This study aimed at evaluating how encapsulation in a regular nanocarrier (NC) (providing extended circulation time) or in a brain-targeting NC (providing prolonged circulation time and increased brain uptake) may influence the therapeutic index compared with the unformulated drug and to explore the key parameters affecting therapeutic performance using a model-based approach. Pharmacokinetic (PK) models were built with chosen PK parameters. For a scenario where central effect depends on area under the unbound brain concentration curve and peripheral toxicity relates to peak unbound plasma concentration, dose-effect and drug-side effect curves were constructed, and the therapeutic index was evaluated. Regular NC improved the therapeutic index compared with the unformulated drug due to reduced peripheral toxicity, while brain-targeting NC enhanced the therapeutic index by lowering peripheral toxicity and increasing central effect. Decreasing drug release rate or systemic clearance of NC with drug still encapsulated could increase the therapeutic index. Also, a drug with shorter half-life would therapeutically benefit more from a NC encapsulation. This work provides insights into how a NC for brain delivery should be optimized to maximize the therapeutic performance and is helpful to predict if and to what extent a drug with certain PK properties would obtain therapeutic benefit from nanoencapsulation.  相似文献

目的 针对中医药大数据平台传统的加密方案效率不高的问题,提出一种高效的基于属性的内积加密的数据脱敏算法。方法 Hash(哈希)算法是应用广泛的高效的数据加密方法,但传统的哈希算法基于单一的控制策略,效率不高。本文提出一种基于属性的内积加密的数据脱敏算法,把批量的敏感数据分割为不同长度数据颗粒度,与特定密文的哈希进行内积处理。结果 在面对中医药大数据平台的海量数据加密的场景,与传统的哈希加密算法相比,本文提供的加密算法具有很好的性能。结论 为了保障个人隐私数据不被泄露,中医药大数据平台中的个人医疗数据需要加密脱敏后,才能进行分析处理或对外发布。本文提出的算法具备灵活的数据颗粒度、策略和高效的性能表现,适用于海量的中医药数据脱敏。  相似文献

ObjectiveMethamphetamine is used extensively around the world as a psychostimulant. The complications related to methamphetamine include methamphetamine-induced neurotoxicity, mainly involving intraneuronal processes, such as oxidative stress and excitotoxicity. Curcumin is effective against neuronal injury due to its antioxidant, anti-inflammatory effects. In this study, we examined the protective effects of curcumin against methamphetamine neurotoxicity.MethodsSixty male Wistar rats were divided into the following groups: control (n = 12), DMSO (n = 12), methamphetamine (n = 12), and methamphetamine + curcumin (100 and 200 mg/kg, respectively, intraperitoneal [IP]; n = 12). Neurotoxicity was induced by 40 mg/kg of methamphetamine administrated through 4 injections (4 × 10 mg/kg, q2h, IP). Curcumin (100 and 200 mg/kg) was administered at 7 days after the last methamphetamine injection. By using a Morris water maze task, the hippocampus-dependent memory and spatial learning were evaluated 1 day after the last curcumin injection. Then, the animal brains were isolated for biochemical measurements, as well as glial fibrillary acidic protein (GFAP), ionized calcium-binding adaptor protein-1(Iba-1) and caspase-3 immunohistochemical staining.ResultsThe current study demonstrated that administration of curcumin significantly attenuates spatial memory impairment (P < 0.01) following methamphetamine neurotoxicity. Curcumin caused a significant increase in the levels of superoxide dismutase and glutathione peroxidase (P < 0.05). However, it decreased tumor necrosis factor (TNF-α) (P < 0.05) and malondialdehyde (P < 0.01) levels as compared to the methamphetamine group. Also, curcumin significantly reduced Iba-1 (P < 0. 01), GFAP and caspase-3 positive cells in the hippocampus (P < 0.001).ConclusionCurcumin exerted neuroprotective effects on methamphetamine neurotoxicity because of its antioxidant and anti-inflammatory effect.  相似文献

中药(单体/复方)药效物质基础研究是中医药现代研究的关键问题之一。然而,目前运用的生化和分子生物学方法很难诠释其多靶点、整体性和动态性的整合调节作用。课题组提出的"证治代谢组学"假说理论指出,不同的证候存在"证相关代谢谱群"和"证相关生物标志物",这可能是中药药效物质基础所在,辨证论治后偏离的代谢网络功能呈现回归趋势。基于"证治代谢组学"假说开展中药(单体/复方)药效物质基础研究,这将为阐释中药(单体/复方)防病治病和养生保健的科学内涵及其对疾病个体的整合调节作用提供新的思路。本文以冠心病为研究载体、气阴虚血瘀证为切入点、活血保心丸为干预措施,就该研究思路的内涵与总体模式、提出的背景与依据、实践的方式与可行性、创新与特色及其研究意义等进行了简要介绍。  相似文献

IntroductionThis study evaluated free water loss–induced residual strain with and without axial compressive loading and assessed the mechanical effect of cyclic loading in fully hydrated and partially dehydrated root dentin.MethodsRoot dentin sections prepared from freshly extracted human premolars were used. Customized 3-dimensional digital image correlation was used to qualitatively and quantitatively analyze the residual strain induced by 2 hours of free water loss in different regions of root dentin. Residual strain in partially dehydrated root dentin during axial compressive loading was also analyzed using 3-dimensional digital image correlation. The effect of cyclic loading on load to fracture in fully hydrated and partially dehydrated dentin and their fractography were analyzed using micro–computed tomographic imaging.ResultsFree water loss resulted in a heterogeneous distribution of residual strain and an overall formation of residual compressive strain with areas of tensile strain localized to the root canal and outer dentin. More residual compressive strain was observed in the apical dentin compared with the cervical dentin (P < .05), and more residual shear strain was observed in outer dentin compared with inner dentin (P < .05). Axial loading resulted in an increase in the load-induced compressive strain in the direction perpendicular to dentinal tubules (P < .05). Fully hydrated roots displayed a higher mean (P < .05) and median (P < .05) number of cycles to fracture with microcracks characteristic of toughness.ConclusionsAfter free water loss, root dentin displayed an increased formation of heterogenous residual strain, which resulted in increased axial compressive load-induced strain and a decreased resistance to fatigue failure. The effect of free water loss in the loss of mechanical integrity of root-filled teeth needs further investigation.  相似文献

Influenza vaccination is considered the most valuable means to prevent and control seasonal influenza infections, which causes various clinical symptoms, ranging from mild cough and fever to even death. Among various influenza vaccine types, the inactivated subunit type is known to provide improved safety with reduced reactogenicity. However, there are some drawbacks associated with inactivated subunit type vaccines, with the main ones being its low immunogenicity and the induction of Th2-biased immune responses. In this study, we investigated the role of a single-stranded RNA (ssRNA) derived from the intergenic region in the internal ribosome entry site of the Cricket paralysis virus as an adjuvant rather than the universal vaccine for a seasonal inactivated subunit influenza vaccine. The ssRNA adjuvant stimulated not only well-balanced cellular (indicated by IgG2a, IFN-γ, IL-2, and TNF-α) and humoral (indicated by IgG1 and IL-4) immune responses but also a mucosal immune response (indicated by IgA), a key protector against respiratory virus infections. It also increases the HI titer, the surrogate marker of influenza vaccine efficacy. Furthermore, ssRNA adjuvant confers cross-protective immune responses against heterologous influenza virus infection while promoting enhanced viral clearance. Moreover, ssRNA adjuvant increases the number of memory CD4+ and CD8+ T cells, which can be expected to induce long-term immune responses. Therefore, this ssRNA-adjuvanted seasonal inactivated subunit influenza vaccine might be the best influenza vaccine generating robust humoral and cellular immune responses and conferring cross-protective and long-term immunity.  相似文献

分子生物学(molecular biology)是从分子水平阐明生命现象和本质的科学,其发展为传统生药学的研究提供了新的生物技术和方法。金银花作为常用大宗药材之一,国内外学者在深入研究传统方法的基础上,采用分子生物学手段对其展开真伪鉴别、品质评价和控制等方面的相关研究,并取得了一定成果。该文主要综述了近年来分子生物学技术方法在金银花鉴别、有效成分生物合成的分子机制以及胁迫条件下次生代谢产物积累的分子机制研究,并针对基于杂交技术的标记(RFLP)、基于PCR的分子标记(RAPD,AFLP,SSR,ISSR)和基于DNA序列分析的SNP及DNA条形码对金银花的多样性识别、诊断、鉴定等方面进行了详细的总结,同时提出可以采用多组学技术,构建系统生物学技术和平台,建立次生代谢产物生物合成的相关模型,从而更好地研究金银花活性成分生物合成的分子机制以及药用植物在环境胁迫下的相关代谢产物的合成和积累等生命活动规律并进行调控,为进一步推动金银花现代化及其他中药资源的开发利用提供支撑与参考。  相似文献

背景 现有研究表明,抑郁是影响大学生心理健康的重要原因之一。因此,有必要采取有效的方法干预大学生抑郁情绪。目的 探究循经冥想训练干预大学生抑郁情绪的效果。方法 2018年4月,在以循经冥想课题组的名义招募的某大学本科生中,发放抑郁自评量表(SDS),筛选出自愿接受循经冥想训练的60人。根据受试者性别比例,采用随机数字表法随机分为循冥组和静息组。其中循冥组30人进行4周循经冥想训练,在开始循经冥想训练前和循经冥想训练结束后填写SDS并进行E-prime情绪图片评价任务。静息组30人进行静息训练,其余与循经冥想组相同。比较两组前测、后测及前后测差值有无差异。结果 两组前测积极、中性、消极情绪图片评分比较,差异均无统计学意义(P>0.05);两组后测积极、中性、消极情绪图片评分比较,差异均无统计学意义(P>0.05);两组积极、中性、消极情绪图片评分前测与后测差值比较,差异均无统计学意义(P>0.05)。静息组前测与后测积极、中性、消极情绪图片评分比较,差异均无统计学意义(P>0.05)。循冥组后测积极情绪图片评分低于前测、消极情绪图片评分高于前测(P<0.05)。两组前测SDS评分比较,差异无统计学意义(P>0.05)。循冥组后测SDS评分低于静息组,SDS评分前测与后测差值高于静息组(P<0.05)。循冥组和静息组后测SDS评分均低于前测(P<0.05)。结论 循经冥想能降低大学生抑郁水平,可能是由于其能够改变人的情绪认知评价和睡眠质量,使人变得平和。  相似文献
