In an effort to settle the conflicting views on the proliferation kinetics of Kupffer cells (Kc), we performed 2/3 partial hepatectomy on rats injected with Pelikan ink. Using an anti-rat macrophage monoclonal antibody, ED 2, we evaluated the numerical changes in total, carbon-positive ED 2+ cells and carbon-negative ED 2+ cells in the portal and central area. We also analyzed the ultrastructure and peroxidase cytochemistry of various types of cells observed during regeneration. The total numbers of ED 2+ cells in the remaining liver increased rapidly from day 2 to 5, and the number of dividing ED 2+ cells reached a maximum on day 2. Thus, the numerical increase in ED 2+ cells corresponded to the division phase. In contrast, the carbon-labeling experiment showed a continuous increase of carbon negative ED 2+ cells from day 2 to 7. In the central area where division was less frequent, the proportion of carbon-positive cells decreased markedly to 50% on day 7, as against 97% in control rats. These findings suggest the possibility of an influx of carbon-negative Kc in addition to cell division. Ultrastructurally, the presence of carbon-negative "small Kc" and "immature Kc" with morphological features different from those of carbon-positive Kc was demonstrated. Such carbon-negative Kc with a high nucleus-to-cytoplasm ratio and rather few phagosomes, were not observed in control rats. Furthermore, we demonstrated two types of possible precursor cell, i.e. "transitional" forms between monocytes and Kc, and "immature macrophages". The former showed peroxidase activity in some lysosomes as well as in the rough endoplasmic reticulum and nuclear envelope. Our result indicated that the proliferation kinetics of Kc depend upon both local proliferation and influx.  相似文献

采用4天抑制试验法,观察青蒿提取物和青蒿素对伯氏疟原虫超微结构的影响。结果显示,青蒿提取物和青蒿素均使原虫滋养体膜结构损伤,表现为虫体表膜、食物泡膜、限制膜、线粒体膜、内质网、核膜呈现肿胀及膜间隙增宽。有些虫体表膜、食物泡膜呈现多层螺纹膜样改变。严重病变的原虫滋养体退变崩解,结构消失,残留退变的食物泡和吞噬泡散在红细胞内。此外,还观察到青蒿提取物和青蒿素对疟原虫纳虫泡内裂殖子发育有抑制作用。试验证明,青蒿提取物和青蒿素对伯氏疟原虫滋养体的超微结构损伤部位和病变特征基本相同;并提示对疟原虫纳虫泡内裂殖子发育有一定影响。  相似文献

Carcinoid tumor of the pancreas is extremely rare. This article describes the case of a 22-year-old woman who manifested a typical carcinoid syndrome; a definitive diagnosis of a metastatic carcinoid tumor was made from needle aspiration of a supraclavicular lymph node using morphologic, immunocytochemical, and ultrastructural criteria. The carcinoid tumor was subsequently shown to be of pancreatic origin.  相似文献

Summary Ultrastructural changes in central nervous system (CNS) white matter of three goats affected with-mannosidosis were analyzed to further define characteristics and pathogenesis of axonal and myelin abnormalities. The variations in myelin association and contents of axonal spheroids were delineated. The occurrence of spheroids in a 96/150-day fetus documented the early development of these axonal lesions. In regions of severe myelin deficits, the presence of apparently normal axons and a reduction in the number of oligodendrocytes were confirmed. Many remaining cells in myelin-deficient regions were characterized by dark, vacuolated cytoplasm. The occurrence of internodes with myelin sheaths adjacent to internodes without myelin sheaths suggested that an axonal defect is not primarily responsible for the absence of myelin sheaths. A mild myelin deficit in the spinal cord was indicated by the presence of unmyelinated axons. Except for occasional mild cytoplasmic vacuolation, the spinal cord glial cells appeared relatively normal. The findings presented here are consistent with the hypothesis that an oligodendrocyte defect, expressed by regional differences, is a major factor in the pathogenesis of myelin deficiency in-mannosidosis.Supported by NIH grant NS-16886 to MZJ and BRSG funds from the College of Osteopathic Medicine, Michigan State University, to KLL  相似文献

Summary The aim of this electron microscopy study was to further investigate the effects of cis-dichlorodiammineplatinum (cis-DDP) on the cerebellum of the immature rat. Ten-day-old animals were treated with cis-DDP subcutaneously and killed after 1, 7, 15 or 21 days. On postinjection day 1, cis-DDP effects were evident mainly in the external granular layer, with nuclear damage in many dividing cells, while their cytoplasm appeared to be less affected. Some binucleate cells were also present. On the contrary, in postmitotic or more differentiated cells, only cytoplasmic alterations were found. At later stages (postinjection day 7), the frequency of damaged cells in the external granular layer decreased, but there was a cellular deficit in the internal granular layer. Many postmitotic neurons underwent coagulative necrosis. Finally (postinjection days 15 and 21), the cellular deficit was partly compensated for by reactive structures, e.g., glial cell fibers, which underwent hypertrophy after initial edema. 目的:电镜下观察老龄大鼠嗅球内神经元超微结构改变。方法:2%多聚甲醛、戊二醛混合固定液灌流大鼠,取嗅球,制备超薄切片,电镜观察。结果:突触小球、小球周细胞、僧帽细胞和刷细胞数量减少,核膜断裂,染色质外溢,粗面内质网、高尔基复合体减少。线粒体固缩或形成髓鞘样小体。结论:嗅球内突触小球和神经元减少,细胞器老化可能是导致老年性嗅觉障碍的主要原因。  相似文献

目的:观察弓形虫包囊的超微结构,鉴定体外细胞培养所形成的包囊。方法:体外用HeLa细胞作载体培养PP株弓形虫形成包囊;体内用感染Fukaya株弓形虫的小鼠脑包囊,分别做电镜标本并观察。结果:两者都观察到包囊的特征性超微结构且结果相类似。结论:弓形虫在体外一定培养条件下可以形成包囊,并与体内包囊的超微结构相似。  相似文献

目的:了解角膜内皮细胞损伤的超微结构特征及其临床意义。方法:对20只成年灰兔进行定量角膜内皮细胞损伤,伤后定期进行眼部裂隙灯检查、角膜厚度及眼压测定,预期取角膜材料进行透射电镜和扫描电镜检查。结果:角膜内皮细胞损伤后1天可见细胞膜破损,细胞肿胀,细胞器颗粒粗大;伤后3天内皮细胞崩解,胞质脱落,胞核裸露,伤后5天出现细胞缺损区,膜脂降解聚积而形成脂质球,附着于细胞缺损区,角膜上皮细胞音隙增宽;伤后7天角膜内皮细胞移行修复细胞缺损区;伤后9天后内皮细胞异形明显。上皮细胞出现变性。角膜混浊与角膜厚度呈正相关,眼压增高者角膜厚度增加,内皮细胞损伤加重。结论:超微结构结果提示,角膜内皮细胞受损后细胞崩解似科是必然的结局。动态测定角膜厚度可以判断角膜内皮细胞损伤的程度,控制眼压是预防内皮细胞损伤加重的重要措施之一。  相似文献

对70例不同胎龄及生后不同发育阶段Wistar大鼠睾丸支持细胞的发育进行了光镜、电镜研究。结果:1.依形态学特点,将支持细胞的发育过程初步分为4期:分化前期(胚胎11~15d)、分化期(胚胎15d~生后1至2周)、成熟前期(生后2周~青春期前)、成熟期(青春期后)。2.胚胎及新生时,在支持细胞间见到两侧胞膜紧密相贴及不典型的桥粒连接等;自生后4周,见到紧密连接。3.自胚胎17d至成熟以后,在支持细胞与其周围的生殖细胞间见到大量类似桥粒和类似中间连接等不典型的细胞连接。4.胚胎19d及新生期,睾丸索内除支持细胞及生殖细胞外,尚可见一种特殊的细胞。  相似文献

目的 观察大鸨胰腺超微结构 ,探讨大鸨胰腺功能。 方法 透射电镜观察 3例大鸨胰腺。 结果 大鸨的胰外分泌部为管泡状腺 ,腺小叶由于缺乏叶间结缔组织而界限不如哺乳动物的明显。叶间隙似导管 ,小叶间导管管壁简化以至由胰腺分泌细胞代替。仅在 3个小叶间能见到单个长梭状结缔组织细胞 ,其胞质变细伸展进入两叶间隙中间构成 1条中等密度线。外分泌细胞可分为明暗两种。胰的内分泌细胞呈岛状或单个散布于胰外分泌腺中。胰岛分A和B两种 ;A胰岛仅由A细胞构成。B胰岛主要由B和D两种细胞构成 ,其中B细胞数量多 ;D细胞含量少。 结论 大鸨胰腺具有一些不同于其他动物及适应于飞翔的结构特点  相似文献
