Juvenile Idiopathic Arthritis (JIA) is a multi-factorial disease influenced both by environmental and genetic factors. Progressive pseudorheumatoid dysplasia (PPD) is a rare autosomal recessive genetic disorder affecting multiple joints, mimicking JIA. Aim of the work: to reveal the frequency of HLA-DR types among the studied patients and to correlate the different allele variations clinically. Patients and methods: Thirty JIA patients, in addition to 15 molecularly diagnosed PPD patients were subjected to full history taking and clinical examination. HLA-DRB1 typing was performed to 24/30 JIA and 12/15 PPD cases and thirty healthy age and sex matched children who were included as a control group. Results: The JIA patients were 22 females and 8 males with mean age of 15.8 ± 1.96 years and disease duration 5.3 ± 4.4 years. PPD patients were 8 males and 7 females with mean age of 8.7 ± 3.06 years and disease duration 3.95 ± 2.68 years. A significant frequency of HLA-DRB 04 (p = 0.049) among JIA patients was present in comparison to the controls (OR = 2.81, CI:1.02–7.75), other risky alleles were HLA-DRB 10, 13 and 15. However, HLA-DRB 01, 03, 07, 11 and 14 were found to be protective. HLA-DRB 01, 04, 10 and 13 were found to be risky alleles in PPD. However, HLA 03, 07, 11 and 15 were found to be protective alleles among PPD patients. Conclusion: HLA-DRB 04 was found in a higher frequency in JIA patients with a significant difference in comparison to the controls, denoting that it may play a role in the genetic pathogenesis of JIA. 相似文献

目的研究某院耐碳青霉烯类肠杆菌科细菌(CRE)在细菌耐药方面的分子流行病学特征,为CRE的防控提供依据。方法收集某院2013—2017年细菌室保存的CRE,对其进行多位点序列分型(MLST)、药敏试验、全基因序列测定,选取部分CRE中携带的碳青霉烯耐药基因进行基因环境分析。结果共收集62株CRE,成功复活51株;其中耐碳青霉烯类肺炎克雷伯菌(CRKP)30株,耐碳青霉烯类大肠埃希菌(CREC)9株,耐碳青霉烯类阴沟肠杆菌(CRECL)6株,耐碳青霉烯类其他肠杆菌6株。CRKP MLST主要包括3株ST147、2株ST11;CREC MLST主要包括3株ST167;CRECL MLST主要包括3株ST93、2株ST88。51株CRE对氨苄西林、头孢噻肟的耐药率最高,均为100%。耐碳青霉烯类耐药基因分布:1株携带blaKPC-2,14株携带blaIMP-4,18株携带blaNDM-1,22株携带blaNDM-5,2株携带blaNDM-9,10株携带blaOXA-1,10株携带blaOXA-10,2株携带blaOXA-23,2株携带blaOXA-66。分析blaNDM-1、blaNDM-5、blaNDM-9、blaIMP-4不同菌种的基因环境,发现几种耐药基因各自的基因环境都与已报道的基因环境相似,无明显的菌种间差异性。结论耐药基因通过水平传播能稳定存在于不同的CRE菌株中,对医院感染防控造成一定的威胁。 相似文献

BackgroundTo accurately identify ABO blood typing in pre-transfusion testing is very important to ensure blood transfusion safely, which is a major responsibility of blood station.MethodsEighty-one blood donors samples with ABO blood group typing discrepancy was collected among 61952 donor samples in our blood station from January 2019 to July 2020. Blood group serological method was used to detect ABO blood group. DNA Sequencing was used to determine the genotype. The antibody screening test detects antibodies other than ABO.ResultsIn total, 61,952 donor samples were analysed for ABO typing discrepancies. The incidence among blood donors was 0.13% (81/61952). The most common reason of ABO typing discrepancies was due to specific antibody or non-specific agglutination (54.32%, 44/81), mainly anti-M antibody, cold autoantibody, anti-D antibody, anti-N antibody and anti-Lea antibody. The major cause of forward typing discrepancies among blood donors was ABO subgroups (25.93%, 21/81), including 10 cases of A subtype (1 case of A2, 2 cases of A3, 2 cases of Ax, 3 cases of AxB, 1 case of Ael, 1 case of Ahm), 6 cases of B subtype (2 cases of B3, 1 case of Bel, 3 cases of AB3), 2 cases of B subtype (A), 1 case of cisAB, and 2 cases of acquired B. The serum antibody was weakened in 16 cases (19.75%).ConclusionsThe blood types should be correctly identified by combining serology with gene sequencing to ensure the safety of clinical blood transfusion, when the forward and reverse typing discrepancies among the blood donors. 相似文献

Y-chromosomal SNP (Y-SNP), with its stable inheritance and low mutation, can provide Supplementary information in forensic investigation. While commonly used Y-chromosomal STR haplotypes show their limitations, typing of Y-SNP would become a powerful complement. In this study, a 16-plex Y-SNP typing system based on allele-specific PCR (AS-PCR) was developed to discriminate four dominant Y-chromosomal haplogroups (C-M130, D-CTS3946, N-M231, and O-M175) and 12 predominant sub-haplogroups of O-M175 (O1a-M119, O1a1a1a-CTS3265, O1b-M268, O1b1a2-Page59, O2-M122, O2a1-L127.1, O2a1b-F240, O2a1b1a1-CTS5820, O2a2-P201, O2a2b1a1-M177, O2a2b1a1a1a-Y17728, O2a2b1a2-F114). A series of experimental validation studies including sensitivity, species specificity, male-female mixture and inhibition were performed. The discrimination of the typing system was preliminarily proved with a haplogroup diversity of 0.9239. Altogether, the Y-SNP typing system based on AS-PCR should be capable of distinguishing China’s dominant Y-chromosomal haplogroups in a rapid and reliable manner, thus can be employed as a useful complement in forensic casework. 相似文献

目的 了解广东省肇庆市肺炎克雷伯菌(Klebsiella pneumoniae, KP)临床分离株的抗生素耐药情况及多位点序列分型(multilocus sequence typing, MLST)特征。方法 收集肇庆市两家医院KP临床分离株63株,采用肉汤微量稀释法进行30种抗生素的体外药物敏感性试验,并对所有菌株进行多位点序列分型。结果 63株KP临床分离株的耐药谱特点可分为4类,即全敏感的高毒力肺炎克雷伯菌(hypervirulent K. pneumoniae, hvKP)、产超广谱β-内酰胺酶肺炎克雷伯菌(extended spectrum β-lactamases K. pneumoniae, ESBLsKP)、耐碳青霉烯类肺炎克雷伯菌(carbapenem-resistant K. pneumoniae, CRKP)和其他类型的多重耐药肺炎克雷伯菌(multidrug-resistant K. neumoniae,MDRKP);MLST分型结果显示63株临床分离株可分成41个ST型。其中,ESBLsKP以ST37型(9株,39.13%)为主,CRKP以ST11型(5株,62.50%)为主,hvKP以ST65(5株,25.00%)型和ST23(3株,15.00%)型为主,而其他类型的多重耐药KP的ST型(12株9种ST型)则呈现明显的多态性。结论 我市肺炎克雷伯菌临床分离株的耐药情况比较严重,要加强院内感染监测,警惕其成为优势菌型并引发院内感染的风险。 相似文献
