黄花倒水莲总苷预防性给药对高脂血症家兔的调脂作用

摘    要:目的:利用家兔饵食性高血脂症模型研究黄花倒水莲总苷调脂作用。方法:连续给予新西兰家兔30d高脂饲料,使形成高脂模型,造模同时ig给予黄花倒水莲总苷(PIS)25,50,100mg/kg。停止高脂饲料喂养后继续给药30d。分别于给药前,给药后第30天和第60天测定血清中TC、TG、HDL-C和LDL-C。于第60天测定血清中SOD活性、MDA、ApoAl、ApoB含量并取肝组织测定TC、TG含量。结果:高脂饵食造模后第30天,第60天血清中TC,TG,和LDL-C及MDA,肝组织中TC,TG含量升高,血清中HDL-C含量和SOD活性降低。PTS能降低血清中TC0,TG,LDL-C及MDA,肝组织中TC、TG,升高血清中HDL-C含量和SOD活性。结论:高脂饲料喂养新西兰家兔30d即可形成高脂模型;黄花倒水莲总皂苷预防性给药能降低新西兰家兔血清中TC,TG,LDL-C及MDA含量,肝组织中TC,TG含量并升高血清中HDL-C含量和SOD活性并呈剂量相关性。ApoAl随HDL-C升高而升高,ApoB则随LDL-C减少而减少。

Abstract:AIM:The lipid regulating effect of PTS was studied in rabbits fed with high lipid diet.METHOD:the rabbits were fed with high fatty diet for 30 consecutive days as hyperlipidemia model.Meanwhile they were administered with the different concentrations(25,50,100 mg/kg) of PTS. After stopping high fatty diet the rubbits were continued to adminster for 30 d. The concentration of TC (total cholesterol),TG (triglyceride),low-density lipoprotein cholesterol (LDL-C),and high-density lipoprotein cholesterol (HDL-C)in serum was determined before feeding with high fatty diet,and at the 30 th day and the 60 th day after feeding respectively .The concentration of TC,TG in live,ApoA1,ApoB,MDA (malondialdehyde) and activity of SOD were determined at the 60 th day after feeding.RESULT: After 30d and 60d the concentration of TC,TG,LDL-C and MDA in serum,and TC,TG in liver increased,but HDL-C and the activity of SOD in serum decreased. PTS can decrease the serum TC, TG, LDL-C, ApoB and MDA,but can increase the serum HDL-C, ApoA1 and activity of SOD. CONCLUSION: The rabbit hyperlipidemia model was established after feeding with high fatty diet for 30 days; The results suggested that PTS could decrease the concentration of TC,TG, LDL-C,MDA in serum and TC, TG in liver,but increase the concentration of HDL-C and the activity of SOD in serum. All of these have interrelation with the dosage. ApoA1 increases with the HDL-C increase, ApoB decreases with the LDL-C decrease.
