Acute and prolonged bone complications associated with radiation and chemotherapy in cancer survivors underscore the importance of establishing a laboratory-based complementary dual-isotope tool to evaluate short- as well as long-term bone remodeling in an in vivo model. To address this need, a liquid scintillation dual-label method was investigated using different scintillation cocktails for quantitative measurement of (3)H-tetracycline ((3)H-TC) and (45)Ca as markers of bone turnover in mice. Individual samples were prepared over a wide range of known (45)Ca/(3)H activity ratios. Results showed that (45)Ca/(3)H activity ratios determined experimentally by the dual-label method were comparable to the known activity ratios (percentage difference ~2%), but large variations were found in samples with (45)Ca/(3)H activity ratios in range of 2-10 (percentage difference ~20-30%). Urine and fecal samples from mice administered with both (3)H-TC and (45)Ca were analyzed with the dual-label method. Positive correlations between (3)H and (45)Ca in urine (R=0.93) and feces (R=0.83) indicate that (3)H-TC and (45)Ca can be interchangeably used to monitor longitudinal in vivo skeletal remodeling.  相似文献

观察了三磷酸腺苷(ATP)、过氧化氢酶(CAT)及异搏停(VER)对活性氧作用下的肾上皮细胞(REC)的保护效果,以探讨其可能机制。结果显示预先应用ATP、CAT和VER均使处于活性氧作用下肾上皮细胞培养基中的乳酸脱氢酶(LDH)活性及细胞活力维持或接近正常水准。提示能量危机、氧自由基(OFR)毒性和钙超载可能是细胞缺血/再灌注(I/R)损伤病理生理中的重要环节,向细胞供能、清除OFR及维持钙稳态是防治肾I/R损伤的重要措施。  相似文献

目的:探讨不同来源的细胞内Ca2+([Ca2+]i)对心肌成纤维细胞(fibroblasts,PBs)丝裂素活化蛋白激酶(MAPK)介导的增殖反应的作用。方法:以培养的大鼠FBs为模型,用血管紧张素Ⅱ(AngⅡ)刺激FBs细胞外Ca2+跨膜内流、三磷酸肌醇(IP3)刺激胞内Ca2+释放,应用钙荧光指剂Fura-2/AM动态观测心肌细胞[ca2+]i浓度,γ-32P-ATP掺入法和免疫印迹(westen blot)测MAPK活性及蛋白含量,氚-亮氨酸(3H-Leu)、氚-胸腺嘧定(3H-TdR)掺入量作为FBs增殖的指标。结果:AngⅡ、IP3均能显著增加FBs[Ca2+]j浓度、MAPK活性及蛋白含量,并提高3H-Leu、3H-TdR掺入量,与对照组FBs相比差异显著,P<0.01。结论:激活[Ca2+]i使MAPK活性及含量明显增加而促进FBs的增殖,FBs的增殖与[ca2+]i浓度增加有关,与[Ca2+]i的来源无关。  相似文献

目的 观察酚妥拉明和地塞米松对重度吸入性损伤大鼠肝脏损害的保护作用及机制。方法 将 72只大鼠随机分为A、B、C 3组。对照组 (C组 )腹腔注射生理盐水 ,A组腹腔注射酚妥拉明 ,B组腹腔注射地塞米松。另取 6只非致伤大鼠做伤前值测定 ,再取 6只致伤大鼠 ,伤后 30分钟做伤后值测定。结果 C组谷丙转氨酶 (GPT)、总胆红素 (TBIL)、丙二醛 (MDA)、MDA/超氧化物歧化酶 (SOD)及组织含水量 (WCT)均明显增高 ,组织中一氧化氮 (NO)水平较伤前值变化不大 ,SOD、ATP水平降低。A组和B组中TBIL、GPT、MDA、MDA/SOD、NO明显低于C组 (P <0 .0 1 ) ,A组ATP和SOD明显高于C组 (P <0 .0 1 ) ;B组SOD高于C组 (P<0 .0 1 ) ,B组的ATP则与C组差别不显著 (P >0 .0 5 ) ,B组肝组织含水量低于对照组 (P <0 .0 1 )。结论 酚妥拉明与地塞米松均可能通过调节机体的NO水平进而提高机体抗氧化能力和能量代谢 ,从而对重度吸入性损伤后大鼠的肝脏起保护作用。在改善能量供应和降低MDA方面酚妥拉明效果优于地塞米松  相似文献

目的观察腺苷A1受体拮抗剂DPCPX对脑神经元低氧复氧(H/R)损伤的影响及其机制。方法通过建立体外培养大鼠大脑皮质神经元H/R损伤模型,观察终浓度分别为0(对照组)、25、50、100nmol/L的DPCPX对常氧(低氧0h)和低氧暴露8、12、24h后复氧24h的H/R神经元乳酸脱氢酶(LDH)释放量的影响,并检测了100nmol/LDPCPX对低氧暴露12h的H/R神经元丙二醛(MDA)含量及黄嘌呤氧化酶(XO)、Ca2 -ATP酶活性的影响。结果与对照组比较,100nmol/LDPCPX显著增加低氧暴露12h的H/R神经元LDH释放量(P<0·05),明显升高其MDA含量(P<0·01)和XO活性(P<0·05),明显降低其Ca2 -ATP酶活性(P<0·05)。结论DPCPX可加重培养神经元H/R损伤,其机制可能包括降低神经元抗氧化能力及Ca2 -ATP酶活性。  相似文献

目的:探讨促红细胞生成素(Erythropoietin,Epo)对肾缺血再灌注损伤的保护作用。方法:观察Epo对肾缺血再灌注损伤大鼠血丙二醛(MDA)、肌酐(Cr)、尿素氮(BUN)和肾组织匀浆超氧化物歧化酶(SoD)、白介素-6(IL-6)变化。结果:Epo治疗组血浆MDA、Cr、BUN水平较缺血再灌注组显著下降(P〈0.05);肾组织匀浆SOD显著升高、IL-6显著降低(P〈0.05)。结论:Epo可提高SOD,降低IL-6对肾缺血再灌注损伤具有保护作用.  相似文献

目的 研究低剂量辐射能否诱导免疫适应性应。方法 3HTdR 掺入法检测淋巴细胞增殖率;CTLL依赖株检测IL2 活性;单克隆抗体免疫荧光标记,流式细胞术(FCM) 检测淋巴细胞亚组及膜受体表达;PI标记,FCM 检测细胞周期及细胞凋亡;双抗体夹心间接ELISA 法测定可溶性白介素2 受体;Fura2 负载,双波长荧光测定法检测细胞内[Ca2+ ]i;传代培养细胞测定细胞倍增时间。结果 低剂量辐射预照射可明显减轻其后大剂量照射对诸多淋巴细胞功能的抑制作用。结论 低剂量X 射线可诱导多参数免疫适应性反应。  相似文献

It is shown that paraquat treatment (1 h, 1 x 10(-5) mol/l) increases the activity of superoxide dismutase (SOD) in L5178Y (LY) R and S cells by about three times. When combined with X-irradiation, 0.5 h of treatment preceding irradiation increased the SOD activity two-fold and the alpha/beta ratio three-fold, as estimated from the X-ray survival curves. LY-S cells were more sensitive than LY-R cells to treatment with paraquat alone. These results indicate that SOD may be a radioprotective enzyme in LY strains and that LY-S cells are particularly sensitive to superoxide radicals as a result of a relatively low SOD activity. This explains their sensitivity to paraquat, which generates O2-, and to X-rays. The low SOD level may also explain the higher initial DNA damage in X-irradiated LY-S than LY-R cells.  相似文献

目的 :探讨中药扶正解毒汤 (FJD)防治硫酸镍致睾丸组织损伤的作用机制。方法 :6 0只Wister大鼠随机分为 6组 :空白对照组、NiSO4染毒组、FJD不同剂量治疗组和FJD不同剂量预防组。中药治疗 4周后 ,测定精子细胞中Ca2 + ,睾丸组织中丙二醛(Maleicdialdehyde ,MDA)、超氧化物歧化酶 (Superoxidedismutase ,SOD)含量、以及谷胱甘肽过氧化物酶 (GlutathionePeroxidase ,GSH -PX)活力。结果 :染毒组Ca2 + 、MDA含量升高 ,SOD含量和GSH -PX活力下降 ,与对照组比较P <0 .0 5 ;中药治疗后各组Ca2 + 、MDA含量降低 ,SOD含量和GSH -PX活力升高 ,与染毒组比较P <0 .0 5。结论 :硫酸镍可导致大鼠精子细胞钙超载 ,睾丸组织脂质过氧化损伤 ,FJD具有调节钙稳态和抗睾丸组织脂质过氧化损伤的作用。  相似文献

In order to obtain more information concerning the mechanism of Gallium (67Ga) accumulation in malignant tissue, an investigation was carried out using Ehrlich ascites tumor cells. Chlorpromazine, a phospholipid stabilizer decreased the uptake of 67Ga and Calcium 45 (45Ca) by the cells at low dose, but increased them at high dose. On the other hand, the uptake of both radionuclides by the cells was inhibited by ruthenium red, a Ca ATPase inhibitor in a dose dependent manner. The time course of 67Ga and 45Ca uptake were quite different from each other. Moreover, the subcellular distribution patterns of 67Ga and 45Ca were also different from each other; 67Ga accumulated in the lysosomal fraction and 45Ca mainly in the mitochondrial fraction. These results suggest that there may be a commonality for 67Ga and 45Ca uptake by the tumor cells, whereas the behaviour of these two radionuclides in the cells is dissimilar.  相似文献
