Objective: To investigate whether ginsenoside-Rb1(Gs-Rb1) improves the CoCl2-induced autophagy of cardiomyocytes via upregulation of adenosine 5'-monophosphate-activated protein kinase(AMPK) pathway. Methods: Ventricles from 1-to 3-day-old Wistar rats were sequentially digested, separated and incubated in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum for 3 days followed by synchronization. Neonatal rat cardiomyocytes were randomly divided into 7 groups: control group(normal level oxygen), hypoxia group(500 μmol/L CoCl_2), Gs-Rb1 group(200 μmol/L Gs-Rb1 + 500 μmol/L CoCl_2), Ara A group(500 μmol/L Ara A + 500 μmol/L CoCl_2), Ara A+ Gs-Rb1 group(500 μmol/L Ara A + 200 μmol/L Gs-Rb1 + 500 μmol/L CoCl_2), AICAR group [1 mmol/L 5-aminoimidazole-4-carboxamide ribonucleotide(AICAR) + 500 μmol/L CoCl_2], and AICAR+Gs-Rb1 group(1 mmol/L AICAR + 200 μmol/L Gs-Rb1 + 500 μmol/L CoCl_2). Cel s were treated for 12 h and cell viability was determined by methylthiazolyldiphenyl-tetrazolium bromide(MTT) assay and cardiac troponin I(cTnI) levels were detected by enzyme-linked immunosorbent assay(ELISA). AMPK activity was assessed by 2',7'-dichlorofluorescein diacetate(DCFH-DA) ELISA assay. The protein expressions of Atg4 B, Atg5, Atg6, Atg7, microtubule-associated protein 1 A/1 B-light chain 3(LC3), P62, and active-cathepsin B were measured by Western blot. Results: Gs-Rb1 significantly improved the cell viability of hypoxia cardiomyocytes(P0.01). However, the viability of hypoxia-treated cardiomyocytes was significantly inhibited by Ara A(P0.01). Gs-Rb1 increased the AMPK activity of hypoxia-treated cardiomyocytes. The AMPK activity of hypoxia-treated cadiomyocytes was inhibited by Ara A(P0.01) and was not affected by AICAR(P=0.983). Gs-Rb1 up-regulated Atg4B, Atg5, Beclin-1, Atg7, LC3B Ⅱ, the LC3BⅡ/Ⅰ ratio and cathepsin B activity of hypoxia cardiomyocytes(P0.05), each of these protein levels was significantly enhanced by Ara A(all P0.01), but was not affected by AICAR(all P0.05). Gs-Rb1 significantly down-regulated P62 levels of hypoxic cardiomyocytes(P0.05). The P62 levels of hypoxic cardiomyocytes were inhibited by Ara A(P0.05) and were not affected by AICAR(P=0.871). Conclusion: Gs-Rb1 may improve the viability of hypoxia cardiomyocytes by ameliorating cell autophagy via the upregulation of AMPK pathway.  相似文献

目的 探讨山竹提取物对人肝癌细胞HepG2增殖及凋亡的影响及其作用机制.方法 采用MTT法检测不同浓度山竹提取物对HepG2细胞增殖的影响;分别应用Annexin-V/PI双重染色、PI单染法检测山竹提取物对HepG2凋亡和细胞周期的影响;天冬氨酸特异性半胱氨酸蛋白酶-3(Caspase-3)试剂盒检测山竹提取物对HepG2细胞Caspase-3酶活化的影响.结果 不同浓度(100 μmol/L、200μmol/L、400 μmol/L、600 μmol/L、800 μmol/L)山竹提取物均可抑制人肝癌细胞HepG2的增殖活性,且随着山竹提取物浓度及其作用时间的增加,细胞抑制率升高(P<0.05);山竹提取物浓度为256.67 μmol/L时可诱导人肝癌细胞HepG2出现早期凋亡,并且随着山竹提取物作用时间的增加,凋亡早期癌细胞的比率增高.细胞周期分析结果显示随着山竹提取物的浓度升高(0 μmol/L、200 μmol/L、400 μmol/L、600 μmol/L、800 μmol/L),G1期HepG2细胞比例升高,而S期细胞比例下降(P<0.05).与对照组(0 μmol/L)比较,各浓度(200 μmol/L、400 μmol/L、600 μmol/L、800 μmol/L)山竹提取物组HepG2细胞的Caspase-3酶活性均升高(P<0.05),给药组的酶活力单位随给药浓度的增加而明显增加(P<0.05).结论 山竹提取物对人肝癌细胞HepG2有抑制增殖和促进凋亡的作用,其作用机制可能与抑制HepG2细胞进入S期和激活Caspase-3有关.  相似文献

目的:研究三氧化二砷(As2O3)对人恶性黑色素瘤A375细胞株凋亡的诱导作用.方法:采用流式细胞术(FCM)检测不同浓度的三氧化二砷诱导A375细胞的早期凋亡率和晚期凋亡率.结果:As2O3浓度分别为5 μmol/L、10μmol/L、20 μmol/L时,A375细胞的早期凋亡率分别为9.75%、50.9%和43.7%,晚期凋亡率分别为2.10%、4.61%和11.2%,总凋亡率分别为11.85%、55.51%和54.9%.结论:As2O3可诱导A375细胞早期凋亡和晚期凋亡,10 μmol/L和20μmol/L的As2O3可明显增加A375细胞总凋亡率.  相似文献

目的:探讨铁调节蛋白1(IRP1)及铁调节蛋白2(IRP2)在芒果甙保护柔红霉素致大鼠心肌细胞毒性作用中的表达及其意义.方法:以4 μmol/L柔红霉素单用或联合应用25~200 μmol/L芒果甙为处理因素作用于大鼠心肌细胞24,48 h及72 h,应用qRT-PCR法检测各组心肌细胞IRP1、IRP2 mRNA表达.结果:25~200 μmol/L 芒果甙模型组在干预24 h后IRP1、IRP2表达水平较柔红霉素组(4 μmol/L)降低,干预48,72 h后,二者表达较柔红霉素组升高.结论:芒果甙能改变柔红霉素致大鼠心肌毒细胞中铁调节蛋白的表达水平,对细胞内铁代谢产生影响.  相似文献

OBJECTIVE: Saikokeishito(TJ-10) is a Kampo(traditional Japanese herbal) medicine, clinically used for hundreds of years in East Asia. Among its various mechanisms elucidated so far, TJ-10 inhibits the production of transforming growth factor-β1(TGF-β1) and development of pancreatic fi brosis in vivo. Oxidative damage of normal human dermal fi broblasts(NHDFs) in the corium is a cause of human dermal senescence. Our aim was to determine whether TJ-10 protects NHDFs from premature senescence by hydrogen peroxide(H2O2). METHODS: Premature senescence was induced in NHDFs by 200 μmol/L H2O2 for 4 h. Cell viability and the expressions of p53, AMP-activated protein kinase α1(AMPKα1), AMPKα2, and 14-3-3 protein sigma(14-3-3 σ) were measured in NHDFs treated with TJ-10 for 48 h before exposure to H2O2 for 4 h. RESULTS: Cell viability after treatment with 200 μmol/L H2O2 for 4 h was similar(about 80%) to after pre-treatment with TJ-10. Ascorbic acid as a control did not protect NHDFs from damage by 200 μmol/L H2O2. Treatment with 200 μmol/L H2O2 tended to up-regulate p53 and to down-regulate SIRT1 and AMPKα1, but had no effect on AMPKα2 and 14-3-3 σ expression. Pretreatment with TJ-10 inhibited H2O2-induced up-regulation of p53 and enhanced AMPKα1 expression. CONCLUSION: It is suggested that Saikokeishito has a protective effect on oxidative stressinduced senescence of NHDFs.  相似文献

目的 研究二苯乙烯苷(TSG)对过氧化氢( H2O2)诱导损伤的人脐静脉内皮细胞单核细胞趋化蛋白1( MCP-1)表达的影响. 方法 体外培养人脐静脉内皮细胞,分为空白对照组、H2O2组(200 μmol/L H2O2)、辛伐他汀组(5 μmol/L辛伐他汀+200 μmol/L H2O2)、TSG组(1μmol/L TSG +200 μmol/L H2O2),按分组处理24h细胞后,运用逆转录聚合酶链式反应和酶联免疫吸附试验分别检测MCP-1 mRNA与其蛋白的表达. 结果 200 μmol/L的H2O2作用内皮细胞24h后,MCP-1的mRNA和蛋白水平均较空白对照组显著升高,P<0.01.TSG预处理细胞后,MCP-1的mRNA和蛋白水平较H2O2组降低,差异有显著性(P<0.05). 结论 TSG能抑制H2O2诱导损伤的人脐静脉内皮细胞MCP-1的表达.  相似文献

目的 探讨尼莫地平对H2O2引起的血管内皮细胞凋亡的影响.方法 以体外培养的人脐静脉内皮细胞为研究对象,通过TUNEL法、流式细胞术来检测H2O2对血管内皮细胞凋亡的影响以及尼莫地平的干预作用.结果 H2O2 (100 μmoI/L、200 μmol/L、400 μmol/L) 明显增加内皮细胞的凋亡率,而尼莫地平(1.2 μg/ml)能拮抗H2O2(200 μmol/L)引起内皮细胞的凋亡作用.结论 尼莫地平对H2O2引起的血管内皮细胞凋亡有拮抗作用.  相似文献

目的:探讨氯化锰(MnCl2)对SD大鼠体外培养睾丸间质细胞(Leydig cells)睾酮合成的影响.方法:建立SD大鼠睾丸Leydig细胞体外原代培养模型.MnCl2染毒剂量组分为对照(0 μmol/L)、1.0 μmol/L、2.5 μmol/L、5.0 μmol/L、10.0 μmol/L、25.0 μmol/L、 50.0 μmol/L、100.0 μmol/L,培养24 h后,采用台盼蓝染色法测定细胞存活率.同法分组,采用放射免疫法检测加入人绒毛膜促性腺激素(HCG)与不加HCG条件下,上述剂量MnCl2对Leydig细胞合成睾酮的影响.结果:随着MnCl2染毒剂量的升高,Leydig细胞存活率逐渐下降.MnCl2剂量≥5.0 μmol/L时,细胞存活率均低于对照组(P<0.05).基础状态下(不加HCG)MnCl2剂量≥5.0 μmol/L时睾酮水平均低于对照组(P<0.05);HCG刺激状态下MnCl2剂量≥10.0 μmol/L时睾酮水平低于对照组(P<0.05).结论:锰可以直接影响原代培养Leydig细胞睾酮的合成.  相似文献

近年新生儿、婴儿、成人麻疹患者逐年增加,临床表现一般仍较典型,成年人麻疹患者全身中毒症状较重。麻疹抗体检测结果阳性是主要的诊断依据。麻疹发病的双相移位的机理可能是,免疫保护力不足,婴儿出生时麻疹抗体力低。孕期母传胎的麻疹抗体减弱,母经乳汁传给婴儿的抗体减弱,成人麻疹抗体水平逐年下降。预防措施是怀孕前给予育龄妇女麻疹疫苗接种,鼓励母乳喂养,麻疹疫苗计划免疫适当提前,在成人追加麻疹疫苗的免疫,加强病毒变异的研究等。  相似文献

目的解决腰椎间盘突出症手术中神经压迫。方法对1980~1998年再手术资料进行统计分析,讨论分析再手术原因,再次手术前影像学检查,观察病理变化以确定再手术方法。结果对11例随访6个月~1年,优7例(68.4%),良3例(36.8%),差1例(2.8%)。结论初次手术前详细查体和分析X线片,术中用导尿管和神经剥离探查,尽量避免髓核遗留,手术范围不宜太大,尽量减少对软组织和脊柱结构的破坏,避免形成硬膜囊与神经根粘连而致单纯形疤痕。  相似文献

目的 探讨降钙素原变化率的测定对细菌性肺炎的评估价值.方法 选取住院治疗的细菌性肺炎患者72例,根据治疗效果分为治疗有效组(有效组)47例和治疗无效组(无效组)25例,对治疗有效组和无效组中的降钙素酶原变化及影响肺炎治疗效果的危险因素进行分析.结果 有效组治疗后3天及治疗后7天的降钙素酶原显著低于无效组的患者(P<0.05);年龄>65岁、心功能≥3级、COPD、糖尿病、肺部双侧受累(X线示)、菌血症、感染性休克和3天内降钙素酶原变化率<30%是影响细菌性肺炎治疗效果的危险因素;感染性休克、肺部双侧受累、3天内降钙素酶原变化率<30%及心功能≥3级是影响细菌性肺炎治疗效果的独立危险因素.结论 测定降钙素原变化率对细菌性肺炎的疗效评估具有重要临床意义,可在临床推广应用.  相似文献

以^3氢-胸腺嘧啶核苷放射自显影法及HE染色,观察并分别测定了18例正常子宫内膜增殖中期,15例增殖晚期的腺上皮细胞或间质细胞的标记指数、分裂指数。结果显示:子宫内膜增殖晚期腺上皮细胞或间质细胞之LI均明显高于增殖中期。同时,增殖晚间质细胞之MI也明显高于增殖中期,即此两种细胞在增殖晚期中增生明显,其增生状态初步获得了定位定量测定的正常值。  相似文献
