冠心病不同类型间斑块稳定性相关指标的比较研究   总被引:10,自引:0,他引:10  
目的 比较血清抗氧化低密度脂蛋白 (oxLDL)抗体、细胞间黏附分子 1(ICAM 1)、血管细胞黏附分子 1(VCAM 1)和E 选择素水平对不同类型冠心病斑块稳定性的评价意义。方法 用酶联免疫吸附法 (ELISA)检测了 10 0例冠心病患者 (17例急性心肌梗死 ,4 1例不稳定性心绞痛 ,4 2例稳定性心绞痛 )及 31例正常健康对照者血清ICAM 1、VCAM 1、E 选择素和血清抗oxLDL抗体水平 ,并比较上述各指标水平与不同类型冠心病斑块稳定性之间的关系。结果 血清ICAM 1水平在急性心肌梗死组 (76 4± 111)ng/ml和不稳定性心绞痛组 (70 9± 10 0 )ng/ml明显高于稳定性心绞痛组 (6 0 7± 83)ng/ml,和正常对照组 (6 0 3± 90 )ng/ml,P <0 0 1;血清VCAM 1水平在急性心肌梗死组 (185 5± 6 6 6 )ng/ml和不稳定性心绞痛组 (172 4± 5 5 5 )ng/ml明显高于稳定性心绞痛组 (136 0± 36 0 )ng/ml,和正常对照组 (10 39± 319)ng/ml,P <0 0 1;血清E 选择素水平在急性心肌梗死组 (5 4± 19)ng/ml和不稳定性心绞痛组 (5 3± 2 2 )ng/ml明显高于稳定性心绞痛组 (39± 19)ng/ml,和正常对照组 (38± 14 )ng/ml,P <0 0 5 ;血清抗oxLDL抗体水平在急性心肌梗死组 (1 39± 0 6 8)和不稳定性心绞痛组 (1 35± 0 6 2 )明显高于稳定性心绞痛组 (0  相似文献

目的 探讨高血压病 (EH)患者心肌纤维化的无创性检测指标。方法 采用放免法测定 30例正常人和 6 0例EH患者 (伴心肌肥厚者 35例和不伴心肌肥厚者 2 5例 )血清Ⅰ型前胶原 (PCⅠ )和Ⅲ型前胶原 (PCⅢ )的浓度 ,用M型超声测算左室重量指数 (LVMI) ,用多普勒超声测定二尖瓣口舒张早期和晚期最大血流速度 (VE 和VA)。结果 ①EH患者血清PCⅠ和PCⅢ均明显高于正常对照组 [(4 6 6 5± 11 0 1)μg/Lvs (34 31± 5 91) μg/L ,P <0 0 5和 (146 0 0± 2 9 35 ) μg/Lvs (96 2 4± 2 1 18) μg/L ,P <0 0 1]。②EH非左室肥厚组血清PCⅢ浓度明显高于正常对照组 [(12 0 6 2± 16 2 3) μg/Lvs(96 2 4± 2 1 18) μg/L ,P <0 0 1],而血清PCⅠ与正常对照组无差别 [(33 73± 6 83) μg/Lvs (34 31± 5 91) μg/L ,P >0 5 ];EH左室肥厚组血清PCⅢ高于非肥厚组 [(16 4 14±2 2 2 8) μg/Lvs (12 0 6 2± 16 2 3) μg/L ,P <0 0 1]和正常对照组 [(16 4 14± 2 2 2 8) μg/Lvs (96 2 4± 2 1 18) μg/L ,P <0 0 1];血清PCⅠ亦高于非肥厚组 [(5 5 88± 12 86 ) μg/Lvs(33 73± 6 83) μg/L ,P <0 0 1]和正常对照组 [(5 5 88±12 86 ) μg/Lvs (34 31± 5 91) μg/L ,P <0 0 1]。③血清PCⅢ与V  相似文献

目的探讨血清可溶性血管细胞黏附分子-1(sVCAM-1)与2型糖尿病及其并发症的关系.方法采用酶联免疫吸附法测定61例合并高血压和微血管并发症的糖尿病患者及36例正常人血清中sVCAM-1水平.结果2型糖尿病患者的sVCAM-1水平[(973.25±73.38)μg/L]明显高于非糖尿病患者[(230.45±12.47)μg/L],P<0.001.合并高血压患者sVCAM-1[(1263.99±87.00)μg/L]较微血管病变者[(726.57±95.40)μg/L]增高更明显(P<0.001).结论sVCAM-1参与了糖尿病血管并发症的发生和发展.  相似文献

目的 检测急性髓性白血病 (AML)细胞与内皮细胞的黏附及细胞黏附分子 1(ICAM 1)及其配体淋巴细胞功能相关抗原 1(LFA 1)在黏附中的作用。方法 观察AML细胞与静止内皮细胞和肿瘤坏死因子α(TNFα)激活的内皮细胞的黏附 ;AML细胞与内皮细胞混合培养 2 4h的黏附 ;正常中性粒细胞与AML细胞培养上清作用 2 4h后的内皮细胞的黏附 ;流式和ELISA方法检测AML细胞培养上清作用后内皮细胞ICAM 1及可溶性ICAM 1的表达 ;并用ICAM 1和LFA 1的抗体进行阻滞黏附试验。结果 AML细胞与静止的内皮细胞黏附较少 (2 4 33± 2 87) % ,AML细胞与TNFα激活的内皮细胞的黏附 ,与内皮细胞混合培养 2 4h后的黏附以及正常中性粒细胞与AML细胞培养上清作用后内皮细胞的黏附明显增加 ,分别为 (81 87± 4 0 8) % ,(82 0 6± 7 0 5 ) % ,(83 99± 3 86 ) % (n =2 1,P <0 0 0 1) ;静止的内皮细胞ICAM 1及可溶性ICAM 1的表达分别为 (5 5 81± 4 11) %和 (0 839± 0 2 36 )μg/L ;AML细胞培养上清作用后内皮细胞ICAM 1及可溶性ICAM 1的表达明显增加 ,分别为 (6 5 36±5 97) %和 (1 4 2 4± 0 4 6 9) μg/L(n =2 1,P <0 0 5 ) ;用ICAM 1及LFA 1的抗体进行黏附阻滞后 ,AML细胞与TNFα激活的内皮细胞的黏附下降为 (2 0 12±  相似文献

目的 检测肥胖青少年血清瘦素、胰岛素、胰岛素原水平的变化 ,探讨青少年肥胖与代谢综合征的关系。方法 从年龄 14~ 16岁的 2 2 17例学生中筛选出体重指数 (BMI)≥ 2 5kg/m2 的肥胖学生 (肥胖组 ) 198例 ,BMI在 18 5~ 2 3 0kg/m2 之间的体重正常学生 (正常组 ) 78例 ,用放射免疫方法测定血清瘦素、胰岛素和胰岛素原水平 ,同时测定血糖及血脂水平 ,比较两组间差异。结果 血清瘦素水平女生明显高于同龄男生 [(18 5 3± 1 4 1) μg/L比 (6 33± 1 79) μg/L]。肥胖组血清瘦素、胰岛素和胰岛素原水平均高于同龄体重正常者 [分别为 (19 94± 1 91) μg/L比 (11 2 7± 2 0 4 ) μg/L ,(15 34± 1 6 6 ) μIU/L比 (13 17± 1 4 3) μIU/L ,(16 19± 1 6 4 )pmol/L比 (11 79± 1 70 )pmol/L ],血糖、甘油三酯 (TG)和高密度脂蛋白胆固醇 (HDL C)水平虽然在正常范围内 ,但肥胖者血糖和TG水平高于同龄体重正常者 [分别为 (4 6 3± 0 5 0 )mmol/L比 (4 13± 0 33)mmol/L ,(1 2 0± 0 5 6 )mmol/L比 (0 90±0 32 )mmol/L],HDL C水平低于同龄体重正常者 [(1 14± 0 2 4 )mmol/L比 (1 38± 0 2 6 )mmol/L]。结论 肥胖青少年可能存在瘦素抵抗、胰岛素抵抗及潜在的糖代谢和脂代谢异常等代谢综合征改变 ,  相似文献

目的 探讨扩张型心肌病患者血清可溶性胞间黏附因子-1(sICAM-1)水平与扩张型心肌病的发生发展的关系及临床意义.方法 应用酶联免疫双抗体夹心法(ELISA)测定25例扩张型心肌病患者和15例健康对照组的血清sICAM-1水平.所有扩张型心肌病患者均经过超声心动图、X线胸片、心电图等多项检查证实,确属不明原因的心脏扩大伴有不同程度的心肌收缩功能减退,左室射血分数(LVEF)<35%者.结果 扩张型心肌病组血清sICAM-1明显高于对照组[(162.46±78.56)ng/ml,69.48 ng/ml,P<0.01];心功能Ⅱ级组血清sICAM-1高于对照组[(99.80±13.49)ng/ml,(69.48±16.86)ng/ml,P<0.05];心功能Ⅲ级组血清sICAM-1水平明显均高于心功能Ⅱ级组[(188.40±27.15)ng/ml,(99.80±13.49)ng/ml,P<0.01],心功能Ⅳ级组血清sICAM-1水平明显高于心功能Ⅲ级组[(370.85±49.97)ng/ml,(188.40±27.15)ng/ml,P<0.01].结论 扩张型心肌病患者血清中sICAM-1高表达,并且随着心衰加重,外周血清中sICAM-1的水平逐渐升高.在DCM中,sICAM-1可以作为一种无创性炎症活动的标记物  相似文献

目的 对比研究缬沙坦和尼群地平对原发性高血压 (EH )患者血压及血清可溶性粘附分子sICAM 1,sVCAM 1水平的影响。方法 随机对照方法设计 ,将 6 4例轻中度原发性高血压患者分为两组 ,分别予缬沙坦 (80mg/d)和尼群地平 (10~ 30mg/d)治疗 6周 ,治疗前后抽血 ,用酶联免疫方法 (ELISA)测定外周血清中可溶性粘附分子sICAM 1,sVCAM 1水平。另选择 2 5例健康人作为对照组。结果 治疗前与对照组比较 ,高血压患者血清中可溶性粘附分子sICAM 1,sVCAM 1水平升高 ,分别为 [(318 2± 2 7 5 ) μg/Lvs (2 71 5± 2 6 8) μg/L ,P <0 0 5 ]和 [(5 90 6± 4 0 1) μg/Lvs (4 17 9± 38 7) μg/L ,P <0 0 1]。治疗后两组的血压均明显下降 (P <0 0 5 ) ,两组间无差异。缬沙坦组可溶性粘附分子sICAM 1,sVCAM 1水平均较治疗前明显下降 ,分别为[(32 0 5± 2 3 6 ) μg/Lvs (2 80 2± 2 5 4 ) μg/L ,P <0 0 5 ]和[(5 86 2± 4 2 5 ) μg/Lvs (4 5 1 2± 38 9) μg/L ,P <0 0 1]。而尼群地平组可溶性粘附分子sICAM 1,sVCAM 1水平较治疗前无明显下降 (P >0 0 5 )。结论 与尼群地平比较 ,缬沙坦不仅能有效降低血压 ,还能抑制血管壁的炎症反应 ,有利于预防和延缓动脉粥样硬化的发生和发展。  相似文献

目的 :探讨血浆中性粒细胞弹性蛋白酶 (neutrophilelastase,NE)与冠心病 (CHD)的关系。方法 :对85例观察对象进行分组 ,根据冠状动脉造影结果分为 :简单病变组 2 4例 ,复杂病变组 5 1例 ,正常对照组 10例。根据CHD病情分为 :急性心肌梗死 (AMI)组 9例 ,不稳定型心绞痛 (UAP)组 4 5例 ,稳定型心绞痛 (SAP)组 2 1例 ,正常对照组 10例。用酶联免疫吸附法 (ELASA)检测血浆NE含量 ,比较不同组间NE含量。结果 :血浆NE含量 :复杂病变组 [(6 3.4 2± 13.6 2 ) μg/L]高于简单病变组 [(4 4 .5 0± 12 .73) μg/L],差异有统计学意义 (P <0 .0 1)。AMI组 [(71.4 8± 10 .16 ) μg/L]高于UAP组 [(6 1.2 7± 14 .5 7) μg/L],但差异无统计学意义 (P >0 .0 5 ) ,明显高于SAP组 [(4 0 .95± 10 .0 9) μg/L]及正常对照组 [(36 .6 3± 12 .35 ) μg/L],差异均有统计学意义 (P <0 .0 1) ,后两组间比较差异无统计学意义 (P >0 .0 5 )。UAP组明显高于SAP组及正常对照组 ,差异有统计学意义 (P <0 .0 1)。结论 :血浆NE含量与冠状动脉病变稳定性及CHD病情严重性相关 ,NE可能是冠状动脉病变活动性的标志物。  相似文献

目的 探讨转导反义内皮素转换酶 (ECE)核酸的气道上皮细胞对尘螨过敏性患者外周血单个核细胞释放Th1/Th2相关细胞因子的影响。方法 尘螨过敏患者 2 1例 ,分离其外周血单个核细胞 (PBMC) ,根据实验要求分为 :未刺激组 (A组 )、反义ECE上皮细胞 +PBMCs组 (A1组 )、对照组上皮细胞 +PBMCs组 (A2 组 )及螨刺激组 (B组 )、反义ECE上皮细胞 +PBMCs+尘螨提取液组 (B1组 )、对照组上皮细胞 +PBMCs+尘螨提取液组 (B2 组 ) ,共培养 72h ,螨刺激组同时加入尘螨提取液 2 0 μg/ml。培养上清用酶联免疫吸附 (ELISA)法检测白细胞介素 5 (IL 5 )、γ干扰素 (IFN γ)。结果2 1例患者中有 12例患者经尘螨刺激后IL 5释放明显增加。将 12例患者进行统计学分析发现 ,未经尘螨刺激时 ,A1组IL 5和IFN γ水平分别为 [(6 0± 1 3)× 10 -9g/L]和 [(6 3± 2 6 )× 10 -9g/L],A2 组为 [(7 5± 1 1)× 10 -9g/L]、[(70± 5 2 )× 10 -9g/L],两组比较差异均无显著性 (P >0 0 5 ) ;尘螨刺激后 ,B1组的IL 5水平和IFN γ分别为 [(8 2± 1 6 )× 10 -9g/L]、[(10 0± 4 1)× 10 -9g/L],B2 组分别为[(12 0± 1 8)× 10 -9g/L]、[(15 3± 71)× 10 -9g/L],两组IL 5比较差异有显著性 (P =0 0 4 7) ,而IFN γ水平比较差异无显著性 (P >  相似文献

目的 通过测定类风湿关节炎 (RA)患者血清可溶性细胞黏附分子 1(sICAM 1)和E 选择素 ,分析ICAM 1、E 选择素在慢作用抗风湿药治疗前后的变化以及与肿瘤坏死因子 (TNF) α、白细胞介素 (IL) 1β和临床资料的相关性 ,以了解黏附分子在RA发病机制中的作用和临床意义。方法 收集 30例活动期RA患者和 15名正常人血清 ,17例患者服用甲氨蝶呤 (MTX) ,13例服用阿克他利 (ACT) ,连续半年。采用酶联免疫吸附测定 (ELISA)法检测ICAM 1、E 选择素、IL 1β和TNF α,并测定血沉、类风湿因子 ,同时记录RA患者采血时的临床资料。结果  30例RA患者中男性 4例 ,女性 2 6例 ,年龄 18~ 6 2岁 ,平均 (4 3± 12 )岁 ,病程 3个月~ 8年 ,平均 (4± 5 )年。关节功能Ⅱ级 18例 ,Ⅲ级 12例。X线分期 :Ⅰ期 10例 ,Ⅱ期 11例 ,Ⅲ期 9例。治疗前RA患者血清ICAM 1和E 选择素均明显高于正常对照组 (P <0 0 0 1) ;治疗半年后RA病情缓解 ,血清ICAM 1和E 选择素较治疗前明显下降 (P <0 0 0 1) ,MTX组较ACT组下降明显 ,但统计学处理差异无显著性 ;ICAM 1、E 选择素和TNF α、IL 1β之间呈显著正相关 (P <0 0 1) ,且与血沉 (ESR)明显相关。 结论 ICAM 1和E 选择素参与RA发病 ,慢作用抗风湿药治疗后可使其水平下降 ,不论治疗前后均  相似文献

目的 研究糖尿病患者血清可溶性细胞间粘附分子 - 1(s ICAM- 1)和可溶性血管细胞粘附分子 - 1(s VCAM- 1)水平的变化。方法 应用酶联免疫吸附法 (EL ISA)检测了 18例 1型糖尿病和 47例 2型糖尿病患者及 2 0例健康对照者的血清 s ICAM- 1、s VCAM- 1以及甘油三酯 (TG)水平。结果  1血清 s ICAM- 1和 s VCAM- 1水平 ,1型、2型糖尿病患者组均显著高于健康对照组 (P<0 .0 5~ 0 .0 1) ,2型糖尿病有微血管病变组又高于无微血管病变组 (P<0 .0 5~ 0 .0 1) ,1型糖尿病与 2型糖尿病组之间无显著性差异 ;2血清 s VCAM- 1水平 ,2型糖尿病的高甘油三酯血症组 (A组 )、高甘油三酯血症合并高血压组 (B组 )高于单纯高血压组 (C组 )和甘油三酯、血压正常组 (D组 ) (P均 <0 .0 5 ) ;3 1型糖尿病和 2型糖尿病患者血清 s ICAM- 1和 s VCAM- 1之间均呈正相关关系 (r=0 .83、0 .5 3,P均 <0 .0 1)。结论  12型糖尿病患者血清 s VCAM- 1升高与高甘油三酯血症有关 ;2血清 s ICAM- 1和 s VCAM- 1参与了糖尿病微血管病变的发病过程 ,并可作为糖尿病病情变化的监测指标  相似文献

AIM To elucidate the biological and clinical significance of sICAM-1 and sVCAM-1 in patients with gastric cancer.METHODS The serum levels of soluble ICAM-1 and VCAM-1 were measured with sandwith enzyme immunoassay.RESULTS In gastric cancer patients, soluble ICAM-1 and VCAM-1 concentrations were significantly elevated in comparision with those of healthy subjects (289.23μg/L±32.69μg/L vs 190.44μ/L±35.92μg/L,1430.88μg/L±421.71μg/L vs 727.24μg/L±157.68μg/L, respectively, P<0.01). The increment in serum sICAM-1 and sVCAM-1 concentrations correlated well with the staging of gastric cancer. The serum levels of sICAM-1 and sVCAM-1 in patients of Ⅲ-Ⅳ stages were higher than those of Ⅰ-Ⅱ stages (346.60μg/L±92.10μg/L vs 257.54μg/L±32.77μg/L, 1800.60μg/L±510.76μg/L vs 1262.81μg/L±236.73μg/L). The levels of sICAM-1 and sVCAM-1 were correlated significantly (r=0.49,P<0.01). The sICAM-1 and sVCAM-1 levels correlated positively with alkaline phophatase (r=0.63,0.71,P<0.001) and white cell count (r=0.52,0.43, P<0.01); but correlated negatively with serum albumin (r=-0.41, -0.49, P<0.01).CONCLUSION The measurement of circulating ICAM-1 and VCAM-1 may bring additional prognostic information for patients with gastric cancer in varying stages.INTRODUCTIONTumor growth and metastasis involves a variety of cell-cell and cell-extracellular matrix interactions mediated by cell adhesion molecules. Currently, a number of cell adhesion molecules, such as intercellular adhesion molecules-1 (ICAM-1), vascular cell adhesion molecules-1 (VCAM-1), etc. have been found.ICAM-1 and VCAM-1 are members of the immunoglobulin supergene family which are cytokine-induced glycoproteins (IL-1, TNFα and IFNγ). Both of them have five or seven extracellular immunoglobulin-like domains, a single transmembranous domain and a short cytoplasmic tail[1,2]. The natural ligand of ICAM-1 or VCAM-1 is LFA-1 (CD11a) and Mac-1 (CD11b) or VLA-4, respectively[3]. ICAM-1 is a widely distributed protein on a variety of tissues, and can be detected in many cells such as macrophage, T- and B-cells, or fibroblasts, endothelial and epithelial cells. VCAM-1 is also a widely distributed protein and is constitutively expressed on tissue macrophage, dentritic cells in lymphoid tissue and skin, as well as on bone marrow fibroblasts and epithelial cells. Expression of VCAM-1 is inducible on vascular endothelial cells under pathological conditions[4].Recently, soluble forms of several adhesion molecules including ICAM-1 and VCAM-1 were found in serum of normal donors[5]. Abnormally high levels of them have been described in some solid malignant tumors, leukemia, autoimmune disease, infectious disease, etc.The present study was carried out to measure the circulating levels of sICAM-1 and sVCAM-1 in gastric cancer before treatment was given and to study their correlation with clinical, histological and routine laboratory parameters.  相似文献

BACKGROUND: The aetiology of idiopathic portal hypertension (IPH) is unknown. However, some evidence of immunological abnormalities in IPH patients has been reported. METHODS: As adhesion molecules are important in the interaction between lymphocytes and accessory and target cells, the expression and release of the soluble form of vascular cell adhesion molecule-1 (VCAM-1) and intercellular adhesion molecule (ICAM-1) were examined in this study. RESULTS: In IPH patients, the serum level of soluble VCAM-1 was found to be increased, compared with that of healthy subjects, fatty liver patients and chronic hepatitis patients. The level of soluble ICAM-1 of IPH patients was found to be slightly increased, compared with that of healthy subjects; however, it was not different from the level in patients with other diseases. The expression of VCAM-1 was observed in the sinusoidal lining cells and endothelial cells around the liver vessels of several IPH patients. In contrast, ICAM-1 was weakly expressed in sinusoidal lining cells and hepatocytes in the liver tissue of only one of four IPH patients. CONCLUSIONS: This differential pattern of VCAM-1 and ICAM-1 was found in IPH patients and it was suggested that VCAM-1 might be an important molecule in the occurrence of IPH.  相似文献

目的探讨血清炎性标志物与急性冠脉综合征(ACS)的关系。方法检测48例ACS患者血清C-反应蛋白(CRP)、可溶性血管内皮细胞黏附分子-1(sVCAM-1)和可溶性细胞间黏附分子-1(sICAM-1)等炎性因子变化,与冠脉造影结果和临床预后进行对比分析。结果1ACS组血清CRP与sICAM-1、sVCAM-1水平均高于稳定性心绞痛(SA)组和正常对照组;2sVCAM-1≥870ng/ml者3个月内主要冠脉不良事件(MACE)发生率明显高于<870ng/ml者;3血清肌钙蛋白阳性者CRP、sICAM-1、sVCAM-1浓度均明显高于阴性者;4冠脉狭窄程度在ACS组与SA组无统计学差异;冠脉狭窄程度积分与TC水平呈正相关,而与血清CRP、sICAM-1、sV-CAM-1及血浆纤维蛋白原(Fig)水平均无相关性;5sVCAM-1、Fig和肌钙蛋白预测3个月内MACE发生的敏感性分别为67.5%、82.4%、57.1%;特异性分别为54.8%、58.1%、81.5%。结论血清炎性标志物对ACS患者临床危险分层和判断预后有重要意义。  相似文献

AIMS: Inflammatory mechanisms have been implicated in the pathogenesis of atherosclerosis. Cell adhesion molecules, expressed on endothelial cells and leukocytes, mediate transendothelial migration of leukocytes into the vessel wall, but also circulate in soluble forms. In the present study we related soluble cell adhesion molecules to the risk of suffering a cardiovascular death or a non-fatal myocardial infarction (cardiovascular death/myocardial infarction) in a substudy to the Angina Prognosis Study in Stockholm (APSIS). METHODS AND RESULTS: Soluble intercellular adhesion molecule-1, vascular adhesion molecule-1 and E-selectin were measured in serum collected on inclusion in the APSIS study. During follow-up, seven patients suffered non-fatal myocardial infarction or cardiovascular death, whereas 86 patients were event-free. Cardiovascular death/myocardial infarction was associated with elevated intercellular adhesion molecule-1 (354+/-142 vs 282+/-62ng x ml-1;P<0.01) and vascular adhesion molecule-1 (538+/-138 vs 433+/-135ng x ml-1;P =0.05), and E-selectin levels tended to be higher (72+/-54 vs 49+/-20ng x ml-1). Clinical risk factors (history of hypertension, previous myocardial infarction, diabetes and smoking) were more abundant in the event group. Subgroup analyses showed that hypertension, smoking or male sex were associated with elevated intercellular adhesion molecule-1, whereas previous myocardial infarction or male sex were associated with elevated vascular adhesion molecule-1. CONCLUSION: Patients with stable angina pectoris who developed cardiovascular death/myocardial infarction had elevated serum levels of soluble cell adhesion molecules, indicating increased inflammatory activity. The value of soluble cell adhesion molecules as prognostic markers in patients with stable ischaemic heart disease merits further study.  相似文献

目的探讨通心络超微粉对高脂饮食兔胸主动脉NF-κB、胞间黏附分子1(ICAM-1)及血管细胞黏附分子1(VCAM-1)表达的影响。方法健康雄性新西兰白兔32只,随机分为空白对照组、模型组、阿托伐他汀组、通心络组四组。空白对照组,饲以普通饲料;模型组,饲以高脂饲料;阿托伐他汀组,饲以高脂饲料同时阿托伐他汀(3mg·kg^-1·d^-1)灌胃;通心络组,饲以高脂饲料同时通心络超微粉(0.31g·kg^-1·d^-1)灌胃,连续给药,于6周末免疫组织化学染色法检测主动脉壁中NF-κB核转位情况、ICAM-1及VCAM-1蛋白表达情况,RT—PCR法检测ICAM-1 mRNA及VCAM-1 mRNA表达。结果与空白对照组相比,模型组家兔主动脉壁中NF—KB核转位明显增加。ICAM-1、VCAM-1基因及蛋白表达明显增多(P〈O.01)。与模型组相比,通心络组与阿托伐他汀组家兔主动脉壁中NF-κB核转位明显减少、ICAM-1、VCAM-1基因及蛋白表达明显减少(P〈0.01或P〈0.05),通心络组显著少于阿托伐他汀组(P〈0.01)。结论通心络超微粉通过抑制NF-κB核转位进而降低ICAM-1、VCAM-1基因及蛋白表达,减轻动脉粥样硬化病理改变。  相似文献

目的观察蜂胶水提物对损伤血管内皮细胞的保护作用,探讨蜂胶抗动脉粥样硬化的作用及其机制。方法用50 μg/L TNF-α诱导体外培养脐静脉内皮细胞损伤,用50、100、200 mg/L蜂胶水提物分别干预6、12、24 h,分为对照组、模型组、蜂胶低浓度组、蜂胶中浓度组、蜂胶高浓度组、氟伐他汀钠组、联合组,采用流式细胞仪检测细胞间黏附分子1(ICAM-1)和血管细胞黏附分子1(VCAM-1)的表达。结果与对照组比较,模型组ICAM-1和VCAM-1表达明显升高;与模型组比较,蜂胶低浓度组、蜂胶中浓度组和蜂胶高浓度组ICAM-1和VCAM-1明显降低(P0.01)。12 h时与氟伐他汀钠组比较,联合组ICAM-1和VCAM-1表达明显降低(P0.01)。结论蜂胶水提物能降低ICAM-1和VCAM-1的表达。与氟伐他汀钠联合应用,对血管内皮细胞损伤有协同保护作用。  相似文献

目的 观察氟伐他汀 (Flu)和辛伐他汀 (Sim)对肿瘤坏死因子 (TNF)α诱导的人脐静脉内皮细胞 (HUVEC)细胞间黏附分子 (ICAM) 1表达的影响 ,探讨他汀类药物可能的非调脂抗动脉粥样硬化作用。方法 体外培养HUVEC ,加TNFα (10 0U/ml)及 1× 10 -8~ 1× 10 -5mol/L浓度Flu或Sim ,共孵育 6、12和 2 4h ,采用细胞ELISA、流式细胞技术和逆转录 聚合酶链反应测定ICAM 1蛋白及mRNA水平表达。结果 Flu和Sim皆对TNFα诱导的ICAM 1蛋白表达及ICAM 1mRNA表达有抑制作用 ,呈浓度依赖性和时间依赖性特征。结论 Flu和Sim抑制TNFα诱导的HUVECICAM 1表达 ,可能对阻止血单核细胞向血管内皮细胞募集和黏附、延缓动脉粥样硬化的发生和发展有一定作用。  相似文献

目的通过动物实验,探讨细胞间黏附因子-1(ICAM-1)在病毒性心肌炎(VMC)心肌组织中的动态变化及意义.方法成年Balb/c小鼠分为实验组和对照组.实验组分为4组,分别于接种柯萨奇B3病毒(CVB3)后第3、5、8、15天应用免疫组织化学方法检测心肌中ICAM-1及NF-KB.对照组给予不含CVB3的Eagle'液,作为总体对照.结果Balb/c小鼠注射CVB3后第5天小鼠心肌ICAM-1增加,第8天达高峰,持续到第15天仍明显.心肌ICAM-1及心肌病理改变半定量结果经直线相关分析二者具相关性.注射CVB3后第8~15天见胞质中NF-KB(P65)增强及在部分细胞核上存在.结论提示ICAM-1产生与NF-KB活化有关,ICAM-1参与VMC的病理损伤过程,可能作为VMC病变严重程度的免疫组化指标.  相似文献
