Objective To investigate the role of opioid receptors in the protective effects of isoflurane-induced delayed preconditioning against myocardial ischemia-reperfusion (I/R) injury in rabbits. Methods Forty male New Zealand white rabbits weighing 2.0-2.5 kg were randomly assigned into 4 groups ( n = 10 each) : group I sham operation (S); group II I/R; group Ⅲ isoflurane + I/R (Iso) and group IV Iso + naloxone + I/R (Nal). Myocardial I/R was induced by 40 min occlusion of left anterior descending branch (LAD) of coronary artery followed by 120 min reperfusion. In group Ⅲ (Iso) 2% isoflurane in 100% O2 was inhaled for 2 h and I/R was produced 24 h later. In group IV (Nal) naloxone 6 mg/kg was given iv 10 min before 2 h of 2% isoflurane inhalation and I/R was produced 24 h later. At the end of 120 min reperfusion, infarct size (IS) and area at risk (AAR) were determined by Evan's blue and TTC staining. Myocardial ultrastructure was examined by electron microscopy. The phosphorylated p38MAPK protein expression in myocardium was determined by Western blot. Results The IS was significantly smaller in group Iso ( Ⅲ ) ( 19.7% ± 2.8%) than in I/R group ( II ) (37.8% ±1.7%) (P<0.05). The phosphorylated p38MAPK protein expression in myocardium was significantly lower in group Iso than in group I/R. Microscopic examination showed less myocardial damage in Iso group than in group I/R. The protective effects of delayed preconditioning by isoflurane was prevented by naloxone pretreatment. ConclusionOpioid receptors may be involved in the protective effects of delayed preconditioning by isoflurane against myocardial I/R injury.  相似文献

Objective To investigate the role of cerebral opioid receptors in the protective effects of intracerebro-ventricular (ICV) morphine preconditioning (MPC) against myocardial ischemia-reperfusion (I/R) injury in rats. Methods Male SD rats weighing 320-370 g were used in this study. A needle was inserted through a surgically created hole into the cerebro-ventricle using a stereotactic instrument and fixed. Sixty male SD rats in which ICV needle was successfully placed without complication were randomly divided into 10 groups of 6 animals each. In group I sham operation was performed (S). In group]] myocardial I/R was produced (I/R) . In group Ⅲ (ischemic preconditioning), the animals were subjected to 3 episodes of 5 min myocardial ischemia at 5 min intervals before ischemia (IPC) . In group IV morphine was given ICV in 3 repeated doses of 1 μg/kg at 5 min intervals before ischemia (MPC). Three types of opioid receptor antagonists -nor-binaltorphimine (nor-BNI) (κ receptor antagonist), D-Phe-Cys-Tyr-D-Trp-Orn-Thr-Pen-Thr-NH2(CTOP) (μ receptor antagonist) and naltrindole (NTD) (S receptor antagonist) 15 nmol were given ICV in group V . VI and VIⅡI respectively at 10 min before MPC. In group VIII,IX and X , nor-BNI, CTOP and NTD 15 nmol were given ICV respectively at 40 min before ischemia. Myocardial I/R was produced by occlusion of left anterior descending branch of coronary artery for 30 min followed by 120 min reperfusion. At the end of 120 min, femoral venous blood samples were taken for determination of lactate dehydrogenase ( LDH) and creatine kinase (CK) activities and calcitonin gene-related peptide (CGRP) concentration. The animals were then killed and hearts removed for measurement of area at risk (AAR) and infarct area (IA) . IA/AAR ratio was calculated. Results The size of IA was smaller and IA/AAR ratio lower and significantly less LDH and CK and more CGRP were released in group IPC and MPC ( group Ⅲ and IV) than in group I/R (group II ) . The protective effects of MPC were abolished by pretreatment with nor-BNI, CTOP and NTD. Conclusion Cerebral μ, k and δ opioid receptors are involved in the protective effects of ICV morphine preconditioning against myocardial I/R injury through CGRP released from peptidergic nerve fibers of heart.  相似文献

目的 评价杭州健康女性骨超声速度(SOS)值随增龄减少和骨质疏松患病率,建立杭州地区女性骨超声速度值参考数据库。方法 定量超声法测定1208例杭州地区健康女性桡骨远端(RAD),第3指骨近节(PLX),第V跖骨(MTR)和胫骨中段(TIB)的超声速度值。结果 RAD、PLX、MTR和TIBSOS峰值(Peak of SOS)均出现在40-45岁,TJB的SOS峰值出现在35—40岁,此后随年龄增长而下降。绝经后妇女在绝经后早期和晚期各有1个SOS快速减少期,前见于桡骨近端,平均年减少率为2.4%,后见于胫骨中段,平均年减少率为1.8%。各部位骨SOS累积减少率随年龄增长而增加,到85岁4部位累积减少为13%-18%。60岁以后骨质疏松性症(OP)检出率为45%-70%,OP检出率以桡骨远端最高,60-70岁平均为67%,第3指骨近端次之约50%,胫骨中段最低为36%;75岁以后分别为70%,65%和45%。结论 全身各部位骨超声速度值到达峰值的年龄不同,峰值也各有差异。绝经后妇女骨超声速度值随年龄增加减少较快,应予激素和补钙治疗,桡骨远端为本地区SOS检测和OP检出的敏感部位。  相似文献

目的 评价脊髓胶质细胞在大鼠炎性痛形成中的作用.方法 清洁Ⅱ级成年雄性SD大鼠,体重180~220 g,取蛛网膜下腔置管成功的大鼠65只,随机分为5组(n=13),生理盐水组(NS组):右后肢踝关节外侧皮下注射NS 50μl;炎性痛组(IP组):采用右后肢踝关节外侧皮下注射完全弗氏佐剂50μl的方法制备炎性痛模型;氟代柠檬酸组(FC组):经蛛网膜下腔导管注射FC 1 nmol/10 μl,15 min后右后肢踝关节外侧皮下注射NS 50 μl;NS+IP组:经蛛网膜下腔导管注射NS 10 μl,15 min后制备炎性痛模型;FC+IP组:经蛛网膜下腔导管注射FC 1 nmol/10 μ,15 min后制备炎性痛模型.于模型制备前2 d(T_0)、皮下注射药物前(T_1)和注射药物后2、4、6、8、10、12、24、26 h(T_(2~9))时测定机械缩足阈值(MWT)和热缩足潜伏期(TWL).皮下注射药物后8 h时采用免疫组化法测定脊髓背角星形胶质细胞标记物(GFAP)和小胶质细胞标记物(OX-42)的表达水平.结果 与NS组比较,IP组和NS+IP组T_(3~9)时MWT和TWL降低,FC+IP组T_(3~9)时MWT降低,T_(8,9)时TWL降低,IP组、NS+EP组和FC+EP组脊髓GFAP和OX-42的表达水平均上调(P<0.05);与IP组比较,FC组T_(3~9)时MWT和TWL升高,FC+IP组T_(3~7)时MWT和TWL升高,2组脊髓GFAP和OX-42的表达水平均下调(P<0.05或0.01).结论 脊髓胶质细胞的活化参与了大鼠炎性痛的形成.  相似文献

目的 评价中脑导水管周围灰质小胶质细胞活化在大鼠神经病理性痛中的作用.方法 雄性SD大鼠176只,体重200 ~ 250 g,9周龄,采用随机数字法,将其分为4组:假手术组(S组,n=40)、神经病理性痛组(NP组,n=40)、生理盐水组(NS组,n=48)和米诺环素组(M组,n=48).NP组、NS组和M组采用慢性坐骨神经缩窄性损伤法制备大鼠神经病理性痛模型;S组仅暴露坐骨神经,而不结扎.术后第7天时,NS组和M组分别于中脑导水管周围灰质的腹外侧区注射生理盐水或米诺环素0.5μl.取8只大鼠,分别于术前1 d(T0)、术后第3天(T1)、第7天给药前30 min(T2)、第7天给药后30 min(T3)、第14天(T4)和第21天(T5)时测定机械痛阈.于T1-5时各处死8只大鼠,取脑组织,行小胶质细胞计数.结果 与S组比较,NP组、NS组和M组T1-5时机械痛阈降低,小胶质细胞计数升高(P<0.05);NP组和NS组各时点机械痛阈和小胶质细胞计数差异无统计学意义(P>0.05);与NP组和NS组比较,M组T3时机械痛阈升高,小胶质细胞计数降低(P<0.05).结论 中脑导水管周围灰质小胶质细胞的活化参与了大鼠神经病理性痛中的形成与维持.  相似文献

目的 评价脊髓胶质细胞在小鼠骨癌痛形成中的作用.方法 健康雄性C3H/He小鼠40只,周龄8~10周,体重18~22 g,随机分为4组(n=10):假手术组(S组)、骨癌痛组(B组)、PBS组(P组)和米诺环素组(M组).S组跟骨骨髓腔内注射PBS 10 μl;余3组跟骨骨髓腔内注射含2×105个骨纤维肉瘤细胞的PBS 10 μl制备骨癌痛模型,于造模前即刻开始PBS组鞘内注射PBS 5μl,M组鞘内注射米诺环素(用PBS溶解为0.2 mmol/L)5μl,1次/d,连续11 d.于造模前1 d、造模后即刻、3、5、7、9、11 d时测定机械痛阈;于造模后3、7、9、11 d机械痛阈测定结束后测定冷痛阈.痛阈测定结束后处死小鼠,取脊髓组织,测定神经胶质纤维酸性蛋白(GFAP)和CD11b的表达水平.结果 与S组比较,B组和P组造模后3-11 d时、M组造模后3、5 d时机械痛阈升高,B组、P组和M组造模后7~11 d时冷痛阈升高,脊髓CD11b和GFAP表达上调(P<0.05).与B组比较,M组造模后3-11 d时机械痛阈降低,造模后7-11 d时冷痛阈降低,脊髓CD11b和GFAP表达下调(P<0.05).结论 脊髓胶质细胞(星形胶质细胞和小胶质细胞)的激活参与了小鼠骨癌痛的形成.  相似文献

为找出沈阳地区髋部骨折发生男性多于女性的原因,探索该病在不发达国家或地区的流行特点,我们再次通过查阅病例记录,对沈阳市1994年50岁以上人口的部分髋部骨折病发生的原因进行了较详细的调查分析。共调查分析266髋部骨折病例,其中男163例,女103例。损伤原因记为单纯摔倒(滑倒或绊倒)、骑自行车摔倒、自行车撞倒、机动车事故和高位跌下(滚楼梯或从较高位置掉下)。结果表明:男女在髋部骨折伤因构成上有差别(P=0.004)。女性髋部骨折的大多数(70%)是由单纯摔倒引起,而在男性则不足一半(49%),即男性髋部骨折的一半以上不是由于单纯摔倒而是由各种意外事故造成的(P=0.0008)。在各种意外事故中,男性骑自行车摔倒引起骨折的频率(28%)明显高于女性(10%)。除了骑自行车摔倒外,男性由自行车撞倒和高位跌下引起骨折的频率稍高于女性,但无太大差别。机动车事故造成骨折的频率男女基本一致。此结果在一定的程度上说明,1994年沈阳50岁以上的男性髋部骨折发病率高是由于男性发生的各种意外事故多,尤其是骑自行车引起的事故造成的。  相似文献

目的:探讨脊柱侧凸患者顶椎区椎间盘纤维环中多聚蛋白多糖的表达及其意义。方法:采集2003年7月至2004年9月间行前路松解或矫形手术的40例脊柱侧凸患者侧凸顶椎区椎间盘组织。青少年特发性脊柱侧凸患者(adolescent idiopathic scoliosis,AIS)25例,胸椎椎间盘11例,腰椎椎间盘14例;先天性脊柱侧凸患者(congenital scoliosis,CS)15例,胸椎椎间盘6例,腰椎椎间盘9例。利用RT—PCR扩增多聚蛋白多糖(Aguecan),琼脂糖凝胶电泳,在UVP(紫外光测定法)成像系统进行扫描.GelWork图像分析系统中进行灰度测定半定量分析。分别计算AIS组和CS组凹侧、凸侧纤维环中Aggrecan含量,并对CS组和AIS组胸椎和腰椎以及椎间盘的凹侧和凸侧进行比较。结果:AIS组和CS组椎间盘凹侧纤维环中Aggrecan含量低于凸侧.差异有显著性(P〈0.01),胸椎椎间盘纤维环中Aggrecan的含量低于腰椎,但无统计学差异。AIS组Aggrecan的含量和CS组相应部位Aggrecan的含量无明显差别。结论:AIS椎间盘纤维环凹凸侧存在Aggrecan代谢差异.并且可能是脊柱侧凸所致的继发改变.但也可能是脊柱侧凸发生、发展中的重要因素。  相似文献

目的 在现有的研究生培训体系中,术前访视能力缺少有效的评估方法。本研究拟评价标准化患者(standardized patients, SP)情景模拟在术前访视能力评估中的可行性和有效性。方法 将SP情景模拟整合到多站式考核的术前访视考核站点中,对我院第二、第三年麻醉临床型研究生进行考核。考核过程中由SP和2名现场考官分别对考生进行评分,评分方法采用一个核查表评估术前访视的内容,采用一个量表评估病史采集技巧、交流技巧及人文素养的能力,同时由现场考官对SP的表现进行打分。考核结束后,另一名考官通过回顾视频对考生和SP的表现进行打分。SP与考官打分采用Pearson相关性分析。结果 在量表评估中,病史采集技巧评分(r=0.701,P<0.001)、交流技巧评分(r=0.752,P<0.001)和人文素养评分(r=0.595,P<0.001)在SP与考官之间均存在良好的相关性。在核查表评估中,SP评分与考官评分也具有良好的相关性(r=0.718,P<0.001)。结论 标准化患者情景模拟能够为麻醉临床型研究生术前访视过程中非技术技能的考核提供一种直观可靠的方法。  相似文献

目的 探讨脊髓背角卡配因在大鼠足底炎性痛形成中的作用.方法 雄性SD大鼠48只,6周龄,体重160~200 g,采用随机数字表法,将其随机分为3组:正常对照组(C组,n=8)、PBS组(n=16)和酵母多糖诱发足底炎性痛组(Z组,n=24).Z组于大鼠左侧后足足底皮下注射酵母多糖1.25 mg,制备酵母多糖诱发足底炎性痛模型,PBS组给予等容量PBS 100μl.分别于给药前(T0)、给药后30 min(T1)、1 h(T2)、2 h(T3)、4 h(T4)、8 h(T5)、24 h(T6)和48 h(T7)时测定左侧后足机械刺激缩足阈值(MWT)、热缩足反应潜伏期(PWTL)和左侧后足足底最大厚度.PBS组于T4时处死8只大鼠,Z组分别于T4、T6和T7时各处死8只大鼠,取左侧脊髓L4~6节段,采用Western blot法测定脊髓背角spectrin αⅡ降解产物、IκBα、环氧化酶-2(COX-2)的表达和NF-κB活性.结果 与C组比较,Z组MWT降低,PWTL缩短,足底最大厚度增厚,脊髓背角spectrin αⅡ降解产物和COX-2的表达上调,IκBα表达下调,NF-κB活性升高(P<0.05或0.01),PBS组上述指标差异无统计学意义(P>0.05).结论 脊髓背角卡配因活化参与了大鼠足底炎性痛的形成,其机制与激活NF-κB,上调COX-2表达有关.  相似文献
