鞣花酸对小鼠酒精性肝损伤的保护作用

摘    要:目的:观察鞣花酸对小鼠酒精性肝损伤的保护作用,并探讨其作用机制。方法: 50只昆明小鼠随机分为空白对照组,肝损伤模型组和低、中、高剂量鞣花酸组,每组10只。空白对照组和肝损伤模型组小鼠给予羧甲基纤维素钠溶剂灌胃,不同剂量鞣花酸组小鼠分别以160、320和480 mg·kg-1鞣花酸灌胃,连续15 d。末次灌胃后,空白对照组小鼠灌胃等量蒸馏水,其余组小鼠均灌胃50%乙醇12 mL·kg-1,禁食不禁水,16 h后各组小鼠眼球取血,处死,取小鼠肝脏组织和血清。计算各组小鼠肝脏系数;测定小鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性和甘油三酯(TG)水平;测定小鼠肝组织中超氧化物歧化酶(SOD)、谷胱甘肽(GSH)活性和丙二醛(MDA)水平。结果:与空白对照组比较,肝损伤模型组小鼠肝脏系数、血清ALT和AST活性及TG水平明显升高(P<0.05或P<0.01),肝组织中SOD和GSH活性及MDA水平明显降低(P<0.05或P<0.01);与肝损伤模型组比较,低、中和高剂量鞣花酸组小鼠肝脏系数、血清ALT和AST活性明显降低(P<0.05或P<0.01),肝组织中SOD活性明显升高(P<0.05或P<0.01),MDA水平明显降低(P<0.01);中和高剂量鞣花酸组小鼠肝组织GSH活性明显升高(P<0.01);高剂量鞣花酸组小鼠血清TG水平明显降低(P<0.05)。结论:鞣花酸对小鼠酒精性肝损伤具有保护作用,以480 mg·kg-1剂量时作用最为明显;鞣花酸可能通过抗氧化作用实现其对肝细胞损伤的保护作用。

Abstract:Objective To investigate the protecive effects of ellagic acid on alcoholic liver injury in the mice,and to explore its mechanisms. Methods 50 KM mice were randomly divided into control group,model group,low,medium and high doses of ellagic acid groups (n=10). The mice in control group and model group were intragastricly administed with sodium carboxymethyl cellulose solvent;the mice in ellagic acid groups were administed with 160,320 and 480 mg·kg-1 ellagic acid daily,respectively.After administrated for 15 d,the liver injury model was induced with 12 mL·kg-1 50% ethanol.16 h later,all the mice were taken blood from the eyes and killed,and the activities of serum aspartate aminotransferase (AST),alanine aminotransferase (ALT), and triglycerides (TG) levels in serum and activities of liver superoxide dismutase (SOD), glutathione peroxidase(GSH) were determined as well as liver malondialdehyde (MDA) levels in liver tissue. Results Compared with control group,the liver index,the activities of ALT,AST,and the level of TG in serum and the level of MDA in liver tissue of the mice were significantly increased(P<0.05 or P<0.01),and the activities of SOD and GSH in liver tissue were significantly decreased in model group(P<0.05 or P<0.01). Compared with model group,the liver index,the activities of ALT,AST in serum and the level of MDA in liver tissue of the mice were significantly decreased,the activities of SOD in liver tissue were significantly increased in low,medium and high doses of ellagic acid groups(P<0.05 or P<0.01), and the activities of GSH in liver tissue were significantly increased in medium and high dose of ellagic acid groups(P<0.01),and the level of TG in serum were significantly decreased in high dose of ellagic acid group(P<0.05). Conclusion Ellagic acid has the protective effect on alcoholic liver injury in the mice and the most effective dose is 480 mg·kg-1.
