目的:建立舒心颗粒中有效成分丹参素钠的含量测定方法.方法:用HPLC法测定.以十八烷基硅烷键合硅胶为填充剂,色谱柱Dirmonsil C18(4.6mm×250mm,5μm),流动相甲醇-1%磷酸(15:85),流速0.8mL·min-1;检测波长280nm.柱温30℃.结果:丹参素钠在0.374~3.366μg与其峰面积值呈良好的线性关系(r=0.9999),精密度RSD为0.43%,平均回收率为97.77%,RSD为1.22%.结论:该方法简便易行,结果准确可靠,可用于舒心颗粒产品质量控制.  相似文献

目的:探讨注射用核糖核酸Ⅱ安全与合理性专项评价研究。方法:本研究按月份分层随机抽取某院2016年9月-2017年8月临床使用核糖核酸Ⅱ的病例270份,药品说明书定量评价、药物利用研究及药品不良反应/事件Logistic回归分析综合评价方法,初步评价核糖核酸Ⅱ的安全合理性。结果:依据药品说明书评价结果提示合理性较高,其中给药剂量(100%),溶媒选择(100%),溶媒剂量(72.59%),给药浓度(74.07%),给药频次(98.15%),给药时间(100%),6项指标12个月内波动幅度与均值保持一致(90.80%);药物利用研究结果提示,存在超剂量、超浓度给药情况(aDDDs=5.68,aDDCs=7.37,dDUI=1.19,cDUI=1.53)。研究期内发现4例不良反应,均为一般不良反应,经Logistic回归分析提示与药物使用浓度有关。结论:此种评价方法可有效评价药物临床使用的安全合理性,应加强我院核糖核酸Ⅱ临床监测工作,尤其是超剂量、超浓度给药情况。  相似文献

The present study was designed to investigate the effects of p-cymene on lipopolysaccharide (LPS)-induced inflammatory cytokine production both in vitro and in vivo. The production of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), interleukin-6 (IL-6), and interleukin-10 (IL-10) in LPS-stimulated RAW 264.7 cells and C57BL/6 mice was evaluated by sandwich ELISA. Meanwhile, the mRNA levels of cytokine genes were examined in vitro by semiquantitative RT-PCR. In a further study, we analyzed the activation of nuclear factor-κB (NF-κB) and mitogen-activated protein kinase (MAPK) signaling pathways by western blotting. We found that p-cymene significantly regulated TNF-α, IL-1β, and IL-6 production in LPS-stimulated RAW 264.7 cells. Furthermore, the levels of relative mRNAs were also found to be downregulated. In in vivo trail, p-cymene markedly suppressed the production of TNF-α and IL-1β and increased IL-10 secretion. We also found that p-cymene inhibited LPS-induced activation of extracellular signal receptor-activated kinase 1/2, p38, c-Jun N-terminal kinase, and IκBα. These results suggest that p-cymene may have a potential anti-inflammatory action on cytokine production by blocking NF-κB and MAPK signaling pathways.  相似文献

Despite of the variety of combined modality treatments for laryngeal carcinoma have been introduced, the distance recurrence rate and 5-year overall survival rate over the past decades are still the major issues, underlining the importance to better understand the biological bases that contribute to disease progression. Here, we reported that miR-423-3p overexpressed in primary laryngeal carcinoma cell line where it plays a critical role in tumor progression. Suppression of miR-423-3p expression resulted in decreasing cell proliferation, clonogenicity, cell migration and invasion. By using in silico prediction algorithms for target identification, AdipoR2 (adiponectin receptor 2) and DUSP4 (MAP kinase phosphatase 2) were identified to be potential targets of miR-423-3p. Overexpression of miR-423-3p was associated with epigenetic silencing of AdipoR2 in human laryngeal carcinoma samples, which have been previously implicated in suppression of tumor proliferation and angiogenesis. Luciferase reporter assays and western blot further confirmed the direct interaction of miR-423-3p with AdipoR2. Our findings have demonstrated that miR-423-3p plays an important oncogenic role in laryngeal carcinoma progression, and further suggest that suppression of miR-423-3p expression might be useful for its clinical management.  相似文献

Chronic long-term exposure to cuprizone causes severe brain demyelination in mice,which leads to changes in locomotion,working memory and anxiety.These findings suggest the importance of intact myelin for these behaviors.This study aimed to investigate the possible behavioral changes in mice with mild oligodendrocyte/myelin damage that parallels the white matter changes seen in the brains of patients with psychiatric disporders.We used the cuprizonetreated mouse model to test both tissue changes and behavioral functions(locomotor activity,anxiety status,and spatial working memory).The results showed that mice given cuprizone in their diet for 7 days had no significant myelin breakdown as evaluated by immunohistochemical staining for myelin basic protein,while the number of mature oligodendrocytes was reduced.The number and length of Caspr protein clusters,a structural marker of the node of Ranvier,did not change.The locomotor activity of the cuprizonetreated mice increased whereas their anxiety levels were lower than in normal controls;spatial working memory,however,did not change.These results,for the first time,link emotion-related behavior with mild white matter damage in cuprizone-treated mice.  相似文献

Brachytherapy (BT) delivers integrated boost doses to the central tumor while sparing the surrounding organs at risk (OARs) efficiently. It’s a mandatory treatment component for locally advanced cervical cancer (LACC) because it results in excellent overall survival and local control compared with other dose boosting modalities. Currently, BT is undergoing a transition from 2-dimensional (2D) to 3-dimensional (3D) treatment planning. Imaging-guided BT (IGBT) employing computed tomography (CT) or magnetic resonance imaging (MRI) can provide exact individual delineation of target and OARs meanwhile prescribe the dose to the target volume instead of “point A” for X-ray-based BT. There are three main techniques for BT: intracavitary (IC), interstitial (IS), and intracavitary/interstitial (IC/IS) combination. The applicator choice depends on the specific tumor extension. The real-time transabdominal ultrasound (US)-guided applicator placement technique is strongly recommended to ensure ideal applicator positioning. MRI is the ideal standard imaging for BT owing to its superior soft tissue visualization than CT. However, CT-based BT is more often performed because of the availability. In developing countries, US-based BT can be adopted. For treatment planning, the applicator reconstruction is easier on CT than on MRI, because the applicator image is more clearly visible. Individual treatment planning should be performed for every single applicator insertion to ensure dose accuracy. In this review article, we explain the main clinical process and common techniques, including the applicator choice and placement, imaging techniques, target delineation, and treatment planning; asthose will help to improve the efficiency of 3D BT.  相似文献

腹腔间隔综合征(abdominal compartment syndrome,ACS)是开腹手术较为罕见的并发症,死亡率高。结合1例宫颈恶性肿瘤根治术后ACS合并切口裂开的病例,强调对于高危患者准确监测腹内压和及时剖腹手术的重要性。该患者极度肥胖,体质量指数(BMI)=40 kg/m^2,两次新辅助化疗后行宫颈癌根治术(经腹广泛子宫、双侧卵巢、双侧输卵管切除术,盆腔淋巴结切除术),术后第1天,持续腹胀咳嗽;术后第4天出现ACS,行开腹减压术。术后恢复良好,经过10个月的随访,未见并发症。  相似文献

Previous studies reported that sciadopitysin (Sc), a type of biflavonoids, protects reactive oxygen species (ROS)-mediated osteoblast dysfunction, but its role in osteoclastogenesis remains unclear. In this study, we observed that Sc dose-dependently suppressed RANKL-induced osteoclastogenesis and bone resorption. Our results indicated that Sc treatment strongly reduced RANKL-induced osteoclast-specific genes expression, including cathepsin K (CTSK), tartrate-resistant acid phosphatase (TRAP) and MMP-9. Furthermore, Sc apparently attenuated RANKL-increased expressions of c-Fos and NFATc1. Meanwhile, Sc also strikingly inhibited the activation of NF-κB without altering the phosphorylation of MAPKs (p38, JNK and ERK1/2). Finally, our study demonstrated that Sc administration could reverse the bone loss in LPS-induced mice model. This study suggests that Sc inhibits RANKL-induced osteoclastogenesis and bone loss by inhibiting NF-κB activation and reducing the expression of c-Fos and NFATc1. Therefore, Sc might be benefit for RANKL-mediated osteolytic bone diseases.  相似文献
