摘 要 目的:考察不同浓度乙醇对当归保肝活性成分超声提取的影响。方法: 以阿魏酸含量和当归多糖含量作为考察指标,采用单因素试验分别考察20%,30%,40%,50%,60%及70%梯度浓度的乙醇对当归保肝活性成分超声提取的影响。结果: 以30%乙醇为溶剂时取得了较好的效果,超声提取最佳条件为加12倍的30%乙醇,60℃超声提取4次,每次1.5 h。结论:以30%乙醇作为溶剂超声提取当归保肝活性成分最佳。 相似文献

Characterization of principal compositions in the roots of Angelica sinensis by HPLC-ESI-MSn and chemical comparison of its different parts 下载免费PDF全文
Angelicae Sinensis Radix, Danggui in Chinese, is an important traditional Chinese medicine derived from the roots of Angelica sinensis (Oliv.) Diels. HPLC-ESI-MSn was applied to analyze the chemical constituents in the methanol extract of Danggui. A total of 24 compounds were identified or tentatively characterized based on their UV and mass spectra, containing 11 phenolic compounds, 12 phthalides and 1 amino acid. Three dicaffeoylquinic acids, 5-feruoylquinic acid and ferulic acid hexoside were detected in A. sinensis for the first time. According to Chinese medicine theories, the three parts of Danggui (Guitou, Guishen, Guiwei) have different therapeutic effects and their substantial evidence has not been clearly elucidated yet. In the present study, the chemical compositions of different parts of Danggui were investigated by using the established HPLC method. The result indicated that the content of tryptophan was getting lower and the contents of low polarity constituents like phthalides trended to get higher from Guitou to Guiwei. This might partly explain the therapeutic effects of different parts of Danggui. 相似文献

目的:研究黄芪、当归单用或伍用为当归补血汤后,动物血浆中黄芪甲苷、阿魏酸药动学参数的变化规律,以探索当归补血汤配伍的合理性。方法:将黄芪、当归及当归补血汤制成水煎液,分别灌胃给予新西兰大耳白兔或SD大鼠。采用HPLC法测定给予当归补血汤或当归后兔血浆中阿魏酸的血药浓度;采用HPLC/MS/MS法测定给予当归补血汤或黄芪后大鼠血浆中黄芪甲苷的血药浓度,并计算主要药动学参数。结果:与单用当归比较,给予当归补血汤后新西兰大耳白兔血浆中阿魏酸的药动学参数几乎没有改变;与单用黄芪比较,大鼠口服当归补血汤后血浆中黄芪甲苷的达峰时间提前一倍[tmax由(4.00±1.54)h到(2.00±0.95)h],达峰浓度提高一倍[Cmax由(60±7)μg/L到(146±27)μg/L]。结论:合用后当归增强了黄芪中黄芪甲苷在体内的活力,提前了黄芪甲苷发挥功效的进程,体现了当归臣药的辅佐之功。以上结果从药动学角度验证了当归补血汤的配伍合理性。 相似文献

目的建立一种快速可靠测定当归、川芎中活性成份阿魏酸松柏酯的HPLC方法。方法色谱条件:ZorbaxODSC18色谱柱(250mm×4.6mmID,μm),流动相采用1%醋酸与乙腈1:1等度洗脱,检测波长318nm。结果建立的HPLC方法日内和日间精密度分别为0.22%–1.16%和0.86%–2.62%,阿魏酸松柏酯在0.380–0.038mg·mL^–1范围内线形关系良好(r^2=0.9995),加样回收率为105.3%,RSD为2.7%。结论该方法能够准确、快速定量测定当归、川芎中阿魏酸松柏酯的含量,有利于提高对当归、川芎的质量控制。 相似文献

The screening and analysis of bioactive components in traditional Chinese medicines (TCMs) is very important not only for the quality control of crude drugs but also for elucidating the therapeutic principle. In this study, a method for screening potential active components from TCMs was developed by using biomembrane extraction and high performance liquid chromatography. Based on the methodology, aqueous extract of Angelica sinensis (WEAS) was used, and four compounds were detected by HPLC in the desorption eluate of red cell membrane extraction for WEAS. The compounds were identified as ferulic acid, ligustilide, senkyunolide H and senkyunolide I based on their UV, MS and NMR spectra. Actually, ferulic acid and ligustilide are considered as major active components in Angelica sinensis. Therefore, this method may be applied to predict the potential bioactivities of multiple compounds in TCMs simultaneously. 相似文献

目的建立当归养血丸中阿魏酸含量测定方法。方法采用高效液相色谱法。色谱条件:色谱柱为HypersilC18柱(250mm×4.6mm,5μm);流动相:甲醇-1%冰乙酸溶液(25∶75);柱温:25°C;流速:1.0ml/min;检测波长为320nm;理论塔板数按阿魏酸峰计算不低于3000。结果阿魏酸在0.03~0.48μg范围内线性关系良好,r=0.9997。阿魏酸平均回收率为98.90%,RSD为0.94%。结论该方法操作简单、准确、重复性好,可用于当归养血丸的质量控制。 相似文献
