新药金龙蛇颗粒体外抗肿瘤作用研究

摘    要:目的观察新药金龙蛇颗粒的体外抗肿瘤作用。方法采用MTT比色法和软琼脂克隆形成实验,观察金龙蛇颗粒体外对人胃癌细胞MKN-45、人肺癌细胞SPCA-1的作用。结果 MTT法结果表明,对MKN-45细胞金龙蛇低、中、高剂量组24h抑制率为30.69%、32.67%、43.56%,48h为36.99%、39.04%、43.84%,对SPCA-1细胞24h抑制率则为23.33%、24.44%、37.78%,48h为28.46%、33.33%、46.34%,皆与空白组存在统计学差异,并且体现了剂量依赖效应,抑制率随剂量增加而增高。软琼脂克隆形成实验结果显示,对MKN-45细胞金龙蛇低、中、高剂量组100个细胞克隆形成率为25.67%、22.17%、19.83%,200个细胞为22.09%、19.75%、17.67%,对SP-CA-1细胞100个细胞克隆形成率为33.50%、28.17%、24.17%,200个细胞为24.59%、20.25%、18.25%,同样与空白组存在差异,并体现出了剂量效应。结论体外实验观察新药金龙蛇颗粒具有一定的抗肿瘤作用。

Abstract:Objective To investigate the in vitro anti-tumor effect of new Chinese herbal medicine jinlongshe granules.Methods MTT assay and the soft agar colony formation assay were used to observe the effects of jinlongshe on MKN-45 cells of stomach cancer and SPCA-1 cells of lung cancer in human.Results MMT assay indicated that the inhibitory rates in 24 h to MKN-45 cells were 30.69%,32.67% and 43.56% and those in 48 h were 36.99%,39.04% and 43.84%;the inhibitory rates of 24 h to SPCA-1 cells were 23.33%,24.44% and 37.78%,and those of 48 h were 28.46%,33.33%,46.34% in jinlongshe groups of low,middle and high doses separately,all of which presented the statistical differences as compared with blank group and explained the dose-dependence effect,meaning the inhibitory rate increased while the dose was higher.The results of soft agar colony formation assay showed that the colony formation rates of 100 MKN cells were 25.67%,22.17% and 19.83%;and those of 200 cells were 22.09%,19.75% and 17.67%;the colony formation rates of 100 SPCA-1 cells were 33.50%,28.17% and 24.17% and those of 200 cells were 24.59%,20.25% and 18.25% in jinlongshe groups of low,middle and high doses separately;all of which presented the differences as compared with blank group and explained the dose effect.Conclusion The in vitro experiment has observed that new Chinese herbal medicine,jinlongshe granules act on anti-tumor to a certain extent.
