目的观察并探究补阳还五汤对脑出血大鼠脑组织磷脂酰肌醇3激酶/丝氨酸/苏氨酸蛋白激酶(PI3K/AKT)信号转导通路的影响及其神经保护作用的可能机制。方法 72只SD大鼠随机分为四组:假手术组、模型组、补阳还五汤组、银杏叶片组,每组18只。其中模型组、补阳还五汤组、银杏叶片组采用Rosenberg法制作脑出血大鼠模型。Garcia法检测大鼠神经功能评分,电镜观察神经元线粒体超微结构,western blot法检测磷酸化蛋白激酶(p-AKT)蛋白表达,TUNEL法检测细胞凋亡变化,免疫组化法检测B细胞淋巴瘤基因-2(bcl-2)蛋白、Bcl-2相关X蛋白(bax)的表达,甲酰胺法检测血脑屏障(BBB)通透性,干湿重法检测脑组织含水量。结果与脑出血模型组相比,补阳还五汤治疗明显提高神经功能学评分(P0.05),降低BBB通透性,减少脑组织水含量(P0.05),上调p-AKT、bcl-2蛋白表达,下调bax蛋白表达,减轻线粒体损伤,抑制神经元凋亡。结论补阳还五汤抗脑出血引起脑损伤的作用机制可能与其激活PI3K/AKT信号途径,抑制神经元凋亡,降低BBB通透性,减轻脑水肿有关。  相似文献

糖尿病视网膜病变是糖尿病常见的并发症,作为微血管病变被广泛报道。研究发现视网膜神经元退行性病变在糖尿病视网膜病变早期已经发生,且在糖尿病视网膜病变进展过程中扮演重要角色。神经元退行性病变以神经元凋亡和胶质反应为主要特征,其发生机制包括高血糖、氧化应激、谷氨酸毒性、炎症等。糖尿病视网膜神经退行性病变与微血管病变的发生发展密切相关。  相似文献

An ultra-sensitive T2-weighted MR imaging contrast agent was prepared based on Fe3O4 nanoparticles and Gd3+ ions (Fe3O4@Gd). Amino modified Fe3O4 nanoparticles were conjugated to diethylenetriamine pentaacetic acid, and finally coordinated with Gd3+ ions. The nanoparticles had a uniform morphology with a size of 100 nm and a Gd/Fe mass ratio of 1/110. The r2 (transverse relaxivity) of the Fe3O4 nanoparticles increased from 131.89 mM−1 s−1 to 202.06 mM−1 s−1 after coordination with Gd3+ ions. MR measurements showed that the aqueous dispersion of Fe3O4@Gd nanoparticles had an obvious concentration-dependent negative contrast enhancement. Hepatoma cells were selected to test the cytotoxicity and MR imaging effect. The application of Fe3O4@Gd nanoparticles as contrast agents was also exploited in vivo for T2-weighted MR imaging of rat livers. All the results showed the effectiveness of the nanoparticles in MR diagnosis.

AIM: To distinguish subtypes of gastric signet ring cell (SRC) carcinoma by investigating the expression of gastric and intestinal phenotypic markers, and to study the significance of phenotypic classification in predicting tumor progression and outcome. METHODS: Immunohistochemistry was performed in 66 cases of SRC carcinoma with MUC2, VILLIN, CDX2, Li-cadherin antibodies as intestinal phenotype markers and MUC5AC, HGM, MUC6 antibodies as gastric phenotype markers, and the relationship was analyzed between the phenotypic expression pattern and clinicopathologic parameters, as well as the 3-year survival rate. RESULTS: Expression of intestinal phenotypic markers was positively associated with tumor size, wall invasion, vascular invasion, lymph node metastasis and tumor-node-metastasis (TNM) stage. Cases expressing one or more intestinal markers had a significant lower survival rate than cases expressing none of the intestinal markers. CONCLUSION: The SRC carcinomas expressing intestinal phenotype markers exhibited a high pro-liferative potential, bad biological behaviors and poor prognosis. Examination of phenotype expression may be useful in distinguishing histological type and in predicting the prognosis of gastric SRC carcinoma.  相似文献

BACKGROUNDStress-related gastric mucosal damage or ulcer remains an unsolved issue for critically ill patients. Stress ulcer prophylaxis has been part of routine intensive care, but uncertainty and controversy still exist. Co-secreted with mucins, intestinal trefoil factor (ITF) is reported to promote restitution and regeneration of intestinal mucosal epithelium, although the mechanism remains unknown.AIMTo elucidate the protective effects of ITF on gastric mucosa and explore the possible mechanisms.METHODSWe used a rat model of gastric mucosal damage induced by water immersion restraint stress and lipopolysaccharide-treated human gastric epithelial cell line to investigate the potential effects of ITF on damaged gastric mucosa both in vivo and in vitro.RESULTSITF promoted the proliferation and migration and inhibited necrosis of gastric mucosal epithelia in vitro. It also preserved the integrity of gastric mucosa by upregulating expressions of occludin and zonula occludens-1. In the rat model, pretreatment with ITF ameliorated the gastric mucosal epithelial damage and facilitated mucosal repair. The protective effects of ITF were confirmed to be exerted via activation of Akt signaling, and the specific inhibitor of Akt signaling LY249002 reversed the protective effects.CONCLUSIONITF might be a promising candidate for prevention and treatment of stress-induced gastric mucosal damage, and further studies should be undertaken to verify its clinical feasibility.  相似文献

目的:评价新型Ziemer LDV Z6纳焦飞秒激光机(简称“Z6飞秒”)制作角膜瓣的LASIK手术效果。
  方法:随机抽取我院2015-06/12飞秒制瓣LASIK手术患者200例400眼,其中Z6飞秒制瓣200眼( Z6组),传统Ziemer LDV CrystalLine纳焦飞秒激光机(简称“CrystalLine飞秒”)制瓣200眼( CrystalLine组)。比较两组患者在术前、术后不同时间的视力、屈光度、Sim-K平均值、裂隙灯表现、眼压、非侵袭平均泪膜破裂时间( NIAvg-BUT )、手术操作难易度等方面的差异。
  结果:两组患者在术前和术后6 mo时的视力、屈光度、Sim-K平均值、眼压、NIAvg-BUT等方面无统计学差异(P>0.05)。 Z6组的部分患者在术中掀瓣方面有一定难度,术中不透明气泡层和不完全切割的发生率较高(P<0.05),但无严重并发症发生。术后裂隙灯下观察,Z6组患者角膜瓣边界比CrystalLine组更容易辨认。
  结论:新型Z6飞秒比传统CrystalLine飞秒在术中角膜瓣处理方面的要求更为精细和严格,需要引起屈光手术医师的重视。  相似文献

Glucocorticoid (GC) hormone has been commonly used to treat systemic inflammation and immune disorders. However, the side effects associated with long-term use of high-dose GC hormone limit its clinical application seriously. GC hormone that can specifically target the lung might decrease the effective dosage and thus reduce GC-associated side effects. In this study, we successfully prepared human lung-targeting liposomal methylprednisolone crosslinked with nanobody (MPS-NSSLs-SPANb). Our findings indicate that MPS-NSSLs-SPANb may reduce the effective therapeutic dosage of MPS, achieve better efficacy, and reduce GC-associated side effects. In addition, MPS-NSSLs-SPANb showed higher efficacy and lower toxicity than conventional MPS.  相似文献

目的 了解新冠疫情下本科护生职业认同现状及其影响因素。方法 采用一般情况调查表、职业认同量表、护生专业承诺量表对安徽某院校的588名本科护生进行调查。结果 职业认同总分为(58.17±11.27);护生专业承诺总分为(86.55±20.01);职业认同与专业承诺呈显著正相关(r=0.790,P<0.05);专业承诺总分、“有无您崇拜的护理前辈或老师”、“如果有机会,您是否会调换专业”两条目得分对职业认同有影响。结论 新冠疫情下本科护生职业认同处于中等水平且受多种因素影响。  相似文献
