Somatostatin (SRIF) in the central nervous system is mostly concentrated in the median eminence (ME). Immunocytochemical methods have revealed high densities of SRIF-positive perikarya between the preoptic area and the periventricular nucleus of the hypothalamus (NPE). The aim of the present study was to define more precisely the specific pathways of SRIF neurons from NPE to the ME. SRIF levels were measured by radioimmunoassay, following various hypothalamic transections. Frontal periventricular sections decreased SRIF-ME content by 70% (P less than 0.01), when located at the anterior end of the ME but no diminution was observed when the cuts were located anteriorly or posteriorly. Parasaggital transections decreased SRIF-ME levels by 50% (P less than 0.05) when located at the outer border of the ventromedial and premammillary nucleus, but the decrease was not significant when cuts were located anteriorly. Taken together, our data indicate that most SRIF-containing neurons, originating in the NPE, do not reach the ME directly along the border of the 3rd ventricle; instead they form a loop across the medial forebrain bundle before re-entering the mediobasal hypothalamus at the ME level.  相似文献

用放射免疫法(RIA)测定甲地孕酮(MA)血清浓度并计算了在家兔和大鼠静注或口服的药物动力学参数。MA口服的生物利用度,家兔为64%,大鼠为56%。MA肝微粒体酶代谢实验提示有极性较MA为大的代谢产物形成,~3H-MA十二指肠给药后30min内门静脉血中放射性远高于外周血,也有极性代谢产物形成。  相似文献

采用电刺激减压神经及牵拉颈动脉窦的方法兴奋压力感受器,观察在产生减压效应时,血浆及某些脑区内神经降压素含量的改变,以探讨其在家兔压力感受性反射中的作用.结果表明,电刺激减压神经及牵拉颈动脉窦产生明显的减压效应时,家兔延髓、垂体、下丘脑及血浆内神经降压素含量明显增高.提示内源性神经降压素可能参与家兔压力感受性反射的调节过程.  相似文献

The characteristic distribution of calcitonin gone-related peptide(CGRP)inthe small intestine of rats and its changes in acute intestinal radiation sickness(AIRS)were studied with immunocytochemistry(whole mount stretch preparations of the smallintestine and cryostat sections)and radio-immunoassay.It was found that in all the lay-ers of the intestinal walls,there were large amounts of CGRP immunoreactive(CGRP-I)nerve fibers which existed in especiaUy high density in the myenteric,submucosal andmucosal plexuses.There was also a rather high density of the nerves around the smallvessels of the small intestine and the intestinal crypts.Some CGRP-I neurons were seenin the myenteric and submucosal plexuses.In AIRS,the intestinal CGRP showed a dip-hasic change,in a lower level in the 24th h and a higher level in the 48th and 72nd h af-ter irradiation.The results indicate that CGRP may be related to the regulation of the motility,se-cretion,absorption,sensation,and regional blood flow of the gastrointestinal tract.Pro-bably,CGRP is released under the stress of AIRS and participates in the mechanism ofinjury through many ways especially through the influence on the regional blood flowand the increase of the permeability of blood vessels.  相似文献

选用标化成年雄性Wistar大鼠、以放射免疫测定法(RIA),研究了大鼠由平原引入高海拔环境后,在24小时急性司服期,垂体及各脑区内β-内啡肪样免疫活性物质(β-EPLI)的含量变化。结果表明,大鼠引入高海拔环境后脑内β-EP含量发生明显的动态变化。高海拔实验组与平原对照组相比,垂体内β-EP含量降低非常显著(P<0.01)。纹状体、下丘脑、丘脑、桥延、海马、皮层内β-EP含量增加非常显著(P<0.01)。中脑内β-EP含量虽然也增加,统计学处理无显著性意义(P>0.05)。据此推测:大鼠急进高海拔低氧环境后垂体β-EP的分泌与释放受到影响。提示,脑内β-EP参与低氧的应激反应过程,可能作为神经递质和调质,通过神经──内分泌环路在应激反应中起介导作用,β-EP可能为中枢神经系统内调节应激反应的主要应激激素之一。  相似文献

①目的 探讨电化学发光免疫分析 (ECLIA)法测定血清胰岛素的效果及临床应用价值。②方法采用ECLIA法和放射免疫分析 (RIA)法分别测定糖尿病病人的血清胰岛素含量 ,并用酶联免疫法 (ELISA)测定C 肽含量 ,并对测定结果进行分析比较。③结果 ECLIA法测定胰岛素的批内与批间变异系数分别为 1.97% ,2 .5 6 % ,平均回收率是 99.5 % ;RIA法的批内与批间变异系数分别为 10 .18% ,13.2 1% ,平均回收率是 93.4 % ;E CLIA及RIA法所测胰岛素含量与ELISA法所测C 肽含量均呈正相关 (r =0 .94 ,0 .91,P均 <0 .0 1)。④结论ECLIA法测胰岛素的各技术参数优于RIA法 ,该方法能较好评价胰岛B细胞功能 ,且操作简便 ,检测速度快 ,具有重要临床应用价值。  相似文献

The C-terminal flanking peptide of preprocholecystokinin (preproCCK) has been identified in extracts of rat brain using a novel radioimmunoassay. There is a single form of immunoreactive material on gel filtration, ion exchange and reversed-phase HPLC. The C-terminal preproCCK immunoreactivity had a similar pattern of distribution to CCK8 in different regions of rat brain. This assay should help in studies of neuronal CCK biosynthesis.  相似文献

In a study to elucidate molecular mechanisms in pain, substance P-like immunoreactivity (SP-LI) was measured in lumbar CSF from 75 patients with lower back pain. Two samples – one before and one after lidocaine treatment – were obtained from each patient, and total SP-LI was measured in unfractionated (no HPLC) samples. SP-LI data were separated into three categories: placebo responders, pharmacological responders, and pharmacological non-responders. A significant difference was observed between the total SP-LI measurement of first and second samples of pharmacological non-responders. Distribution of SP-LI immunoreactive molecular species in two CSF patient samples (no ODS) was analyzed with a combination of reversed phase (RP) HPLC and RIA. Immunoreactivity in collected HPLC fractions was measured at calibrated retention times of synthetic SP-sulfoxide (SP-O), SP, and SP5–11. Qualitative and quantitative differences in those HPLC-RIA metabolic profiles were observed within and between those two patients' samples. These data indicate that the type and amount of SP metabolism and SP precursor-processing differs in CSF between these two representative patients and within the short amount of time elapsed between acquiring these two samples.  相似文献

用放免法检测乙型慢性活动性肝炎病人的红细胞c3b受体(KBCCR1).结果:病人RBCCR1明显低于献血员(P<0.05);抗-HBs特异性免疫复合物阳性病人RBCCR1明显低于阴性病人(P<0.01),与红细胞C3b受体花环试验检测结果一致。表明乙型慢性活动性肝炎病人RBCCR1数量减少,活性下降。其原因可能是特异性循环免疫复合物占据了RBCCR1空位,使CR1活性下降。  相似文献
