目的:探讨黄芩素调控miR-625-5p表达对支气管哮喘小鼠气道平滑肌细胞(ASMCs)增殖、迁移及炎性因子的影响。方法:培养小鼠原代ASMCs细胞,作为对照组,重组小鼠血小板衍生生长因子(PDGF)诱导ASMCs增殖,作为PDGF组; 分别采用10、20、40 μmol/L的黄芩素处理PDGF诱导ASMCs细胞。将miR-NC、miR-625-5p转染至PDGF诱导的ASMCs细胞,作为PDGF+miR-NC、PDGF+miR-625-5p组; 将anti-miR-NC、anti-miR-625-5p转染至PDGF诱导ASMCs细胞,以40 μmol/L的黄芩素处理,作为PDGF+anti-miR-NC+40 μmol/L组、PDGF+anti-miR-625-5p+40 μmol/L组。CCK-8法检测细胞增殖; Transwell实验检测细胞迁移; 酶联免疫吸附实验检测细胞IL-4、IL-6、IL-8水平; 实时荧光定量PCR检测细胞miR-625-5p表达水平。结果:与对照组比较,PDGF组细胞增殖率、迁移率以及IL-4、IL-6、IL-8水平显著升高(均P<0.05); 与PDGF组比较,黄芩素低、中、高剂量组细胞增殖率、迁移率以及IL-4、IL-6、IL-8水平显著降低(均P<0.05)。与对照组比较,PDGF组细胞miR-625-5p表达显著降低(P<0.05); 与PDGF组比较,黄芩素低、中、高剂量组细胞miR-625-5p表达显著升高(均P<0.05)。与PDGF+miR-NC组比较,PDGF+miR-625-5p组细胞增殖率、迁移率以及IL-4、IL-6、IL-8水平显著降低(均P<0.05)。与PDGF+anti-miR-NC+40 μmol/L组比较,PDGF+anti-miR-625-5p+40 μmol/L组细胞miR-625-5p表达水平显著降低,细胞增殖率、迁移率以及IL-4、IL-6、IL-8水平显著升高(均P<0.05)。结论:黄芩素通过上调miR-625-5p表达抑制支气管哮喘小鼠的细胞增殖率、迁移率以及炎性因子水平。  相似文献

目的:研究木棉花提取物对骨关节炎软骨细胞损伤的影响及潜在的作用机制。方法:用浓度分别为15、30、60 mg/L的木棉花提取物处理骨关节炎软骨细胞作为木棉花提取物组,以无木棉花提取物处理的细胞作为对照组;将miR-NC、miR-30b-3p、anti-miR-NC、anti-miR-30b-3p、si-NC、si-PDCD5质粒转染至骨关节炎软骨细胞中,分别记为miR-NC组、miR-30b-3p组、anti-miR-NC组、anti-miR-30b-3p组、si-NC组、si-PDCD5组;将anti-miR-NC、anti-miR-30b-3p质粒转染至骨关节炎软骨细胞后再用60 mg/L的木棉花提取物处理48 h,记为木棉花提取物+anti-miR-NC组和木棉花提取物+anti-miR-30b-3p组;转染均采用脂质体法。qRT-PCR检测骨关节炎软骨细胞中miR-30b-3p、PDCD5 mRNA表达;Western blot检测Bcl-2、Bax、PDCD5蛋白表达;ELISA法检测上清液中IL-6、IL-1β含量;流式细胞术检测细胞凋亡;双荧光素酶报告基因检测荧光活性。结果:与对照组比较,随着木棉花提取物浓度的升高,培养上清中IL-6、IL-1β含量及骨关节炎软骨细胞的凋亡率逐渐降低;miR-30b-3p、Bcl-2表达水平逐渐升高,PDCD5、Bax表达水平逐渐降低(P0.05)。miR-30b-3p过表达和抑制PDCD5表达可显著降低培养上清中IL-6、IL-1β含量,降低骨关节炎软骨细胞的凋亡率(P0.05)。miR-30b-3p靶向调控PDCD5的表达,抑制miR-30b-3p表达逆转了木棉花提取物对骨关节炎软骨细胞的凋亡抑制作用。结论:木棉花提取物可抑制骨关节炎软骨细胞IL-6、IL-1β的分泌及其凋亡,其机制可能与调控miR-30b-3p/PDCD5的表达有关。  相似文献

目的:探讨五味子乙素(Sch B) 调控miR-22-3p/runt 相关转录因子3(RUNX3) 分子轴对重症急性胰腺炎(SAP)腺泡细胞损伤的影响。方法:将大鼠胰腺腺泡细胞AR42J 分为Con 组、SAP 组、SAP+Sch-L 组、SAP+Sch-M 组、SAP+Sch-H组、SAP+anti-miR-NC 组、SAP+anti-miR-22-3p 组、SAP+Sch+miR-NC 组、SAP+Sch+miR-22-3p 组。除Con 组外,其余各组采用10 nmol/L 雨蛙素联合10 mg/L 脂多糖刺激AR42J 细胞24 h 构建SAP 细胞模型。SAP+Sch-L 组、SAP+Sch-M 组、SAP+Sch-H 组采用浓度为10、20、40 μmol/L 的Sch B 处理细胞;SAP+anti-miR-NC 组、SAP+anti-miR-22-3p 组为分别转染antimiR-NC、anti-miR-22-3p;SAP+Sch+miR-NC 组、SAP+Sch+miR-22-3p 组为分别转染miR-NC、miR-22-3p mimics 并用40 μmol/L的Sch B 干预。酶联免疫吸附法测定细胞培养液上清中白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α) 表达水平;流式细胞术检测细胞凋亡率;实时荧光定量PCR 检测miR-22-3p 和RUNX3 mRNA 表达水平;BCA 法检测RUNX3、B 细胞淋巴瘤-2 基因(Bcl-2)、Bcl-2 相关X 蛋白(Bax)、活化的含半胱氨酸的天冬氨酸蛋白水解酶3(cleaved-caspase3) 表达水平;双荧光素酶报告基因实验检测miR-22-3p 与RUNX3 的靶向结合关系。结果:与Con 组比较,SAP 组AR42J 细胞IL-6、TNF-α、miR-22-3p、Bax、cleaved-caspase3 的表达及细胞凋亡率升高(P<0.05),RUNX3、Bcl-2 的表达降低(P<0.05)。与SAP 组比较,SAP+Sch-L组、SAP+Sch-M 组、SAP+Sch-H 组IL-L、TNF-α、miR-22-3P、Bcl-2 表达降低(P<0.05),Bax、cleaved-caspase3、RUNX3 表达及细胞凋亡率升高(P<0.05),且呈剂量依赖性(P<0.05)。与SAP+anti-miR-NC 组比较,SAP+anti-miR-22-3p 组细胞IL-6、TNF-α、Bcl-2 的表达降低(P<0.05),细胞凋亡率及Bax、cleaved-caspase3、RUNX3 的表达升高(P<0.05)。与SAP+Sch+miR-NC 组比较,SAP+Sch+miR-22-3p 组细胞IL-6、TNF-α、Bcl-2 的表达升高(P<0.05),细胞凋亡率及Bax、cleavedcaspase3、RUNX3 的表达降低(P<0.05)。结论:Sch B 可通过下调miR-22-3p/RUNX3 分子轴减轻SAP 腺泡细胞损伤。  相似文献

目的通过研究溃疡性结肠炎(UC)模型大鼠结肠粘膜组织Bcl-2、Bax的变化,探讨葛根芩连汤对UC大鼠细胞凋亡的的影响。方法根据体重将60只SD大鼠随机分为正常组、模型组、对照药组、葛根芩连汤组;采用免疫诱导法(第1、10、17、24d于大鼠双侧足趾、双侧腹股沟及背部皮下注射抗原乳化液)复制UC大鼠模型;第30d观察大鼠结肠黏膜组织损伤程度并评分,ELISA法分析结肠黏膜组织匀浆中Bcl-2、Bax水平变化。结果 UC模型大鼠结肠溃疡形成,可伴有炎症、充血、水肿等病理改变;UC模型大鼠结肠黏膜Bcl-2水平显著降低,Bax水平显著升高(P<0.05);葛根芩连汤组结肠黏膜Bcl-2水平显著升高,Bax水平显著降低(P<0.05)。结论 UC模型大鼠结肠黏膜损伤,可能与细胞凋亡因子Bcl-2、Bax的变化有关;葛根芩连汤可上调UC模型大鼠Bcl-2水平、下调Bax水平。  相似文献

目的 观察葛根芩连汤对幽门螺杆菌(Hp)感染人正常胃黏膜上皮(GES-1)细胞增殖与凋亡的影响及相关机制。方法 按照葛根芩连汤干预剂量的不同、是否有Hp感染将人GES-1细胞分为空白对照组、模型组、葛根芩连汤低剂量组(10μmol/L)、葛根芩连汤中剂量组(20μmol/L)、葛根芩连汤高剂量组(40μmol/L)及阳性药对照组(阿莫西林5μmol/L)。培养结束后,采用细胞增值检测分析法(CCK-8法)检测细胞存活率,酶联免疫吸附法(ELISA)检测上清液肿瘤坏死因子α(TNF-α)、白细胞介素1β(IL-1β)和IL-6水平,流式细胞术检测细胞凋亡率,实时荧光定量聚合酶链式反应(RT-qPCR)检测细胞p38、丝裂原活化蛋白激酶2(MK2)mRNA表达,蛋白免疫印迹(Western bloting)法检测细胞p38、MK2蛋白表达。结果 与空白对照组比较,模型组人GES-1细胞存活率降低(P<0.05);与模型组比较,葛根芩连汤各剂量组与阳性药对照组人GES-1细胞存活率均升高(P<0.05),且葛根芩连汤各剂量组效应呈剂量依赖性(P<0.05);阳性药对照组与葛...  相似文献

目的 探讨异紫堇碱对肺癌A549细胞增殖、迁移和侵袭的影响及其分子机制。方法 将A549细胞分为对照组,异紫堇碱低、中、高剂量组,miR-NC组,miR-1296-5p组,anti-miR-NC组,anti-miR-1296-5p组,异紫堇碱+anti-miR-NC组,异紫堇碱+anti-miR-1296-5p组。MTT法检测细胞增殖抑制率,Transwell实验检测细胞迁移和侵袭能力,RT-qPCR法检测miR-1296-5p和花斑抑制因子同源物1(SUV39H1)mRNA表达,Western blot法检测相关蛋白表达,双荧光素酶报告实验验证miR-1296-5p和SUV39H1的靶向关系。结果 不同剂量异紫堇碱处理A549细胞后,细胞增殖抑制率升高,迁移、侵袭细胞数降低,miR-1296-5p表达升高,SUV39H1 mRNA和蛋白表达降低,并呈剂量依赖性(P<0.05)。miR-1296-5p靶向调控SUV39H1,过表达miR-1296-5p可抑制肺癌细胞增殖、迁移和侵袭。结论 异紫堇碱可能通过调控miR-1296-5p/SUV39H1抑制肺癌细胞增殖、迁移和侵袭。  相似文献

目的:探讨石榴皮多酚在肺癌中的抗肿瘤作用及可能机制。方法:RT-qPCR 检测肺癌组织中 miR-4316 表达水平。将肺癌细胞A549 分为石榴皮多酚-低组、石榴皮多酚-中组、石榴皮多酚-高组、对照 组、miR-NC 组、miR-4316 组、石榴皮多酚+anti-miR-NC 组、石榴皮多酚+anti-miR-4316 组。细胞迁移、增 殖及侵袭分别采用划痕愈合实验、平板克隆实验和CCK-8 法、Transwell 实验检测。结果:肺癌组织中miR- 4316 表达水平显著降低(P<0.05)。石榴皮多酚(低、中、高) 组A549 细胞抑制率、miR-4316 水平较对照 组高,克隆形成数、划痕愈合率、侵袭数较对照组低(P<0.05)。与miR-NC 组比较,miR-4316 组较miRNC 组A549 细胞抑制率更高(P<0.05),克隆形成数、划痕愈合率及侵袭数miR-4316 组较miR-NC 组 低(P<0.05)。石榴皮多酚+anti-miR-4316 组较石榴皮多酚+anti-miR-NC 组A549 细胞抑制率更低(P< 0.05),克隆形成数、划痕愈合率及侵袭数石榴皮多酚+anti-miR-4316 组较石榴皮多酚+anti-miR-NC 组 高(P<0.05)。结论:石榴皮多酚通过上调miR-4316 表达来抑制肺癌细胞增殖、迁移及侵袭。  相似文献

目的探讨黄芩苷对缺氧缺糖(oxygen-glucosedeprivation,OGD)诱导的神经元细胞增殖、凋亡、炎症、氧化应激的影响及其对miR-190的调控作用。方法采用OGD处理小鼠海马神经元HT22细胞建立细胞损伤模型,使用不同剂量(10、30mg/L)的黄芩苷处理HT22细胞,分别将miR-NC、miR-190mimics转染至HT22细胞后进行OGD处理(OGD+miR-NC组、OGD+miR-190组);分别将anti-miR-NC、anti-miR-190转染至HT22细胞后使用黄芩苷处理24h,随后进行OGD处理(OGD+黄芩苷+anti-miR-NC组、OGD+黄芩苷+anti-miR-190组);采用MTT法检测细胞增殖情况;采用流式细胞术检测细胞凋亡率;采用ELISA法检测白细胞介素-1β(interleukin-1β,IL-1β)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的水平;采用2,4-二硝基苯肼显色法检测乳酸脱氢酶(lactate dehydrogenase,LDH)的活性;采用黄嘌呤氧化酶法检测超氧化物歧化酶(superoxide dismutase,SOD)的含量;采用qRT-PCR法检测miR-190的表达量;Western blotting法检测细胞周期蛋白1(Cyclin D1)、未剪切的半胱天冬酶-3(pro-cysteinyl aspartate-specific protease-3,pro-Caspase-3)、p21、活化的含半胱氨酸的天冬氨酸蛋白水解酶3(cleaved-cysteinylaspartate-specificprotease-3,cleaved-Caspase-3)蛋白表达量。结果 OGD可明显降低HT22细胞活力及CyclinD1、pro-Caspase-3蛋白水平(P0.05),升高细胞凋亡率及p21、cleavedCaspase-3蛋白水平(P0.05),并可提高IL-1β、TNF-α的水平及LDH的活性(P0.05),降低SOD的含量(P0.05);黄芩苷可明显提高OGD诱导的HT22细胞活力及CyclinD1、pro-Caspase-3蛋白水平(P0.05),降低细胞凋亡率及p21、cleaved-Caspase-3蛋白水平(P0.05),并可降低IL-1β、TNF-α的水平及LDH的活性(P0.05),升高SOD的含量(P0.05);OGD可明显降低HT22细胞中miR-190的表达水平(P0.05),而黄芩苷可明显升高miR-190的表达水平(P0.05);miR-190过表达对OGD诱导的HT22细胞增殖、凋亡、炎症及氧化应激的作用与黄芩苷的作用相似;抑制miR-190表达可明显逆转黄芩苷对OGD诱导的HT22细胞增殖、凋亡、炎症及氧化应激的作用。结论黄芩苷可通过上调miR-190的表达而促进细胞增殖及抑制炎症反应、氧化应激、细胞凋亡从而减轻OGD诱导的神经元细胞损伤。  相似文献

目的建立微波消解-电感耦合等离子体质谱(ICP-MS)直接稀释测定脉络宁注射液中25种矿物质元素(Mg、Ca、Fe、Cu、Zn、Mn、Al、B、Ba、Co、Cr、K、Li、Mo、Na、Ni、P、Pb、Sr、Th、Ti、V、As、Cd和Hg)的方法。方法分别对微波消解条件和测试条件进行考察;样品经微波消解后,采用电感耦合质谱仪测定25种矿物质元素,并对测定方法学进行考察。结果确定最佳消解条件为3步缓慢升温:400 W 80℃升温10 min,保留5 min;600 W 120℃升温10 min,保留5 min;900 W 200℃升温20 min,保留20 min;25种矿物质元素在各自的线性范围内线性关系良好,r≥0.999 6,精密度、稳定性和重复性试验的RSD均符合定量分析要求;加标回收率为94.7%~106.1%,RSD在0.34%~2.79%。脉络宁注射液中检测出Mg、Ca、Fe、Cu、Zn、Mn、Al、B、Ba、Co、Cr、K、Li、Mo、Na、Ni、P、Pb、Sr、Th、Ti、V,未检出As、Cd和Hg。结论该方法简便、迅速、准确,适用于脉络宁注射液中25种矿物质元素的同时测定。  相似文献

This paper deals with an interdisciplinary study covering historic, botanical, phytochemical, pharmacological and clinical aspects of rhubarb and related species, to lay stress on the correlation between plant phylogeny, chemical constituents and purgative activity.

It was found that the official rhubarbs were exclusively restricted in the Sect. Palmata e.g. Rheum palmatum R. palmatum var. tanguticum R. officinale; the following criteria may serve as their standard, viz., the presence of sennoside derivatives and rhein, the occurrence of the reduced form of rhein and aloe-emodin, the leaves with any kind of palmate incision. Comprehensive multivariate analyses showed that there is a very close relationship between the leaf incision, existence of sennosides or rhein and purgative activity.  相似文献

Background

At present, the state-of-the-art medication in treating idiopathic taste disorder (gustatory dysfunction) is zinc. If zinc medication was unsuccessful, not tolerated or if it is contraindicated the persons affected can hardly be helped.

Objectives

In a randomized placebo-controlled clinical trial an attempt was made to determine the efficacy of acupuncture.

Methods

The treatment group was treated with acupuncture needles, the placebo group with an inactive acupuncture-laser. Endpoints of the study were changes in the taste test, the quality of life and the severity of symptoms of depression (BDI) as well as mood changes (ZMS). Satisfaction with the success of the respective therapy was also evaluated. Data were collected before and after therapy and again for the treatment group 10 weeks after completion of the treatment period.

Findings

Treatment with real acupuncture showed a significant improvement in gustatory function as well as a significant improvement in psychological symptoms. Greater satisfaction with the treatment results was evident in the treatment group. The treatment results of acupuncture remained stable over a period of ten weeks after completion of the treatment.

Conclusion

Acupuncture was effective in the therapy of idiopathic taste disorders and therefore it is an effective alternative when zinc medication is not tolerated or unsuccessful. But it is also suitable as a therapy of first choice. Acupuncture can be considered as an extension of the therapeutic spectrum in the difficult treatment of taste disorders.  相似文献

Background

Experimental and clinical studies postulate a neurobiological mechanism of acupuncture by modulating central pain systems, neurohormones and -transmitters. It seems to be quite likely that the autonomic nervous system (ANS) plays an important mediating role in this neuromodulation.

Aim

In measuring heart rate variability (HRV), as a non-invasive approach to the ANS-function, the object of this study was to monitor probable acupuncture-induced changes of autonomic balance. The innovative technology of laser-needle acupuncture offers for the first time a stimulation method which fulfils the demands on a placebo-controlled acupuncture trial not requiring controversial sham acupuncture.

Method

19 healthy voluntary subjects underwent 3 treatments in randomized sequence, consisting of placebo-, 1-point- (PC 6) and simultaneous 3-point- (PC 6, LR 3, SI 3) laser-needle-stimulation over 20 minutes. Before, during and after placebo-/verum laser-needle acupuncture, HRV-data for normalized HF(high frequency)- and LF(low frequency)-Power (nHF, nLF) and their quotient nLF/nHF obtained by power spectrum analysis (FFT), were measured and analyzed statistically at 5 different time points (ANOVA repeated measures, p < 0,05).

Results

During both verum applications, the 1-point- (PC 6) as well as the simultaneous 3-point- (PC 6, LR 3, SI 3) laser-needle stimulation, significant changes of the ANS-activity were found in comparison to the placebo application. The most significant modification of HRV was an increase of nHF (p < 0,05 (PC 6), p < 0,0001 (PC 6, LR 3, SI 3)) as an expression of growing vagal activity during acupuncture.

Conclusion

Laser-needle acupuncture causes in 1-point-stimulation (PC 6) as well as in simultaneous 3-point-stimulation (PC 6, LR 3, SI 3) significant changes in HRV in comparison to placebo application. HRV-monitoring seems to be a suitable approach in exploration of acupuncture-induced changes of ANS and could possibly be established in combination with laser needle acupuncture as a standard for further scientific and clinical acupuncture investigations which are greatly needed.  相似文献

目的采用HPLC梯度洗脱法同时测定沉香化滞丸中沉香四醇、柚皮苷、橙皮苷、新橙皮苷、和厚朴酚、大黄素、厚朴酚、木香烃内酯、去氢木香内酯、大黄酚、大黄素甲醚11种成分。方法采用Thermo Syncronis C18色谱柱(250 mm×4.6 mm,5μm),流动相为水-乙腈,梯度洗脱:0~10 min,20%乙腈;10~20 min,20%~40%乙腈;20~24 min,40%乙腈;24~26 min,40%~52%乙腈;26~30 min,52%乙腈;30~31 min,52%~90%乙腈;31~35 min,90%乙腈;35~40 min,90%~100%乙腈;40~43min,100%乙腈;43~45min,100%~20%乙腈;检测波长215nm,体积流量1.0m L/min,柱温30℃,进样量20μL。结果各成分在43 min内分离良好,沉香四醇、柚皮苷、橙皮苷、新橙皮苷、和厚朴酚、大黄素、厚朴酚、木香烃内酯、去氢木香内酯、大黄酚、大黄素甲醚的线性范围分别为1.4~13.6、10.0~200.0、31.5~315.0、1.0~120.1、1.8~50.6、0.93~10.1、1.8~30.0、0.2~40.3、1.8~18.1、1.7~25.0、0.45~10.70μg/mL;样品中各成分的平均回收率均在98.90%~100.87%;11种成分精密度RSD在0.55%~1.54%;供试品溶液在30 h内稳定性良好,RSD在0.75%~1.94%;重复性RSD在0.39%~1.73%。6批次样品中沉香四醇、柚皮苷、橙皮苷、新橙皮苷、和厚朴酚、大黄素、厚朴酚、木香烃内酯、去氢木香内酯、大黄酚、大黄素甲醚质量分数分别为92.0~201.0、511.5~9 033.0、5 475.0~12 635.5、54.5~5 095.5、192.0~2 137.5、117.0~391.5、106.5~1 281.5、13.0~136.5、93.5~199.0、177.0~1 207.0、33.5~251.5μg/g。结论本方法准确、快速、简便,重复性好,精密度高,适用于沉香化滞丸中多种活性成分的定量分析。  相似文献
