Male endocrine functions in workers with moderate exposure to lead   总被引:4,自引:0,他引:4  
Evidence for the effect of occupational exposure to lead on the male endocrine system is conflicting. This study evaluated the primary (testicular) and secondary (hypothalamo pituitary testicular) effects of exposure to lead in 122 current lead workers and 49 non-exposed workers. The mean current blood lead concentration was 35.2 (range 9.6-77.4) micrograms/dl in the exposed workers, and 8.3 (range 2.6-14.8) micrograms/dl in the non-exposed workers. Concentrations of plasma luteinising hormone (LH) and follicle stimulating hormone (FSH) were both significantly higher in the exposed workers, but testosterone (T) was not significantly different between the two groups. In older exposed workers, however (greater than or equal to 40 years), plasma T concentrations were significantly lower, but LH and FSH concentrations were not significantly different. Compared with non-exposed workers, those exposed for less than 10 years had significantly raised LH and FSH and normal T concentrations whereas those exposed for 10 or more years had significantly lower T, and normal LH and FSH concentrations. The concentrations of LH and FSH showed a moderate increase in relation to blood lead concentrations in the range of 10 micrograms/dl to 40 micrograms/dl and thereafter reached a plateau or declined. No apparent trend for plasma T concentrations occurred. No significant difference in prolactin (PRL) concentration was noted. It is concluded that moderate exposure to lead was associated in dose related fashion with small but measurable changes in male endocrine functions that reflected both primary and secondary effects of lead on the testes and the hypothalamo pituitary testicular axis.  相似文献

为评价铅接触对下丘脑-垂体-性腺轴的影响,选取健康男性工人189人,其中接触男工97名(血铅2.12±1.39μmol/L),非接触男工92名(血铅1.10±0.56μmol/L),以放射免疫法测定其血浆睾酮及血清LH、FSH。接触组工人平均血浆睾酮浓度为561.12ng/dl,明显低于对照组的725.29ng/dl(P<0.01)而血清LH水平则明显高于对照组工人,前者为19.17mIU/ml,后者为15.19mIU/ml(P<0.05);两组工人血清FSH含量无显著性差异,分别为12.21和11.03mIU/ml(P>0.05)。多元逐步回归分析表明,血清LH及血浆睾酮水平的改变与接触者的工龄明显相关,其次与吸烟也有一定关系。提示男工接触铅后可影响下丘脑-垂体-睾丸轴内分泌功能,引起血浆睾酮及血清LH浓度的改变。  相似文献

In this study of the effects of lead on the endocrine system, 77 secondary lead-smelter workers (i.e., 62 active and 15 retired) were compared with 26 referents. Lead concentrations were determined in plasma with inductively coupled plasma mass spectrometry (i.e., index of recent exposure), in blood with atomic absorption spectrophotometry, and in fingerbone with K x-ray fluorescence technique (i.e., index of long-term exposure). In addition, pituitary hormones were determined in serum by fluoroimmunoassay, and thyroid hormones and testosterone in serum were determined with radioimmunoassay. Nine lead workers and 11 referents were challenged with gonadotrophin-releasing hormone and thyrotrophin-releasing hormone, followed by measurements of stimulated pituitary hormone levels in serum. Median levels of lead in plasma were 0.14 microg/dl (range = 0.04-3.7 microg/dl) in active lead workers, 0.08 microg/dl (range = 0.05-0.4 microg/dl) in retired lead workers, and 0.03 microg/dl (range = 0.02-0.04 microg/dl) in referents (1 microg/dl = 48.3 nmol/l). Corresponding blood lead concentrations were 33.2 microg/dl (range = 8.3-93.2 microg/dl), 18.6 microg/dl (range = 10.4-49.7 microg/dl), and 4.1 microg/dl (range 0.8-6.2 microg/dl), respectively. Respective bone lead levels were 21 microg/gm (range = -13 to 99 microg/gm), 55 microg/gm (range = 3-88 microg/gm), and 2 microg/gm (range = -21 to 14 microg/gm). Concentrations of basal serum hormone (i.e., free thyroid hormones, thyrotrophin, sex hormone binding globulin, and testosterone) were similar in the 3 groups. There were no significant associations between the hormones mentioned herein and blood plasma, blood lead, and bone lead levels. In the challenge test, stimulated follicle-stimulating hormone levels were significantly lower in lead workers (p = .014) than in referents, indicating an effect of lead at the pituitary level. Also, there was a tendency toward lower basal stimulated follicle-stimulating hormone concentrations in lead workers (p = .08). This effect, however, was not associated with blood plasma level, blood lead level, or bone lead level. In conclusion, a moderate exposure to lead was associated with only minor changes in the male endocrine function, particularly affecting the hypothalamic-pituitary axis. Given that sperm parameters were not studied, the authors could not draw conclusions about fertility consequences.  相似文献

目的探讨铅对男性工人性激素水平的改变及其影响因素。方法选153名职业性接触铅男工为暴露组,同时选择年龄和经济条件相近,距离冶炼车间1.5km煤场中47名不接触铅男工为非铅暴露组,用石墨炉原子吸收光谱法测定血铅,ELISA方法测定血清中睾酮(T)、黄体生成素(FSH)、卵泡刺激素(LH)、雌二醇(E2)和抑制素B水平。结果暴露组男工血铅水平为370.2μg/L,高于非暴露组221.2μg/L(P0.01);暴露组男工血清中T的平均水平为8.20μg/L,低于非暴露组9.65μg/L(P0.05);抑制素B平均水平为240.5pg/ml高于非暴露组211.1pg/ml(P0.05);黄体生成素(FSH)、卵泡刺激素(LH)、雌二醇(E2)水平两组间差异无显著性(P0.05)。通过Logistic回归分析,发现影响血清T水平异常变化的因素主要是年龄和铅暴露,而抑制素B水平的改变主要与铅暴露有关。结论职业性铅暴露能诱导男工血清中T和抑制素B水平的改变,提示接触铅可损伤内分泌和睾丸支持细胞功能。  相似文献

OBJECTIVES--To assess neurobehavioural effects of low exposure to lead, 43 workers from a lead smelter and 45 workers from a glass factory were evaluated with the World Health Organisation neurobehavioural core test battery (NCTB) in a cross sectional study. METHODS--The NCTB comprises a questionnaire and seven tests that measure simple reaction time, short-term memory (digit span, Benton), mood (profile of mood states), eye-hand coordination (Santa Ana pegboard, pursuit aiming II), and perceptual speed (digit-symbol). RESULTS--Smelter workers were employed on average for four years, and had a mean blood lead concentration of 2.0 mumol/l (42 micrograms/dl). Glass factory workers had a mean of 0.72 mumol/l (15 micrograms/dl). Historical blood lead concentrations were used to classify exposure based on current, peak, and time weighted average. Although the exposed workers performed less well than the non-exposed in 10 of 14 response variables, only profile of mood states tension-anxiety, hostility, and depression mood scales showed a significantly poorer dose-response relation with blood lead concentration in multiple linear regression models that included age, education, and alcohol intake as covariates. The frequency of symptoms of anger, depression, fatigue, and joint pain were also significantly increased in the exposed group. CONCLUSION--This study is consistent with the larger body of neurobehavioural research of low occupational exposure to lead. The small effects found in this study occurred at blood lead concentrations slightly lower than those reported in several previous studies.  相似文献

No adverse effects of lead on renal function in lead-exposed workers   总被引:3,自引:0,他引:3  
A cross-sectional study was performed on 165 male lead-exposed workers to clarify the quantitative relationships between less severe exposure to lead and its effects on renal function in 1985. Mean and range of blood lead concentration (Pb-B) were 36.5 micrograms/dl and 6-73 micrograms/dl, respectively. Duration of lead exposure was 0.1 to 26.3 years. No lead-related changes were detected in serum creatinine concentration, beta-2 microglobulin in urine, creatinine clearance, beta-2 microglobulin clearance, and uric acid clearance. Twenty of the 165 workers had been exposed to lead for more than 10 years with mean duration of 21.0 years. Average concentrations of Pb-B in each individual during 1972 to 1984 were 26.1-66.6 micrograms/dl. Renal function indices of these 20 workers were not different from those of remaining lead-exposed workers whose lead exposure duration were 10 years or less. These results suggest that long-term less severe exposure to lead up to 70 micrograms/dl of Pb-B may not cause adverse effects on renal glomerular function and proximal tubular function.  相似文献

OBJECTIVE--To evaluate lead exposure among lead-acid battery workers in Korea, to evaluate in more detail the erythrocyte pyrimidine 5'-nucleotidase (P5N) test for lead exposure, and to evaluate the abnormal accumulation of erythrocyte pyrimidine nucleotides in the battery workers. METHODS--Activity of P5N and other biological variables were examined in 66 exposed workers in a lead-acid battery factory and in 26 non-exposed workers in Korea. RESULTS--At the factory the time-weighted average of 13 (72%) of 18 air samples for lead exceeded 0.05 (range 0.012-0.468) mg/m3. Blood lead concentration (PbB) in 39 of the 66 exposed workers was above 40 micrograms/dl, and the mean (SD) PbB in the exposed group was 45.7 (15.7) micrograms/dl. Compared with the nonexposed group, free erythrocyte protoporphyrin in the exposed group was significantly increased, whereas erythrocyte P5N activity and activity of erythrocyte delta-aminolevulinic acid dehydratase (ALAD) were significantly inhibited. Erythrocyte P5N activity had valid correlation biologically with PbB and with other biological variables, such as ALAD activity. In 28 exposed workers, the concentration of erythrocyte pyrimidine nucleotides (uridine 5'-diphosphate-glucose and cytidine 5'-triphosphate) correlated inversely with P5N activity and positively with PbB. CONCLUSIONS--These findings show that the depression of erythrocyte P5N activity by lead exposure results in the accumulation of erythrocyte pyrimidine nucleotides. In general, the standard analysis of PbB performed in laboratories around the world remains the most useful index of recent exposure. The results indicate that the erythrocyte P5N activity test provides supporting evidence of lead exposure and shows the effect of lead on nucleotide metabolism.  相似文献

OBJECTIVE: To study the association in a group of battery manufacturing workers between computerised postural sway parameters and present concentrations of blood lead (PPb), index of cumulative blood lead years (CBI), and cumulative blood lead at different years of exposure (CPbYs). METHODS: Postural stability was investigated with a computerised postural sway measurement system in 60 workers exposed to lead with exposure duration of 84 (range 3-366) months and in 60 control subjects. An index of CBIs in 55 workers (previous blood lead results of five workers were not available) and CPbYs were computed for each worker by calculating the area under the curve of concentrations of blood lead against time. RESULTS: The mean (SD) PPb was 36.0 (11.7) (range 6.4 to 64.5) micrograms/dl for the exposed workers and 6.3 (2.4) (range 3.1-10.9) micrograms/dl for the 14 randomly selected control subjects. Significant differences between groups for the postural sway parameters obtained when the eyes were closed were found for length of sway path (L); mean velocity of the centre of pressure along its path (Vel); area included within the path of the centre of pressure (Ao); 95% confidence elliptical area (Ae). The Romberg ratio (the relation between eyes closed and open) for the Vel, L, Ao, and Ae of the exposed group were also significantly different from those of the controls. The postural sway parameters (eyes closed) were not significantly correlated with PPb or CBI. However, the cumulative blood lead for the past two years before the postural sway assessment, CPbY2, was significantly correlated with all the postural sway parameters. CONCLUSION: The study showed that workers exposed to lead had significantly poorer postural stability than a control group. Lead may affect certain parts of the somatosensory system resulting in postural instability when the visual input is cut off. The CPbY2 was significantly positively correlated with most of the postural sway parameters. Effects of lead on postural stability may be related to recent increases in blood lead concentration among the exposed workers rather than to cumulative body burden.  相似文献

BACKGROUND: Secondary exposure to lead has been identified as a public health problem since the late 1940s; we investigate the risk of lead exposure among families of radiator repair workers. METHODS: A sample of the wives and children, aged 6 months to 6 years (exposed children) (n = 19), of radiator repair workers and a sample of children whose parents were not occupationally exposed to lead (non-exposed children) (n = 29) were matched for age and residence; their geometric mean blood lead levels are compared. Blood samples were obtained by the finger stick method and environmental dust samples by the wipe method; both were analyzed using a portable anodic stripping voltameter. RESULTS: Dust lead levels were significantly higher in the houses of exposed children (143.8 vs. 3.9 microg/g; P < 0.01). In crude analyses, the highest lead levels were observed among children whose fathers worked in home-based workshops (22.4 microg/dl)(n = 6). Children whose fathers worked in an external workshop (n = 13) also had high levels (14.2 microg/dl) (P < 0.01), while blood lead levels in non-exposed children were significantly lower (5.6 microg/dl)(P < 0.01). The observed differences remained significant after adjustment for age and gender. CONCLUSIONS: This study confirms that children of radiator repair workers are at increased risk of lead exposure and public health interventions are needed to protect them.  相似文献

目的 分析焊工作业中锰接触对作业人员血清免疫球蛋白和性激素的影响。
方法 2016年10月以某人防工程公司96名男性焊工为焊工组, 同时选择96名男性非焊工人员作为对照组, 以全自动生化仪检测两组人员的血清免疫球蛋白IgA、IgM、IgG水平, 以石墨炉原子吸收法检测两组人员的尿锰水平; 以化学发光免疫分析法检测促黄体生成激素(LH)、促卵泡生成激素(FSH)、睾酮(T)、黄体酮(P)、雌二醇(E2)和泌乳素(PRL), 并对结果进行分析。
结果 焊工组尿锰水平为(1.59 ±0.67)μg/L, 高于对照组的(0.45 ±0.08)μg/L, 差异有统计学意义(P < 0.01)。焊工组的血清IgG、T和P水平低于对照组, FSH水平高于对照组, 差异有统计学意义(P < 0.05或0.01)。焊工组的IgA与T水平呈负相关(r=-0.271, P < 0.05), IgM和E2、尿锰, 尿锰和FSH、PRL均呈正相关(r=0.209、0.220、0.487、0.250, P < 0.01)。
结论 电焊作业增加了锰接触的机会, 可能会降低作业人员的免疫能力并影响其内分泌系统。

Organophosphorous pesticides (OPs) are suspected of altering reproductive function by reducing brain acetylcholinesterase activity and monoamine levels, thus impairing hypothalamic and/or pituitary endocrine functions and gonadal processes. Our objective was to evaluate in a longitudinal study the association between OP exposure and serum levels of pituitary and sex hormones. Urinary OP metabolite levels were measured by gas-liquid chromatography, and serum pituitary and sex hormone levels by enzymatic immunoassay and radioimmunoassay in 64 men. A total of 147 urine and blood samples were analyzed for each parameter. More than 80% of the participants had at least one OP metabolite in their urine samples. The most frequent metabolite found was diethylthiophosphate (DETP; 55%), followed by diethylphosphate (DEP; 46%), dimethylthiophosphate (DMTP; 32%), and dimethyldithiophosphate (DMDTP; 31%). However, the metabolites detected at higher concentrations were DMTP, DEP, DMDTP, and dimethylphosphate. There was a high proportion of individuals with follicle-stimulating hormone (FSH) concentrations outside the range of normality (48%). The average FSH serum levels were higher during the heavy pesticide spraying season. However, a multivariate analysis of data collected in all periods showed that serum FSH levels were negatively associated with urinary concentrations of both DMTP and DMDTP, whereas luteinizing hormone (LH) was negatively associated with DMTP. We observed no significant associations between estradiol or testosterone serum levels with OP metabolites. The hormonal disruption in agricultural workers presented here, together with results from experimental animal studies, suggests that OP exposure disrupts the hypothalamic-pituitary endocrine function and also indicates that FSH and LH are the hormones most affected.  相似文献

目的 探讨二苯甲撑二异氰酸酯(MDI)对作业男工体内性激素水平的影响。方法 对某化工厂84名接触MDI的男工和96名对照男工血清睾酮(T)、卵泡刺激素(FSH)和黄体生成素(LH)水平进行检测和比较。结果 MDI作业男工血清T水平显著低于对照组(P<0.01);FSH及LH水平显著高于对照组(P<0.01);高浓度接触组男工血清T显著低于中浓度组和低浓度组(P<0.01);而LH及FSH却显著高于中浓度组和低浓度组(P<0.01);MDI作业男工性功能与对照组比显著减退(P<0.01);吸烟饮酒可导致血清T水平显著低于未吸烟饮酒组(P<0.01),而LH及FS未吸烟饮酒组无明显差异(P>0.05),且MDI与吸烟饮酒三者存在协同作用的关系。结论 MDI具有一定的生殖毒性。  相似文献

The aim of the present study is to determine the effect of blood lead on the plasma levels of amino acids and serum liver enzymes in industrial workers in United Arab Emirates (UAE). This comparison study consisted of 100 industrial workers (exposed) and 100 non-industrial workers (non-exposed), matched for age, sex and nationality selected from Al-Ain, Abu-Dhabi Emirates. Industrial workers had higher proportion (19%) of smokers than non-industrial workers (11%) which was not considered to be statistically significantly different. Industrial workers had significantly higher mean of blood lead level (77.5 +/- 42.8 miccrog x dl(-1)) than non-industrial workers (19.8 +/- 12.3 microg x dl(-1)). The amino acid analysis showed higher values among industrial than non-industrial workers for histidine, isoleucine, leucine, threonine, lysine, valine, methionine and arginine (essential amino acids, (p<0.0001). Ornithine, taurine, glutamic acid, serine, glycine, proline and alanine (non-essential amino acids) showed significantly higher values in industrial when compared with the non-industrial workers (p<0.0001). Plasma liver function test, cardiac enzymes and renal function test were carried out on industrial and non-industrial workers. The results revealed alkaline phosphatase (p=0.012) and lactate dehydrogenase (p=0.029) were significantly higher in industrial than in non-industrial workers. On the basis of this study, it can be concluded that a substantial difference in amino acid profiles, blood lead and LFT between exposed and non-exposed was found. These results might be related to lead exposure and might have affects on the kidneys or liver.  相似文献

The workers' blood lead concentrations ranged from 22 to 59 micrograms/dl (mean 38 micrograms/dl (1.8 mumol/l]. The 24 hour urinary excretion of metals after CaEDTA administration (mobilisation yield) was on average 13 times the background excretion for lead, 11 times for zinc, 3.8 times for manganese, 3.4 times for cadmium, 1.3 times for copper, and 1.1 times for chromium; no significant increase was found for mercury. The mobilisation yield of lead (MPb) was significantly correlated with whole blood and erythrocyte concentrations and the urinary excretion of lead but not with its plasma concentration; similarly, the mobilisation yield of cadmium was significantly correlated with its erythrocyte concentration. In addition, MPb was significantly correlated with intra-erythrocytic enzyme delta-aminolaevulinic acid dehydratase activity and urinary coproporphyrin excretion. The relation between the mobilisation yield of heavy metals and their body burden (and toxic signs) is discussed in the light of these findings.  相似文献

Exposure-effect and exposure-response relation between exposure to lead and delta-aminolaevulinic acid concentration in blood (ALA-B) were examined in 238 male workers exposed to lead. Concentrations of ALA-B ranged from 26 to 352 micrograms/l and lead concentrations in blood (Pb-B) from 7.1 to 86.0 micrograms/dl. Concentrations of ALA-B correlated closely with concentrations of Pb-B (r = 0.74), and increased ALA-B concentration occurred at Pb-B concentrations of around 30 micrograms/dl. Exposure-response curves indicated that the 50 percentile response doses were roughly 30, 40, and 50 micrograms/dl Pb-B when cut off points of ALA-B were set at 50, 60, and 70 micrograms/l respectively. The sensitivity and specificity of measurements of ALA-B concentrations for health screening were sufficiently high when the health based exposure limits of lead were set at 30-50 micrograms/dl. Moreover, a pronounced increase in ALA-B concentrations occurred when the inhibition rate of erythrocyte ALA dehydratase exceeded 85%. These findings suggest that ALA-B is a useful indicator for assessing the early effects of exposure to lead on haem biosynthesis.  相似文献

OBJECTIVE--To detect, with a computerised postural sway measurement system, any significant deviation of postural sway parameters among lead workers compared with a control group. METHODS--Postural stability was investigated with a computerised postural sway measurement system in 60 workers exposed to lead with a duration of exposure of 84 (range 3-366) months and 60 controls. Sway parameter data were collected with an Advanced Mechanical Technology computerised biomechanics platform system. RESULTS--The mean current blood lead concentration was 36.0 (range 6.4 to 64.5) micrograms/dl for the exposed workers and 6.3 (range 3.1 to 10.9) micrograms/dl for the 14 controls. There was no significant difference between the groups for the postural sway parameters obtained when the eyes were open. Significant differences were found for Xs, Ys (SDs of the coordinates of the centre of pressure); Xm, Ym (mean deviation of the coordinates of the centre of pressure); Rm (average displacement of Xi, Yi, from Xo, Yo); L (length of sway path); Vel (mean velocity of the centre of pressure along its path); Ao (area included within the path of the centre of pressure); Ae (95% confidence elliptical area). The Romberg ratios (the relation between eyes closed and open) for the Vel, L, Ao and Ae of the exposed workers were also significantly different from the controls. CONCLUSION--The study showed that workers exposed to lead had significantly poorer postural stability than the controls. Lead may affect certain parts of the whole sensory neural axis resulting in postural instability when the visual input is cut off. Based on current blood lead concentrations, we were not able to obtain any significant association with the postural sway parameters.  相似文献

OBJECTIVE--The aim was to study whether functional impairment of the pituitary, thyroid, testes, and adrenal glands of humans occupationally exposed to mercury (Hg) vapour can be shown as a result of accumulation of Hg in these glands. METHODS--Basal concentrations of thyrotrophin (TSH), prolactin, free thyroxine (free T4), free 3,5,3'-triiodothyronine (free T3), antibodies against thyroperoxidase, and testosterone in serum, as well as cortisol in morning urine were measured in 41 chloralkali workers exposed (10 years on average) to Hg vapour, and in 41 age matched occupationally unexposed referents. The chloralkali workers had a mean urinary Hg concentration (U-Hg) of 15 nmol/mmol (27 micrograms/g) creatinine, and a mean blood Hg concentration (B-Hg) of 46 nmol/l. For the reference group U-Hg and B-Hg were 1.9 nmol/mmol (3.3 micrograms/g) creatinine and 17 nmol/l respectively. RESULTS--The serum free T4 concentration and the ratio free T4/free T3 were slightly, but significantly, higher in the subgroups with the highest exposure, and the serum free T3 was inversely associated with cumulative Hg exposure. This indicates a possible inhibitory effect of mercury on 5'-deiodinases, which are responsible for the conversion of T4 to the active hormone T3. Serum total testosterone, but not free testosterone, was positively correlated with cumulative Hg exposure. Prolactin, TSH and urinary cortisol concentrations were not significantly associated to exposure. CONCLUSION--Apart from inhibition of the deiodination of T4 to T3, the endocrine functions studied seem not to be affected by exposure to Hg vapour at the exposure levels of the present study. 对52名接触苯乙烯和109名非接触苯乙烯男工的生殖功能进行了回顾性调查,并应用放射免疫分析方法(RIA)测定了男工和苯乙烯(剂量分别为0.3g/kg、1.0g/kg和2.5g/kg)染毒雄性大鼠血清的性激素和促性腺激素含量。实验结果指出,接触组男工表现为性功能障碍,其妻子的妊娠合并症增多。接触组男工血清睾酮(T)和促卵泡生成素(FSH)明显降低(P<0.05),大鼠血清T含量低于对照组,FSH高于对照组(P<0.05),但促间质细胞激素(ICSH或LH)无改变。本实验结果说明苯乙烯干扰下丘脑-垂体-睾丸轴系统的神经内分泌调节功能,导致生殖功能障碍。  相似文献
