A radioimmunoassay (RIA) using polystyrene beads as the solid phase for cytomegalovirus (CMV) antigen and iodinated immunosorbent purified anti-human IgG, IgM, and IgA as indicator antibodies was developed for the detection of immunoglobulin class-specific antibodies to CMV. An antigen prepared from extracellular virus was essential for reliable results, and a preparation ultracentrifuged and sonicated twice was better than a crude antigen. The optimal antigen gave low cpm values with a negative reference serum, resulting in cpm ratios of 10 or higher between early convalescent phase serum and negative reference serum. Of six patients with an increase in CMV CF titres, all six had an increase in RIA IgG titres, four had an increase in IgA titres, and all had IgM antibodies. The IgG titres were high, up to 1/64,000. In a group of 17 infants negative in CMV CF test, 14 had CMV IgG antibodies in RIA test, indicating mainly low levels of maternal antibodies. In six of seven patients with CMV isolations from urine specimens, an increase in IgG or IgA titres or the presence of IgM antibodies was found, and only one of these patients had an increase in CMV CF titre. The specificity of the developed CMV RIA test was further demonstrated by detecting no significant increase in RIA titres in serum specimens of patients with primary herpes simplex infection, chickenpox, herpes zoster, or infectious mononucleosis.  相似文献

Cancer metastasis involves distinct steps that depend on complicated tumor–host interactions. The hematogenous dissemination of tumor cells may be facilitated by factors that promote the arrest and adherence of cancer cells in capillaries. We examined whether anti-tumor monoclonal immunoglobulin M (IgM) antibodies promoted the hematogenous dissemination of B16 melanoma cells in syngeneic mice. IgM monoclonal antibodies were generated that selectively bind to B16 melanoma cells as compared to syngeneic fibroblasts, lymphocytes or Lewis lung carcinoma cells. Incubation of B16-BL6 or B16-F0 melanoma cells with these IgM anti-tumor antibodies significantly increased the number of lung colonies as compared with control antibodies. Moreover, intraperitoneal injection of specific antibody also significantly increased lung colonization. All anti-tumor antibodies promoted the aggregation of B16 melanoma cells. A chemically generated immunoglobulin G (IgG)-like fragment of an anti-tumor IgM antibody displayed greatly reduced tumor aggregation and, in contrast to intact IgM, did not significantly increase lung colonization of B16 melanoma cells. Neither intact IgM nor the IgG-like fragment enhanced the in vitro invasiveness of B16 melanoma cells across Matrigel-coated membranes. Our results, therefore, suggest that besides their beneficial anti-tumor effects, anti-tumor IgM antibodies may also promote the hematogenous dissemination of cancer cells. To develop improved reagents for mumps serology a high-level yeast expression system was employed to produce recombinant mumps nucleoprotein (rNP). The rNP was purified by CsCl gradient centrifugation and yielded approximately 15 mg/l of yeast culture. Electron microscopy of the rNP revealed characteristic herring-bone structures. The electrophoretic mobility of rNP in yeast cells was similar to native NP in SDS-PAGE. Monoclonal antibodies to rNP reacted with native mumps virus nucleoprotein by immunofluorescence assay. A monoclonal antibody to native mumps virus NP reacted with rNP by Western blot assay. The rNP was investigated as antigen in an IgM capture enzyme immunoassay (EIA) using a horseradish peroxidase conjugate of monoclonal antibody to the rNP. Eighteen sera previously found to be positive by IgM capture radioimmunoassay (MACRIA) and 30 sera that were mumps IgM negative by MACRIA were tested by mumps IgM capture EIA. The results for the two test were concordant. In addition, 26 rheumatoid factor positive sera and 35 sera that were IgM positive for measles, rubella or parvovirus B19 were tested. Fifty-nine sera were negative by mumps IgM capture EIA but two sera collected from two infants 3 and 6 weeks after mumps, measles and rubella vaccination were positive. Mumps MACRIA confirmed these results. Compared to MACRIA the overall sensitivity was 100% (20/20) and specificity was 96.8% (30/31). The yeast expressed rNP was highly immunogenic and suitable for use in IgM capture EIA for the diagnosis of mumps.  相似文献

Competitive-inhibition enzyme immunoassays for the measurement of human IgG, IgA and IgM are described. These assays can be readily performed with commercial antisera and a recently developed method for purifying human IgA and IgM with high yield. The assays described are specific, with undetectable (<0.5%) cross-reactivity between the immunoglobulin classes in all systems, except with purified IgM, which cross-reacted to 1.9% with the IgG enzyme immunoassay.Minimal detectable concentrations of 2.5±0.8 ng/ml for IgG 4.2±0.9 ng/ml for IgA and 7.2±1.4 ng/ml for IgM were recorded, indicating that these assays are particularly sensitive. There is little within-assay variation (mean coefficient of variation = 3.9%), although the between-assay variation was substantially greater (mean coefficient of variation = 23.5%). These assay systems appear to be particularly suited to the measurement of immunoglobulin production by lymphocytes in culture. In such studies the assay must be specific, sensitive and be capable of discriminating between levels of immunoglobulin produced in response to various experimental treatments.  相似文献

目的了解丙型肝炎病毒(HCV)感染及病毒血症存在情况。方法用酶联免疫吸附试验(ELISA)和聚合酶链反应(PCR)对不同人群的血清标本做了抗-HCV、抗-HCVIgM和HCVRNA的检测,并对三项指标间的关系进行了对比分析。结果抗-HCV在普通成年人、献血员、急性肝炎和肝硬化患者中的检出率分别是357%,858%,625%和4838%;与HCVRNA的符合率分别是1143%,6111%,800%和7333%。相同人群抗-HCVIgM与HCVRNA的符合率分别是75%,909%,8181%和100%。结论抗-HCVIgM比抗-HCV能更客观地反映HCV病毒血症的情况,个别抗-HCV阴性血清检测到了抗-HCVIgM和HCVRNA。  相似文献

目的探讨IgM相关性肾小球疾病的治疗。方法53例患者分为雷公藤多甙片组(A组)和雷公藤多甙片+中药益肾活血(B组)治疗,疗程2个月。结果两组治疗前后尿蛋白均下降(P<0.05),组间比较则B组降低更明显(P<0.05)。尿红细胞均有减少,A组P>0.05,B组P<0.05;组间比较P<0.05。结论雷公藤多甙片+中药益肾活血法治疗IgM相关性肾小球疾病较单用雷公藤多甙片疗效显著提高。认为辨病辨证相结合组方用药,能够提高疗效。  相似文献

目的 为对麻疹疑似病例作出科学诊断。方法 采用酶联免疫吸附试验(ELISA),对麻疹疑似病例的血清标本进行麻疹特异性IgM抗体检测。结果 1992年以来收集到的187例麻疹疑似病例检测IgM抗体阳性率为45.5%,其中2~6岁阳性率最高(52.6%),19例麻疹疑似病例进行风疹IgM抗体检测,结果风疹IgM抗体阳性率为20.1%,说明麻疹疑似病例中有相当部分为风疹病例。阳性病例中无免疫史的占21.2%。结论 麻疹疑似病例不能单以临床症状进行诊断,应通过实验室检测IgM抗体才能准确诊断。进一步提高人群免疫水平是防止病例发生的最经济有效的措施。  相似文献

目的评价麻疹IgG抗体亲和力试验对判定麻疹病例的价值。方法以中国疾病预防控制信息系统2013-2015年天津市麻疹实验室确诊病例和麻疹排除病例为研究对象。回顾性追溯保存的病例血清,开展麻疹IgG抗体亲和力试验,重新对麻疹排除病例进行归类。结果共收集到326例麻疹病例血标本,其中实验室确诊病例267例,排除病例59例,≥20岁病例占92.33%(301/326)。麻疹IgG抗体亲和力试验显示,确诊病例和排除病例中麻疹IgG高亲和力抗体的比例分别为66.95%(158/236)和91.23%(52/57),差异有统计学意义(χ2=13.33,P<0.001)。根据判定标准,15.25%(9/59)排除病例被重新判定为麻疹病例,其中8例是高亲和力抗体,有含麻疹成分疫苗(MCV)免疫史,判定为继发性免疫失败病例;1例为低亲和力抗体,有典型的麻疹临床症状,无MCV免疫史。结论麻疹IgG抗体亲和力试验能够提供有参考意义的血清学证据,可以减少麻疹急性期血清学诊断中由于IgM抗体假阴性而造成的错误排除。  相似文献

Receptors for IgM were detected on peripheral blood human eosinophils by a rosette technique with ox red blood cells coated with the IgM fraction of the specific immunserum. Between 14 % and 43 % (mean 27 %) FcµR positive cells were found after an overnight incubation period at 37°C by using this technique. The specificity of the receptors for IgM was assessed by studying the inhibitory capacity of purified human IgM in the rosette assay. From an ultrastructural point of view, the EAM rosette-forming cells are mature eosinophlic granulocytes characterized by a nucleus with a variable number of lobes and a certain number of «first type» granules partially or totally devoid of their content.  相似文献

本文ELLSA检测,对55例HBV感染者血清中的抗-HBcIgM随访研究其动态变化。结果发现,急性乙肝35例抗-HBcIgM的阳性率在入院时,6个月,12个月分别为100%(35/35),86%(31/36),及34%(6/18),提示抗-HBcIgM在6个月大都保持阳性,至12个月大部分消失;“无症状”携带者20例抗-HBcIgM阳性率在首检时,6个月,24个月都为100%(各为20/20,17/17,及10/10),提示“无症状”携带者的抗-HBcIgM很难自然消失,同时把抗-HBcIgM和HBsAg检测对比,提示HBsAg阴性不能排除急性乙肝的诊断。  相似文献
