应用断裂冷冻法体外标记肾小球成份,定位WGA受体和阴电荷点。组织经冷冻断裂和冻融后,标记物无阻碍地到达细胞表面和基质横断面可直接比较结合活性,使用的示踪物可以不管缓冲液的pH,用阳离子铁蛋白(CF,PI 8.3)测定肾小球阴电荷点,所有细胞表面、基底膜和系膜基质断裂面呈连续标记,在所有细胞表面和断裂基质见到WGA—金胶复合物。神经氨酸酶处理后,WGA不再与足细胞和基底膜结合,但与内皮细胞、系膜细胞和系膜基质断裂面结合,提示在足细胞WGA结合NeuNAc,在内皮细胞、系膜细胞和系膜基质结合GIcNAc,在基底膜结合NeuNAc和GIcNAc。 相似文献

用人A型红细胞作为抗原免疫BALB/C小鼠,取其脾细胞与小鼠骨髓瘤细胞NS-1进行细胞杂交。从五批细胞融合中选取五个克隆细胞株,其中一株命名C_(11)细胞株,经七次克隆化培养,体外传代培养达八个多月,生长良好,继续分泌单克隆抗体。其单克隆抗体与不同血型红细胞(凝集法)和正常人外周血T、B淋巴细胞以及三个正常人B淋巴母细胞系补体依赖微量细胞毒方法作交叉试验,结果C_(11)细胞株产生的单克隆抗体只对A型红细胞发生反应与淋巴细胞抗原亦无交叉反应。 相似文献

作者等对一个Becker型肌营养不良症家系中的3名患者与3名递体进行了肌肉活组织检查。3例递体均有肌肉病理改变。肌核变化比收缩部份变化明显,其病变随年龄增长而加重,提示递体的肌肉病变也是缓慢进行的,因病变不重,范围不广,而不足以引起肌力减退。从而或可将递体认为是亚临床病例。患者的病肌变化包括肌纤维粗细不匀、变性、肌管形成、坏死、被噬、纤维化与脂肪浸润等。肌核变化也明显。包括肌核增生,中央肌核,链状排列与成簇等。递体的肌肉,早期出现肌核变化。寻找病损的最早部位,有利于了解病机,在这方面,除肌膜外尚需注意肌核。 相似文献

nephrotic syndrome were observed in this study.

globulin G and immunoglobulin M were analysed

various pairing systems according to the relation
with the 3 proteins of different molecular size
and also to assess the role the 3 proteins played
in connection with the results of treatment,

Four brothers from a family are suffered from hypertension, the two of of them are due to the Liddle's syndrome. Their monther was hypertensive and attacked with apoplexy two times, she died of coronary heart disease. The renal biopsy was taken in one patient and examined under optical and electronic microscope. The erythrocytes of the two patients were examined by scan electronic microscope. The treatment with triamterene combined to hyposodium regime was imperative, the arterial tension and biological abnormalities of the patients became normal rapidly. 相似文献

本文采用多聚赖氨酸阳离子膜制备人类血细胞的扫描电镜观察样品。由于血细胞的多阴离子表面和多聚赖氨酸的多阳离子表面之间的相互作用,各种细胞可以牢固地吸附到用多聚赖氯酸(Poly-L-lysine)预先处理过的玻璃片上。当细胞与表面接触时,则立即发生细胞的附着。被吸附的细胞伸展好,单层排列,分布均匀,画面清晰,便于观察而不失去细胞原有的鉴别性形态特征。被吸附的生物材料不因制备样品过程中的固定、脱水和临界点干燥等处理而脱落。应用该法制备了人外周血淋巴细胞,建系的淋巴细胞的扫描电镜的观察样品。这些细胞都显示了清晰而立体感强的扫描图象。这对研究各种血细胞及其他的游离细胞的表面超微结构和功能的改变具有一定意义。 相似文献
