AIM To study the clinical significance of alterations of serum hyaluronic acid in rats with carbontetrachioride-induced liver fibrosis.METHODS Rat liver fibrosis model was induced by carbon tetrachloride (CC14). The rats were divided intofive groups; group 1 (control): 0 week with no CCl4-inducing; group 2, 3, 4 and 5: 3, 6, 9 and 12 weeksafter CCl4-induction respectively. Serum HA level was analysed among various liver fibrosis groups andcontrol, and then compared the HA findings with the hepatic histopathology.RESULTS During rat liver fibrosis, serum HA levels of the liver fibrosis groups (group 2: 7.98ng/mL;group 3: 20.10 ng/mL; group 4:229.73 ng/mL; group 5:324,74 ng/mL) were significantly higher thanthat of control group (group 1:0.21 ng/mL) (P<0.01), in which group 4 and group 5 are much higher1094 times (229.73ng/mL/0.21 ng/mL) and 1546 times (324.74 ng/mL/0.21 ng/mL) than group 1respectively. When compared with each other, the serum HA levels are 38 times (7.98ng/mL/0.21 ng/mL; P<0.01, group 2 vs group 1); 2.5 times (20.10ng/mL/7.98 ng/mL; P<0.01, group 3 vsgroup 2); 11.4 times (229.73 ng/mL/20.10 ng/mL; P<0.01, group 4 vs group 3); 1.4 times (324.74 ng/mL/229.73 ng/mL; P<0.01, group 5 vs group 4) respectively.CONCLUSION The results demonstrated that the dynamic alterations of serum HA play an important rolein the early clinical diagnosis and staging of liver cirrhosis.  相似文献

目的:观察复肝丸对四氯化碳诱导的小鼠肝纤维化和二甲基亚硝胺诱导的大鼠肝纤维化的防治作用。方法:小鼠随机分为正常组、模型组、复肝丸组和培哚普利,除正常组外,其余组腹腔注射四氯化碳复制肝纤维化模型;自造模之日起各组灌胃4周。大鼠随机分为正常组、模型组、复肝丸组和培哚普利,除正常组外,其余各组腹腔注射二甲基亚硝胺4周复制肝纤维化模型;造模结束后各组灌胃4周。称重测量肝体比、脾体比,比色法测定血清谷丙转氨酶和谷草转氨酶活性,溴甲酚绿法测定血清白蛋白水平,碱消化法测定肝组织羟脯氨酸含量,HE染色观察肝组织炎性变化、天狼猩红胶原染色观察肝组织病理性胶原沉积变化。结果:与模型组相比,复肝丸10g生药/kg小鼠体重预防用药或6g/kg大鼠体重治疗用药均能降低大鼠和小鼠血清谷丙转氨酶和谷草转氨酶活性,减轻肝脏炎性病理,降低肝组织羟脯氨酸水平,改善肝组织病理性胶原沉积增生。结论:复肝丸对大鼠和小鼠肝纤维化具有防治作用。  相似文献

目的 研究四氯化碳(CCl4)致大鼠急性肝损伤模型大鼠血浆内源性代谢物变化特点和二至丸对异常代谢的调节作用,寻找二至丸治疗急性肝损伤可能的代谢生物标志物,以探索二至丸治疗肝损伤的作用机制。方法 运用快速分离液相色谱-串联四极杆飞行时间质谱(RRLC-Q-TOF/MS)技术,对大鼠血浆中内源性代谢物进行测定。结果 定性鉴定出9种潜在代谢生物标志物,通过独特通路分析(IPA),找到2条肝损伤代谢通路。结论 二至丸可以使造模后大鼠的体内代谢物含量有不同程度的恢复,提示其治疗作用可能与2条肝损伤代谢通路的调节有关。独特通路分析能有效的识别药物靶标,这有助于进一步了解药物作用机制,提高研究效率。  相似文献

Aim of the study

The study was aimed to investigate the protective effect against hepatic injury induced by CCl4 for the ethanolic extract of FZJ.

Materials and methods

The alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were detected as biomarker in blood of hepatic injury. Product of lipid peroxidation (MDA), activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px), and reduced glutathione (GSH) contents were evaluated for oxidative stress in hepatic injury. Moreover, histopathological observation was assayed at the degree of hepatic injury.

Results

After administrated the FZJ, the dose of 200 mg/kg significantly decreased ALT and AST, and attenuated histopathology of hepatic injury, and ameliorated the oxidative stress in hepatic tissue. Partly assayed indexes were ameliorated after administrated FZJ at the dose of 100 mg/kg.

Conclusion

These results indicated that hepatic protective effects of FZJ were very relevant to modulate the oxidative stress in hepatic injury.  相似文献

The objective of the present study was to evaluate hepatoprotective activity of methanolic extract of Hiptage bengalensis (L.) kurz (MEHB) in rats. Hepatic damage was induced by administration of carbontetrachloride(1 ml/kg, b.w, p.o.) in combination with liquid paraffin (1:1) as a single dose on 7th day. The extent of liver damage was studied by estimating biochemical parameters. Administration of MEHB (200 mg & 400 mg/kg) for 6 days before carbontetrachloride administration showed a significant reduction (p < 0.01) of serum liver damage enzymes markers-aspartate transaminase, alanine transaminase, total bilirubin and alkaline phosphatase (ALP). Histopathological changes of hepatic tissue were also evaluated in control and MEHB treated groups. Results also indicated that MEHB possessed potential antioxidant effect by increasing the levels of glutathione and also possessed free radical scavenging activities. The hepatoprotective effect of Hiptage bengalensis (L.) kurz was comparable to standard drug silymarin (50 mg/kg).  相似文献
