目的:探讨桂千金子提取物对金黄色葡萄球菌的体外抗菌活性及作用机制.方法:通过体外抑菌试验,测定桂千金子乙醇提取物及其乙酸乙酯、正丁醇萃取物对金黄色葡萄球菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC),比较标准菌株金黄色葡萄球菌(SA)及临床分离的金黄色葡萄球菌(MSSA)和耐甲氧西林金黄色葡萄球菌(MRSA)的抗菌活性大小.测定桂千金子乙醇提取物作用后MRSA碱性磷酸酶(AKP)活性、外流DNA/RNA大分子含量、胞内蛋白含量的变化,采用电镜观察MRSA细胞形态.结果:桂千金子提取物对3种供试菌株的抑菌圈直径均大于15mm,MIC为1.56~12.50mg/mL,MBC为3.13~25.00 mg/mL,抗菌强度排序为乙醇提取物≈乙酸乙酯萃取物>正丁醇萃取物;与空白对照组比较,实验组AKP活性升高(P<0.05),各实验组与空白组外流DNA/RNA大分子含量和胞内蛋白含量比较,差异均无统计学意义(均P>0.05);电镜下MRSA菌体细胞变形,表面粗糙有颗粒附着.结论:桂千金子提取物对金黄色葡萄球菌及其耐药菌株均具有显著的抗菌活性,其抗菌作用可能与细胞壁结构破损致通透性发生改变有关.  相似文献

目的:比较广西产石韦和柔软石韦的抗炎镇痛疗效.方法:雄性昆明种小鼠80只,随机分为8组:模型组(蒸馏水),阳性对照组(地塞米松0.01 mg/kg),石韦及柔软石韦给药高(40 g生药/kg)、中(20 g生药/kg)、低(10 g生药/kg)剂量组,每组10只,各组小鼠灌胃给药,每天1次,连续7d,于末次给药后30 min,采用巴豆油混合物致小鼠耳廓肿胀及对角叉菜胶致足趾肿胀方法,观察石韦和柔软石韦对小鼠耳廓肿胀及对角叉菜胶致足趾肿胀的影响;雌性及雌雄并用昆明种小鼠80只,随机分为8组:空白对照组(蒸馏水)、阳性对照组(阿司匹林0.2 g/kg),石韦及柔软石韦给药高、中、低剂量组,给药方法同上,于末次给药后30 min,采用小鼠热板试验和醋酸扭体试验方法,观察石韦和柔软石韦对小鼠热板痛阈和扭体次数的影响.结果:与模型组比较,石韦和柔软石韦高、中剂量组均可明显减轻小鼠耳廓肿胀度和足趾肿胀度(P<0.01或P<0.05).石韦和柔软石韦比较,石韦高、中剂量组明显减轻足趾肿胀度(P<0.01),石韦中剂量组明显减轻小鼠耳廓肿胀度(P<0.05);与空白对照组比较,石韦和柔软石韦高、中剂量组的痛阈值和痛阈提高百分率均有明显的提高(P <0.01或P<0.05),石韦和柔软石韦高、中、低剂量组明显减少小鼠扭体次数(P <0.01或P<0.05).石韦和柔软石韦比较,石韦高、中剂量组明显提高小鼠热板痛阈值(P <0.01),石韦和柔软石韦对小鼠扭体次数无明显差异(P>0.05).结论:石韦和柔软石韦均有明显的抗炎镇痛作用,石韦的抗炎镇痛作用强于柔软石韦.  相似文献

Aim To prepare the sea cucumber enzy¬molysis fermentation liquid (SCEFL) by enzymatic hydrolysis of protease and fermentation of probiotics and to investigate the effect of SCEFL on the immunosup-pression induced by cyclophosphamide in mice and to explore its mechanism by metabomic method. Methods The immunosuppressive model was induced by in-traperitoneal injection of cyclophosphamide. C57BL/6J mice were randomly divided into normal group, model group, Levamisole group, SCEFL groups (at low, medium and high doses). The pathological changes of spleen were observed by HE staining. The proportion of CD4+and CD8+T lymphocyte subsets and the pro-portion of CD4 /CD8 T lymphocyte subsets in peripheral blood were detected by flow cytometry. The con¬tents of IL-2 ( interleukin-2 ), IgG ( immunoglobulin G) and IgM (immunoglobulin G) in serum were detected by ELISA. The changes of endogenous metabolites in mouse serum were analyzed by UPLC-Q/TOF-MS metabolomics, and the related metabolic pathways were explored. Results SCEFL could improve the pathological changes of immunosuppressed mice, in¬crease the proportion of CD4 T cells and the proportion of CD4+/CD8+T lymphocyte subsets,and increase the contents of IL-2, IgG and IgM in serum. The immune function of mice was regulated mainly through six related metabolic pathways, including glyc-erophospholipid metabolism, sphingolipid metabolism, linoleic acid metabolism, a-linoleic acid metabolism, glycophosphatidylinositol-anchored biosynthesis and arachidonic acid metabolism.
