目的探讨中药鸡血藤Spatholobus suberectus Dunn活性成分儿茶素对小鼠骨髓细胞增殖周期及其脾细胞内造血生长因子IL-6和GM-CSF mRNA表达的影响,以初步阐明其造血调控作用机理。方法用流式细胞仪检测技术观察儿茶素对正常及骨髓抑制小鼠骨髓细胞增殖周期的影响,同时采用RT-PCR技术,检测在儿茶素刺激条件下,小鼠脾细胞内IL-6和GM-CSF mRNA表达的变化。结果儿茶素可促使正常及骨髓抑制小鼠骨髓细胞G0/G1期细胞比例下降,S+G2/M期细胞比例增加,并可使正常及骨髓抑制小鼠脾细胞内IL-6 mRNA和GM-CSF mRNA表达显著上调。结论儿茶素可通过诱导脾细胞内IL-6 mRNA和GM-CSF mRNA的表达,促进正常小鼠骨髓细胞进入增殖周期,并促使骨髓抑制小鼠的骨髓细胞跳出“G1期阻滞”,进入细胞增殖周期,从而加速造血干/祖细胞的增殖和分化。  相似文献

The effect of the proteolytic enzyme, trypsin, on bovine retinal rod outer segments has been investigated.Electron microscopy showed that trypsin rapidly digests the outer plasma membrane releasing free and apparently intact discs. That the discs were intact was confirmed by their ability to swell in hypotonic solutions.The visual pigment, rhodopsin, which accounts for some 85% of the total protein of the discs appears not to be digested by trypsin: neither its characteristic absorption spectrum, its ability to regenerate following bleaching by light, nor its molecular weight, were altered by the enzyme.It is suggested that the incubation of rod outer segments with trypsin is a gentle and rapid means of isolating free, viable discs.  相似文献

目的:利用家兔饵食性高血脂症模型研究黄花倒水莲总苷调脂作用。方法:连续给予新西兰家兔30d高脂饲料,使形成高脂模型,造模同时ig给予黄花倒水莲总苷(PIS)25,50,100mg/kg。停止高脂饲料喂养后继续给药30d。分别于给药前,给药后第30天和第60天测定血清中TC、TG、HDL-C和LDL-C。于第60天测定血清中SOD活性、MDA、ApoAl、ApoB含量并取肝组织测定TC、TG含量。结果:高脂饵食造模后第30天,第60天血清中TC,TG,和LDL-C及MDA,肝组织中TC,TG含量升高,血清中HDL-C含量和SOD活性降低。PTS能降低血清中TC0,TG,LDL-C及MDA,肝组织中TC、TG,升高血清中HDL-C含量和SOD活性。结论:高脂饲料喂养新西兰家兔30d即可形成高脂模型;黄花倒水莲总皂苷预防性给药能降低新西兰家兔血清中TC,TG,LDL-C及MDA含量,肝组织中TC,TG含量并升高血清中HDL-C含量和SOD活性并呈剂量相关性。ApoAl随HDL-C升高而升高,ApoB则随LDL-C减少而减少。  相似文献

目的:观察黄花倒水莲总苷对大鼠和家兔血液流变性的影响。方法:用高分子右旋糖酐造成大鼠急性血瘀模型,观察黄花倒水莲总苷对大鼠急性血瘀模型血液流变学各指标的影响。用高脂饲料造成家兔高脂血症模型,观察黄花倒水莲总苷对血液流变学各指标的影响。结果:黄花倒水莲总苷高,中,低剂量组均能明显防止急性血瘀模型大鼠和高脂饲料喂养家兔的全血黏度、纤维蛋白原含量的升高。结论:黄花倒水莲总苷能抑制高分子右旋糖酐血瘀模型和高脂血症造成血液流变学指标的改变,降低血液黏度,改善血液循环。  相似文献

目的 研究十齿花的化学成分.方法 采用硅胶柱色谱及Sephadex LH-20等色谱技术分离纯化,根据理化性质及波谱数据鉴定结构.结果 从十齿花的乙醇提取物中共分离得到9个化合物,分别为正三十醇(Ⅰ)、β-谷甾醇(Ⅱ)、邻苯二酚(Ⅲ)、对苯二酚(Ⅳ)、松柏醛(Ⅴ)、香草酸(Ⅵ)、麦角甾醇过氧化物(Ⅶ)、β-谷甾醇-3-O-β-D-葡萄糖-3'-O-三十三烷酸酯(Ⅷ)、胡萝卜苷(Ⅸ).结论 9个化合物均为首次从该植物中分得,其中化合物Ⅷ为新化合物,命名为十齿花素.  相似文献

Background Hematopoietic growth factor (HGF) is indispensable to hematopoiesis in the body. The proliferation and differentiation of hematopoietic cells must rely on the existence and stimulation of HGF. This study investigated the effect of catechin, an active component extracted from Spatholobus suberectus Dunn (SSD), on bioactivity of granulocyte-macrophage colony-stimulating activity (GM-CSA), burst-promoting activity (BPA) and megakaryocyte colony-stimulating activity (MK-CSA) in spleen condition medium (SPCM) of mice to clarify the hematopoietic mechanism of catechin and SSD.
Methods Spleen cells of mice were separated and spleen condition medium (SPCM) was prepared from spleen cell culture. Bone marrow cells of mice were separated and cultured in a culture system including 10% (v/v) SPCM (induced by catechin in vivo or ex vivo) for 6 days. Granulocyte-macrophage colony forming units (CFU-GM), erythrocyte burst-colony-forming units (BFU-E) and megakaryocyte colony-forming units (CFU-Meg) formation were employed to assay the effects of different treatment on the bioactivity of GM-CSA, BPA and MK-CSA in SPCM.
Results SPCM induced by 100 mg/L catechin ex vivo could promote the growth of CFU-GM, BFU-E and CFU-Meg, which indicated that catechin could stimulate the production of GM-CSA, BPA and MK-CSA in SPCM. SPCM prepared at the fourth day of spleen cell culture showed the best stimulating activity. The bioactivity of GM-CSA, BPA and MK-CSA in the SPCM prepared after intraperitoneally injecting catechin into mice was also increased. The number of CFU-GM, BFU-E and CFU-Meg gradually increased as the dose of catechin increased and the time of administration prolonged. CFU-GM, BFU-E and CFU-Meg of the high-dose catechin group were significantly higher than those of the control group (P〈0.01) and reached the maximum at the seventh day after administration.

BackgroundDietary self-monitoring (DSM) of foods and beverages is associated with weight loss in behavioral interventions; however, DSM may be burdensome, and adherence may decrease over time. Novel methods of DSM, including apps that track food using photographs, may decrease burden, increase DSM adherence, and improve weight loss.ObjectiveThe objective was to test a mobile photo DSM app compared to a calorie-tracking DSM app on tracking frequency and weight loss in a remotely delivered behavioral weight-loss intervention.DesignThis was a 6-month (October 2016 to April 2017) randomized trial.Participants/settingParticipants were adults (n=41) classified as overweight or obese (body mass index 25 to 49.9) from South Carolina.InterventionParticipants received remotely delivered twice-weekly behavioral weight-loss podcasts and tracked diet using a calorie-tracking DSM app (Calorie Group) or a photo DSM app (Photo Group).Main outcome measuresMain outcomes were the number of days diet was tracked, podcasts downloaded, and weight change at 6 weeks and 6 months.Statistical analysesResearchers used nonparametric Wilcoxon rank sum tests and χ2 analysis to test for differences between groups at baseline; repeated-measures models to estimate weight change and Spearman correlations to determine relationships between DSM frequency, podcasts downloaded, and weight change at 6 months.ResultsThere were no differences between groups for the number of days that diet was recorded (P=0.18), which was low overall (<30% of days) but was statistically significantly and strongly correlated with weight change for all participants pooled (r=0.63; P<0.001) and for the calorie tracking group (r=0.70; P=0.004), but not the photo tracking group (r=0.51; P=0.06). Participants in both groups had significant weight loss at 6 months (Photo Group, –2.5±0.9 kg; P=0.008; Calorie Group –2.4±0.9 kg; P=0.007), with no differences between groups at either 6 weeks (P=0.66) or at 6 months (P=0.74).ConclusionsAs part of a remotely delivered weight loss intervention, frequency of DSM was significantly associated with overall weight loss for participants using a calorie DSM app but not a photo DSM app. DSM was low regardless of group and weight loss was significant, although minimal. Increasing user engagement with any DSM may be important to increase self-monitoring and improve weight loss.  相似文献

目的 系统研究白花前胡香豆素合成途径关键酶基因的蛋白结构特征,并明确关键酶基因在盐胁迫下的表达模式。方法 通过白花前胡前期转录组测序,确定前胡香豆素生物合成关键酶基因的序列,利用蛋白质结构分析预测软件对关键酶蛋白进行系统预测分析,并通过qRT-PCR分析盐胁迫下关键酶基因的变化规律。结果 关键酶基因编码蛋白都有着各自特殊的保守结构域和三级结构;0.15 mol/L NaCl处理4 h对PAL和4CL基因的表达水平影响最高,比对照组上调1倍;而COMT-S和GXM在不同处理时间下,基因表达水平均明显下降。结论 受盐胁迫后上调表达的香豆素合成途径关键酶基因,可以提高植物对逆境胁迫的耐受性。  相似文献

Alcoholic extracts of blood collected for measurement of circulating bradykinin were analysed for co-extracted kininogen which would artificially elevate measured bradykinin levels. The blood extract contained a glycoprotein with identical chromatographic properties to kininogen. Bradykinin immunoreactivity in the extract eluted as a single peak from an immunoaffinity column of anti-bradykinin IgG bound to Sepharose identically to both bradykinin and kininogen showing immunoidentity of these substances. Affinity chromatography on Concanavalin A-Sepharose or ion-exchange chromatography on CM-Sephadex C25 separated bradykinin and the glycoprotein which again eluted identically to kininogen. The decrease in measured values of blood bradykinin after purification of the extract on CM-Sephadex C25 was a similar amount to that calculated as cross-reactivity with the amount of co-extracted kininogen. Hence radioimmunoassay of bradykinin in ethanolic extracts of blood is inaccurate owing to the presence of co-extracted kininogen, and additional purification steps such as chromatography on CM-Sephadex C25 are mandatory for accurate assay by ligand binding techniques.  相似文献
