汉防已甲素治疗博莱霉素诱导的大鼠肺间质纤维化实验研究

目的　观察中西药钙拮抗剂对肺间质纤维化的治疗作用。方法　将 72只SD大鼠一次性气管内注入博莱霉素A5制备肺纤维化模型 ,次日随机给药 ,并测定分析各组给后d7、d14、d2 8肺组织内羟脯氨酸、脂质过氧化物含量、组织病理学变化以及比较各组d2 8肺系数。结果　汉防己甲素组及维拉帕米组各期肺组织脂质过氧化物及羟脯氨酸含量均较模型组明显降低 (p <0 .0 5 ,p<0 .0 1) ,肺系数明显减小 (p <0 .0 1,p<0 .0 1) ,组织病理学显示肺泡炎及肺纤维化程度均明显减轻 ,其中汉防己甲素优于维拉帕米。结论　中药钙拮抗剂汉防己甲素治疗肺间质纤维化疗效满意 ,值得进一步研究。

Experimental study on tetrandrine treating pulmonary interstitial fibrosis induced by bleomycin in rats

Objective To observe the curative effect of the calcium antagonist (both traditional Chinese medicine and western medicine) on pulmonary fibrosis.Methods Seventy-two SD rats were replicated into pulmonary fibrosis models by intratracheal injection in a single of bleomycin A 5 and were divided into 4 groups randomly.Then,normal saline ig in untreated model group (n=24),tetrandrine ig in tetradrine group(n=24) , verapamil ig in verapamil group (n=24) and control group (n=15) were performed until the end of experimental . The hydroxy prolines (HP) and lipid peroxides(LP) contents in lung tissues and the histopathologic changes of the lung of the rats in each group were assayed on the 7th, 14th, and 28th day after the injection, and the lung coefficients of the rats of each group on the 28th day were compared between groups.Results The HP and LP contents in tetrandrine group and verapamil group were obviously lower than those of model group at all phases (p<0.05 or p<0.01),and so were the lung coefficients of the two group (p<0.001 or p<0.01).The degree of alveolitis and fibrosis of the lung studied by histopathology in the two groups was lower than that in the model group,and the tetrandrine was superior to verapamil. Conclusions The tetrandrine has satisfactory curative effect on pulmonary fibrosis and it is worth research further.
