Mutations in PAX6/Pax6 lead to a variety of ocular anomalies in humans and mice. The aim of the study was to characterise the ocular abnormalities caused by the missense Pax6Leca4 mutation and compare them to published observations on Pax6 alleles that are functionally equivalent to Pax6 null alleles (such as Pax6Sey and Pax6Sey-Neu) and human inherited eye diseases. Ocular features of homozygous Pax6Leca4/Leca4 and heterozygous Pax6Leca4/+ embryos at E12.5-E18.5, heterozygous Pax6Leca4/+ young mice at P18 and heterozygous Pax6Leca4/+ adults at 12 weeks were analysed histologically with their wild-type Pax6+/+ littermates. Homozygous Pax6Leca4/Leca4 fetuses died perinatally with no eyes although an optic cup rudiment with pigmented cells developed. Pax6Leca4/+ mice were microphthalmic and a range of other severe ocular phenotypes affected both the anterior and the posterior segments. In contrast to Pax6+/−, the Pax6Leca4/+ eyes had no goblet cells in the corneal epithelium, the iris was not hypoplastic and there was no lens-corneal epithelial plug. However, microphthalmia was more severe, corneal vascularisation occurred earlier (during fetal stages), pigmented cells were present in the vitreous and corneal stroma and the ciliary body was malformed or abnormal. These results show that, although Pax6Leca4/+ lacked some eye abnormalities commonly seen in Pax6Sey/+ and Pax6Sey-Neu/+ eyes, in most respects their eyes were more severely affected. These differences probably reflect both differences between the Pax6Leca4 and the Pax6Sey-Neu mutations and differences in modifier gene expression in different genetic backgrounds. The presence of pigmented cells in the cornea is a novel observation. Some Pax6Leca4/+ ocular abnormalities were similar to those present in human Peters' anomaly and persistent hyperplastic primary vitreous (PHPV) so Pax6Leca4/+ mice provide a useful model for some inherited eye diseases.  相似文献

目的 研究甲醛对小鼠的致突变作用。方法 选择健康昆明种小鼠 5 0只 (雌雄各半 ) ,随机分为 3个剂量组 :甲醛高剂量组 (2 0 0 0mg/kg·bw)、中剂量组 (2 0 0mg/kg·bw)、低剂量组 (0 2 0mg/kg·bw) ,1个阴性对照组 (生理盐水 )和 1个阳性对照组 (环磷酰胺 5 0mg/kg) ,采用腹腔注射染毒 ,每天 1次 ,连续 5d。于第六天处死雌性小鼠进行骨髓细胞微核试验 ,于第三十五天处死雄性小鼠进行精子畸形试验 ,显微镜下观察并计数微核及畸形精子数。结果 甲醛中、高剂量组骨髓细胞微核数显著高于阴性对照组 (P <0 0 5 ;) ,同时低、中、高剂量组精子畸形率显著高于阴性对照组 (P <0 0 1)。结论 甲醛对小鼠具有致突变作用  相似文献

目地研究双眼视网膜母细胞瘤患者Rb1基因杂合性突变的分子生物学特性。方法应用PCR—SSCP直接测序技术检测双眼视网膜母细胞瘤患者白细胞DNA中Rb1基因杂合性突变。结果50例证实有Rb1基因杂合性突变的病例中有1例发生于第16内含子中可以用3种定位方法解释、具有相同序列的21个碱基缺失。结论这种极为少见的Rb1基因突变方式可能是由于破坏了正常拼接位点的结构而激活了“隐蔽拼接位点”,导致异常的Rb1基因mRNA产生或由此影响整个拼接过程。  相似文献

We have identified the A3243G heteroplasmic point mutation in mitochondrial DNA from a female patient with headache as the main clinical feature. The mitochondrial origin of her disease was only suspected because of her brother with MELAS syndrome. Morphological and biochemical studies failed to reveal mitochondrial respiratory chain dysfunction in her muscle which contained 65% of mutated mitochondrial DNA molecules. Molecular studies performed among four generations (in the blood of seven subjects) showed the variable transmission of mutated molecules and pointed out the difficulty in giving genetic counsel.  相似文献

We performed an immunohistochemical analysis to investigate the expression of p53 protein in a panel of 18 laryngeal squamous cell carcinomas, 15 primary tumours and three in relapse, previously analysed by us for the presence of p53 gene mutations. Dysplastic and/or normal surrounding mucosa was evaluated in 15 different tumours. The results of our study are the following: (1) expression of p53 protein was observed in one out of five tumours positive for p53 gene mutations (20%) and in 10 out of 13 (80%) negative cases; (2), p53 protein over-expression was frequently observed in normal and/or dysplastic mucosa surrounding either wild-type (7/11) or mutated p53 tumours (2/4); (3), p53 immunoreactive cells showed a pattern of distribution in normal and mildly/moderately dysplastic mucosa (basal layers), different from that in severely dysplastic mucosa (whole thickness). These data further support the hypothesis that p53 protein over-expression may be a marker of the earliest phases of multistep tumorigenesis in laryngeal squamous cell carcinoma.  相似文献

【目的】调查线粒体NADH脱氢酶亚单位 1(ND1)基因的T3394C位点突变与中国人糖尿病的发病情况。【方法】随机收集 338例无血缘关系的糖尿病患者及 2 6 2例正常对照组 ,用聚合酶链反应扩增、限制性内切酶HaeⅢ消化进行突变筛查 ,DNA序列分析确认。【结果】糖尿病患者中T3394C突变者共 8例 (2 37%) ,正常对照组中只有 1例 (0 38%,P <0 0 1)。糖尿病突变患者中 4例有糖尿病家族史 ,4例病人由于胰岛素分泌功能下降和 /或出现并发症 ,需用胰岛素治疗。【结论】ND1/T3394C突变在糖尿病患者中的发生率明显高于正常对组 ,提示这个位点突变可能是导致线粒体糖尿病的易感基因之一。  相似文献

目的探索可溶性HLAG1分子结构对功能的影响,为其临床应用打下基础。方法通过分子克隆技术,去掉HLAG1重链分子α1功能区氨基端24肽,然后与轻链蛋白在体外与九肽共折叠复性形成复合物,并检测复合物对NK细胞杀伤活性的影响。结果成功构建突变的HLAG1重链分子的原核表达载体,表达的重链蛋白与轻链蛋白形成复合物,经Westernblot鉴定可与HLAⅠ类分子的单抗W6/32结合,并且可明显抑制NK细胞对K562细胞的细胞毒作用。结论α1功能区氨基端缺失24肽的HLAG1分子,可抑制NK细胞的细胞毒作用。  相似文献

BACKGROUND: Gitelman's syndrome (GS) is an autosomal recessive disorder resulting from inactivating mutations in the thiazide-sensitive Na-Cl co-transporter (NCCT) gene. To date, almost 90 mutations have been identified. It is possible that there is a population-specific distribution of mutations. In this study, we analysed mutations in the NCCT gene of seven Japanese patients with GS. METHODS: Peripheral blood mononuclear cells were isolated from patients with GS, their family members and healthy control subjects. A mutation analysis of the NCCT gene was performed completely by direct automated sequencing of polymerase chain reaction-amplified DNA products. In patients with a deletion or splice site mutation, we undertook cDNA sequence analysis. RESULTS: We identified nine mutations. Five of them [c.185C>T (Thr60Met), c.1712C>T (Ala569Val), c.1930C>T (Arg642Cys), c.2552T>A (Leu849His) and c.1932delC] have been reported in Japanese patients, but not in GS patients from other ethnic groups. The remaining four mutations [c.7A>T (Met1Leu), c.1181_1186+20del26, c.1811_1812delAT and IVS16+1G>A] were novel. In cDNA derived from a patient with c.1181_1186+20del26, a deletion of exon 9 and a frameshift at the start of exon 10 were observed. In cDNA derived from patients with IVS16+1G>A, an additional 96 bp insertion between exons 16 and 17 was observed. Six out of seven patients were compound heterozygotes, and the remaining one carried a single heterozygous mutation. CONCLUSIONS: We found four novel mutations in the NCCT gene in seven Japanese patients with GS. Moreover, our study suggests that the distribution of mutations in the NCCT gene in Japanese GS patients potentially differs from that in other populations.  相似文献
