核糖核酸的酶消化方法直接顺序分析

应用硷基特异性核酸内切酶进行RNA的顺序分析。以~(32)P标记RNA分子的5′或3′末端的四种硷基能被酶试剂部分裂解。放射性的水解产物经胶电泳按其大小而分离。放射自显影后它的核苷酸顺序从带谱上可以直接读出。核糖核酸酶(RNasc)T_1(G特异性)、U_2(A特异性)、Phy M(U+A特异性)和B.cereus(C+U特异性)是最常用于顺序RNA的四种酶。     

血液胆碱酯酶活力测定方法的进一步研究Ⅰ．红细胞／全血胆碱酯酶活力比例及红细胞计数校正酶活力值的研究

应用硫代乙酰胆碱－联硫代双硝基苯甲酸（ＡＳＣｈ－ＤＴＮＢ）比色测定法，测定了４０名健康者及５名贫血者血样的全血胆碱酯酶（ｂｌ－ＣｈＥ）活力值、红细胞胆碱酯酶（ｅ－ＣｈＥ）活力值及血浆胆碱酯酶（ｐ－ＣｈＥ）活力值，探讨它们之间的比例关系并用红细胞计数值（ＲＢＣ）校正酶活力。结果表明：ｅ－ＣｈＥ与ｂ１－ＣｈＥ之间关系密切（ｒ＝０．９４８，Ｐ＜０．００１），ｅ－ＣｈＥ稳定地占ｂｌ－ＣｈＥ的８４．９７％，在质和量两方面验证了以ｂｌ－ＣｈＥ值表示ｅ－ＣｈＥ值的可靠性和可信程度，血液胆碱酯酶（ｂ１ＣｈＥ）活力主要取决于ｅ－ＣｈＥ活力，ＲＢＣ值的离散对ｅ－ＣｈＥ值有极大影响，故用ＲＢＣ值校正ｅ－ＣｈＥ活力，这样可缩小人群测定值的变异度，得到较难确的群体均值；消除男女之间测定值的差异，建立更合理和通用的临界参比值，极大地方便了使用，使血液胆碱酯酶（ＣＨＥ）活力测定法在防治有机磷农药中毒中的应用，更为敏感和特异。     

医院老年病房四季全膳食中十五种矿物质元素的测定及评价

对我院老年病房四季１１２份全膳食中的１５种元素（钾、钠、钙、磷、镁、锌、铜、铁、锰、硒、铝、钴、铬、钼、镍）进行了实测。结果表明：钾的平均日摄入量为１８０９＋３８ｍｇ，接近ＲＤＡ值的下限；钠的平均日摄入量为５９７２±１４６ｍｇ远高于１１００～３３００ｍｇ的供给标准。钙、磷、镁的日摄入量已达到了ＲＤＡ值的标准。全膳食中锌、铜的含量分别为１１．６８±０．１９ｍｇ和１．７２±０．１０ｒｎｇ，略低于ＲＤＡ值（１５ｍｇ及２～３ｍｇ）的标准，锰含量为３．４１±０．０９ｍｇ硒为９８±２．５μｇ，都处于安全与适宜摄入范围（２．５～５．０ｍｇ和５０～２００μ９）。钴、铬、钼、镍均为人体必需的微量元素、目前我国尚未建立有关营养供给量标准。该项研究结果可为今后确定其推荐供给量标准提供必要的基础资料．     

中药结合心理干预治疗不安障碍40例

[目的]探索不安障碍治疗的有效途径。[方法]采用中药结合心理干预治疗不安障碍患者40例。[结果]显效31例(77·5%)、有效7例(17·5%)无效2例(5·0%)。SDS、CMI有显著性差异。[结论]中药结合心理干预治疗不安障碍疗效显著。     

流行性乙型脑炎病毒结构蛋白编码基因的重组构建

目的:建立流行性乙型脑炎病毒(JEV)C、prME,E等结构蛋白编码基因克隆.方法:病毒感染C6/36细胞,RT-PCR法依次获取该病毒结构蛋白基因并进行DNA测序分析,PCR克隆法将PCR产物分别构建于真核表达载体pcDNA 3.1的BamHI与EcoRI酶切位点并依次命名为pJC、pJME与pJE,通过DNA测序、电泳以及限制性内切酶分析加以鉴定.结果:JEV C、prME与E基因片段分别为380、2001以及1 500bp,各基因测序结果与发表的JEV JaGAr-01株相对应序列相一致,从重组质粒释出的插入子分别与各基因大小相符合.结论:pJC、pJME与pJE重组质粒分别含有C、prME与E蛋白编码基因.     

Whole-heart cine MRI using real-time respiratory self-gating.

Two-dimensional (2D) breath-hold cine MRI is used to assess cardiac anatomy and function. However, this technique requires cooperation from the patient, and in some cases the scan planning is complicated. Isotropic nonangulated three-dimensional (3D) cardiac MR can overcome some of these problems because it requires minimal planning and can be reformatted in any plane. However, current methods, even those that use undersampling techniques, involve breath-holding for periods that are too long for many patients. Free-breathing respiratory gating sequences represent a possible solution for realizing 3D cine imaging. A real-time respiratory self-gating technique for whole-heart cine MRI is presented. The technique enables assessment of cardiac anatomy and function with minimum planning or patient cooperation. Nonangulated isotropic 3D data were acquired from five healthy volunteers and then reformatted into 2D clinical views. The respiratory self-gating technique is shown to improve image quality in free-breathing scanning. In addition, ventricular volumetric data obtained using the 3D approach were comparable to those acquired with the conventional multislice 2D approach.     

植入后全胚胎培养的培养基pH值改变对大鼠胚胎生长发育的影响

目的　探讨植入后全胚胎培养培养基最适pH值。方法　分离 9.5天龄Wistar大鼠胚胎 ,分别在实验组 (即不同pH :5 .5 ,6 .0 ,6 .5 ,7.0 ,7.5 ,8.0 ,8.5 ,9.0 )和对照组 (pH为 8.2 )的培养基 (1 0 0 %大鼠即刻离心血清 )中体外旋转培养4 8小时 ,观察pH值的改变对胚胎生长发育的影响。结果　与对照组比较 ,当培养基pH为 6 .5、6 .0和 9.0时培养胚胎的生长发育明显受到影响 (P <0 .0 5 ) ,胚胎畸形率和死亡率增高 (P <0 .0 5 )。当培养基pH为 5 .5时 ,培养胚胎死亡率达1 0 0 %。结论　全胚胎培养培养基pH条件改变 (过低或过高 )能够影响培养胚胎的生长发育。植入后全胚胎培养培养基最适pH值为 7.5～ 8.2。     

Distribution and spatial geometry of dopamine interplexiform cells in the retina. II. External arborizations in the adult rat and monkey

The morphology and distribution of dopaminergic interplexiform cells in adult rat and monkey retinas were analyzed to determine any correlation with the function of dopamine in the outer retinal layers. The retinas were processed as whole mounts for tyrosine hydroxylase immunohistochemistry. There was a network formed by the sclerally directed processes of interplexiform cells in the inner nuclear, outer plexiform, and outer nuclear layers running throughout the retina. Their density was higher in the superior retina than in the inferior retina of the rat and was especially high in the superior temporal quadrant. The external network in this quadrant was significantly less dense in the monkey than in the rat, as are the interplexiform cells. The somata of interplexiform and other dopaminergic cells were about the same size in both rats and monkeys. Computer-assisted reconstruction of external arborizations of individual cells showed that external processes lay very close to horizontal and photoreceptor cells and also to blood capillaries. Because they were long, thin, and highly varicose; branched at right angles; and often arose from an axon hillock, the external processes were identified as axons. Therefore, we define the dopaminergic interplexiform cells as multiaxonal neurons, with at least one outwardly directed axon that reaches the outer plexiform layer. The function of the network of external processes from the interplexiform dopaminergic cells is discussed in terms of modulating the release of dopamine to external layers.     

成人全唾液与血浆中醛固酮含量测定

本文采用直接放射免疫测定法,固定条件,按标准程序测得30名正常成人全唾液及血浆中的醛固酮(Aldosterone,Aldo)浓度。均值唾液为6.6±1.9nmol/L,血浆为32.5±8.0nmol/L,两者呈平行关系(r=0.4,P<0.05,=11.29+0.79X),浓度比平均为20.2％。结果进一步证实唾液作为临床测定Aldo标本的有效性与实用性,有助于对一些全身及口腔疾病进行诊断及估计预后,并可据以评价皮质类固醇治疗效果及用药的合理性。
