目的 探讨经鼻给予神经生长因子(NGF)对沙林染毒大鼠脑组织梨状皮质区神经元的影响.方法 建立大鼠沙林染毒模型,常规治疗后经鼻给予NGF或生理盐水,24 h后采用HE染色和免疫组织化学染色观察梨状皮质区神经元的变化.结果 与空白对照组相比,经鼻给予生理盐水的大鼠梨状皮质区可见较多变性、坏死的神经元,神经元数量[(404.75±25.17)个/mm2]明显减少(39.44%);而经鼻给予NGF组变性、坏死的神经元较少,神经元数量[(651.94±36.02)个/mm2]减少不明显.结论 经鼻给予NGF可以减轻沙林染毒大鼠梨状皮质区损伤程度.经鼻给予NGF有可能成为针对沙林所致脑损伤的有效治疗手段. 相似文献

We identified and characterized a neurodifferentiation compound from the marine brown alga Sargassum fulvellum collected from the Japanese coastline. Several instrumental analyses revealed the compound to be pheophytin a. Pheophytin a did not itself promote neurite outgrowth of PC12 cells. However, when PC12 cells were treated with a low concentration of pheophytin a (3.9 microg/ml) in the presence of a low level of nerve growth factor (10 ng/ml), the compound produced neurite outgrowth similar to that produced by a high level of nerve growth factor (50 ng/ml). Pheophytin a also enhanced signal transduction in the mitogen-activated protein kinase signaling pathway, which is also induced by nerve growth factor. The effect of pheophytin a on neurite outgrowth of PC12 cells was completely blocked by U0126, a representative mitogen-activated protein kinase kinase inhibitor. These results suggest that pheophytin a enhances the neurodifferentiation of PC12 cells in the presence of a low level of nerve growth factor and that this effect is mediated by activation of a mitogen-activated protein kinase signaling pathway. 相似文献

目的:评价神经生长因子(nerve growth factor,NGF)几丁质管在修复兔面神经缺损中的作用。方法:在16只新西兰兔的两侧面神经上颊支上分别造成8mm缺损,左侧用管腔内注入NGF的几丁质管修复,右侧用自体神经移植修复作对照。术后8周和16周分别取8只动物进行电生理和组织学检查及计算机图像分析。结果:术后8周,实验组的再生神经已通过近远中吻合口,神经纤维密集成束;术后16周,再生神经纤维的数量增加,神经纤维束增粗,形态接近于正常神经。电生理检测和图像分析显示实验组和对照组的神经传导速度和有髓神经轴突总截面积均无显著差异(P>0.05)。结论:NGF几丁质管可为免面神经缺损提供良好的修复环境。 相似文献

Summary The long term results of 21 cases of cross-face nerve grafting are presented and analyzed to evaluate the value of the procedure. The follow-up period extended beyond 3 years in all cases as the final assessment of the facial reanimation is only possible after a very long time interval. A satisfactory or good reanimation of the face occurred in 80% of the patients. In well defined cases the procedure may be considered a valuable tool in the rehabilitation of the paralyzed face. 相似文献

Central activation of excitatory amino acid receptors has been implicated in neuropathic pain following nerve injury. In a rat model of painful peripheral mononeuropathy, we compared the effects of non-competitive NMDA receptor antagonists (MK 801 and HA966) and a non-NMDA receptor antagonist (CNQX) on induction and maintenance of thermal hyperalgesia induced by chronic constrictive injury (CCI) of the rat common sciatic nerve. Thermal hyperalgesia to radiant heat was assessed by using a foot-withdrawal test and NMDA/non-NMDA receptor antagonists were administered intrathecally onto the lumbar spinal cord before and after nerve injury. Four daily single treatments with 20 nmol HA966 or CNQX beginning 15 min prior to nerve ligation (pre-injury treatment), reliably reduced thermal hyperalgesia in CCI rats on days 3, 5, 7 and 10 after nerve ligation. Thermal hyperalgesia was also reduced in CCI rats receiving a single post-injury treatment with HA966 (20 or 80 nmol) or MK 801 (5 or 20 nmol) on day 3 after nerve ligation when thermal hyperalgesia was well developed. In contrast, a single post-injury CNQX (20 or 80 nmol) treatment failed to reduce thermal hyperalgesia or to potentiate effects of HA966 or MK 801 (5 or 20 nmol) on thermal hyperalgesia in CCI rats. Moreover, multiple post-injury CNQX treatments utilizing the same dose regime as employed for the pre-injury treatment attenuated thermal hyperalgesia but only when the treatment began 1 or 24 h (but not 72 h) after nerve ligation.(ABSTRACT TRUNCATED AT 250 WORDS) 相似文献

The effects of intraventricular nerve growth factor (NGF) or saline treatments on extracellular acetylcholine (ACh), dopamine (DA) and adenosine (Ade) levels in the cortex and striatum of rats with unilateral devascularizing cortical lesions were studied in vivo with microdialysis. The devascularizing cortical lesion produced a decrease in extracellular ACh levels in both cortex and striatum as compared to those in normal rats, while the NGF treatment produced a significant increase in ACh levels in both regions. NGF could even increase cortical ACh levels in normal rats. The cortical lesion produced a decrease in extracellular DA in the cortex, while the NGF treatment appeared to reverse this effect. No significant changes in DA were observed in the striatum. The present study gives evidence that a unilateral cortical devascularizing lesion leads to changes in extracellular ACh and DA levels in cortex and striatum and that these changes could be reversed with intraventricular NGF treatment. 相似文献
