神经纤毛蛋白-1(Neuropilin-1,NRP-1)是一种非酪氨酸激酶跨膜蛋白,可表达于多种肿瘤细胞及免疫细胞,与不同配体结合后参与轴突形成、血管发育、免疫调节和肿瘤发生等过程。最近,NRP-1在肿瘤免疫学领域引起广泛关注,其在肿瘤免疫微环境中发挥着强大的双重作用,不仅可协调免疫细胞介导的免疫抑制作用,同时可限制抗肿瘤免疫反应,与肿瘤的发生发展密切相关。越来越多的研究表明,NRP-1是肿瘤免疫治疗中独特的免疫调节分子,是最有前景的免疫检查点分子,深入探讨NRP-1在肿瘤免疫调节中的作用机制,有望为以NRP-1为靶点的免疫治疗提供新策略。本文就国内外关于NRP-1在肿瘤免疫调节作用中的最新研究进展作一综述。  相似文献

目的:探讨P50型EX-PRESS引流钉结合生物羊膜植入术在原发性开角型青光眼(POAG)中的治疗效果。

方法:选择2013-03/2019-01收治的POAG患者96例96眼为研究对象,以随机数字表法将患者分为观察组和对照组各48例,对照组单纯植入P50型EX-PRESS引流钉,观察组在对照组基础上结合生物羊膜植入术治疗,观察两组滤过泡情况,术前、术后7d,1、3mo的眼压、视网膜神经纤维厚度(RNFLT)变化,并发症发生情况及手术成功率。

结果:观察组术后3mo功能性滤过泡占比显著高于同期对照组(P<0.05); 两组术前眼压、RNFLT均无差异(P>0.05),观察组术后7d,1、3mo的眼压及RNFLT均明显低于同期对照组(P<0.05); 观察组与对照组并发症发生率分别为12%、25%(P>0.05); 观察组与对照组手术成功率分别为96%、75%(P<0.05)。

结论:POAG应用P50型EX-PRESS引流钉结合生物羊膜植入术治疗,疗效满意,能改善术后滤过泡情况,降低眼压及RNFLT,安全性佳,手术成功率高。  相似文献

Semaphorin-3A (Sema3A) and vascular endothelial growth factor (VEGF165) are ligands of neuropilin-1 (NRP-1 or CD304) and are related to immunoregulation and tumor angiogenesis, respectively. However, possible interactions between NRP-1 and Sema3A and VEGF165 in acute leukemia remain unclear, especially whether Sema3A plays a role in acute leukemia. In this study, both of the proportion of regulatory T cells (Tregs) and their expression of NRP-1 were found to increase in acute leukemia patients compared with healthy controls. In contrast, lower mRNA and plasma levels of Sema3A were detected in the acute leukemia patients. In vitro, the addition of exogenous Sema3A inhibited the expression of NRP-1 on Tregs and it promoted apoptosis of leukemia cells. However, in the presence of anti-Sema3A antibody, the effect of rhSema3A on NRP-1 expression was reversed. These results suggest that Sema3A promotes apoptosis in leukemia cells by inhibiting expression of NRP-1, and thus, represents a tumor suppressor protein with a role in the pathogenesis of acute leukemia. Consequently, NRP-1/Sema3A signaling may represent a novel target for the treatment of acute leukemia and should be further studied. 摘 要目的:探讨基因重组牛碱性成纤维细胞生长因子(rbFGF)促进烧伤创面愈合的临床效果。 方法:将 2019 年 7 月至 2021 年 4 月湛江中心人民医院收治的烧伤患者 60 例作为研究对象,根据随机数字表法分为观察组与对照组,各 30 例。 对照组接受清创后无菌凡士林纱布外敷治疗,观察组接受清创后 rbFGF 治疗,对患者治疗 1 周、2 周、3 周后的愈合率、总 愈合时间、浅Ⅱ度创面愈合时间、深 Ⅱ 度创面愈合时间情况、治疗 3 周后的总有效率进行观察。 结果:与对照组患者相比, 观察组治疗 1 周、2 周、3 周的愈合率、治疗 3 周后的总有效率明显较高,总愈合时间、浅Ⅱ度创面愈合时间、深Ⅱ度创面 愈合时间明显较短,差异均具有统计学意义(P < 0.05)。 结论:rbFGF 对促进烧伤创面愈合具有良好的效果。  相似文献

目的:探讨调节性T细胞(Tregs)及Th1/Th2型细胞因子在急性白血病发病中的作用。方法:采用流式细胞术检测37例急性白血病患者[包括急性髓细胞性白血病(acute myeloid leukemia,AML)24例,急性淋巴细胞白血病(acute lymphocytic leukemia,ALL)13例]及28例健康对照者外周血CD4+CD25+Foxp3+Tregs的比例,酶联免疫吸附法(ELISA)法检测血浆IL-2、IFN-γ、IL-10、IL-4和TGF-β水平。结果:AML和ALL患者外周血CD4+CD25+Foxp3+Tregs比例均高于健康对照组 (P<0.05);AML及ALL患者血浆IL-4、IL-10和TGF-β水平均较健康对照组升高 (P<0.05);IFN-γ水平较健康对照组降低 (P<0.05);IL-2水平与健康对照组比较无明显差异 (P>0.05)。结论:Tregs与Th1/Th2细胞因子同时参与了急性白血病的发生;急性白血病患者机体Th1/Th2细胞因子失衡,Th2占优势状态,这可能是导致急性白血病细胞免疫逃逸的原因之一;Tregs可能通过影响Th1/Th2细胞因子平衡参与急性白血病的发病。  相似文献

BackgroundAutologous hematopoietic stem cell transplantation is an effective therapeutic strategy for lymphoma patients. However, some patients have to give up receiving transplantation because of failing to obtain sufficient CD34+ cells yields. Therefore, we ex vivo expanded HSCs of lymphoma patients using UM171 to solve the problem of HSCs deficiency.MethodsMobilized peripheral blood-derived CD34+ cells from lymphoma patients were cultured for 10 days with or without UM171. The fold of cell expansion and the immunophenotype of expanded cells were assessed by flow cytometry. RNA-seq experiment was performed to identify the mechanism by which UM171 promoted HSCs expansion.ResultsUM171 treatment increased the proportion of CD34+ (68.97 ± 6.91%), CD34+ CD38 cells (44.10 ± 9.20%) and CD34+CD38CD45RACD90+ LT-HSCs (3.05 ± 2.08%) compared to vehicle treatment (36.08 ± 11.14%, 18.30 ± 9.49% and 0.56 ± 0.45%, respectively). UM171 treatment led to an 85.08-fold increase in LT-HSC numbers relative to initial cells. Importantly, UM171 promoted expansion of LT-HSCs achieved 138.57-fold in patients with poor mobilization. RNA-seq data showed that UM171 upregulated expression of HSC-, mast cell-specific genes and non-canonical Wnt signaling related genes, and inhibited genes expression of erythroid, megakaryocyte and inflammatory mediated chemokine.ConclusionsOur study shows that UM171 can efficiently promote ex vivo expansion of HSCs from lymphoma patients, especially for poorly mobilizing patients. In terms of mechanism, UM171 upregulate HSC-specific genes expression and suppress erythroid and megakaryocytic differentiation, as well as activate non-classical Wnt signaling.  相似文献

甘草毛状根中甘草总黄酮和甘草酸的检测和分析   总被引:2,自引:1,他引:1  
目的检测甘草毛状根中总黄酮和甘草酸的含量。方法采用紫外分光光度法检测甘草总黄酮含量,采用HPLC检测甘草酸含量。结果甘草毛状根中的总黄酮含量在不同根系中有很大差异,最高可达0.803%,接近生药根;大多数根系高于正常组培根;总黄酮含量随着培养时间的延长呈增加趋势,至9周时增加了68.5%。不同甘草毛状根系中的甘草酸含量差异显著,部分根系未检出甘草酸,能检出的根系最高干重含量为0.69mg·g^-1,远低于生药根(干重含量32.13mg·g^-1),且含量不受培养时间影响。结论甘草毛状根在目前情况下不适于作为甘草酸的获取资源,但作为甘草总黄酮获取的新资源,是值得进一步研究的。  相似文献

To compare the potency of recipient-derived, antigen-specific regulatory T cells induced by different dendritic cells (DCs; iTregs) and freshly isolated natural regulatory T cells (nTregs) in preventing mouse graft-versus-host disease (GVHD) after allogeneic bone marrow transplantation (BMT). CD4+ T cells from recipient BALB/c mice were stimulated with DCs from recipient BALB/c (syn-DCs), donor B6 (allo-DCs), and third-party C3H (third-party-DCs) mice to induce different iTregs. In parallel, nTregs were isolated from spleen cells of recipient BALB/c (syn-nTregs) and donor B6 (allo-nTregs) mice using magnetic-activated cell sorting. Mixed lymphocyte reaction (MLR) assays were performed to evaluate the suppressive ability of these various regulatory T cells (Tregs). Both the iTregs and nTregs were transfused to GVHD mice on Days 0, 1, 3, and 5. Body weight, GVHD score, and survival time were monitored. Peripheral Tregs were subsequently examined on Days 7, 14, 21, and 28 after BMT, while chimerism was evaluated on Days 14 and 60. Histopathology of colon, liver, and spleen were also performed. DCs markedly induced CD25+ and Foxp3+ expression on CD4+ T cells. The allo-DC-induced Tregs (allo-iTregs) suppressed the proliferation of alloreactive T cells better than the other iTregs/nTregs in MLR assays (P < 0.05). Meanwhile, transfusion of the allo-iTregs reduced the severity of GVHD (P < 0.05), increased survival time compared with the GVHD group (P < 0.05), and enhanced the chimerism proportion. On Day 28 after BMT, the allo-iTregs group had the highest frequency of peripheral Tregs (P < 0.05). Recipient-derived allo-iTregs induced by donor DCs included predominant clones that specifically recognized donor antigens. These allo-iTregs not only prevented GVHD by suppressing the proliferation of donor-alloreactive T cells, but also promoted engraftment, and prolonged the survival of GVHD mice.
