建立家兔系膜增殖性肾炎模型,研究API0134对家兔血脂、超氧歧化酶(SOD)及丙二醛(MDA)作用以及对肾脏组织的影响,并探讨了可能的机制。结果表明,API0134用药组血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)低于模型组,高密度脂蛋白胆固醇(HDL-C)及HDL-C/LDL-C高于模型组(P<0.05),SOD活性明显高于模型组(P<0.01),而MDA正常。用药组肾组织损害较轻。研究证实,API0134能够预防兔的高脂血症,具有抗氧化作用,能够抑制系膜细胞及基质的增殖,保护肾组织。 相似文献

目的 观察穿心莲成分 API0 1 34 (API)抗血小板活化和聚集的机制。方法 建立高脂血症家兔动脉血栓形成模型。观察穿心莲成分 API0 1 34 (API)对闭塞性血栓形成时间 (OT)、血小板聚集、血液血小板活化因子 (PAF)含量、血小板内三磷酸肌醇 (IP3)含量和 IP3受体 (IP3R)表达的影响。结果 API能够显著延长 OT和抑制血小板聚集 ,降低血液 PAF和血小板内 IP3含量 ,抑制血小板内 IP3R蛋白的表达。API 5 0 mg/ kg的抑制作用明显强于 API 5 m g/ kg。结论 API具有较强的抗血小板聚集和抗血栓形成作用 ,API对血小板 PAF- IP3/ IP3R信号途径的抑制作用 ,是 API抗血小板活化和聚集的机制之一 相似文献

The ameliorative properties of bioactive compound andrographolide (ANDRO), aqueous extract of Andrographis paniculata (AE-AP) and vitamin E (vit.E) were tested against nicotine induced liver, kidney, heart, lung and spleen toxicity. A group of male Wistar rats were intraperitoneally administered vehicle, nicotine (1 mg/kg body weight/day), nicotine + ANDRO (250 mg/kg body weight/day), nicotine + AE-AP (250 mg/kg body weight/day) and nicotine + vit.E (50 mg/kg body weight/day) for the period of 7 days. The significantly increased levels of lipid peroxidation, protein oxidation and the decreased antioxidant enzyme status were noted in nicotine treated group as compared to vehicle treated group. ANDRO, AE-AP and vit.E significantly reduced the lipid peroxidation, protein oxidation and increased the antioxidant enzyme status. This indicates A. paniculata and vit.E may act as putative protective agent against nicotine induced tissue injury and may pave a new path to develop suitable drug therapy. 相似文献

Context: Andrographis paniculata (Burm.f.) Nees (Acanthaceae) is widely used in tribal medicine in India and some other countries for multiple clinical applications. It contains andrographolide (AG) (diterpenoid lactone), a major phytomarker which probably accounts for its medicinal properties.Objective: This study investigates the site-specific distribution of AG in different tissues of rats and its pharmacokinetic parameter evaluation by using a validated liquid chromatography tandem mass spectrometry (LC-MS/MS) method.Materials and methods: A simple and sensitive LC-MS/MS method has been developed and validated to quantify the presence of AG in plasma and various tissues of rat following oral administration of A. paniculata extract and AG in a dose of 133.33 and 100?mg/kg/day, respectively, for four weeks.Results: The present study showed that the highest concentration of AG was in kidney (156.12?ng/g) followed by liver, spleen and brain while almost same concentration was found in heart and lung. The apparent Cmax, Tmax, elimination half-life and total exposure (AUC0–α) were 115.81?ng/ml, 0.75, 2.45 and 278.44?ngh/ml, respectively.Conclusion: This was an attempt to determine the presence of AG (a known biomarker) in tissues such as kidney, heart, lungs, brain and plasma of rats using a validated LC-MS/MS method. 目的:探讨穿心莲和当归对小鼠红细胞血红蛋白再释放的影响。方法:将实验用小鼠雌雄各半按体重均匀分为7组(6个用药组和1个对照组),按实验设计分别对穿心莲和当归高、中、低浓度用药组小鼠用穿心莲和当归水煎液进行灌胃,对照组用生理盐水灌胃,每天1次,连续灌胃15 d,然后分别采血分离红细胞观察其血红蛋白再释放的情况,并用BandScan扫描定量,进行统计分析。结果:中、高浓度的穿心莲和高浓度的当归都能使小鼠红细胞血红蛋白的再释放增强。结论:穿心莲和当归水煎液均能影响实验小鼠的血红蛋白再释放,但影响程度随浓度不同而有差异。 相似文献
