使细胞阳染程度降低.细胞在HA和含有HA的复合材料颗粒的成骨诱导培养液中的ALP活性均显著高于常规培养基质及纯ZrO2组(P<0.05).RT-PCR结果显示复合材料颗粒能够促进collagen Ⅰ和osteocalcin基因的表达.结论 HA/ZrO2复合材料颗粒可促进骨髓间充质干细胞增殖及成骨分化.Abstract: Objective To evaluate the effect of the HA/ZrO2 composite particle on proliferation and osteogenesis of rabbit mesenchymal stem cells (MSCs) by using molecular biology methods.Methods The HA/ZrO2 composite particles were sintered at high temperature using the powder of HA and ZrO2 with different proportions.MSCs were isolated from rabbits and cultured.The effect of the composite particles on promoting cell proliferation of rabbit MSCs was detected using MTT method.Alkaline phosphatase activities were measured with ALP method.RT-PCR method was applied to measure the expression of collagen Ⅰ , osteocalcin and osteopontin mRNA.Results Pure HA pauicles and composite particles which contain HA promoted MSCs proliferation.Vonkossa staining showed that both pure HA particks and composite particles decreased the degree of positive stain of MSCs.ALP test showed that cell activity in the culture medium that contained pure HA particles or composite particles was significantly higher than that in the conventional culture medium and that contained pure ZrO2 particles (P < 0.05=.RT-PCR results showed that composite particles promoted the expression of collagen Ⅰ and osteocalcin gene.Conclusion The HA/ZrO2 composite particles promote proliferation and osteogenesis of MSCs. 相似文献

目的探讨改良经皮椎体后凸成形术(PKP)治疗老年骨质疏松性压缩性骨折(OVCF)的疗效。方法 38例OVCF患者根据手术方法不同分为常规PKP组(20例)及改良PKP组(18例),比较两组疗效。结果常规组术后4例椎体出现骨水泥渗漏,改良组3例;两组各1例出现神经根症状。两组术前、术后3 d、术后2周、术后4周及术后24周视觉疼痛模拟评分(VAS)差异均无统计学意义(均>0.05),术中及术后12周差异均有统计学意义(均<0.05)。两组间术后12周Cobb角差异有统计学意义(<0.05)。两组术后椎体前、后缘高度差异均有统计学意义(<0.05)。结论改良PKP组治疗新鲜OVCF疗效满意,术后疼痛评分及Cobb角恢复明显改善。 相似文献

股骨近端解剖锁定钢板治疗伴外侧壁骨折的股骨转子间骨折中期疗效随访 总被引:1,自引:1,他引:0
目的 :对股骨近端解剖型钢板治疗伴外侧壁骨折的转子间骨折中期疗效进行回顾性研究。方法 :2010年6月至2013年1月采用股骨近端解剖型钢板固定治疗转子间伴外侧壁骨折18例,男8例,女10例;年龄19~83岁,平均75.5岁;车祸伤8例,摔伤6例,高处坠落伤4例;受伤至手术时间为1~14 d,平均4.5 d;随访时拍摄股骨正侧位X线片。记录患者手术时间、术中出血量、住院天数,术后疗效评价采用Parker Palmer mobility score(PPMS)及Harris髋关节功能评分标准。结果:18例患者均获得随访,时间36~68个月,平均(44.8±8.8)个月。手术时间(61.02±38.28)min,术中出血量(226.00±162.52)ml,住院时间(10.8±9.2)d。所有患者未出现局部感染、深静脉血栓、螺钉切割股骨头及断钉,2例患者出现不同程度髋内翻短缩畸形,无骨折不愈合患者。骨折愈合时间为4~10个月,平均6.2个月。末次随访时,PPMS评分为(7.22±2.36)分;Harris评分为(79.46±11.02)分,其中优5例,良9例,可2例。结论:解剖型锁定钢板治疗伴外侧壁骨折的转子间骨折中期疗效显著,术中可避免外侧壁二次损伤,尤其对外侧壁严重粉碎、髓内钉操作困难以及高龄骨质疏松者,术后需严格随访,不应强调过早负重。 相似文献

目的 评价羟基磷灰石( hydroxyapatite,HA)、二氧化锆(zirconia,ZrO2)梯度复合材料与骨界面的结合情况及修复骨缺损的能力. 方法 在24只Beagle犬腰椎椎体制作骨缺损模型,依次植入HA/ZrO2梯度复合材料(A组)、HA/ZrO2单层复合材料(B组)、单纯ZrO2材料(C组)、单纯HA材料(D组),分别于术后6,12,16周分批处死动物取材,6,12周处死动物前肌肉注射盐酸四环素行荧光标记,并通过X线片、组织形态学、生物力学测试,观察其界面结合及骨缺损修复情况. 结果 X线片显示A组随着植入时间的延长,骨痂生成较多,骨修复愈合较好,其次为B组和D组,C组相对较差.组织形态学观察结果显示,6~12周A组材料的荧光标记逐渐增强,并从材料的边缘向材料内部长入,且与材料结合紧密,提示成骨活跃,新骨生成量较多.而B、C、D组材料的荧光标记均集中在材料的边缘,材料内部未见荧光标记.6,12周时四种材料的矿化率差异均有统计学意义(P<0.05),骨性结合率在6,12,16周时均以A组为最高,16周时达(90.26±3.82)%(P<0.05).生物力学测试结果显示,6,12,16周时四组材料的最大抗剪切强度均以A组最大,分别为(2.64±0.16) MPa、(2.95±0.19) MPa、(3.45±0.23)MPa(P <0.05). 结论 HA/ZrO2梯度复合材料与骨界面结合良好,具有较强的骨缺损修复能力,是一种理想的新型骨缺损修复材料. 相似文献

目的: 采用体外微核法评价HA/ZrO2复合材料颗粒的基因毒性。方法: 按不同比例将羟基磷灰石与二氧化锆粉体混合在高温高压下烧结制备HA/ZrO2复合材料颗粒,纯HA和纯ZrO2颗粒作为对照。制备复合材料颗粒的悬液,分离和培养兔间充质干细胞。MTT法检测复合材料颗粒悬液对兔间充质干细胞增殖促进作用,体外微核试验(MNT) 检测复合材料颗粒悬液对兔间充质干细胞的基因毒性。结果: MTT实验显示:纯HA和含HA的复合材料颗粒均对间充质干细胞增殖有一定促进作用,而纯ZrO2颗粒对间充质干细胞增殖没有促进作用,差异显著(P<0.05)。MNT实验显示:HA组与阴性对照组相比差异不显著(P>0.05),与阳性对照组比差异显著(P<0.05); ZrO2组与阴性对照组比差异显著(P<0.01),与阳性对照组比差异不显著(P>0.05)。结论: HA/ZrO2复合材料颗粒的基因毒性随着ZrO2比例和复合材料浓度的增加而增加,30%wtHA/70%wtZrO2在200 mg/L时具有显著基因毒性(P<0.01)。 相似文献

目的:分离培养兔骨髓间充质干细胞并成骨诱导分化。方法:从兔骨髓中分离培养原代MSCs并进行传代培养,绘制生长曲线。取第3代细胞进行成骨诱导分化,进行Vonkossa染色,碱性磷酸酶染色,碱性磷酸酶(ALP)活性测定,总蛋白检测,提取细胞总RNA及RT-PCR法检测Collagen I,osteocalcin及osteopontin基因表达情况。结果:MSCs贴壁生长,呈放射样、爆花样或漩涡样,可传20代以上。未经成骨诱导的MSCs不发生矿化沉积,而经成骨诱导分化后,细胞可发生显著的矿质沉积。未经成骨诱导的MSCs可微量表达CollagenI,但不表达osteocalcin和osteopontin,而经成骨诱导分化后,细胞表达CollagenI增强,并出现osteocalcin和osteopontin等成骨标志物的表达。结论:本实验成功分离培养MSCs并成骨诱导分化。 相似文献
